        Morphological and functional evaluation of an animal model for the retinal degeneration induced by N-methyl-N-nitrosourea

        The retinal degeneration (RD) is a general cause of blindness. To study its pathophysiology and evaluate the effects of new therapeutic agents before clinical trials it is essential to establish reliable and stable animal models. Th is study evaluated a RD animal model in which blindness was induced by N-methyl-N-nitrosourea (MNU) a potent retinotoxin leading to apoptosis of photoreceptors. MNU was applied to the Sprague-Dawley rats by a single intraperitoneal injection in different doses (40 50 and 60 ㎎/㎏). Th e retinal functions were examined at 1 week after MNU injection by electroretinogram (ERG). Afterwards each retina was examined by hematoxylin and eosin stain and immunohistochemistry with anti-glial fibrillary acidic protein antibody. Upon MNU injection of 40 50 and 60 ㎎/㎏ the ERG amplitude of a-waves showed significant reductions of 7 26 and 44% respectively when compared to that of normal a-waves. The b-wave amplitudes were about 89 65 and 58% of normal b-waves in the response to scotopic light stimulus. At 1 week 2 weeks and 4 weeks aft er MNU injection (50 ㎎/㎏) all scotopic ERG components decreased progressively. In addition degeneration of retinal neurons was observed in a time- and dose-dependent manner after MNU injection. Taken together functional reduction following RD induced by MNU correlates with morphological changes. Thus this RD rat model may be a useful model to study its pathophysiology and to evaluate the effects of new therapeutic agents before clinical trials.

        Capnocytophaga endodontalis sp. nov., Isolated from a Human Refractory Periapical Abscess

        <P>A novel Gram-negative, capnophilic, fusiform bacterium, designated strain ChDC OS43(T), was isolated from a human refractory periapical abscess in the left mandibular second molar and was characterized by polyphasic taxonomic analysis. The 16S rRNA gene sequence revealed that the strain belongs to the genus Capnocytophaga, as it showed sequence similarities to Capnocytophaga ochracea ATCC 27872(T) (96.30%) and C. sputigena ATCC 33612(T) (96.16%). The prevalent fatty acids of strain ChDC OS43(T) were isoC(15:0) (57.54%), C-16:0 (5.93%), C-16:0 3OH (5.72%), and C-18:1 cis 9 (4.41%). The complete genome of strain ChDC OS43(T) was 3,412,686 bp, and the G+C content was 38.2 mol%. The average nucleotide identity (ANI) value between strain ChDC OS43(T) and C. ochracea ATCC 27872(T) or C. sputigena ATCC 33612(T) was > 92.01%. The genome-to-genome distance (GGD) value between strain ChDC OS43(T) and C. ochracea ATCC 27872(T) or C. sputigena ATCC 33612(T) was 32.0 and 45.7%, respectively. Based on the results of phenotypic, chemotaxonomic, and phylogenetic analysis, strain ChDC OS43(T) (= KCOM 1579(T) = KCTC 5562(T) = KCCM 42841(T) = JCM 32133(T)) should be classified as the type strain of a novel species of genus Capnocytophaga, for which the name Capnocytophaga endodontalis sp. nov. is proposed.</P>

        일반논문 : 중학교 과학 생명 영역의 스토리텔링 학습 자료 개발

        본 연구에서는 학생의 과학에 대한 흥미와 학습 동기를 향상시키기 위해서 스토리텔링을 적용하여 중학교 과학 생명 영역의 학습 자료를 개발하였다. Ellis & Brewster의 스토리텔링 모형에 근거하여 학습 자료의 초안을 개발하였고, 생물 교육 전공자들의 검토를 거쳐서 수정·보완하였다. 이 자료는 중학생들을 대상으로 예비 적용되었으며, 그 결과를 반영하여 최종적으로 학습 자료를 완성하였다. 또한 개발한 자료에 대하여 중학교 과학 교사 28명의 의견을 조사하였다. 본 연구에서 개발한 학습 자료는 2009 개정 중학교 과학 생명 영역의 5개의 단원에 대하여 각각 2개씩 총 10개로 구성되어 있다. ``광합성`` 단원에서는 ``식물로의 여행``이라는 주제로 가상 상황을 제시하여 이야기를 구성하였고, ``소화·순환·호흡·배설`` 단원에서는 ``체육대회``라는 주제로 우리 몸의 각 기관들을 등장인물로 설정하여 시나리오 형식으로 구성하였다. ``자극과 반응`` 단원에서는 우리나라 전래동화인 해와 달이 된 오누이, 콩쥐 팥쥐 이야기를 과학 개념과 연결하여 재구성하였다. ``생식과 발생`` 단원에서는 정자와 수정란을 주인공으로 하여 아기가 되기까지의 과정을 이야기 형식으로 구성하였고, ``유전과 진화`` 단원에서는 멘델과 다윈의 일화를 일기 형식으로 구성하였다. 본 연구에서 개발한 학습 자료에 대한 교사들의 응답을 살펴본 결과, 중학생의 학습 동기나 흥미 유발 도움 정도에 대해서는 5점 만점에 4.16, 학교 수업에서의 적용 가능성에 대해서는 5점 만점에 3.97로 나타났다. 그리고 28명 중에서 23명 이상이 본인 수업에서 이 자료를 활용할 의사가 있다고 응답하였다. 따라서 본 연구에서 개발한 스토리텔링 학습 자료는 대단원의 주요 학습 내용을 다루면서 단원별로 다양한 소재와 형식으로 이야기가 구성되어 있고, 학생들의 과학 학습 동기와 흥미를 유발시키는 데 도움을 줄 수 있는 자료라고 생각한다. In this study, learning materials were developed in life area of middle school science applying storytelling to improve the students` interest and motivation on science. In order to do this, the draft of learning materials were developed based on the storytelling model of Ellis & Brewster, and revised through the review of biology education majors. The pilot test of this materials were implemented on middle school students, and then the result was reflected to complete the learning materials. In addition, 28 middle school science teachers were surveyed to get comment on the developed materials. The learning materials developed in this study are composed of the contents on life area of middle school science according to 2009 revised curriculum. In the ``Photosynthesis`` unit, the story is constructed that the students go into the plant by the bus and observe the structure of the plant and the process of photosynthesis. In the ``Digestion·Respiration·Circulation·Excretion`` unit, the story is constructed on the form of scenario that the organs of human body are characterized and talk about the change after running or lunch. In the ``Stimulus and Response`` unit, the brother and sister who became the sun and the moon and the Kongjwi and Patjwi, which are Korean fairy tales, are reconstructed by connecting with scientific concepts. In the ``Reproduction and Development`` unit, the story is constructed on the process that is from the sperm and egg to the baby, with characterizing the sperm and fertilized egg. In the ``Heredity and Evolution`` unit, the story is constructed on the form of diary including the anecdotes of Mendel and Darwin. The result of teachers` response is as follows. In the respect of students` interest and motivation on science, the score is 4.16, and in that of appropriateness to apply in an actual class, the score is 3.97. And more than 23 teachers are willing to use these materials. In conclusion, the storytelling based learning materials developed in this study would be helpful to improve students` science learning motivation and interest.

      • Effects of strobe light stimulation on postnatal developing rat retina

        <P>The nature and intensity of visual stimuli have changed in recent years because of television and other dynamic light sources. Although light stimuli accompanied by contrast and strength changes are thought to have an influence on visual system development, little information is available on the effects of dynamic light stimuli such as a strobe light on visual system development. Thus, this study was designed to evaluate changes caused by dynamic light stimuli during retinal development. This study used 80 Sprague-Dawley rats. From eye opening (postnatal day 14), half of the rats were maintained on a daily 12-h light/dark cycle (control group) and the remaining animals were raised under a 12-h strobe light (2 Hz)/dark cycle (strobe light-reared group). Morphological analyses and electroretinogram (ERG) were performed at postnatal weeks 3, 4, 6, 8, and 10. Among retinal neurons, tyrosine hydroxylase-immunoreactive (TH-IR, dopaminergic amacrine cells) cells showed marked plastic changes, such as variations in numbers and soma sizes. In whole-mount preparations at 6, 8, and 10 weeks, type I TH-IR cells showed a decreased number and larger somata, while type II TH-IR cells showed an increased number in strobe-reared animals. Functional assessment by scotopic ERG showed that a-wave and b-wave amplitudes increased at 6 and 8 weeks in strobe-reared animals. These results show that exposure to a strobe light during development causes changes in TH-IR cell number and morphology, leading to a disturbance in normal visual functions.</P>

      • Cyanidin-3-glucoside Extracted from Mulberry Fruit Can Reduce <i>N</i>-methyl-<i>N</i>-nitrosourea-Induced Retinal Degeneration in Rats

        <P><I>Purpose</I>: To investigate the effect of cyanidin-3-<I>O</I>-glucoside (C3G) on a rat retinal degeneration (RD) model.</P><P><I>Materials and methods</I>: Experimental RD was induced in rats by the intraperitoneal injection of <I>N</I>-methyl-<I>N</I>-nitrosourea (MNU) at 50 mg/kg. C3G extracted from mulberry (<I>Morus alba </I>L.) fruit (50 mg/kg) was orally administered, daily for 1, 2 and 4 weeks after MNU injection. The effects of C3G administration on MNU-induced RD retinas were histologically and functionally assessed by hematoxylin and eosin staining and electroretinography (ERG), respectively. The degree of retinal injury in C3G-administered RD rats was evaluated by immunohistochemistry with an antibody against glial fibrillary acidic protein (GFAP). The preferential protective effect of C3G on scotopic vision was examined by western blot analysis.</P><P><I>Result</I>s: Marked loss of photoreceptors in the outer nuclear layer (ONL) was observed in RD rats at 2 and 4 weeks after MNU injection, while the ONL in the MNU-induced RD rats given C3G was relatively well preserved. Immunohistochemistry with anti-GFAP showed that retinal injury was also reduced in the retinas of the rats given C3G. Functional assessment by using ERG recordings showed that scotopic ERG responses were significantly increased in RD rats given C3G for 4 weeks (<I>p</I> < 0.01) compared with that of untreated RD rats. In the RD rats given short-term C3G (for 1 and 2 weeks), the increase in ERG responses was not significant. In addition, western blot analysis showed that rhodopsin level in the C3G-administered RD retinas significantly increased compared to that in the non-administered RD retinas (<I>p</I> < 0.05), whereas red/green opsin level did not show any significant difference.</P><P><I>Conclusions</I>: Long-term administration of C3G extracted from mulberry fruit could structurally reduce photoreceptor damage and functionally improve scotopic visual functions in the RD rat model induced by MNU.</P>
