        Overexpression of cholinergic receptor nicotinic gamma subunit inhibits proliferation and differentiation of bovine preadipocytes

        Objective: Muscle acetylcholine receptors have five alpha subunits (α, β, δ, ε, or γ), and cholinergic receptor nicotinic gamma subunit (CHRNG) is the γ subunit. It may also play an essential role in biological processes, including cell differentiation, growth, and survival, while the role of CHRNG has not been studied in the literature. Therefore, the purpose of this study is to clarify the effect of CHRNG on the proliferation and differentiation of bovine preadipocytes. Methods: We constructed a CHRNG overexpression adenovirus vector and successfully overexpressed it on bovine preadipocytes. The effects of CHRNG on bovine preadipocyte proliferation were detected by Edu assay, cell counting Kit-8 (CCK-8), real-time fluorescence quantitative polymerase chain reaction (RT-qPCR), Western blot and other techniques. We also performed oil red O, RT-qPCR, Western blot to explore its effect on the differentiation of preadipocytes. Results: The results of Edu proliferation experiments showed that the number of EDUpositive cells in the overexpression group was significantly less. CCK-8 experiments found that the optical density values of the cells in the overexpression group were lower than those of the control group, the mRNA levels of proliferating cell nuclear antigen (PCNA), cyclin A2 (CCNA2), cyclin B1 (CCNB1), cyclin D2 (CCND2) decreased significantly after CHRNG gene overexpression, the mRNA levels of cyclin dependent kinase inhibitor 1A (CDKN1A) increased significantly, and the protein levels of PCNA, CCNB1, CCND2 decreased significantly. Overexpression of CHRNG inhibited the differentiation of bovine preadipocytes. The results of oil red O and triglyceride determination showed that the size and speed of lipid droplets accumulation in the overexpression group were significantly lower. The mRNA and protein levels of peroxisome proliferator activated receptor gamma (PPARγ), CCAAT enhancer binding protein alpha (CEBPα), fatty acid binding protein 4 (FABP4), fatty acid synthase (FASN) decreased significantly. Conclusion: Overexpression of CHRNG in bovine preadipocytes inhibits the proliferation and differentiation of bovine preadipocytes.

        Investigation on the Influence of Temperature on Starting Torque of Magnetic Fluid Seal

        The widest application of magnetic fluid is in magnetic fluid seals. The starting torque varying with temperature of magnetic fluid seal has limited its application in special seal field such as military industry. This paper firstly measure the starting torque of magnetic fluid seals under different temperature conditions from −55 oC to 70 ℃ after holding for 2.5 hours in a closed environment with uniform temperature. And the law of starting torque with temperature change is studied experimentally. The results show that the starting torque of the magnetic fluid seal changes unobvious at 25 ℃ or higher temperature. The starting toque of magnetic fluid seal increases with the decrease of temperature, and the causes of which are analyzed. The results of this study provide experimental and theoretical reference for reducing the staring torque of magnetic fluid seal in low temperature environment.

        Experimental Study on Lime-Treated Waste Soil Based on Water Transfer Mechanism

        A large amount of waste soil (mud) is being produced from the construction of civil and infrastructures. As a result, resource utilization of the waste soil has become even more important in geotechnical engineering community. This study quantitatively investigates the underlying modification mechanism of lime-treated waste soil based on water transfer mechanism. Content of different types of water was first measured, and content of each hydration product and the Atterberg limits of soils before and after the lime treatment were then analyzed. It is found the mechanism of lime treatment is to transfer free water with low potential energy to bound water and hydration water with high potential energy. In addition, the formation of hydrates gel mainly improves soil plasticity by affecting the limit of soil plasticity. The hydration water can be used to evaluate the effectiveness of treatment and the growth potential.

        A novel and effective approach to enhance the interfacial interactions of meta-aramid fibers

        Meta-aramid paper exhibits some unique properties, including high temperature resistance, electricalinsulation. However, due to the inherent chemical inertness of aramid fibers, the interfacial bondingbetween fibers is weak, which negatively affect the properties of aramid paper. Herein, we report a simple,effective and scalable process for substantially improving the interfacial bonding between aramidfibers, thus, mechanical and insulating properties of the meta-aramid composite paper, and it wasachieved by surface coating of aramid paper with meta-aramid stock solution that contains N, N dimethylacetamide(DMAc)-CaCl2 and high molecular weight poly (m-phenylene isophthalamide) polymers. Results show that the tensile strength, modulus, internal bond strength and tearing index of the resultantall-PMIA paper increased by 83 %, 58 %, 173 % and 89 %, respectively, in comparison with those of the control. The breakdown strength of the improved aramid paper is 190 % higher than that of the control paper,attaining 26.46 KV/mm. This work provides a simple, economical, efficient, and scalable method toimprove the overall performance of meta-aramid paper, which has great potential to be implementedat the industrial scale.
