      • Korean Dictionaries and Some Model Dictionaries of the World : Critical Essays toward the Plan for a New Korean Dictionary

        After briefly reviewing the history of lexcoigraphy in modem Korea, Nam Kishim tackles many problems created by the general deficiency of current Korean monolingual dictionaries in phonological morphological and semantic points. For example, most Korean dictionaries do not treat such phonological changes as the shortening of the stem vowel ㅓ in 더운, the prenominal form of the adjective 덥다, whose stem vowel ㅓ is pronounced long. He also takes issue with the practice of Korean dictionaries of including all kinds of proper names on little reliable principles : a typical dictionary enters a 600m-high hill while excluding better-known mountains higher than that. Hong Chai-song concentrates on the often misguiding information on transitive / intransitive verbs in current dictionaries. For example, a representative dictionary designates the verb 뛰다 simply as intransitive. But it is quite often used as an transitive also. In the sentence, 철수는 운동장을 뛰었다, 운동장 is the object of the transitive verb 뛰다. How could the compilers miss such an obvious fact? Hong suggests that they may have not been interested in being grammatically informative. A historical survey of the Oxford English Dictionary(OED) is given by Lee Sang-sup, with special emphasis on the vast amount of quotations from real sources contributed by thousands of volunteer readers through several decades and on its unwavering historical principle. A comparison between the OED's definition of Kimchi and a Korean dictionary's 김치 is given, showing the difference of principle in treating the same lexical item. Ahn Sam-huan presents a detailed account of Grimm's Deutsches Wo¨rterbuch which took generations of German lexicographers 133 years from its inception to its completion in 1971. On the basis of his study, Ahn suggests that future dictionary makers should cooperate as a group on a long-range comprehensive project. A moltivolume dictionary is an all-important cultural event for the whole nation, not to be influenced by private commercial interests. Choi Young-ai's survey of Chinese lexicography highlights the now on-going publication of 漢語字典 and 漢語詞典, admirable results of the cooperative effort of hundreds of Chinese scholars, a modus operandi for the Yonsei University graze to adopt in planning a new dictionary of the Korean language.

      • Effects of <i>Ginkgo biloba</i> Extract on Promotion of Neurogenesis in the Hippocampal Dentate Gyrus in C57BL/6 Mice

        <P><I>Ginkgo biloba </I>leaf extract (Gb) has been known to improve blood flow and preclude the tissue from free radical damage. Effects of Gb were examined by using Ki67, a specific proliferative marker for cellular proliferation, and doublecortin (DCX), a marker for immature neurons, indicating degree of neuroblast differentiation in the hippocampal dentate gyrus (DG) of adult C57BL/6 mice. The mice were fed with Gb at 40 and 100 mg/kg once daily for 28 days. The increase of Ki67- and DCX-immunoreactive cells in the DG was increased in a dose-dependent manner. Especially, the group having 100 mg/kg Gb showed a significant increase of DCX-immunoreactive neuroblasts with well-developed tertiary dendrites. Expression of DCX protein in the Gb groups was also significantly increased upon compared with the vehicle group. The results suggested that repeated intake of Gb would enhance cell proliferation and neuroblast differentiation in the mouse DG.</P>

      • Effects of pyridoxine on a high‐fat diet‐induced reduction of cell proliferation and neuroblast differentiation depend on cyclic adenosine monophosphate response element binding protein in the mouse dentate gyrus

        <P><B>Abstract</B></P><P>In this study, we challenged pyridoxine to mice fed a high‐fat diet (HFD) and investigated the effects of pyridoxine on HFD‐induced phenotypes such as blood glucose, reduction of cell proliferation and neuroblast differentiation in the dentate gyrus using Ki67 and doublecortin (DCX), respectively. Mice were fed a commercially available low‐fat diet (LFD) as control diet or HFD (60% fat) for 8 weeks. After 5 weeks of LFD or HFD treatment, 350 mg/kg pyridoxine was administered for 3 weeks. The administration of pyridoxine significantly decreased body weight in the HFD‐treated group. In addition, there were no significant differences in hepatic histology and pancreatic insulin‐immunoreactive (‐ir) and glucagon‐ir cells of the HFD‐treated group after pyridoxine treatment. In the HFD‐fed group, Ki67‐positive nuclei and DCX‐ir neuroblasts were significantly decreased in the dentate gyrus compared with those in the LFD‐fed mice. However, the administration of pyridoxine significantly increased Ki67‐positive nuclei and DCX‐ir neuroblasts in the dentate gyrus in both LFD‐ and HFD‐fed mice. In addition, the administration of pyridoxine significantly increased the protein levels of glutamic acid decarboxylase 67 (GAD67) and brain‐derived neurotrophic factor (BDNF) and the immunoreactivity of phosphorylated cyclic AMP response element binding protein (pCREB) compared with the vehicle‐treated LFD‐ and HFD‐fed mice. In contrast, the administration of pyridoxine significantly decreased HFD‐induced malondialdehyde (MDA) levels in the hippocampus. These results showed that pyridoxine supplement reduced the HFD‐induced reduction of cell proliferation and neuroblast differentiation in the dentate gyrus via controlling the levels of GAD67, pCREB, BDNF, and MDA. © 2012 Wiley Periodicals, Inc.</P>

        Substance P restores normal skin architecture and reduces epidermal infiltration of sensory nerve fiber in TNCB-induced atopic dermatitis-like lesions in NC/Nga mice

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Atopic dermatitis (AD) is a chronic inflammatory skin disorder characterized by intense pruritus and eczematous lesion. Substance P (SP) is an 11-amino-acid endogenous neuropeptide that belongs to the tachykinin family and several reports recently have supported the anti-inflammatory and tissue repairing roles of SP.</P> <P><B>Objective</B></P> <P>In this study, we investigated whether SP can improve AD symptoms, especially the impaired skin barrier function, in 2, 4, 6-trinitrochlorobenzene (TNCB)-induced chronic dermatitis of NC/Nga mice or not.</P> <P><B>Method</B></P> <P>AD-like dermatitis was induced in NC/Nga mice by repeated sensitization with TNCB for 5 weeks. The experimental group designations and topical treatments were as follows: vehicle group (AD-VE); SP group (AD-SP); and SP with NK1R antagonist CP99994 (AD-SP-A) group. Histological analysis was performed to evaluate epidermal differentiation, dermal integrity, and epidermal nerve innervation in AD-like lesions. The skin barrier functions and pruritus of NC/Nga mice were evaluated by measuring transepidermal water loss (TEWL) and scratching behavior, respectively.</P> <P><B>Result</B></P> <P>Topical SP treatment resulted in significant down-regulation of Ki67 and the abnormal-type keratins (K) K6, K16, and K17, restoration of filaggrin and claudin-1, marked reduction of TEWL, and restoration of basement membrane and dermal collagen deposition, even under continuous sensitization of low dose TNCB. In addition, SP significantly reduced innervation of itch-evoking nerve fibers, gelatinase activity and nerve growth factor (NGF) expression in the epidermis but upregulated semaphorin-3A (Sema3A) expression in the epidermis, along with reduced scratching behavior in TNCB-treated NC/Nga mice. All of these effects were completely reversed by co-treatment with the NK1R antagonist CP99994. In cultured human keratinocytes, SP treatment reduced expression of TGF-α, but upregulated TGF-β and Sema3A.</P> <P><B>Conclusion</B></P> <P>Topically administered SP can restore normal skin barrier function, reduce epidermal infiltration of itch-evoking nerve fibers in the AD-like skin lesions, and alleviate scratching behavior.         Purpose: ACR (American College of Radiology) offers variable parameters to PET/CT quality control by using ACR Phantom. ACR Phantom was made to evaluate parameters which are uniformity, attenuation, scatter, contrast and resolution. Manual analysis method wasn't good for the use of QC because values of parameter were changed as it may user and it takes long time to analysis. Ki-Chun Lim, a nuclear scientist in AMC, developed program that automatically analysis values of parameter by using ACR Phantom to overcome above problems. In this study, we evaluated automatic analysis program's usability, through the comparing SUV of each method, reproducibility of SUV when repeated analysis and the time required. Materials and Methods: Using Flangeless Esser PET Phantom, the ideal ratio of 4 : 1 hot cylinder and BKG but it actually showed a ratio of 3.89 to 1 hot cylinder and BKG. SIEMENS Biograph True Point 40 was used in this study. We obtained images using ACR phantom at Fusion WB PET Scan condition (2 min/bed) and 120 kV, 100 mAs CT condition. Using True X method, 3 iterations, 14 subsets, Gaussian filter, FWHM 4 mm and Zoom Factor 1.0, $168{\times}168$ image size. We obtained Max. & Min. SUV and SUV Mean values at Cylinder (8, 12, 16, 25 mm, Air, Bone, Water, BKG) by automatic program and obtained SUV by manual method. After that, we compared manual and automatic method. we estimate the time required from opened the image data to final work sheet was completed. Results: Automatic program always showed same result and same the time required. At 8, 12, 16 and 25 m cylinder, manual method showed 6.69, 3.46, 2.59, 1.24 CV values. The larger cylinder size became, the smaller CV became. In manual method, bone, air, water's CV were over 9.9 except BKG (2.32). Obtained CV of Mean SUV showed BKG was low (0.85) and bone was high (7.52). The time required was 45 second, 882 second respectably. Conclusions: As a result of difference automatic method and manual method, automatic method showed always same result, manual method showed that the smaller hot cylinders became, the lager CV became. Hot cylinders mean region size, the smaller hot cylinder size becomes we had some trouble in doing ROI poison setting. And it means increase in variation of SUV. The Study showed the time required of automatic method was shorten then manual method.

        Low Dose Exposure to Di-2-Ethylhexylphthalate in Juvenile Rats Alters the Expression of Genes Related with Thyroid Hormone Regulation

        Phthalates widely used in the manufacture of plastics have deeply penetrated into our everyday lives. Recently, a concern over the toxicity of phthalates on thyroid, has been raised but in most of cases, the doses employed were unrealistically high. To investigate the effects of phthalates on thyroid, we investigated the effects of the repeated oral exposure to low to high doses (0.3, 3, 30 and 150 mg/kg) di-2-ethylhexylphthalate (DEHP) from weaning to maturity for 90 days in juvenile rats on the thyroid. The histological examination revealed that DEHP significantly induced hyperplasia in the thyroid from the doses of 30 mg/kg, which was confirmed with Ki67 staining. In line with this finding, increased mRNA expression of thyrotropin releasing hormone (Trh) was observed in the thyroid of female at 0.3 mg/kg and 150 mg/kg as determined by RNAseq analysis. Moreover, significantly increased expression of parathyroid hormone (Pth) in the female at 0.3 mg/kg, and thyroglobulin (Tg) and thyroid hormone responsive (Thrsp) in the male at 0.3 mg/kg were noted in the blood, of which changes were substantially attenuated at 150 m/kg, alluding the meaningful effects of low dose DEHP on the thyroid hormone regulation. Urinary excretion of mono-2-ethylhexyl-phthalate (MEHP), a major metabolite of DEHP was determined to be 4.10 and 12.26 ppb in male, 6.65 and 324 ppb in female at 0.3 and 30 mg/kg DEHP, respectively, which fell within reported human urine levels. Collectively, these results suggest a potential adverse effects of low dose phthalates on the thyroid.

      • Eudesmin impairs adipogenic differentiation via inhibition of S6K1 signaling pathway

        <P><B>Abstract</B></P> <P>Eudesmin has been reported to possess diverse therapeutic effects, including anti-tumor, anti-inflammatory, and anti-bacterial activities. However, its molecular action has not been implicated in metabolic disease. In this study, we show that treatment of mesenchymal stem cells (MSCs) with eudesmin disturbs adipogenesis via suppression of S6K1 signaling pathway. Eudesmin treatment inhibited activation and nuclear translocation of S6K1. Consequently, S6K1-mediated phosphorylation of H2B at serine 36 (H2BS36p) was reduced upon eudesmin treatment, further inducing the expression of <I>Wnt6</I>, <I>Wnt10a</I>, and <I>Wnt1</I>0b, which disturbed adipogenic differentiation. Moreover, eudesmin promoted myogenic and osteogenic gene expression in MSCs. Taken together, we found a novel small molecule, eudesmin, to block adipogenesis through down-regulation of S6K1-H2BS36p axis, followed by regulation of cell fate determination genes. This study suggests a promising therapeutic approach with eudesmin to cure obesity and metabolic diseases.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Eudesmin inhibits activation sand nuclear translocation of S6K1. </LI> <LI> Eudesmin promotes <I>Wnt</I> gene expression by reducing S6K1-mediated H2BS36 phosphorylation. </LI> <LI> Eudesmin disturbs adipogenic gene expression and de novo adipocyte generation. </LI> <LI> Eudesmin enhances myogenic and osteogenic gene expression. </LI> </UL> </P>
