        Two groups of S-Layer proteins, SLP1s and SLP2s, in Bacillus thuringiensis co-exist in the S-Layer and in parasporal inclusions

        We screened four B. thuringiensis strains whose parasporal inclusions contained the S-layer protein (SLP), and cloned two slp genes from each strain. Phylogenetic analysis indicated these SLPs could be divided into two groups, SLP1s and SLP2s. To confirm whether SLPs were present in the S-layer or as a parasporal inclusion, strains CTC and BMB1152 were chosen for further study. Western blots with isolated S-layer proteins from strains CTC and BMB1152 in the vegetative phase showed that SLP1s and SLP2s were constituents of the S-layer. Immunofluorescence utilizing spore-inclusion mixtures of strains CTC and BMB1152 in the sporulation phase showed that SLP1s and SLP2s were also constituents of parasporal inclusions. When heterogeneously expressed in the crystal negative strain BMB171, four SLPs from strains CTC and BMB1152 could also form parasporal inclusions. This temporal and spatial expression is not an occasional phenomenon but ubiquitous in B. thuringiensis strains. [BMB reports 2011; 44(5): 323-328]

      • New Model of In-situ Xenograft Lymphangiogenesis by a Human Colonic Adenocarcinoma Cell Line in Nude Mice

        Objective: To explore a new model of in-situ xenograft lymphangiogenesis of human colonic adenocarcinomas in nude mice. Method: On the basis of establishing subcutaneous xenograft lymphangiogenesis model of human colonic adenocarcinoms, in-situ xenografts were established through the in situ growth of the HT-29 human colonic adenocarcinoma cell line in nude mice. The numbers of lymphangiogenic microvessels, the expression of lymphatic endothelial cell markers lymphatic vessel endothelial hyaloronic acid receptor-1 (LYVE-1), D2-40 and the lymphatic endothelial growth factors vascular endothelial growth factor-C (VEGF-C), -D (VEGF-D) and receptor-3 (VEGFR-3) were compared by immunohistochemical staining, Western bolt and quantitative RT-PCR in xenograft in-situ models. Results: Some microlymphatics with thin walls, large and irregular or collapsed cavities and increased LMVD, with strong positive of LYVE-1, D2-40 in immunohistochemistry, were observed, identical with the morphological characteristics of lymphatic vessels and capillaries. Expression of LYVE-1 and D2-40 proteins and mRNAs were significantly higher in xenograpfts in-situ than in the negative control group(both P<0.01). Moreover, the expression of VEGF-C, VEGF-D and VEGFR-3 proteins and mRNAs were significantly higher in xenografts in-situ (both P<0.01), in conformity with the signal regulation of the VEGF-C,-D/VEGFR-3 axis of tumor lymphangiogenesis. Conclusions: In-situ xenografts of a human colonic adenocarcinoma cell line demonstrate tumor lymphangiogenesis. This novel in-situ animal model should be useful for further studying mechanisms of lymph node metastasis, drug intervention and anti-metastasis therapy in colorectal cancer.

        Novel 5-Fluorouracil Derivatives: Synthesis and Cytotoxic Activity of 2-Butoxy-4-Substituted 5-Fluoropyrimidines

        Twenty two new 5-fluorouracil (5-FU) derivatives, 2-butoxy-4-substituted 5-fluoropyrimidines, were synthesized and characterized by IR, $^1H$ NMR, MS, HRMS. All compounds were preliminarily evaluated by MTT assay on human liver BEL-7402 cancer cell line in vitro. Ten compounds were selected to test their cytotoxic activity against A549, HL-60 and MCF-7 cancer cell lines in vitro. These compounds were more sensitive to BEL-7402 than other cell lines, particularly, cytotoxic activity of compounds 6b, 6d-f, 6p, 6s-u were in sub-micromolar scale. The highest cytotoxic potency against A549, HL-60 and MCF-7 was shown by 2-butoxy-4-chloro-5-fluoropyrimidine (5) with $IC_{50}$ values of 0.10, 1.66 and $0.59{\mu}M$, respectively. Compounds 6d and 6e were effective against MCF-7 with $IC_{50}$ $9.73{\mu}M$ and HL-60 with $IC_{50}$ $8.83{\mu}M$, respectively.

        Investigation into performance enhancements of Li–S batteries via oxygen-containing functional groups on activated multi-walled carbon nanotubes using Fourier transform infrared spectroscopy

        It was demonstrated that the electrochemical performance enhancements in KOH-activated carbon materials should be mainly due to the created polar oxygen-containing functional groups (OFGs, such as such as C–O, C–– O, –OH, and O–C–– O), while the role of each OFGs on the electrochemical enhancements is still unclear. In this work, KOH activation treatments were systematically conducted on carbon nanotubes (CNTs) to explore the role of each OFG on the performance enhancements of Li–S batteries (LSBs). Results showed that the capacity of activated-CNT-sulfur (a-CNT-S) cathodes is 33% higher than that of the pristine CNT-S cathodes, and their rate capability and cycling stability are also enhanced. And the electrochemical analysis combining with Fourier transform infrared spectroscopy indicated that the formed C–O bonds are the real factor for the enhanced electrochemical performances of a-CNT-S cathodes. Furthermore, the optimal activation conditions on CNTbased cathodes for LSBs were optimized to be 10 min at 700 ℃.

      • Surfactant-Assisted Synthesis of Fe<sub>2</sub>O<sub>3</sub> Nanoparticles and F-Doped Carbon Modification toward an Improved Fe<sub>3</sub>O<sub>4</sub>@CF<sub><i>x</i></sub>/LiNi<sub>0.5</sub>Mn<sub>1.5</sub>O<sub>4</sub> Battery

        <P>A simple surfactant-assisted reflux method was used in this study for the synthesis of cocklebur-shaped Fe<SUB>2</SUB>O<SUB>3</SUB> nanoparticles (NPs). With this strategy, a series of nanostructured Fe<SUB>2</SUB>O<SUB>3</SUB> NPs with a size distribution ranging from 20 to 120 nm and a tunable surface area were readily controlled by varying reflux temperature and the type of surfactant. Surfactants such as cetyltrimethylammonium bromide (CTAB), polyvinylpyrrolidone (PVP), poly(ethylene glycol)-<I>block</I>-poly(propylene glycol)-<I>block</I>-poly(ethylene glycol) (F127) and sodium dodecyl benzenesulfonate (SDBS) were used to achieve large-scale synthesis of uniform Fe<SUB>2</SUB>O<SUB>3</SUB> NPs with a relatively low cost. A new composite of Fe<SUB>3</SUB>O<SUB>4</SUB>@CF<SUB><I>x</I></SUB> was prepared by coating the primary Fe<SUB>2</SUB>O<SUB>3</SUB> NPs with a layer of F-doped carbon (CF<SUB><I>x</I></SUB>) with a one-step carbonization process. The Fe<SUB>3</SUB>O<SUB>4</SUB>@CF<SUB><I>x</I></SUB> composite was utilized as the anode in a lithium ion battery and exhibited a high reversible capacity of 900 mAh g<SUP>–1</SUP> at a current density of 100 mA g<SUP>–1</SUP> over 100 cycles with 95% capacity retention. In addition, a new Fe<SUB>3</SUB>O<SUB>4</SUB>@CF<SUB><I>x</I></SUB>/LiNi<SUB>0.5</SUB>Mn<SUB>1.5</SUB>O<SUB>4</SUB> battery with a high energy density of 371 Wh kg<SUP>–1</SUP> (vs cathode) was successfully assembled, and more than 300 cycles were easily completed with 66.8% capacity retention at 100 mA g<SUP>–1</SUP>. Even cycled at the high temperature of 45 °C, this full cell also exhibited a relatively high capacity of 91.6 mAh g<SUP>–1</SUP> (vs cathode) at 100 mA g<SUP>–1</SUP> and retained 54.6% of its reversible capacity over 50 cycles.       • Fluorine-doped porous carbon-decorated Fe<sub>3</sub>O<sub>4</sub>-FeF<sub>2</sub> composite versus LiNi<sub>0.5</sub>Mn<sub>1.5</sub>O<sub>4</sub> towards a full battery with robust capability

        <P><B>Abstract</B></P> <P>A new fluorine-doped porous carbon-decorated Fe<SUB>3</SUB>O<SUB>4</SUB>-FeF<SUB>2</SUB> composite, referred to as Fe<SUB>3</SUB>O<SUB>4</SUB>-FeF<SUB>2</SUB>@CF<SUB>x</SUB>, was prepared for the first time. The formation mechanism is discussed, and a new concept of introducing double layers of FeF<SUB>2</SUB> and CF<SUB>x</SUB> into the oxide-based anode is presented for lithium ion batteries. Varying the amount of fluorine precursor, derivatives of Fe<SUB>3</SUB>O<SUB>4</SUB>@CF<SUB>x</SUB> and FeF<SUB>2</SUB>@CF<SUB>x</SUB> were further obtained, allowing an original analysis of their electrochemical behaviors. As-prepared Fe<SUB>3</SUB>O<SUB>4</SUB>-FeF<SUB>2</SUB>@CF<SUB>x</SUB> can deliver a high capacity of 718mAhg<SUP>−1</SUP> at 50mAg<SUP>−1</SUP>. Under a hash rate of 1600mAg<SUP>−1</SUP>, the capacity of Fe<SUB>3</SUB>O<SUB>4</SUB>-FeF<SUB>2</SUB>@CF<SUB>x</SUB> (around 338mAhg<SUP>−1</SUP>) is higher than that (200mAhg<SUP>−1</SUP>) of FeF<SUB>2</SUB>@CF<SUB>x</SUB>. Further, its capacity retention of 97% over 100 cycles is much better than the 59.4% observed for Fe<SUB>3</SUB>O<SUB>4</SUB>@CF<SUB>x</SUB>. The positive effect of the CF<SUB>x</SUB> layer on the electronic conductivity and ionic diffusion ability was confirmed. The role of FeF<SUB>2</SUB> in the stabilization of the structure of CF<SUB>x</SUB> and Fe<SUB>3</SUB>O<SUB>4</SUB> is also discussed.         The Thioacetate-Functionalized Self-Assembled Monolayers on Au: Toward High-Performance Ion-Selective Electrode for Ag<sup>+</sup>

        Two classes of morpholino-substitued thioacetate have been successfully synthesized and their electrochemical properties of self-assembled monolayers (SAMs) on Au electrode are measured by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The barrier property of the SAMs-modified surfaces is evaluated by using potassium ferro/ferri cyanide. The results suggest that the arenethioacetate forms higher-quality close-packed blocking monolayers in comparison with alkanethioacetate. Furthermore, it has shown that the barrier properties of these monolayers can be significantly improved by mixed SAMs formation with decanethiol. From our experimental results we find that the electron transfer reaction of $[Fe(CN)_6]^{3/4-}$ redox couple occurs predominantly through the pinholes and defects present in the SAM and both SAMs show a good and fast capacity in recognition for $Ag^+$. The morphological and elementary composition have also been examined by scanning electron microscope (SEM) and energy dispersive spectrometer (EDS).

        Purification and Characterization of Cathepsin B from the Gut of the Sea Cucumber (Stichopus japonicas)

        Cathepsin B from the gut of sea cucumber (Stichopus japonicas) was purified 81-fold with a 3%recovery by ammonium sulfate fractionation and a series chromatography on DEAE Sepharose CL-6B, Sephadex G-75, and TSK-Gel 3000 SWxl. The purified protein appeared as a single band on Native-PAGE but showed 2bands of 23 and 26 kDa on SDS-PAGE. The optimum activity was found at pH 5.5 and 45°C. The enzyme was stable at pH 4.5-6.0 and the thermal stability was up to 50oC. The enzyme was strongly inhibited by E-64, iodoacetic acid, and antipain, demonstrating it is a cysteine protease containing sulfhydryl groups. Cu^2+, Ni^2+, and Zn^2+ could strongly inhibit the enzyme activity. The amino acid sequences of the purified enzyme were acquired by mass spectrometer, which did not show any homology with previously described cathepsins, suggesting it may be a novel member.

      • DNA Repair Gene Associated with Clinical Outcome of Epithelial Ovarian Cancer Treated with Platinum-based Chemotherapy

        Objective: The nucleotide excision repair (NER) and base excision repair (BER) pathways, two DNA repair pathways, are related to platinum resistance in cancer treatment. In this paper, we studied the association between single nucleotide polymorphisms (SNPs) of involved genes and response to platinum-based chemotherapy in epithelial ovarian cancer. Method: Eight SNPs in XRCC1 (BER), XPC and XPD (NER) were assessed in 213 patients with epithelial ovarian cancer using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and primer-introduced restriction analysis-polymerase chain reaction (PIRA-PCR) techniques. Results: The median progression-free survival (PFS) of patients carrying the Lys/Lys and Lys/Gln+Gln/Gln genotype of the XPC Lys/Gln polymorphism were 25 and 12 months, respectively (P=0.039); and the mean overall survival (OS) of patients was 31.1 and 27.8 months, respectively (P=0.048). Cox's multivariate analysis suggested that patients with epithelial ovarian cancer with the Gln allele had an increased risk of death (HR=1.75; 95% CI=1.06-2.91) compared to those with the Lys/Lys genotype. There are no associations between the XPC PAT+/-, XRCC1 Arg194Trp, Arg280His, Arg399Gln, and XPD Asp312Asn, Lys751Gln polymorphisms and the survival of patients with epithelial ovarian cancer when treated with platinum-based chemotherapy. Conclusion: Our results indicated that the XPC Lys939Gln polymorphism may correlate with clinical outcome of patients with epithelial ovarian cancer when treated with platinum-based chemotherapy in Northern China.
