        Supplementation of retinoic acid alone in MSC culture medium induced germ cell-like cell differentiation

        Background: Germ cells undergo towards male or female pathways to produce spermatozoa or oocyte respectively which is essential for sexual reproduction. Mesenchymal stem cells (MSCs) have the potential of trans-differentiation to the multiple cell lineages. Methods: Herein, rat MSCs were isolated from bone marrow and characterized by their morphological features, expression of MSC surface markers, and in vitro differentiation capability. Results: Thereafter, we induced these cells only by retinoic acid supplementation in MSC medium and, could able to show that bone marrow derived MSCs are capable to trans-differentiate into male germ cell-like cells in vitro. We characterized these cells by morphological changes, the expressions of germ cell specific markers by immunophenotyping and molecular biology tools. Further, we quantified these differentiated cells. Conclusions: This study suggests that only Retinoic acid in culture medium could induce bone marrow MSCs to differentiate germ cell-like cells in vitro . This basic method of germ cell generation might be helpful in the prospective applications of this technology.

        In the near future we know that vehicles will communicate with each other to make Vehicular ad hoc network and gives the concept of intelligent transportation system. In this paper we presented the review of security in VANET. Consequently, several researchers represented the attacks and solutions in vehicular communication We analyzed some of the security problems and proposed solutions to overcome it. We discussed the need for robust Vehicular Ad hoc networks, which is very strongly dependent on their security and privacy features. This paper will review the existing attacks in VANET in the perspective approach of security.We also provided the solutions to the particular attack in VANET.

      • Genetic Variability and Character Association Analysis for Yield-Contributing Traits in Wheat (Triticum aestivum L.)

        The present study was conducted to evaluate the genetic variability and correlation of different characters associated with grain yield in wheat. Forty genotypes were grown in randomized block design with three replications and evaluated for twelve yield and yield-related characters. Highly significant differences and adequate genetic variability were observed among the genotypes for all twelve characters. The heritability showed highest value for plant height, days to 50% flowering, days to maturity, peduncle length, 100 grain weight, number of spikelet per spike and number of grains per spikelet. Highest values of genetic advance were obtained in biological yield, plant height, days to maturity, peduncle length and days to 50% flowering. Correlation studies revealed an intensive selection for number of tillers, days to 50% flowering, days to maturity, number of spikelet per spike, number of grains per spikelet and 100 grain weight will contribute to improve the seed yield in wheat. Path analysis showed that maximum direct effect on grain yield per plant was contributed mostly by biological yield, followed by harvest index, spike length, plant height and days to 50% flowering. Principal components analysis (PCA) was performed and the first five principal components explained 75% of the observed variation. PC1 accounted for 23% of variation and showed the largest loading values for number of tiller`s per plant, biological yield and grain yield per plant. PC2 accounted for 17% and associated with plant height, peduncle length and harvest index. PC3 accounted for 13% and related to the days to 50% flowering and days to maturity. PCA analysis showed that wheat genotypes HPB42, UP2123, PBW527 and HD3024 showed good performance among wheat genotypes on the basis of all traits combined. The identified genotypes may be useful for wheat yield improvement.

        Efficacy of a Cue-Mate Intravaginal Insert and Injection of Prostaglandin F2α for Synchronizing Estrus in Hanwoo Cattle

        The present study was performed on farm animals to test the effectiveness of progesterone-releasing intravaginal device (Cue-Mate® 1.56 g) and injection of prostaglandin F2α (PGF2α) for synchronization estrus in Hanwoo cattle. The cattle were at random stage of the estrus cycle. The cows were artificially inseminated at day 7 after Cue-Mate withdrawal, using commercial semen from Korean native bulls. There was a season effect on the estrus synchronization rate. It was higher in spring (94.3%) followed by winter (93.3%), autumn (90.4%) and summer (67.2%). In summary, The results of this study revealed that season has influences on estrus behavior of cattle with no significant effect on pregnancy rate. In summary, we suggest summer reproductive management to alleviate the effects of heat stress. It should be based on intensive cooling combined with hormonal treatment. Given that different subgroups of cows benefit differently from the treatments, selective hormonal administration should be considered.

        The Effects of Vitamin A Administration to Heifer and Pregnant Cow on the Changes of Hormonal and Body Weight

        The objective of this study was to investigate the interrelationship of weight and hormonal changes through the administration of vitamin A to first time pregnant heifers. A total of 28 Korean heifers was used for this study. The heifers were divided into two groups - with vitamin A (n=14) and without vitamin A (control) (n=14) in the feed. Body weight increased in vitamin A treated heifers starting 9 months until 15 months. After pregnancy, vitamin A treated heifers were found to maintain higher body weights than the control group. Pre-pregnancy and post pregnancy progesterone levels were not different between the two groups. Serum estradiol levels of heifers at different growth stages showed relatively higher E2 levels than the control. Also, the control during pregnancy may show higher serum E2 levels than the vitamin A treated heifers. The growth phase serum estradiol levels in heifers may be relatively higher than the control. During pregnancy it showed a similar trend. Serum levels of vitamin A treated heifers did not differ from pregnant heifers at 5 months of age. However, after 5 months from conception until 8 months of treatment it showed a high level. Serum cholesterol in pregnant cows was higher in the control group than the treatment from beginning until the end of pregnancy. This is considered to be related to fetus development during pregnancy, as well as the mother's health.

        Increase in Growth, Productivity and Nutritional Status of Rice (Oryza sativa L. cv. Basmati) and Enrichment in Soil Fertility Applied with an Organic Matrix Entrapped Urea

        Field experiments were conducted to evaluate the effects of eco-friendly organic matrix entrapped urea (OMEU) on growth, productivity, and yield of rice (Oryza sativa L. cv. Basmati) and soil enrichment in the paddy field at Rohtak (Haryana) located near Delhi. The OMEU prepared in granular form contained cow dung, rice bran (grain cover of Oryza sativa), powder of neem leaves (Azadirachta indica), and clay soil (diameter of particles < 0.02 mm) in 1:1:1:1 ratios and saresh (plant gum of Acacia sp.) as binder along with half of the recommended dose of commercially available soluble urea (free urea; FU). Single basal application of OMEU showed an increase in plant growth in terms of fresh and dry weights, root length, root, leaf and tiller numbers, soluble protein, total N and ammonium in leaves, productivity in terms of grain and straw yield, and nutritional and microbial activities of field soil over free form of urea and no fertilizer application. Nutritional status of rice grains was also improved over the free urea and no fertilizer controls. Our data indicate that OMEU, which is low cost and based on bio-degradable, non-toxic, and locally available agro-waste, can be attempted to replace the conventional use of soluble urea in rice.

        Studies of a Plasticized PEO + NH<sub>4</sub>PF<sub>6</sub> Proton-conducting Polymer Electrolyte System and Its Application in a Proton Battery

        We have synthesized polyethylene oxide (PEO) + NH<sub>4</sub>PF<sub>6</sub> polymer electrolyte films plasticized with ethylene carbonate (EC) and a mixture of ethylene carbonate and propylene carbonate (EC/PC) by using a solution cast technique. X-ray diffraction (XRD) results show a decrease in crystallinity from ∼67% in the unplasticized complex to 39% and 35% in the EC- and the EC/PC-plasticized complexes, respectively. Scanning electron microscopy (SEM) results show the formation of a fringed miscellar structure in the plasticized complexes. An increased coordination between dissociated ions and the ether oxygen of PEO on plasticization is established from the Fourier transform infrared (FTIR) studies. The ionic conductivity shows an enhancement of about two orders of magnitude on plasticization. The room-temperature ionic conductivities of the highest conducting compositions are obtained as ∼1.52 × 10<sup>−5</sup> S cm<sup>−1</sup> and ∼1.03 × 10<sup>−5<sup> S cm<sup>−1</sup> in the EC- and the EC/PC-plasticized complexes, respectively. The highest conducting compositions of plasticized polymer electrolyte films are used to fabricate proton batteries with the configuration Zn/ZnSO<sub>4</sub>.7H<sub>2</sub>O (anode) ∥Polymer electrolyte∥ PbO<sub>2</sub>/V<sub>2</sub>O<sub>5</sub> (cathode), and different battery parameters are reported.

      • Molecular Markers and Their Usefulness in Rice Breeding

        Molecular markers are extensively used for improving and sustaining the rice productivity. A variety of molecular genetic markers, including restriction fragment length polymorphisms (RFLPs), random amplified polymorphic DNAs (RAPDs), amplified fragment length polymorphisms (AFLPs), microsatellites or simple sequence repeats (SSRs), expressed sequence tags (EST) and single nucleotide polymorphism (SNP) have been developed providing new tools for rice breeding. The major advantages of the molecular markers over the other classes of markers are their number is potentially unlimited, spanning across the genome, their expression is unaffected by the environment and their assessment is independent of the stage of plant development. Molecular markers are landmarks in the chromosome maps that can be used to monitor the transfer of specific chromosome segments known to carry useful agronomic traits. Breeders use these molecular markers to increase the precision of selection for the best trait combinations. Molecular markers have large number of applications ranging from diversity analysis to the improvement of rice varieties by marker assisted selection. This review describes the usefulness of some important DNA markers in rice improvement.
