Colorectal Cancer Concealment Predicts a Poor Survival: A Retrospective Study

Objectives: Understanding the situation of cancer awareness which doctors give to patients might lead to prognostic prediction in cases of of colorectal cancer (CRC). Methods: Subsets of 10,779 CRC patients were used to screen the risk factors from the Cancer Registry in Pudong New Area in cancer awareness, age, TNM stage, and gender. Survival of the patients was calculated by the Kaplan-Meier method and assessed by Cox regression analysis. The views of cancer awareness in doctors and patients were surveyed by telephone or household. Results: After a median observation time of 1,616 days (ranging from 0 to 4,083 days) of 10,779 available patients, 2,596 of the 4,561 patients with cancer awareness survived, whereas 2,258 of the 5,469 patients without cancer awareness and 406 of the 749 patients without information on cancer awareness died of the disease. All-cause and cancer-specific survival were poorer for the patients without cancer awareness than those with (P < 0.001 for each, log-rank test). Cox multivariate regression analysis showed that cancer concealment cases had significantly lower cancer-specific survival (hazard ratio (HR) = 1.299; 95 % confidence interval (CI): 1.200-1.407)and all-cause survival (HR = 1.324; 95 % CI: 1.227-1.428). Furthermore, attitudes of cancer awareness between doctors and patients were significantly different (P < 0.001). Conclusion: Cancer concealment, not only late-stage tumor and age, is associated with a poor survival of CRC patients.
MicroRNA-3200-5p Promotes Osteosarcoma Cell Invasion via Suppression of BRMS1

Tumour metastasis is one of the most serious challenges of cancer as it is the major cause of mortality in patients with solid tumours, including osteosarcoma (OS). In this regard, anti-metastatic genes have potential for metastasis inhibition strategies. Recent evidence showed the importance of breast cancer metastasis suppressor 1 (BRMS1) in control of OS invasiveness, but the regulation of BRMS1 in OS remains largely unknown. Here, we used bioinformatics analyses to predict BRMS1-targeting microRNAs (miRNAs), and the functional binding of miRNAs to BRMS1 mRNA was evaluated using a dual luciferase reporter assay. Among all BRMS1-targeting miRNAs, only miR-151b, miR-7-5p and miR-3200-5p showed significant expression in OS specimens. Specifically, we found that only miR-3200-5p significantly inhibited protein translation of BRMS1 via pairing to the 3'-UTR of the BRMS1 mRNA. Moreover, we detected significantly lower BRMS1 and significantly higher miR-3200-5p in the OS specimens compared to the paired adjacent non-tumour bone tissues. Furthermore, BRMS1 and miR-3200-5p levels were inversely correlated to each other. Low BRMS1 was correlated with metastasis and poor patient survival. In vitro, overexpression of miR-3200-5p significantly decreased BRMS1 levels and promoted OS cell invasion and migration, while depletion of miR-3200-5p significantly increased BRMS1 levels and inhibited OS cell invasion and migration. Thus, our study revealed that miR-3200-5p may be a critical regulator of OS cell invasiveness.
Vortex-Induced Vibration Optimization of a Wide Streamline Box Girder by Wind Tunnel Test

Although the streamline box girder exhibits an excellent aerodynamic performance, they are often suffer severe Vortex-Induced Vibrations (VIVs) due to complicated separation and reattachment of air flows. It is necessary to apply control measures on the streamline box girder to suppress VIV. In this study, the VIV optimizations of a super wide streamline box girder were conducted via a series of section model wind tunnel tests. The effects of wind fairings, inspection vehicle rails, guide vanes, traffic barriers and handrails on the heaving VIV performance of the wide streamline box girder were analyzed. The test results show that the inspection vehicle rail and handrail are the main members to induce the heaving VIV of the box girder. A sharper wind fairing without changing the distance of railings is favorable to the heaving VIV performance. However, the countermeasures of changing the position of inspection vehicle rail and installing guide vanes under the girder have slight influences on reducing heaving VIV responses. Inspired by the spanwise sinusoidal perturbation method, the traffic barriers sealed with plates by regular intervals along the bridge deck can suppress the heaving VIV of the box girder successfully. The porosity of handrails has a significant effect on the heaving VIV. By using a sharp wind fairing and circular simplified handrails, the VIV performance of the wide streamline box girder was greatly improved. Their mitigation abilities were verified by a large-scale section model wind tunnel test.

Chemerin exhibits an inhibitory eff ect on hepatocellularcarcinoma; however, the underlying mechanismis unclear. Here, low chemerin expression was confi rmedin samples of liver cancer patients and hepatoma cells. Chemerin altered hepatoma cell morphology but had noeff ect on normal hepatocytes. Chemerin inhibited proliferationof several human hepatoma cell lines. Real-time PCRdetection of hepatocellular carcinoma markers showed thatmRNA levels of albumin and A-type gamma-glutamyltransferase increased whereas those of alpha-fetoprotein,alkaline phosphatase, B-type gamma-glutamyl transferase,insulin-like growth factor II, and human telomerase reversetranscriptase decreased in chemerin-treated SMMC7721cells. Western blotting revealed that chemerin up-regulatedalbumin and vimentin expressions, and downregulated alpha-fetoprotein expression. Phosphorylated STAT3 wassignifi cantly up-regulated, whereas phosphorylated ERKand AKT were signifi cantly downregulated by chemerin. Chemerin decreased phosphorylated ERK and AKT expressionand the cell proliferation induced by PI3K activator 740Y-P but could not signifi cantly alter phosphorylated STAT3expression and the cell growth induced by STAT3 inhibitorNSC74859. In conclusion, chemerin reversed the malignantphenotype and induced SMMC7721 cell diff erentiation byinhibiting MAPK/ERK and PI3K/AKT signaling; growthinhibition by chemerin is not directly related to the JAK/STAT signaling pathway. Our study provides novel evidencethat chemerin could be utilized for liver cancer treatment.

Acarbose, a pseudo-oligosaccharide, is widely used clinically in therapies for non-insulin-dependent diabetes. In the present study, S-adenosylmethionine (SAM) was added to selected media in order to investigate its effect on acarbose fermentation by Actinoplanes utahensis ZJB- 08196. Acarbose titer was seen to increase markedly when concentrations of SAM were added over a period of time. The effects of glucose and maltose on the production of acarbose were investigated in both batch and fed-batch fermentation. Optimal acarbose production was observed at relatively low glucose levels and high maltose levels. Based on these results, a further fed-batch experiment was designed so as to enhance the production of acarbose. Fed-batch fermentation was carried out at an initial glucose level of 10 g/l and an initial maltose level of 60 g/l. Then, 12 h post inoculation, 100 μmol/l SAM was added. In addition, 8 g/l of glucose was added every 24 h, and 20 g/l of maltose was added at 96 h. By way of this novel feeding strategy, the maximum titer of acarbose achieved was 6,113 mg/l at 192 h. To our knowledge, the production level of acarbose achieved in this study is the highest ever reported.

Objectives: To study application of the maximum standardized uptake value (SUVmax), metabolic tumor volume (MTV) and total lesion glycolysis (TLG) with $^{18}F$-FDG PET/CT for predicting prognosis of esophageal squamous cell cancer (ESC) patients. Methods: Eighty-six patients with ESC staged from I to IV were prospectively enrolled. Cisplatin-based chemoradiotherapy (CCRT) or palliative chemoradiotherapy were the main treatment methods and none received surgery. $^{18}F$-FDG PET/CT scans were performed before the treatment. SUVmax, MTV, and TLG were measured for the primary esophageal lesion and regional lymph nodes. Receiver operating characteristic curves (ROCs) were generated to calculate the P value of the predictive ability and the optimal threshold. Results: MTV and TLG proved to be good indexes in the prediction of outcome for the ESC patients. An MTV value of 15.6 ml and a TLG value of 183.5 were optimal threshold to predict the overall survival (OS). The areas under the curve (AUC) for MTV and TLG were 0.74 and 0.70, respectively. Kaplan-Meier analysis showed an MTV less than 15.6 ml and a TLG less than 183.5 to indicate good media survival time (p value <0.05). In the stage III-IV patient group, MTV could better predict the OS (P < 0.001), with a sensitivity and specificity of 0.80 and 0.67, respectively. Conclusions: Pre-treatment MTV and TLG are useful prognostic factors in nonsurgical ESC.
Effect of Matrix Metalloproteinase on Autolysis of Sea Cucumber Stichopus japonicus

To investigate the relationship between matrix metalloproteinase (MMPs) and autolysis of sea cucumber Stichopus japonicus, the dermis homogenate was incubated at 25oC to induce autolysis. EDTA Na2 and 1,10-phenanthroline were used to verify the effect of MMPs on autolysis, which was measured by soluble protein and protein pattern. Soluble protein level increased during a 6-h autolysis process. SDS-PAGE demonstrated obvious protein degradation with the concomitant occurrence of degradation products. The above two indicators could be inhibited significantly by EDTA Na2 and 1,10-phenanthroline, indicating that MMPs might play a significant role in autolysis of sea cucumber.

A new dibenzocyclooctadiene lignan, intermedin A (1), and a new natural bisabolane sesquiterpenoid, intermedin B (2), were isolated from the aerial parts of Schisandra propinqua var. intermedia. Their structures were elucidated on the basis of extensive spectroscopical analysis.
