We evaluated differences in ascorbic acid (AsA) and glutathione (GSH) contents and activities of enzymesinvolved in the AsA metabolism in fruits of 20 cultivars of Chinese persimmon (Diospyros kaki). The total AsAcontent ranged from 3.85 (cv. Yangshuohuoshi) to 83.02 μmol·g-1 FW (cv. Wangmoshuishi), with an average of 36.69μmol·g-1 FW across the 20 cultivars. The total GSH content ranged from 0.205 (cv. Yangshuohuoshi) to 0.815 μmol·g-1 FW(cv. Mimiguan) with an average 0.533 μmol·g-1 FW. There were significant differences in activities of dehydroascorbatereductase (DHAR, EC 1.8.5.1), monodehydroascorbate reductase (MDHAR, EC 1.6.5.4), ascorbate peroxidase (APX,EC 1.11.1.11), and glutathione reductase (GR, EC 1.6.4.2) in fruits among the 20 cultivars. The AsA content wassignificantly positively correlated with MDHAR and DHAR activities, but not with GSH and hydrogen peroxide(H2O2) contents or APX activity. These results indicated that both MDHAR and DHAR, especially MDHAR, playimportant roles in maintaining the AsA redox state by reducing oxidized AsA back to AsA and the AsA level mainlydepends on biosynthesis, rather than recycling.

Non-small-cell lung cancer (NSCLC) is a leading cause of cancer deaths worldwide. Crizotinib has been approved by the U.S. Food and Drug Administration for the treatment of patients with advanced NSCLC. However, understanding of mechanisms of action is still limited. In our studies, we confirmed crizotinib-induced apoptosis in A549 lung cancer cells. In order to assess mechanisms, small molecular docking technology was used as a preliminary simulation of signaling pathways. Interesting, our results of experiments were consistent with the results of computer simulation. This indicates that small molecular docking technology should find wide use for its reliability and convenience.

To investigate the expression of hPOT1 in the HeLa cell line and screen point mutations of hpot1 in different tumor tissues a two step osmotic method was used to extract nuclear proteins. EMSA was performed to determine the expression of hPOT1 in the HeLa cell line. PCR was also employed to amplify the exon14 sequence of the hpot1 gene in various of cancer tissues. A SV gel and PCR clean-up system was performed to enrich PCR products. DNAStar was used to analyse the exon14 sequence of the hpot1 gene. hPOT1 was expressed in the HeLa cell line and the signal was gradually enhanced as the amount of extracted nuclear proteins increased. The DNA fragment of exon14 of hpot1 was successfully amplified in the HeLa cell line and all cancer tissues, point mutations being observed in 2 out of 3 cases of endometrial cancer (66.7%) despite the hpot1 sequence being highly conserved. However, the sequence of hpot1 exon14 do not demonstrate point mutations in most cancer tissues. Since hPOT1 was expressed in HeLa cell and the probability of gene point variants was obviously higher in endometrial cancer than other cancers, it may be involved in the pathogenesis of gynecological cancers, especially in cervix and endometrium.
A Hybrid Proposed Framework for Object Detection and Classification

The object classification using the images' contents is a big challenge in computer vision. The superpixels' information can be used to detect and classify objects in an image based on locations. In this paper, we proposed a methodology to detect and classify the image's pixels' locations using enhanced bag of words (BOW). It calculates the initial positions of each segment of an image using superpixels and then ranks it according to the region score. Further, this information is used to extract local and global features using a hybrid approach of Scale Invariant Feature Transform (SIFT) and GIST, respectively. To enhance the classification accuracy, the feature fusion technique is applied to combine local and global features vectors through weight parameter. The support vector machine classifier is a supervised algorithm is used for classification in order to analyze the proposed methodology. The Pascal Visual Object Classes Challenge 2007 (VOC2007) dataset is used in the experiment to test the results. The proposed approach gave the results in high-quality class for independent objects' locations with a mean average best overlap (MABO) of 0.833 at 1,500 locations resulting in a better detection rate. The results are compared with previous approaches and it is proved that it gave the better classification results for the non-rigid classes.

Chemical Constituents from the Aerial Parts of Isodon coetsa and their Cytotoxicity

Three new compounds (1-3), including a neolignan, a triterpenoid, and a diterpenoid, together with twenty known compounds (4-23), were isolated from the aerial parts of Isodon coetsa. Their structures and relative configurations were elucidated on the basis of spectroscopic data. Compounds 1, 3, 5-9, 11-13, 16-17, and 19-23 were evaluated for their cytotoxicity against HT-29, BEL-7402, and SK-OV-3 human tumor cell lines. Compound 7 showed significant inhibitory effects on all three types of cells, with IC50 values of 2.52, 3.06, 2.14 μM, respectively.
Low temperature oxidation characteristics analysis of ultra-heavy oil by thermal methods

To study the oxidation behavior of heavy oils, low temperature oxidation experiments under differentialtemperature and pressure conditions were carried out for both heavy oils (ordinary heavy and ultraheavyoils) with thermogravimetry (TG–DTG)/differential scanning calorimeter (DSC) analyses. And theresults indicated that the differences of physical properties and oxidation reactions for the heavy oilsascribe to the SARA compositions, H/C ratio and specific oxidation path. Owing to the asphaltenescontent, the ultra-heavy oil after the oxidation has more obvious coke deposition with lower activationenergy demand to make it possible for the in-situ combustion without combustion adjuvants.
The Expression of Aldehyde Dehydrogenase Family in Breast Cancer

Purpose: It is widely accepted that aldehyde dehydrogenase(ALDH) activity is a signature of breast cancer stem cells, andhigh activity has been reported to be associated with poor clinicaloutcome. The aim of this study was to assess the expressionof members of the ALDH family of isozymes in breast cancer tissuesand to evaluate the implications of the results. Methods: Weanalyzed paraffin-embedded tumor tissue from 160 patients withbreast cancer. Immunohistochemistry (IHC) staining was performedon the slides using antibodies against different ALDHfamily members. We collated the IHC results with patient clinicalcharacteristics and determined their prognostic value. In addition,we analyzed normal, hyperplastic, and carcinomatous tissues insitu to check their ALDH distributions. Results: All the testedALDH members were detected in the various tissue types, but atdifferent levels. Only ALDH 1A3 was found to be significantly associatedwith distant metastasis (p=0.001), disease-free survival(p<0.001), and overall survival (p<0.001). Conclusion: The levelof ALDH 1A3 in breast cancer tissue is a predictive marker of apoor clinical outcome.
Guided waves of porous FG nanoplates with four edges clamped

Based on the nonlocal strain gradient (NSG) theory and considering the influence of moment of inertia, the governing equations of motion of porous functionally graded (FG) nanoplates with four edges clamped are established; The Galerkin method is applied to eliminate the spatial variables of the partial differential equation, and the partial differential governing equation is transformed into an ordinary differential equation with time variables. By satisfying the boundary conditions and solving the characteristic equation, the dispersion relations of the porous FG strain gradient nanoplates with four edges fixed are obtained. It is found that when the wave number is very small, the influences of nonlocal parameters and strain gradient parameters on the dispersion relation is very small. However, when the wave number is large, it has a great influence on the group velocity and phase velocity. The nonlocal parameter represents the effect of stiffness softening, and the strain gradient parameter represents the effect of stiffness strengthening. In addition, we also study the influence of power law index parameter and porosity on guided wave propagation.
