      • Quantum Chemical Consideration of CO Adsorption over Cation Exchanged Faujasite

        CNDO/2 calculation for atomic charges Wiberg bond orders and adsorption energies of CO molecules on the cluster model whose Si/Al ratio varied were carried out. The data for the normal cluster and dealuminated cluster were compared. Decationization energies of the canons increased with the charge densities of canon and number of aluminum involved. Adsorption process of CO on the monovalent canons such as , Li^-, Na^+ and divalent canons, Be^(2+), Ca^(2+), and Mg^(2-) was supposed to be occurring by the donation of non-bonded electrons from CO. The decationization energies of canons obviously decreased by the dealumination process. Adsorption energies of CO on the canons generally decreased ache dealumination took place except the case of H^+ and Na^+.

      • Survey of DNA barcoding of Hemiptera

        Recently, hundreds thousands of CO1 barcodes have been produced in many insect groups however, relatively little is known in Hemiptera. In this study, we newly determine 1173 sequences for 374 species distributed among 190 genera and 33 families collected mostly from North America. Average age of the specimen was 12.2 years and about 70% of the specimens produce barcode over 500 bp among examined 1700 specimens. Con-generic divergency calculated by K2P distance was 9.99 which was 11fold higher than the mean intraspecific variation. A steady increase of genetic variation through increasing taxonomic levels was also observed. We found several taxon groups with deep divergency (over 2.0 K2P distance) which were morphologically identified as a same species. Some mixed clusters with several species were also found and suggested more detailed study for these clusters through both morphological and molecular biological method.

        Investigation of Angiotensin Glycosylation by MALDI-TOF and ESI Tandem Mass Spectrometry

        Angiotensin I, a model decapeptide, was glycosylated and partially hydrolyzed with HCl (6 N, 80 $^{\circ}C$, 4 h), aminopeptidase, and carboxypeptidase Y. A single peptide mass map obtained from truncated peptides in the partial acid hydrolysate of angiotensin and its glycosylation product mixture by matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry enabled sequencing of angiotensin by a combinatorial procedure. MALDI-TOF and electrospray ionization (ESI) tandem mass spectrometric results indicate that both the N-terminal amino group of aspartic acid and the guanidinium group of the second residue arginine are glycosylated.

      • Lutibacter crassostreae sp. nov., isolated from oyster.

        <P>A Gram-stain-negative, aerobic, non-spore-forming, non-flagellated, non-gliding and ovoid or rod-shaped bacterial strain, designated TYO-8T, was isolated from an oyster collected from the South Sea in South Korea. Strain TYO-8T grew optimally at 25?C, at pH?7.0-8.0 and in the presence of 2.0-3.0?% (w/v) NaCl. A neighbour-joining phylogenetic tree, based on 16S rRNA gene sequences, revealed that strain TYO-8T fell within the clade comprising the type strains of species of the genus Lutibacter, clustering coherently with the type strain of Lutibacter litoralis with a sequence similarity of 99.3?%. Strain TYO-8T exhibited 16S rRNA gene sequence similarity values of 95.3-97.5?% to the type strains of other species of the genus Lutibacter and of less than 92.9?% to the type strains of other species with validly published names. Strain TYO-8T contained MK-6 as the predominant menaquinone and iso-C15?:?0, iso-C15?:?0 3-OH and iso-C15?:?1 G as the major fatty acids. The major polar lipids of strain TYO-8T were phosphatidylethanolamine and two unidentified lipids. The DNA G+C content of strain TYO-8T was 33.8?mol% and its DNA-DNA relatedness values with the type strains of L. litoralis, Lutibacter aestuarii and Lutibacter flavus were 13-27?%. The differential phenotypic properties, together with the phylogenetic and genetic distinctiveness, revealed that strain TYO-8T is distinct from other species of the genus Lutibacter. On the basis of the data presented, strain TYO-8T is considered to represent a novel species of the genus Lutibacter, for which the name Lutibacter crassostreae sp. nov. is proposed. The type strain is TYO-8T (?=?KCTC 42461T?=?NBRC 110923T).</P>

        Fcγ Receptors Modulate Pulmonary Inflammation by Activating Innate Immune Cells in Murine Hypersensitivity Pneumonitis

        Background: Hypersensitivity pneumonitis (HP) is an interstitial lung disease that develops following repeated exposure to inhaled particulate antigens. The family of $Fc{\gamma}$ receptors ($Fc{\gamma}Rs$) has emerged as central regulators for modulating both pro-and anti-inflammatory responses. However, the role of $Fc{\gamma}Rs$ in the development of HP has not been investigated yet. Methods: To explore the functional roles of $Fc{\gamma}Rs$ in HP, $Fc{\gamma}R^{-/-}$ and B6 mice were challenged with Saccharopolyspora rectivirgula (SR) antigen intranasally, and compared these mice in terms of the histological change, infiltrated immune cells in BALF and in vitro immune responses. Results: $Fc{\gamma}R^{-/-}$ mice exhibited attenuation of HP in terms of histological alterations, and reduced numbers of neutrophils and macrophages in and the increased CD4 : CD8 ratio of bronchoalveolar lavage fluid. The lungs of $Fc{\gamma}R^{-/-}$ mice showed high production of Th2 cytokine such as IL-4 and slightly low production of Th1 cytokine, INF-${\gamma}$ compared to those of B6 mice. However, SR-specific adaptive immune responses of $Fc{\gamma}R^{-/-}$ mice were similar to those of B6 mice. Conclusion: These results demonstrate that activating $Fc{\gamma}$ receptors play an important role in activating neutrophils and macrophages in pulmonary inflammation and inducing Th1 differentiation by regulating cytokine expression in SR-induced HP.

      • Effects of Ultraviolet Light on DNA Replication and Repair in Cultured Myoblast Cells of Chick Embryo

        DNA synthesis, unscheduled DNA synthesis, excision of pyrimidine dimers and phtoreactivation were determined in UV-irradiated differentiating muscle cells at various times of primary culture of 12 day chick embryos and results obtained were as follows. The rates of UV-induced unscheduled DNA synthesis were increased as increase of UV dose. And the rates were gradually decreased as the increase of time after culture, but at higher doses the decreasing tendency was remarkable. The patterns of DNA replication were changed drastically as a function of time so that in the seven day cultures the rate of $^3$H-thymidine incorporation was found to be 0.2% of the original activity. The pattern of inhibition of DNA replication by UV damage demonstrated that in cells of earlier stages there were no remarkable changes, but in cells of later stages there was significant fluctuation. Photoreactivation and the excision of pyrimidine dimer in the one day cultures showed that photoreactivation occurred immediately after UV-irradiation, but excision of pyrimidine dimer was gradually and slowly occurred. These results indicate that the differentiation of embryonic muscle cells accompanies the gradual reduction of DNA replication and unscheduled DNA synthesis, and that the photoreactivation is rapid process compared to excision repair. 발생 12일째의 계배근세포를 약 7일간 배양하면서 자외선에 의한 절제회복, 광재활성 및 DNA 복제억제율을 조사하여 다음과 같은 결과를 얻었다. 자외선에 의한 절제회복은 계배근세포의 분화정도가 진행됨에 따라 감소하였으며, 이는 특히 자외선의 선량이 높을수록 뚜렷하였다. DNA 합성율은 분화가 진행된 세포일수록 현저히 감소하는 경향이었으며 각 분화단계에 따른 DNA 복제억제 현상은 배양 초기 세포인 경우 자외선 조사후 30분에서 1시간반 사이에서 가장 심하게 나타났고, 배양후기의 세포에서는 이러한 억제현상이 뚜렷하지 않았다. 또 배양 1일째 세포에서 광재활성에 의한 피리미딘 이량체의 감소율은 자외선 조사직후에 현저하였다가 그후 시간의 경과에 따른 차이는 보이지 않았다. 그러나, 절제회복만에 의한 이량체의 감소는 조사후 시간이 지남에 따라 서서히 감소하여 광재활성에 의한 이량체의 감소량과 비슷한 수준으로 접근하였다.

      • <i>Phaeobacter aquaemixtae</i> sp. nov., isolated from the junction between the ocean and a freshwater spring

        <P>A Gram-reaction-negative, aerobic, non-spore-forming, non-motile and rod-shaped or ovoid bacterial strain, designated SSK6-1<SUP>T</SUP>, was isolated from the zone where the ocean and a freshwater spring meet at Jeju island, South Korea. Strain SSK6-1<SUP>T</SUP> grew optimally at 30 °C, at pH 7.0–7.5 and in the presence of 2.0 % (w/v) NaCl. Phylogenetic trees based on 16S rRNA gene sequences revealed that strain SSK6-1<SUP>T</SUP> clustered with <I>Phaeobacter daeponensis</I> TF-218<SUP>T</SUP> and <I>Phaeobacter caeruleus</I> LMG 24369<SUP>T</SUP>, exhibiting sequence similarities of 98.6 and 98.3 %, respectively. The novel strain exhibited sequence similarities of 94.6–97.8 % to the type strains of other recognized species of the genera <I>Phaeobacter</I> and <I>Leisingera</I>. Strain SSK6-1<SUP>T</SUP> contained Q-10 as the predominant ubiquinone and C<SUB>18 : 1</SUB>ω7<I>c</I> and 11-methyl C<SUB>18 : 1</SUB>ω7<I>c</I> as the major fatty acids. The major polar lipids of strain SSK6-1<SUP>T</SUP> were phosphatidylcholine, phosphatidylglycerol, phosphatidylethanolamine, one unidentified aminolipid and one unidentified lipid. The DNA G+C content of strain SSK6-1<SUP>T</SUP> was 64.6 mol% and its DNA–DNA relatedness values with <I>P. daeponensis</I> TF-218<SUP>T</SUP> and <I>P. caeruleus</I> were 21 and 25 %, respectively. The differential phenotypic properties, together with phylogenetic and genetic distinctiveness, revealed that strain SSK6-1<SUP>T</SUP> is separate from recognized <I>Phaeobacter</I> species. On the basis of the data presented, strain SSK6-1<SUP>T</SUP> is considered to represent a novel species of the genus <I>Phaeobacter</I>, for which the name <I>Phaeobacter aquaemixtae</I> sp. nov. is proposed. The type strain is SSK6-1<SUP>T</SUP> ( = KCTC 32538<SUP>T</SUP> = CECT 8399<SUP>T</SUP>).</P>

        Association of Deoxyhypusine Hydroxylase with Peroxiredoxin I was Modulated in the Yeast Saccharomyces cerevisiae under Oxidative Stress

        Eukaryotic translation initiation factor 5A (eIF5A) is the only hypusine-containing protein, which is formed by deoxyhypusine synthase (DHS) and deoxyhypusine hydroxylase (DOHH). DOHH is a novel metalloenzyme with HEAT [named for human huntingtin (H), elongation factor 3 (E), a subunit of protein phosphatase 2A (A), and the target of rapamycin (T)]-repeat motifs. Inspite of much progress in determining the roles of iron-containing DOHH holoenzyme as an eIF5A hydroxylase, little is known about iron-free apoenzyme of DOHH under certain stress conditions. For this purpose, we compared cell growth in two yeast strains subjected to oxidative damage. Thus, the existence of more viable cells in the Saccharomyces cerevisiae BY4743 (parental yeast) strain than in the DOHH-null strain after H2O2 treatment indicates the importance of DOHH in protecting yeast cells against oxidative stress. To identify endogenous target proteins influenced by DOHH under oxidative damage, proteomic analysis was applied to the two yeast strains. Of these proteins, the oxidized form of peroxiredoxin I (PrxI) was concomitantly up-regulated in both strains under H2O2 treatment. Two-dimensional electrophoresis (DE) followed by immunoblot analysis shows that the recovery of the oxidized PrxI to the reduced enzyme under H2O2 treatment was found to be much slower in the DOHH-null strain than in the parental strain. Based on the results, we discovered a possible interaction between DOHH and PrxI by immunoprecipitation/immunoblotting in yeast under oxidative stress. Taken together, these results suggest that DOHH might be a candidate protein for protection of yeast cells against oxidative stress in conjunction with PrxI.
