      • Sand Story Counseling for Korean Early Childhood and Young Students: Life History Research on Heesoon Cho's Life Works and their Meanings

        This study centers on the dynamics of psychology theory. Drawing upon Heesoon Cho's life, we aim to extract and analyze sand story counseling in her personal and academic experiences. Additionally, we aim to investigate her process of adapting sand play techniques into Korean sand story counseling and explore how this adaptation of sandplay, along with its dissemination, contributed to early childhood development, counseling, and psychological exploratory behaviors in Korea. To avoid bias, we adopted a life history method, including collaborative interviews and triangulation. The results reflected Cho’s life chronologically, describing significant events in her life. Using existing Western theories, Cho transformed and acculturated the existing sandplay technique into sand story counseling in the Korean context. Cho observed significant positive outcomes through sand story counseling. Sand story counseling theory significantly contributed to professional counseling. This study strongly supports future culture-fit counseling research in Korea, as opposed to adopting Western theories.

      • Development of a real-time internal and external marker tracking system for particle therapy: a phantom study using patient tumor trajectory data

        <P><B>Abstract</B></P><P>Target motion–induced uncertainty in particle therapy is more complicated than that in X-ray therapy, requiring more accurate motion management. Therefore, a hybrid motion-tracking system that can track internal tumor motion and as well as an external surrogate of tumor motion was developed. Recently, many correlation tests between internal and external markers in X-ray therapy have been developed; however, the accuracy of such internal/external marker tracking systems, especially in particle therapy, has not yet been sufficiently tested. In this article, the process of installing an in-house hybrid internal/external motion-tracking system is described and the accuracy level of tracking system was acquired. Our results demonstrated that the developed in-house external/internal combined tracking system has submillimeter accuracy, and can be clinically used as a particle therapy system as well as a simulation system for moving tumor treatment.</P>

      • Association of Herpesvirus Saimiri Tip with Lipid Raft Is Essential for Downregulation of T-Cell Receptor and CD4 Coreceptor

        <B>ABSTRACT</B><P>Lipid rafts are membrane microdomains that are proposed to function as platforms for both receptor signaling and trafficking. Our previous studies have demonstrated that Tip of herpesvirus saimiri (HVS), which is a T-lymphotropic tumor virus, is constitutively targeted to lipid rafts and interacts with cellular Lck tyrosine kinase and p80 WD repeat-containing endosomal protein. Through the interactions with Lck and p80, HVS Tip modulates diverse T-cell functions, which leads to the downregulation of T-cell receptor (TCR) and CD4 coreceptor surface expression, the inhibition of TCR signal transduction, and the activation of STAT3 transcription factor. In this study, we investigated the functional significance of Tip association with lipid rafts. We found that Tip expression remarkably increased lipid raft fractions in human T cells by enhancing the recruitment of lipid raft-resident proteins. Genetic analysis showed that the carboxyl-terminal transmembrane, but not p80 and Lck interaction, of Tip was required for the lipid raft localization and that lipid raft localization of Tip was necessary for the efficient downregulation of TCR and CD4 surface expression. Correlated with this, treatment with Filipin III, a lipid raft-disrupting agent, effectively reversed the downregulation of CD3 and CD4 surface expression induced by Tip. On the other hand, Tip mutants that were no longer present in lipid rafts were still capable of inhibiting TCR signaling and activating STAT3 transcription factor activity as efficiently as wild-type (wt) Tip. These results indicate that the association of Tip with lipid rafts is essential for the downregulation of TCR and CD4 surface expression but not for the inhibition of TCR signal transduction and the activation of STAT3 transcription factor. These results also suggest that the signaling and targeting activities of HVS Tip rely on functionally and genetically separable mechanisms, which may independently modulate T-cell function for viral persistence or pathogenesis.</P>

      • Synthesis and In Vitro Cytotoxicity of 3-or 4-Dialkylaminomethyl-1-azaanthraquinones

        Six 3-dialkylaminomethyl-1-azaanthraquinones and five 4-dialkylaminomethyl-1-azaanthraquinones were synthesized and evaluated in vitro cytotoxicity against four human cancer cell lines. The compounds retained much of their cytotoxic activity against the multi-drug-resistant cell line (KB-V-1) as shown by resistance index.

      • Synthesis and In vitro Evaluation of 4-Substituted-1-azaanthraquinones

        Doxorubicin and daunomycin are the best known members of the anthracycline antibiotics and the most commonly used intercalaing agents in the treatment of cancer(Wakelin and Waring, 1990). Doxorubicin has a broad spectrum of activity, being particularly efficacious against solid tumors. However its clinical usefulness is limited by cardiotoxicity that develop after extended therapy, and the appearance of an acquired resistance (Priebe, 1995; Surato et al 1990). Synthetic analogues, mitoxantrone and ametantrone, resulted from efforts to produce anthracycline analogues that lack cardiac toxicity. Mitoxantrone in particular currently plays an important role in the clinical management of hematological malignancies such as chronic myelogenous leukemia, acute nonlymphoblastic leukemia, and non-Hodgkin's lymphoma, as well as in combination therapy of refractory overian and breast cancers (Wakelin and Waring, 1990). Although mitoxantrone is endowed with an improved tolerability profile compared with doxorubicin and other anthracylines, this drug is not devoid of significant toxic side effects especially those associated with myelosupporession(Gandolf et al., 1995). A number of studies have identified mitoxantrone as an intercalating agent, all indications being that its biological activity is attributable to its DNA-binging properties. DNA intercalation and interference with the DNA-topoisomerase Ⅱ activity resulting in protein associated DNA strand breaks have been proposed as critical events that lead to mitoxantrone-induced cell death(Wakelin and Waring, 1990). The search for new analogues side effects of the anthracycline analogues is of extreme interest and numerous analogues have been reported(Priebe, 1995).

        초고압 전처리한 찹쌀과 현미의 텍스처 특성과 세균 저감 효과

        본 연구에서는 건조 쌀에 적합한 가공적성과 세균 저감화를 위하여 백미 찹쌀(동진벼)과 현미 멥쌀(신동진벼)에 초고압 전처리를 하였다. 백미 찹쌀과 현미 멥쌀을 500MPa의 압력 하에서 5분간 초고압 처리하였을 때, 건조 쌀 제조에 있어 중요한 특성인 백미 찹쌀의 찰기와 현미 멥쌀의 경도가 유의적으로 감소하였는데(p<0.05), 이는 초고압 처리법은 백미 찹쌀의 낱알 분리가 힘든 찰진 특성과 현미 멥쌀의 딱딱한 식감을 효과적으로 개선할 수 있다는 점을 확인해 주었다. 쌀 텍스처 단면의 입자를 FESEM를 이용하여 관찰한 결과, 초고압 처리한 백미 찹쌀과 현미멥쌀의 단면의 텍스처가 처리 전보다 부드러워졌음을 확인할 수 있었다. 600MPa의 압력 하에서 5분간 초고압 처리했을 때, 현미멥쌀에서 생육하는 세균수는 처리 전보다 유의적으로 감소하였지만(p<0.05), 내열성 세균수에는 유의적인 차이가 없었다. Ultra-high pressure (UHP) was applied to glutinous rice (Dongjin) and brown rice (Shindongjin) to determine its effects on the textural characteristics of cooked rice and bacterial reduction in uncooked rice. Hardness and stickiness of the cooked glutinous and brown rice pretreated with 500 MPa for 5 min were significantly attenuated compared to that of the control rice (p<0.05). After the UHP treatment, particles of the glutinous and brown rice were observed to be finer when cross sections of the rice were analyzed using field emission scanning electron microscope. The total bacterial count in the brown rice treated with 600 MPa for 5 min was reduced from 10<SUP>7</SUP> CFU/g to 10<SUP>4</SUP> CFU/g, while the number of thermoduric bacteria in the brown rice was not significantly different after the UHP treatment. These results suggest that UHP may be an effective method for improving texture of dried rice and to reduce bacterial count in raw rice.

      • 4-카바모일옥시메틸-1-아자안트라퀴논 유도체들의 합성 및 세포독성

        In the course of developing novel antitumor intercalating agents, we synthesized 4-carbamoyloxymethyl-1-azaanthraquinones 7-12. incorporating the latent alkylating functionality. These compounds were designed to explore the effect of substituent on the nitrogen of carbamate. The target compounds were prepared by hetero Diels-Alder reaction as a key step followed by functionalization of benzylic methyl to the desired substituents. Growth inhibitory studies of the azaanthraquinones were conducted in vitro against human cancer cell lne (SNU-354: liver and MCF7: breast) and human epidermoid carcinoma cells that are sensitive (KB-3-1) and multidrug-resistant (KB-V-1). the compounds were less potent than doxorubicin against sensitive cell lines. However, the most active compound 12 was not cross-resisitant with doxorubicin against KB-V-1.

        소아의 다발성 치아우식증과 연관된 타액의 생화학적 특성

        Saliva is obviously potential medium to protect the dental caries by not only physical clearing effect, but aggregatiog action of protein with bacteria. Nevertheless, we still do not understand how the dental caries occur and what brings the individual difference in caries prevalence. In the regards of dental caries prevalence, we hypothesized that the composition of salivary protein might be different from caries susceptible group to caries resistant group. The purposes of this experiment were focused on the molecular analysis of salivary proteins from the subjects who were involved in multiple caries. Electrophoretic analysis was eone on the whole saliva collected from the children with and without multiple caries. We found 86.2% of subjects with multiple caries has approximately 120 KDa protein band while 30.4% in the healthy subjemts. And the concentration of the total protein on the subjects with multiple caries is significantly higher than that of the healthy group. However, it turned out that the difference of the salivary composition does vot affect the bacterial adhesion to hydroxyapatite bead. With regards of enzymes in salva, the activity of α-amylase and lactate dehydrogenase does not have any significant difference between both groups. However, the concentrations of Na+ and Cl- in saliva from multiple caries group is higher than that of the control group. Taken all together, it may be concluded that 120 KDa protein in saliva may be associated with the process of dental caries, also the high concentration of protein and Na+, Cl- in saliva may be linked to dental caries development as a cofactors.

      • Deregulation of DNA Damage Signal Transduction by Herpesvirus Latency-Associated M2

        <B>ABSTRACT</B><P>Infected cells recognize viral replication as a DNA damage stress and elicit a DNA damage response that ultimately induces apoptosis as part of host immune surveillance. Here, we demonstrate a novel mechanism where the murine gamma herpesvirus 68 (γHV68) latency-associated, anti-interferon M2 protein inhibits DNA damage-induced apoptosis by interacting with the DDB1/COP9/cullin repair complex and the ATM DNA damage signal transducer. M2 expression constitutively induced DDB1 nuclear localization and ATM kinase activation in the absence of DNA damage. Activated ATM subsequently induced Chk activation and p53 phosphorylation and stabilization without eliciting H2AX phosphorylation and MRN recruitment to foci upon DNA damage. Consequently, M2 expression inhibited DNA repair, rendered cells resistant to DNA damage-induced apoptosis, and induced a G1 cell cycle arrest. Our results suggest that γHV68 M2 blocks apoptosis-mediated intracellular innate immunity, which might ultimately contribute to its role in latent infection.</P>
