Background/Aims: Real-world studies assessing the effectiveness and safety of sofosbuvir/velpatasvir (SOF/VEL) plus ribavirin (RBV) for Child-Pugh B/C hepatitis C virus (HCV)-related cirrhosis are limited. Methods: We included 107 patients with Child-Pugh B/C HCV-related cirrhosis receiving SOF/VEL plus RBV for 12 weeks in Taiwan. The sustained virologic response rates at off-treatment week 12 (SVR<sub>12</sub>) for the evaluable population (EP), modified EP, and per-protocol population (PP) were assessed. The safety profiles were reported. Results: The SVR<sub>12</sub> rates in the EP, modified EP and PP were 89.7% (95% confidence interval [CI], 82.5-94.2%), 94.1% (95% CI, 87.8-97.3%), and 100% (95% CI, 96.2-100%). Number of patients who failed to achieve SVR<sub>12</sub> were attributed to virologic failures. The SVR<sub>12</sub> rates were comparable regardless of patient characteristics. One patient discontinued treatment because of adverse events (AEs). Twenty-four patients had serious AEs and six died, but none were related to SOF/VEL or RBV. Among the 96 patients achieving SVR<sub>12</sub>, 84.4% and 64.6% had improved Child-Pugh and model for endstage liver disease (MELD) scores. Multivariate analysis revealed that a baseline MELD score ≥15 was associated with an improved MELD score of ≥3 (odds ratio, 4.13; 95% CI, 1.16-14.71; P=0.02). Patients with chronic kidney disease (CKD) stage 1 had more significant estimated glomerular filtration rate declines than patients with CKD stage 2 (-0.42 mL/min/1.73 ㎡/month; P=0.01) or stage 3 (-0.56 mL/min/1.73 ㎡/month; P<0.001). Conclusions: SOF/VEL plus RBV for 12 weeks is efficacious and well-tolerated for Child-Pugh B/C HCV-related cirrhosis. (Clin Mol Hepatol 2021;27:575-588)

Objective: Bipolar disorder (BD) is characterized by the poor sleep quality. Whether the striatal dopamine transporter (DAT) availability is related to sleep quality among patients with BD is unclear. Methods: Fifty-three euthymic patients with BD (24 BD-I and 29 BD-II) and sixty-eight healthy controls were enrolled. The Chinese Version of the Pittsburgh Sleep Quality Index (PSQI) was used, and the availability of DAT was assessed by single-photon emission computed tomography (SPECT) using [99mTc] TRODAT-1. Results: The sleep disturbance component of the PSQI was significantly associated with the level of DAT availability among patients with BD. Conclusion: The striatal dopaminergic activity that contributes to resilience to adversity was associated with sleep pattern among patients with BD.

Objective: Hypoactivity in the reward system among patients with attention deficit hyperactivity disorder (ADHD) is a well-known phenomenon. Whether the activity in the reward pathway is related to harm avoidance, such as in sensitivity to punishment, is unclear. Evidence regarding the potential difference between ADHD patients and controls in terms of this association is scarce. Methods: Event-related functional magnetic resonance imaging was conducted on subjects performing the Iowa gambling test. Fourteen adults with ADHD and 14 controls were enrolled in the study. Results: Harm avoidance was found to be positively correlated with the activities of the bilateral orbitofrontal cortex and right insula in individuals with ADHD. A group difference was also confirmed. Conclusion: Understanding the roles of harm avoidance and brain activation during risk tasks is important.

This paper presents a method for stochastic modelling of fatigue crack growth and optimising inspection and maintenance strategy for the structural members of steel bridges. The fatigue crack evolution is considered as a stochastic process with uncertainties, and the Gamma process is adopted to simulate the propagation of fatigue crack in steel bridge members. From the stochastic modelling for fatigue crack growth, the probability of failure caused by fatigue is predicted over the service life of steel bridge members. The remaining fatigue life of steel bridge members is determined by comparing the fatigue crack length with its predetermined threshold. Furthermore, the probability of detection is adopted to consider the uncertainties in detecting fatigue crack by using existing damage detection techniques. A multi-objective optimisation problem is proposed and solved by a genetic algorithm to determine the optimised inspection and maintenance strategy for the fatigue affected steel bridge members. The optimised strategy is achieved by minimizing the life-cycle cost, including the inspection, maintenance and failure costs, and maximizing the service life after necessary intervention. The number of intervention during the service life is also taken into account to investigate the relationship between the service life and the cost for maintenance. The results from numerical examples show that the proposed method can provide a useful approach for cost-effective inspection and maintenance strategy for fatigue affected steel bridges.

Nitrogen absorption in molten metal for stainless steel AISI316 has been investigated by immersion nitrogen blowing through an immersed alumina nozzle with an internal diameter of 3 mm. Based on these experimental data, some kinetic parameters of nitrogen absorption, such as reaction order, rate constant and apparent activation energy of nitrogen absorption reaction, have been obtained. Effect of stirring by immersion nitrogen blowing through an immersed alumina nozzle on nitrogen absorption reaction has been observed. Results show the following: (1) Nitrogen absorption reaction is the −1.5th order reaction. The rate constant N is of the order of 10−5wt%2.5·min −1. Nitrogen absorption reaction for AISI 316 has negative apparent activation energy of −92.40 kJ·mol −1. This indicates that the nitrogen absorption reaction has a complex and multistep reaction mechanism. (2) The rate of nitrogen absorption reaction in molten stainless steel is mixture control by the adsorption of monatomic nitrogen on the surface of molten stainless steel and mass transfer in molten metal. (3) A rate equation of nitrogen absorption reaction has been derived based on a mixed control mechanism by both the -1st order nitrogen absorption reaction and mass transfer in molten metal.

In order to investigate the hysteretic behavior of recycled aggregate concrete with ferronickel slag-filled steel tubular (RAC-FNSFST) columns, the quasi-static loading was implemented on nine specimens with different replacement ratios (RACs), axial load levels, length–diameter ratios, and diameter–thickness ratios. The hysteretic curves, skeleton curves, deformability, energy dissipation capacity, and stiffness degeneration were studied after loading and failure mechanisms were observed, followed by the construction of FE models for parameter analysis. It is demonstrated that the hysteretic loop curve is full, and the hysteretic performance was not dramatically affected by the replacement ratio of RAC. With axial load level increase, ultimate strength at descending stage degrades quickly, stiffness degeneration accelerates, and hysteretic energy dissipation increases. Stiffness degeneration and hysteresis energy dissipation are enhanced as the length–diameter ratio increases. However, when the diameter–thickness ratio decreases, hysteretic energy dissipation increases, and stiffness degeneration accelerates. In addition, a suitable FE model was established and compared with experimental results. Then a wide range of parameter studies was carried out as a supplement to the experimental study. It is shown that the ultimate strength and ductility of specimens are intimately correlated with the RAC strength, yield strength of steel tube, slenderness ratio, axial load level, and steel ratio.

Protein tyrosine phosphatase 1B (PTP1B) is a key factor in negative regulation of the insulin pathway, and is a promising target for the treatment of type-II diabetes, obesity and cancer. Herein, compound ($\mathbf{4}$) was first observed to have moderate inhibitory activity against PTP1B with an $IC_{50}$ value of $13.72{\pm}1.53{\mu}M$. To obtain more potent PTP1B inhibitors, we synthesized a series of chalcone derivatives using compound ($\mathbf{4}$) as the lead compound. Compound $\mathbf{4l}$ ($IC_{50}=3.12{\pm}0.18{\mu}M$) was 4.4-fold more potent than the lead compound $\mathbf{4}$ ($IC_{50}=13.72{\pm}1.53{\mu}M$), and more potent than the positive control, ursolic acid ($IC_{50}=3.40{\pm}0.21{\mu}M$). These results may help to provide suitable drug-like lead compounds for the design of inhibitors of PTP1B as well as other PTPs.

Controlled pore glass bead was modified with bovine serum albumin (BSA), and glutathione (GSH) was immobilized through three kinds of linkers on top of BSA. Bis(3-sulfo-N-hydroxysuccinimide suberate) sodium salt $(BS^3)$, N-hydroxysuccinimide 3-(2-pyridyldithio)propionate (SPDP), or N-hydroxysuccinimide 4-maleimidobutyrate (GMBS) was introduced into the BSA-bound matrix. Subsequently, GSH was immobilized by addition of thiol side chain into the maleimido moiety, replacing a disulfide group, or formation of an amide group upon releasing 3-sulfo-N-hydroxysuccimide group. It was observed that conjugation methodology played a critical role for activity of the immobilized GSH. SDS-PAGE chromatogram showed that the matrix of glutathione immobilized on BSA through GMBS manifested high selectivity towards glutathione-S-transferase (GST) in cell lysate.
Effect of Tissue Engineering Small Vessel Scaffolds in Experimental Animals

Acellular xenogeneic small vessel scaffolds preliminarily eliminate the xenogeneic antigen that could induce an immune rejection. We transplanted the Wistar rat caudal artery scaffolds into Japanese white rabbit ear central arteries, and observed the changes of blood flow and vessel wall. We acquired 30 caudal arteries from 15 Wistar rats, some were directly assigned as caudal artery group, the others were disposed by 1% p-octyl polyethelene glycol phenyl ether, and were assigned as the group of tissue engineered small vessel scaffold. We collected 15 Japanese white rabbits, and utilized a pair of ear central arteries per rabbit as hosts. Through microscopic surgery, the proximal broken end of central artery was overlapped into the donor`s proximal vessel lumen by sleeve anastomosis, while the distal broken end was sutured by classical end-to-end anastomosis. The maximum patency time of small vessel scaffold was 46 hours and 47 minutes, and which of caudal vessel was 14 hours. The poor blood flow primarily was occurred in distal broken end, while proximal end was connected in pedicle graft structure. On the 10th day, tunica intima fibers in vessel scaffold were still arranged in cylindrical conformation regularly without cell adherence. On the 100th day, tunica intima fibers were still preserved completely in proximal end. The results showed that xenogeneic acellular small vessel scaffold can be transplanted into the host, and the sleeve anastomosis was significantly superior to the classical end-to-end anastomosis.

This experiment has investigated the influence of Yamen (Du. 15) point injection on learning and memory dysfunction caused by cerebral ischemia and reprofusion in bilateral cervical general artery combined with bleeding on mouse tail to mimic vascular dementia in human beings. By dividing 40 mice into 4 groups (group1false operation group, group2model group, group3point injection with Cerebrolysin group4point injection with saline.) According to random dividing principles, we observed the influence of Yamen(Du. 15) point injection on the time of swimming the whole course used by model mice which had received treatment for different days in different groups, and the influence of those mice on wrong times they entered blind end. The result showed that point injection with Cerebrolysin and saline could improve learning and memory dysfunction of the mice caused by cerebral ischemia.
