<P>Here, we demonstrate a powerful method to discriminate DNA mismatches at single-nucleotide resolution from 0 to 5 mismatches (<I>χ</I>0 to <I>χ</I>5) using Kelvin probe force microscopy (KPFM). Using our previously developed method, we quantified the surface potentials (SPs) of individual DNA-capped nanoparticles (DCNPs, ∼100 nm). On each DCNP, DNA hybridization occurs between ∼2200 immobilized probe DNA (<I>p</I>DNA) and target DNA with mismatches (<I>t</I>DNA, ∼80 nM). Thus, each DCNP used in the bioassay (each <I>p</I>DNA-<I>t</I>DNA interaction) corresponds to a single ensemble in which a large number of <I>p</I>DNA-<I>t</I>DNA interactions take place. Moreover, one KPFM image can scan at least dozens of ensembles, which allows statistical analysis (<I>i.e.</I>, an ensemble average) of many bioassay cases (ensembles) under the same conditions. We found that as the <I>χ</I>n increased from <I>χ</I>0 to <I>χ</I>5 in the tDNA, the average SP of dozens of ensembles (DCNPs) was attenuated owing to fewer hybridization events between the <I>p</I>DNA and the <I>t</I>DNA. Remarkably, the SP attenuation <I>vs.</I> the <I>χ</I>n showed an inverse-linear correlation, albeit the equilibrium constant for DNA hybridization exponentially decreased asymptotically as the <I>χ</I>n increased. In addition, we observed a cascade reaction at a 100-fold lower concentration of <I>t</I>DNA (∼0.8 nM); the average SP of DCNPs exhibited no significant decrease but rather split into two separate states (no-hybridization <I>vs.</I> full-hybridization). Compared to complementary <I>t</I>DNA (<I>i.e.</I>, <I>χ</I>0), the ratio of no-hybridization/full-hybridization within a given set of DCNPs became ∼1.6 times higher in the presence of tDNA with single mismatches (<I>i.e.</I>, <I>χ</I>1). The results imply that our method opens new avenues not only in the research on the DNA hybridization mechanism in the presence of DNA mismatches but also in the development of a robust technology for DNA mismatch detection.</P>

This paper describes a system for human-robot interaction which is one of the biggest issues recently. We approach the interaction using sound source localization, mapping data and voice recognition. The aim of the system for human-robot interaction is that the robot finds a person speaking in an area and goes to the speaker while the robot avoids obstacles using mapping data obtained before. In addition, the system can get an order such as going to a place through a simple conversation with a person.

Background: Ultrafiltration (UF) would enhance coagulation profiles by concentrating coagulation elements during cardiopulmonary bypass (CPB) for cardiac surgery. Methods: We retrospectively reviewed electronic medical records of 75 patients who had undergone cardiac surgery with rotational thromboelastometry-based coagulation management in a university hospital and analyzed the UF-induced changes in the maximum clot firmness (MCF) of extrinsically activated test with tissue factor (EXTEM) during CPB in 30 patients. Results: The median volume of filtered-free water was 1,350 ml, and median hematocrit was significantly increased from 22.5% to 25.5%. As the primary measure, UF significantly increased the median MCF-EXTEM from 48.0 mm to 50.5 mm (P = 0.015, effect size r = 0.44). The area under the receiver operating characteristic curve pre-UF MCF-EXTEM for discrimination of any increase of MCF-EXTEM after applying UF was 0.89 (95% CI [0.77, 1.00], P < 0.001), and its cut-off value was 50.5 mm (specificity of 81.8% and sensitivity of 84.2% in Youden’s J statistic). In the secondary analyses using the cut-off value, UF significantly increased the median MCF-EXTEM from 40.5 mm to 42.5 mm in 18 patients with pre-UF MCF-EXTEM ≤ 50.5 mm. However, it did not increase MCF-EXTEM in 12 patients with pre-UF MCF-EXTEM > 50.5 mm. There was a significant interaction between pre-UF MCF-EXTEM values and applying UF (P < 0.001 for the subgroup, P = 0.046 for UF, P = 0.003 for interaction).Conclusions: Applying UF improved clot firmness, and the improvement was more pronounced when pre-UF MCF-EXTEM had been reduced during CPB.
Overmodulation Strategy of NPC Type 3-Level Inverter for Traction Drives

This paper proposes an overmodulation method for improving the voltage utilization of NPC type 3-level inverter for traction drives. The conventional railway vehicle has used a vector control to MI=0.907 and a slip-frequency control from MI=0.907 to the six-step mode. The slip-frequency control has less output torque dynamics than the vector control. In this paper, the linear region of NPC type 3-level inverter output voltage is extended to the six-step mode by using overmodulation strategy. As a result, vector control can be adapted in the whole regions and it leads to better output torque dynamics comparing with the conventional slip frequency control in overmodulation region. The overmodulation range is divided into two modes depending on the modulation index (MI), In the overmodulation region Ⅰ, the reference angles are derived from the Fourier series expansion of the reference voltage corresponds to the MI. In the overmodulation region Ⅱ, the holding angles are also derived in the same way. Therefore, it is possible to obtain the linear control and the maximized utilization of PWM inverter output voltage.

<P>We report on how to quantify the binding affinity between a nanoparticle and chemical functional group using various experimental methods such as cantilever assay, PeakForce quantitative nanomechanical property mapping, and lateral force microscopy. For the immobilization of Au nanoparticles (AuNPs) onto a microscale silicon substrate, we have considered two different chemical functional molecules of amine and catecholamine (here, dopamine was used). It is found that catecholamine-modified surface is more effective for the functionalization of AuNPs onto the surface than the amine-modified surface, which has been shown from our various experiments. The dimensionless parameter (i.e., ratio of binding affinity) introduced in this work from such experiments is useful in quantitatively depicting such binding affinity, indicating that the binding affinity and stability between AuNPs and catecholamine is approximately 1.5 times stronger than that between amine and AuNPs. Our study sheds light on the experiment-based quantitative characterization of the binding affinity between nanomaterial and chemical groups, which will eventually provide an insight into how to effectively design the functional material using chemical groups.</P>

Background: Complex regional pain syndrome (CRPS) is an intractable pain disease with various symptoms. Here, we investigated the disease status, work life, sleep problems, medical insurance, economic status, psychological problems, and quality of life (QOL) of CRPS patients. Methods: CRPS patients from 37 university hospitals in South Korea were surveyed. The survey questionnaire consisted of 24 questions on the following aspects of CRPS patients: sex, age, occupation, cause of injury, activities of daily living (ADL), pain severity, sleep disturbance, level of education, economic status, therapeutic effect, and suicidal ideation. Additionally, the abbreviated World Health Organization Quality of Life (WHOQOL-BREF) questionnaire, consisting of 26 questions, was used to identify the status of QOL. Results: A total of 251 patients completed the questionnaire. According to the survey, 54.2% patients could not perform ADL on their own. Over the previous week, the mean pain score was 7.15 ± 1.78 (out of a total of 10 points); 92.1% of patients had sleep disorders and 80.5% had suicidal ideation, with most patients suffering from psychological problems. The average for each domain of WHOQOL-BREF was as follows: 21.74 ± 14.77 for physical, 25.22 ± 17.66 for psychological, 32.02 ± 22.36 for social relationship, and 30.69 ± 15.83 for environmental (out of a total of 100 points each). Occupation, ADL, sleep time, therapeutic effect, and suicidal ideation were statistically correlated with multiple domains. Conclusions: Most patients had moderate to severe pain, economic problems, limitations of their ADL, sleep problems, psychological problems, and a low QOL score.

<P><B>Purpose:</B></P><P>The plasticity of innate lymphoid cells (ILCs) has been reported <I>in vitro</I> and in the microenvironment of the intestine. However, whether ILC plasticity contributes to regulation of the tumor microenvironment remains unknown. In this study, we explored plasticity of ILCs in human lung cancer.</P><P><B>Experimental Design:</B></P><P>We analyzed immune subsets and cytokine expression in lung cancers freshly obtained from 80 patients and explored conversion of ILC1 into ILC3 in coculture with lung cancer cells. Prognostic effects of converted ILC3 and related pathway were evaluated by retrospective cohort composed of 875 patients with lung cancer.</P><P><B>Results:</B></P><P>Low percentages of ILC1, and high percentages of ILC3 were found in pulmonary squamous cell carcinomas (SqCC) but not adenocarcinomas (ADC). In non–small-cell lung cancers, the percentage of ILC3 was associated with IL23 expression in tumor cells but not immune cells. In cocultures, tumor cells of SqCCs converted ILC1 into ILC3 by producing IL23, thus promoting IL17-mediated tumor cell proliferation. Consistently, among IL17<SUP>+</SUP> immune cells, the percentages of ILCs were higher in SqCCs than ADCs. Furthermore, the numbers of CD3<SUP>−</SUP>RORγt<SUP>+</SUP> ILC3, IL17 expression level, and IL23- or IL17RA-expressing tumor cells were associated with short survival of patients with SqCC but not ADC.</P><P><B>Conclusions:</B></P><P>Conversion from ILC1 into ILC3 by IL23-producing SqCCs promotes IL17-mediated tumor progression, resulting in a poor prognosis.</P>
