Additive-assisted preferential crystallization of racemic component: A case of norvaline

Jie Sun,Yaping Wang,Zhenguo Gao,Junbo Gong,Weiwei Tang 한국공업화학회 2022 Journal of Industrial and Engineering Chemistry Vol.110 No.-

Herein, we report a successful resolution of L-norvaline, a racemic compound system, performed near theeutectic composition by coupling preferential crystallization with tailor-made additives. The structures ofenantiomer and racemate were characterized by PXRD, DSC, and the enantiomeric purity were measuredby Chiral HPLC. The preferential crystallization process was designed and established from binary meltingphase diagram and ternary solution phase diagram. The effects of supersaturation, solution enantiomercomposition, and seed quantity on the preferential crystallization process were investigated. However, the significant improvements of enantiomeric purity and product yield were demonstratedby additive-assisted preferential crystallization approach using tailor-made additives. These additivesbearing similar structure motifs to norvaline a highly selective binding to the racemate rather than enantiomerand remarkably suppress nucleation of DL-norvaline, the key factor determining the resultantenantiomer purity and separation efficiency. The underly inhibition principle was revealed due to thecentrosymmetric packing of the racemate while the polarity of enantiomer leads to only partial or slightcrystallization suppression. The interesting inhibition mode by tailor-made additives is also believed tobe applicable for other racemic crystallization systems. Our established additive-assisted preferentialcrystallization has showed great potential in the development of separation technology and resolutionof enantiomer from racemic compounds.

Effects of different dietary ratio of metabolizable glucose and metabolizable protein on growth performance, rumen fermentation, blood biochemical indices and ruminal microbiota of 8 to 10-month-old dairy heifers

        Sun, Jie,Xu, Jinhao,Ge, Rufang,Wang, Mengzhi,Yu, Lihuai,Wang, Hongrong Asian Australasian Association of Animal Productio 2018 Animal Bioscience Vol.31 No.8

Objective: The aim of this experiment was to evaluate the effects of different dietary ratio of metabolizable glucose (MG) to metabolizable protein (MP) on growth performance, blood metabolites, rumen fermentation parameters and the ruminal microbial community of 8 to 10-month-old heifers. Methods: A total of 24 Holstein heifers weighing an average of 282.90 kg (8 month of age) were randomly assigned to four groups of six. The heifers were fed one of four diets of different dietary MG/MP (0.97, 1.07, 1.13, and 1.26). Results: The results showed that the ratio of MG/MP affected the growth performance, blood metabolites, rumen fermentation parameters and the ruminal microbial community of heifers. The average daily gain of heifers was enhanced by increasing the ratio of MG/MP (p<0.05). The concentration of blood urea nitrogen, cholesterol, and low density lipoprotein cholesterol as well as the concentration of total volatile fatty acid in the rumen fluid of heifers decreased with the improvement in the ratio of dietary MG/MP (p<0.05). However, the relative amount of Ruminococcus albus and Butyrivibrio fibrisolvens in the rumen of heifers was increased significantly (p<0.05) when the dietary MG/MP increased. At the same time, with the improvement in dietary MG/MP, the amount of Fibrobacter succinogenes increased (p = 0.08). Conclusion: A diet with an optimal ratio (1.13) of MG/MP was beneficial for the improvement of growth, rumen fermentation, dietary protein and energy utilization of 8 to 10-month-old dairy heifers in this experiment.

Effects of different dietary ratio of physically effective neutral detergent fiber and metabolizable glucose on rumen fermentation, blood metabolites and growth performance of 8 to 10-month-old heifers

        Sun, Jie,Xu, Jinhao,Shen, Yizhao,Wang, Mengzhi,Yu, Lihuai,Wang, Hongrong Asian Australasian Association of Animal Productio 2018 Animal Bioscience Vol.31 No.8

Objective: The present study was undertaken to determine an optimal balance between the amount of physically effective neutral detergent fiber (peNDF) to metabolizable glucose (MG) on rumen fermentation, blood metabolites and growth performance of 8 to 10-month-old heifers. Methods: A total of 15 healthy Holstein heifers weighing an average of 256 kg (8 month of age) were randomly assigned to three groups of five. Treatment diets consisted of the following three $peNDF_{8.0}/MG$ levels: 1.46 (Treatment A), 1.74 (Treatment B), and 2.08 (Treatment C). Results: The results showed that the ratio of $peNDF_{8.0}/MG$ affected rumen fermentation, blood metabolites and growth performance of heifers. The average daily gain of heifers tended to decrease as the ratio of $peNDF_{8.0}/MG$ increased (p = 0.07). The concentrations of blood urea nitrogen, triglyceride, and cholesterol increased significantly (p<0.05), while the high-density lipoprotein concentration decreased (p<0.05). After feeding 2 h and 4 h, insulin concentration in Treatment A was greater than Treatment C (p<0.05). Propionate concentration had decreasing trend (p = 0.07); acetate to propionate ratio and non-glucogenic to glucogenic volatile fatty acid (NGR) increased significantly (p<0.05). In addition, the digestibility of dry matter, crude protein, neutral detergent fiber, and acid detergent fiber decreased significantly (p<0.05). Conclusion: The present investigation indicated that dietary $peNDF_{8.0}/MG$ ratio can affect the growth and development, blood metabolites, rumen fermentation and apparent digestibility of heifers, and the optimal dietary $peNDF_{8.0}/MG$ ratio for 8 to 10-month-old heifers in the present study was 1.46.

Correlation between in vitro fertilization and artificial insemination in Holstein bulls

        Sun Wei,Li Yunxia,Su Jie,Bao Xiangnan,Ding Rui,Zhao Gaoping,Cao Guifang,Hu Shuxiang,Wang Jianguo,Sun Qingyuan,Yu Haiquan,Li Xihe 아세아·태평양축산학회 2021 Animal Bioscience Vol.34 No.12

Objective: Owing to the lack of a breeding index for efficient and quick fertility evaluations of Holstein bulls when using traditional or genome-wide detection methods, this study aimed to determine whether in vitro fertilization (IVF) could be used as an indicator of conception rate of artificial insemination (AI).Methods: Conventional and sexed frozen semen from nine bulls were used for IVF and AI.Results: The IVF and AI conception rates of each bull were confirmed to be positively correlated between the conventional frozen and sexed frozen semen. The correlation coefficient R values of nine bulls between IVF and AI methods were 0.73 and 0.97 for the conventional frozen and sexed frozen semen, respectively. The average conception rate of three bulls undergoing AI was 69.5% and 64.2%, 61.8% and 58.8%, and 48.2% and 46.2% in first-, second-, and third-born cows when conventional frozen and sexed frozen semen were used, respectively, which showed a positive correlation with the fertilization rate in the same parity. We propose an evaluation standard to assess the fertilization ability of bulls based on their IVF test results, which is categorized into three grades: grade one, normal fertility bull with an AI conception rate of 40%±5% and IVF rate of 45% to 60%; grade two, higher fertility bull with an AI conception rate of 50%±5% and IVF rate of 61% to 80%; and grade three, highest fertility bull with an AI conception rate of 60%±5% and IVF rate of >80%.Conclusion: These findings reveal that IVF results can be used as a breeding index for bulls to evaluate their AI conception ability, which may shorten the time required to select bulls for breeding.
