        2-[(4-Cyanophenyl)amino]-3-chloro-1, 4-naphthalenedione (NQ-Y15) was asssayed for its genotoxic potential by using Salmonella typhimurium reversion assay and in vitro chromosome aberration test on Chinese hamster lung cells. In the Ames test, NQ-Y15 induced his+revertants of Salmonella typhimurium TA 98 and TA 1537, reaching levels twice the negative control values. But, NQ-Y15 induced only his+revertants of Salmonella typhimurium TA 1537 more than twice the control values under the condition with metabolic activation system. In the cytogenetic test on chinese hamster lung cells. NQ-Y15 showed significant chromosomal aberrations, but the incidence was significantly reduced in the presence of metabolic activation.

        A New ent-Kaurane type Diterpenoid Glycoside from Inula japonica Thunb

        A new ent-kaurane type diterpenoid glycoside, 17-O-β-D-glucopyranosyl-16α-ent-kauran-19-oic acid (1), together with 17-hydroxy-16α-ent-kauran-19-oic acid (2), 16α,17-dihydroxyl-ent-kauran-19-oic acid (3), and 16α-hydroxy-17-acetoxy-ent- kauran-19-oic acid (4) were isolated from the aerial parts of Inula japonica Thunb. The structure of 1 was determined mainly by use of 1D and 2D NMR spectroscopic techniques including HSQC, 1H-1H COSY, HMBC, and NOESY. In addition, 4 exhibited significant inhibitory activity on NO production in LPS-stimulated RAW264.7 cells with IC50 value of 14.3 μg/mL.

        Phosphorylation of DYNLT1 at Serine 82 Regulates Microtubule Stability and Mitochondrial Permeabilization in Hypoxia

        Hypoxia-induced microtubule disruption and mitochondrial permeability transition (mPT) are crucial events leading to fatal cell damage and recent studies showed that microtubules (MTs) are involved in the modulation of mitochondrial function. Dynein light chain Tctex-type 1 (DYNLT1) is thought to be associated with MTs and mitochondria. Previously we demonstrated that DYNLT1 knockdown aggravates hypoxia-induced mitochondrial permeabilization, which indicates a role of DYNLT1 in hypoxic cytoprotection. But the underlying regulatory mechanism of DYNLT1 remains illusive. Here we aimed to investigate the phosphorylation alteration of DYNLT1 at serine 82 (S82) in hypoxia (1% O2). We therefore constructed recombinant adenoviruses to generate S82E and S82A mutants, used to transfect H9c2 and HeLa cell lines. Development of hypoxia-induced mPT (MMP examining, Cyt c release and mPT pore opening assay), hypoxic energy metabolism (cellular viability and ATP quantification), and stability of MTs were examined. Our results showed that phosph-S82 (S82-P) expression was increased in early hypoxia; S82E mutation (phosphomimic) aggravated mitochondrial damage, ele-vated the free tubulin in cytoplasm and decreased the cellular viability; S82A mutation (dephosphomimic) seemed to diminish the hypoxia-induced injury. These data suggest that DYNLT1 phosphorylation at S82 is involved in MTs and mitochondria regulation, and their interaction and cooperation contribute to the cellular hypoxic tolerance. Thus, we provide new insights into a DYNLT1 mechanism in stabilizing MTs and mitochondria, and propose a potential therapeutic target for hypoxia cytoprotective studies.

        Analysis and Detection of Transmission Lines Based on Twin Reality

        Now life has been transformed and upgraded to the direction of intelligence, digitalization and informatization along with the advancement of scientific and technological information. The traditional detection of transmission lines is completed by the professional maintenance per-sonnel of the power grid through manual detection of lines. This method is inefficient and has a certain threat to the life safety of detection personnel. The development of deep learning and computational vision supplies a fresh idea for detection of transmission lines. Therefore, this paper analyzes the traditional Faster R-CNN based on twin reality scene modeling technology. Then aiming at the shortcomings of traditional algorithms, the Cascade R-CNN algorithm is proposed to complete the detection of transmission lines based on twin reality system. It com-pares the accuracy and other indicators of Cascade R-CNN and Faster R-CNN algorithms. The effectiveness of two algorithms for analysis and detection of transmission lines is verified through experiments. Finally, the results indicates that the system using Cascade R-CNN has higher prediction accuracy, and has better practicability for analysis and detection of transmission lines.

        MiRNA320a Inhibitor-Loaded PLGA-PLL-PEG Nanoparticles Contribute to Bone Regeneration in Trauma-Induced Osteonecrosis Model of the Femoral Head

        BACKGROUND: This study aimed to explore the effect of a nanomaterial-based miR-320a inhibitor sustained release system in trauma-induced osteonecrosis of the femoral head (TIONFH). METHODS: The miR-320a inhibitor-loaded polyethylene glycol (PEG)- Poly(lactic-co-glycolic acid) (PLGA)- Poly-L-lysine (PLL) nanoparticles were constructed using the double emulsion method. The TIONFH rabbit model was established to observe the effects of miR-320a inhibitor nanoparticles in vivo. Hematoxylin–eosin staining and microcomputed tomography scanning were used for bone morphology analysis. Bone marrow mesenchymal stem cells (BMSCs), derived from TIONFH rabbits, were used for in vitro experiments. Cell viability was determined using the MTT assay. RESULTS: High expression of miR-320a inhibited the osteogenic differentiation capacity of BMSCs in vitro by inhibiting the expression of the osteoblastic differentiation markers ALP and RUNX2. MiR-320a inhibitor-loaded PEG-PLGA-PLL nanoparticles were constructed with a mean loading efficiency of 1.414 ± 0.160%, and a mean encapsulation efficiency of 93.45 ± 1.24%, which released 50% of the loaded miR-320a inhibitor at day 12 and 80% on day 18. Then, inhibitor release entered the plateau. After treatment with the miR-320a inhibitor nanoparticle, the empty lacunae were decreased in the femoral head tissue of TIONFH rabbits, and the osteoblast surface/bone surface (Ob.S/BS), osteoblast number/bone perimeter (Ob.N/B.Pm), bone volume fraction, and bone mineral density increased. Additionally, the expression of osteogenic markers RUNX2 and ALP was significantly elevated in the TIONFH rabbit model. CONCLUSION: The miR-320a inhibitor-loaded PEG-PLGA-PLL nanoparticle sustained drug release system significantly contributed to bone regeneration in the TIONFH rabbit model, which might be a promising strategy for the treatment of TIONFH.         Study of Spectral Reflectance Reconstruction Based on an Algorithm for Improved Orthogonal Matching Pursuit

        Spectral reflectance is sparse in space, and while the traditional spectral-reconstruction algorithm doesnot make full use of this characteristic sparseness, the compressive sensing algorithm can make full useof it. In this paper, on the basis of analyzing compressive sensing based on the orthogonal matching pursuitalgorithm, a new algorithm based on the Dice matching criterion is proposed. The Dice similarity coefficientis introduced, to calculate the correlation coefficient of the atoms and the residual error, and is used toselect the atoms from a library. The accuracy of Spectral reconstruction based on the pseudo-inversemethod, Wiener estimation method, OMP algorithm, and DOMP algorithm is compared by simulation onthe MATLAB platform and experimental testing. The result is that spectral-reconstruction accuracy basedon the DOMP algorithm is higher than for the other three methods. The root-mean-square error and colordifference decreases with an increasing number of principal components. The reconstruction error decreasesas the number of iterations increases. Spectral reconstruction based on the DOMP algorithm can improvethe accuracy of color-information replication effectively, and high-accuracy color-information reproductioncan be realized.

      • Predictive and Prognostic Roles of Ribonucleotide Reductase M1 in Patients with Pancreatic Cancer Treated with Gemcitabine: A Meta-analysis

        Increasing scientific evidence suggests that ribonucleotide reductase M1 (RRM1) may be a powerful predictor of survival in patients with pancreatic cancer treated with adjuvant gemcitabine-based chemotherapy after operative resection, but many existing studies have yielded inconclusive results. This meta-analysis aimed to assess the prognostic role of RRM1 in predicting survival in patients with pancreatic cancer treated with gemcitabine. An extensive literature search for relevant studies was conducted on PubMed, Embase, Web of Science, Cochrane Library, and CBM databases from their inception through May 1st, 2013. This meta-analysis was performed using the STATA 12.0 software and crude hazard ratios (HRs) with 95% confidence intervals (CIs) were calculated. Eight clinical studies were included in this meta-analysis with a total of 665 pancreatic cancer patients treated with adjuvant gemcitabine-based chemotherapy, including 373 patients in the high RRM1 expression group and 292 patients in the low RRM1 expression group. Our meta-analysis revealed that high RRM1 expression was associated with improved overall survival (OS) of pancreatic cancer patients (HR=1.56, 95%CI=0.95-2.17, P<0.001). High RRM1 expression also was linked to longer disease-free survival (DFS) than low RRM1 expression (HR=1.37, 95%CI=0.25-2.48, P=0.016). In conclusion, our meta-analysis suggests that high RRM1 expression may be associated with improved OS and DFS of pancreatic cancer patients treated with adjuvant gemcitabine-based chemotherapy. Detection of RRM1 expression may be a promising biomarker for gemcitabine response and prognosis in pancreatic cancer patients.

        Performance investigation of nanoscale thermal cloak by the perforated silicon film

        In recent years, nanoscale thermal cloak, as a representative of nanoscale heat flux regulation devices, has attracted a lot of attention from researchers. However, the existing design methods are relatively complicated and all adopt constant temperature boundaries, the temperature changes constantly in the real environment, which greatly hinders its engineering applications. In this paper, inspired by phonon localization theory, we construct a nanoscale thermal cloak by a perforated silicon membrane and evaluate its cloaking performance and dynamic response. Results show that when the perforated area is fixed, the more the number of holes, the better the cloaking performance. In addition, the nanoscale thermal cloak still exhibits good cloaking performance in the dynamic environment. Finally, the cloaking mechanism is analyzed by calculating the phonon density of states (PDOS) and mode participation rate (MPR), and the reduction of thermal conductivity in the functional region is attributed to phonon localization.

        Chemical Synthesis and Cloning of Panax ginseng Peptide Gene

        The sequence of ginseng peptide gene was designed and synthesized by the solid phase phosphoramidiate method. Synthetic segments were isolated, pllrified and joined to the plasmid pUC19. E.icherichiu coli JM101 cells were transformed with above hybrid plasmids. AmpiciIBin resistant transformants were screened and identified by in situ colony hybridization and Southern blot techniques. Finally the gene sequencing was done by the Sanger dideoxy method using primer extension.
