        Comprehensive profiles and diagnostic value of menopausal-specific gut microbiota in premenopausal breast cancer

        In Western countries, breast cancer tends to occur in older postmenopausal women. However, in Asian countries, the proportion of younger premenopausal breast cancer patients is increasing. Increasing evidence suggests that the gut microbiota plays a critical role in breast cancer. However, studies on the gut microbiota in the context of breast cancer have mainly focused on postmenopausal breast cancer. Little is known about the gut microbiota in the context of premenopausal breast cancer. This study aimed to comprehensively explore the gut microbial profiles, diagnostic value, and functional pathways in premenopausal breast cancer patients. Here, we analyzed 267 breast cancer patients with different menopausal statuses and age-matched female controls. The α-diversity was significantly reduced in premenopausal breast cancer patients, and the β-diversity differed significantly between breast cancer patients and controls. By performing multiple analyses and classification, 14 microbial markers were identified in the different menopausal statuses of breast cancer. Bacteroides fragilis was specifically found in young women of premenopausal statuses and Klebsiella pneumoniae in older women of postmenopausal statuses. In addition, menopausal-specific microbial markers could exhibit excellent discriminatory ability in distinguishing breast cancer patients from controls. Finally, the functional pathways differed between breast cancer patients and controls. Our findings provide the first evidence that the gut microbiota in premenopausal breast cancer patients differs from that in postmenopausal breast cancer patients and shed light on menopausal-specific microbial markers for diagnosis and investigation, ultimately providing a noninvasive approach for breast cancer detection and a novel strategy for preventing premenopausal breast cancer.

        Integrated Hierarchical Cobalt Sulfide/Nickel Selenide Hybrid Nanosheets as an Efficient Three-dimensional Electrode for Electrochemical and Photoelectrochemical Water Splitting

        <P>Developing highly active electrocatalysts for photoelectrochemical water splitting is critical to bring solar/electrical-to-hydrogen energy conversion processes into reality. Herein, we report a three-dimensional (3D) hybrid electrocatalyst that is constructed through in situ anchoring of Co9S8 nanosheets onto the surface of Ni3Se2 nanosheets vertically aligned on an electrochemically exfoliated graphene foil. Benefiting from the synergistic effects between Ni3Se2 and Co9S8, the highly conductive graphene support, and large surface area, the novel 3D hybrid electrode delivers superior electrocatalytic activity toward water reduction in alkaline media, featuring overpotentials of -0.17 and -0.23 V to achieve current densities of 20 and 50 mA cm(-2), respectively, demonstrating an electrocatalytic performance on the top of the Ni3Se2- and Co9S8-based electrocatalysts as reported in literature. Experimental investigations and theoretical calculations confirm that the remarkable activity of the obtained material results from the unique 3D hierarchical architecture and interface reconstruction between Ni3Se2 and Co9S8 through Ni-S bonding, which leads to charge redistribution and thus lowers the energy barrier of hydrogen desorption in the water splitting process. Further integration of the 3D hybrid electrode with a macroporous silicon photocathode enables highly active and sustainable sunlight-driven water splitting in both basic media and real river water. The overall water splitting with 10 mA cm(-2) at a low voltage of 1.62 V is achieved using our hybrid as both anode and cathode catalysts, which surpasses that of the Ir/C-Pt/C couple (1.60 V) for sufficiently high overpotentials.</P>

        The mitogen-activated protein kinase phosphatase-1 (MKP-1) gene is a potential methylation biomarker for malignancy of breast cancer

        The mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1) belongs to the MAPK cascades which are central to cell proliferation and apoptosis. The carcinogenic role of MKP-1 has been reported in many types of cancer but it has rarely been investigated in breast cancer. The present study was designed to evaluate the MKP-1 mRNA expression and its possible regulation by methylation of MKP-1 promoter in the model of several breast cancer cell lines and tissues as well as controls. Our data demonstrate MKP-1mRNA expression significantly decreased in five breast cancer cell lines compared to breast controls (P <0.01). Using the methylation-specific PCR (MSP) analysis,the unmethylated reaction (U) is dominant in both normal cell lines and benign breast tumors (100% vs. 86.2%), whereas the methylated reaction (M) is dominant in both breast cancer cell lines and invasive breast tumors (100% vs. 57.2%). In terms of methylation ratio (M/M+U), methylation level in MKP-1 promoter is significantly higher in the invasive breast tumor tissues (n = 152) than in benign breast tumor tissues (n = 29)(P < 0.0001). Assessing the methylation ratio of the promoter of MKP-1 gene to diagnose the breast malignancy (invasive vs. benign), the area under the receiver-operating characteristic (ROC) curve was 0.809(95% CI: 0.711-0.906, P < 0.001). The best performance for this prediction has a sensitivity of 76.32% and a specificity of 82.76% at the cutoff value of 0.38. Taken together, we firstly demonstrated that the promoter methylation of MKP-1 gene is a potential breast cancer biomarker for breast malignancy.

        Fatigue crack propagation behavior at a notch for needled C/SiC composite under tension-tension loading

        Fatigue test of a needled C/SiC composite with a notch under tension-tension cyclic loading was completed, and the main fatigue crack propagation curve of the needled composite was obtained by the in situ observation of the fatigue process. By analyzing the influence of the failure number and distribution on the tensile loading subjected by 0° fiber bundles, the relationship between the main fatigue crack propagation and the distribution of 0° fiber bundles in the needled composite was established. By observing the fracture microstructure (especially the distribution of 0° fiber bundles) of the needled composite through scanning electron microscopy, the reasons for the varying fatigue resistance of different notched specimens were also explained. In addition, acoustic emission (AE) was also used to analyze the AE energy characteristics during the fatigue crack propagation process of the needled composite.

        Ultrastructural changes of Haematococcus pluvialis (Chlorophyta) in process of astaxanthin accumulation and cell damage under condition of high light with acetate

        Haematococcus pluvialis is a commercial microalga that can produce high quantities of astaxanthin. Under induced conditions, some important changes in the subcellular structures related to astaxanthin accumulation were observable. For example, a large number of astaxanthin granules, oil structures and starch granules appeared in the thick-walled cells; Astaxanthin granules gradually dissolved into the oil structures and spread throughout the entire cell with the fusion and diffusion process of oil structures during the middle and late stages of induction; The plastoglobules were closed to the newly formed structures, and some plastoglobules would abnormally increase in size under stress. Based on observations of cell damage, the degradation of membrane structures, such as chloroplasts, was found to be the primary form of damage during the early stage of induction. During the middle stage of induction, some transparent holes were exposed in the dissolving astaxanthin granules in the cytoplasm. In thick-walled cells, these transparent holes were covered by oil substances dissolving astaxanthin, thereby avoiding further damage to cells. Given the relatively few oil structures, in non-thick-walled cells, the transparent holes expanded to form multiple transparent areas, eventually resulting in the rupture and death of cells. These results suggested that the high level of synthesis and the wide range diffusion of oil explained the expansion of astaxanthin in H. pluvialis.
