Currently, there is growing emphasis on wastewater reclamation and reuse around the world. In this study, a combined coagulation/ceramic membrane ultrafiltration system was used to treat degreasing washing water under high pH/alkalinity conditions containing surfactants, organic matter and other materials resulting from rinsing processes in the plating industry. Coagulant exists as sol or gel species that was not suitable for complexation with smaller surfactants and organic matter under high pH/alkalinity conditions. A pH neutralization process was introduced to improve the removal efficiency of the coagulant prior to coagulation. An optimal coagulant (ferric chloride), optimal coagulation pH 5, and optimal coagulant dosage (700 mg Fe/L) were proposed as a result of coagulation tests. Under slightly acidic conditions, species distribution of ferric chloride consisted of monomers and medium polymers that were favorable for the formation of surfactant-coagulant or organic matter-coagulant complexes. In coagulation/ultrafiltration tests, filtration with in-line coagulation enhanced flux and also reduced resistance caused by internal pore blocking. This phenomenon is due to the rapid formation of a cake layer on the ceramic membrane and this cake layer lessens the adsorption of small foulants in feed water on the membrane. Moreover, filtration with in-line coagulation provided water quality similar to that of filtration of the supernatant. It is concluded that filtration with ferric chloride and in-line coagulation are valuable processes for treating degreasing washing water with reduced capital costs by excluding the need for flocculation and sedimentation zones in a water treatment plant.

Currently, there is a growing emphasis on seawater desalination and wastewater reuse around the world. Reverse osmosis (RO) is typically used for both seawater desalination and wastewater reuse. As an alternative technology of RO, forward osmosis-reverse osmosis (FO-RO) hybrid system has recently attracted attention. To select suitable membrane and feed water for this process, the performance of forward osmosis (FO) membrane according to the types of membrane and feed water quality was evaluated in this study, including three types of FO membrane (membrane I/II/III) and three types of feed water (1st/2nd/3rd wastewater treatment plant effluents, WWTP Efs). Despite high structural parameter, membrane II showed the highest water permeability (5.37LMH/bar) with highest water flux (around 27.1LMH) using draw solution of 0.5M NaCl (feed solution: 5mM NaCl). In fouling test, there was severe water flux decline when 1st WWTP Ef was used as feed solution compared to the case when 2nd/3rd WWTP Efs was supplied. Membrane III showed higher fouling resistance due to its chemistry characteristic compared to membrane I or II. Due to complex feed water characteristics, FO reversibility was lower compared to that of a previous study. Therefore, to optimize FO-RO hybrid system, FO membrane performance should be tested with different membrane characteristics and feed water qualities to develop suitable cleaning or pretreatment method and strategy to increase the reversibility of membranes.

A convenient total synthesis of a new 7-deoxyidarubicinone ivative21,the aglycon of the anticancer anti-biotic idarubicin analogue,is described.Keyfeatures of the synthesis are the Friedel-Crafts acylation and the functionalization of an A-ring si de chain. A synthon 14 for the A and B rings was prepared from intermediate 6 in five steps.

Litter size is an important trait in the pig industry. Therefore, a lot of effort has been put into improving this trait. DNA methylation is an essential epigenetic modification present in unique DNA sequences. Alterations in methylation can affect transcription and phenotypic variation. The purpose of this study was to investigate the effect of DNA methylation on litter size. Methylation-specific restriction enzymes are simple and useful tools for detecting DNA methylation status. We used a pair of methylation-sensitive isoschizomers, which have the same recognition site, HpaII and MspI. Insulin-like growth factor binding protein 4 (IGFBP4) is a key regulator of ovarian follicular development and fetal growth in eutherian mammals. In this study, we discovered that IGFBP4 was hyper-methylated in the uterus tissue of a larger litter size group using bisulfite sequencing, and validated the positive relationship between the methylation status of IGFBP4 and the total number born of pigs using the porcine methylation-specific restriction enzyme polymerase chain reaction (PMP) assay. We suggest that the IGFPB4 gene can be used as a prognostic biomarker for hyperprolific sows and that the PMP assay is a useful tool for methylation status screening.

High-quality meat is of great economic importance to the pig industry. The 1-acylglycerol-3- phosphate-O-acyltransferase 5 (AGPAT5) enzyme converts lysophosphatidic acid to phosphatidic acid in the mitochondrial membrane. In this study, we found that the porcine AGPAT5 gene was highly expressed in muscle tissue, influencing meat characteristics, and we also identified a non-synonymous single-nucleotide polymorphism (nsSNP) (rs196952262, c.673 A>G) in the gene, associated with a change of isoleucine 225 to valine. The presence of this nsSNP was significantly associated with meat color (lightness), lower cooking loss, and lower carcass temperatures 1, 4, and 12 h after slaughter (items T1, T4, and T12 on the recognized quality scale, respectively), and tended to increase backfat thickness and the waterholding capacity. These results suggest that nsSNP (c.673A>G) of the AGPAT5 gene is a potential genetic marker of high meat quality in pigs.

Abstract

Emerging evidence has shown the low levels of glutamate (Glu) and glutamine (Gln) and the hypoactivity in the cortex of patients with depression. The hypoactivity is closely related with low frequency of glutamatergic signaling that is affected by the levels of Glu and Gln. Thus, we hypothesized that there might be a causality among low levels of Glu and Gln, hypoactive glutamatergic neurotransmissions, and depressive behaviors. Here, we found low Glu and Gln levels and low frequency of spontaneous excitatory postsynaptic current (sEPSC) of glutamatergic neurons in the medial prefrontal cortex (mPFC) of chronic immobilization stress (CIS)-induced depressed mice. The depressed mice also showed hypoactive Gln synthetase (GS). Inhibition of GS by methionine sulfoximine (MSO) decreased Glu and Gln levels and increased depressive behaviors with low frequency of sEPSC in the mPFC, indicating that Glu and Gln decrements cause hypoactive glutamatergic neurotransmissions and depressive behaviors. Both Glu and Gln could increase sEPSC of glutamatergic neurons in the mPFC on slice patch, but only Gln overcame MSO to increase sEPSC, suggesting that exogenous Gln would recover CIS-induced low frequency of sEPSC caused by hypoactive GS and act as an antidepressant. Expectedly, Gln supplementation showed antidepressant effects against CIS; it increased glutamatergic neurotransmissions with Glu and Gln increment in the mPFC and attenuated depressive behaviors. Moreover, selective glutamatergic activation in the mPFC by optogenetics decreased depressive behavior. In conclusion, depressive behaviors evoked by chronic stress were due to hypoactive glutamatergic neurons in the mPFC caused by low levels of Glu and Gln, and exogenous Gln can be used as an alternative antidepressant to increase glutamatergic neurotransmission.

Highlights

Chronic stress decreased GS activity and Glu and Gln levels in the mPFC.

Chronic stress reduced glutamatergic sEPSCs but not sIPSCs in the mPFC.

GS activity inhibition resulted in the similar phenotype of chronic stress.

Gln increased glutamatergic sEPSCs regardless of GS inhibition on a slice.

Gln increased Glu and Gln levels and sEPSCs in the mPFC against chronic stress.
