<P>We explored a very interesting gold nanoparticle system—pegylated gold in colloidal solution—and analyzed its uptake by mice colorectal adenocarcinoma CT26 tumor cells and the impact on the cell’s response to x-ray irradiation. We found that exposure to polyethylene glycol (PEG) modified (‘pegylated’) 4.7 ± 2.6 nm gold nanoparticles synthesized by a novel synchrotron-based method enhances the response of CT26 cells to x-ray irradiation. Transmission electron microscopy (TEM) and confocal microscopy revealed that substantial amounts of such nanoparticles are taken up and absorbed by the cells and this conclusion is supported by quantitative induced coupled plasma (ICP) results. Standard tests indicated that the internalized particles are highly biocompatible but strongly enhance the cell damage induced by x-ray irradiation. Synchrotron radiation Fourier transform infrared (SR-FTIR) spectromicroscopy analyzed the chemical aspects of this phenomenon: the appearance of C = O stretching bond spectral features could be used as a marker for cell damage and confirmed the enhancement of the radiation-induced toxicity for cells.</P>

Human pluripotent stem cells (hPSCs), including embryonic stem cells (ESCs) and induced PSCs (iPSCs), represent potentially unlimited cell sources for clinical applications. Previous studies have suggested that hPSCs may benefit from immune privilege and limited immunogenicity, as reflected by the reduced expression of major histocompatibility complex class-related molecules. Here we investigated the global immune-related gene expression profiles of human ESCs, hiPSCs and somatic cells and identified candidate immune-related genes that may alter their immunogenicity. The expression levels of global immune-related genes were determined by comparing undifferentiated and differentiated stem cells and three types of human somatic cells: dermal papilla cells, ovarian granulosa cells and foreskin fibroblast cells. We identified the differentially expressed genes CD24, GATA3, PROM1, THBS2, LY96, IFIT3, CXCR4, IL1R1, FGFR3, IDO1 and KDR, which overlapped with selected immune-related gene lists. In further analyses, mammalian target of rapamycin complex (mTOR) signaling was investigated in the differentiated stem cells following treatment with rapamycin and lentiviral transduction with specific short-hairpin RNAs. We found that the inhibition of mTOR signal pathways significantly downregulated the immunogenicity of differentiated stem cells. We also tested the immune responses induced in differentiated stem cells by mixed lymphocyte reactions. We found that CD24- and GATA3-deficient differentiated stem cells including neural lineage cells had limited abilities to activate human lymphocytes. By analyzing the transcriptome signature of immune-related genes, we observed a tendency of the hPSCs to differentiate toward an immune cell phenotype. Taken together, these data identify candidate immune-related genes that might constitute valuable targets for clinical applications.
Multiple dependent state repetitive sampling plans based on one-sided process capability indices
Yen, Ching-Ho,Chang, Chia-Hao,Aslam, Muhammad,Jun, Chi-Hyuck Informa UK (TaylorFrancis) 2018 Communications in Statistics Vol.47 No.6
<P>In this research, multiple dependent state and repetitive group sampling are used to design a variable sampling plan based on one-sided process capability indices, which consider the quality of the current lot as well as the quality of the preceding lots. The sample size and critical values of the proposed plan are determined by minimizing the average sample number while satisfying the producer's risk and consumer's risk at corresponding quality levels. In addition, comparisons are made with the existing sampling plans [Pearn and Wu (2006a), Yen etal. (2015)] in terms of average sample number and operating characteristic curve. Finally, an example is provided to illustrate the proposed plan.</P>

<P><B>Summary</B></P> <P>Pro-inflammatory cytokines produced in the tumor microenvironment lead to eradication of anti-tumor immunity and enhanced tumor cell survival. In the current study, we identified tumor necrosis factor alpha (TNF-α) as a major factor triggering cancer cell immunosuppression against T cell surveillance via stabilization of programmed cell death-ligand 1 (PD-L1). We demonstrated that COP9 signalosome 5 (CSN5), induced by NF-κB p65, is required for TNF-α-mediated PD-L1 stabilization in cancer cells. CSN5 inhibits the ubiquitination and degradation of PD-L1. Inhibition of CSN5 by curcumin diminished cancer cell PD-L1 expression and sensitized cancer cells to anti-CTLA4 therapy.</P> <P><B>Highlights</B></P> <P> <UL> <LI> TNF-α stabilizes cancer cell PD-L1 in response to chronic inflammation </LI> <LI> Activation of NF-κB by TNF-α induces CSN5 expression leading to PD-L1 stabilization </LI> <LI> CSN5 enzyme activity controls T cell suppression via PD-L1 deubiquitination </LI> <LI> Destabilization of PD-L1 by CSN5 inhibitor curcumin benefits anti-CTLA4 therapy </LI> </UL> </P> <P><B>Graphical Abstract</B></P> <P>[DISPLAY OMISSION]</P>

Fouling with protein mixtures in microfiltration
Ahn, Byunghun,Moon, Dong Ju,Ho, Chia Chi 한국공업화학회 2003 응용화학 Vol.7 No.2
Membrane fouling by protein mixtures during microfiltration has been investigated for binary protein mixtures, including BSA and lysozyme, BSA and casein, and BSA and ovalbumin. Filtration experiments were carried out using 0.2 ㎛ polycarbonate track-etched (PCTE) membrane in a stirred cell under constant transmembrane pressure (14 ㎪) to study the effect of solution pH and mixture composition on filtrate flux decline. Flux decline data were analyzed using a recently developed pore blockage-cake formation model. The model was in good agreement with the data. Protein mixtures may foul more slowly or faster than either of the pure proteins. The rate of flux decline for BSA-lysozyme mixtures is much greater than either of pure proteins. The flux increases as the percentage of lysozyme in the mixture increases. However, the initial flux of BSA-casein mixtures was observed to decrease with increasing casein concentration partially due to the rapid casein adsorption on the membrane surface. It can be concluded that casein plays an important role in the flux decline of protein mixtures. And, the flux decline of the BSA-ovalbumin mixtures increases as BSA increases.
Echocardiographic parameters and indices in 23 healthy Maltese dogs
Chih-Hung Tsai,Chao-Chun Huang,Chia Chi Ho,Marta Claretti 대한수의학회 2021 Journal of Veterinary Science Vol.22 No.5
Background: Echocardiography is a primary tool used by veterinarians to evaluate heart diseases. In recent years, various studies have targeted standard echocardiographic values for different breeds. Reference data are currently lacking in Maltese dogs and it is important to fill this gap as this breed is predisposed to myxomatous mitral valve disease, which is a volume overload disease. Objectives: To establish the normal echocardiographic parameters for Maltese dogs. Methods: In total, 23 healthy Maltese dogs were involved in this study. Blood pressure measurements, thoracic radiography, and complete transthoracic echocardiography were performed. The effects of body weight, age and sex were evaluated, and the correlations between weight and linear and volumetric dimensions were calculated by regression analysis. Results: The mean vertebral heart size was 9.1 ± 0.4. Aside from the ejection fraction, fractional shortening, and the left atrial to aorta root ratio, all the other echocardiographic parameters were significantly correlated with weight. Conclusion: This study describes normal echocardiographic parameters that may be useful in the echocardiographic evaluation of Maltese dogs.
An Internet Based Embedded Network Monitoring System for Renewable Energy Systems
Po-Yen Chen,Se-Kang Ho,Wei-Jen Lee,Chia-Chi Chu,Ching-Tsa Pan 전력전자학회 2007 ICPE(ISPE)논문집 Vol.- No.-
In this paper, an internet based embedded network monitoring system is proposed for renewable energy systems. By using a low cost network communication module (RCM 3700) as a web server, one can achieve better network security, lower power consumption, compact size, and easier to use as compared with a PC based one. Also, Java language is chosen for designing a dynamic webpage to graphically display various real time waveforms of the controlled system for multi-user at the same time. As an illustrative example, a small scale wind power generation system equipped with an EZDSP 2812 controller is adopted for demonstration. In addition, an FPGA EC10 is implemented as a bidirectional communication interface for coordinating the asynchronous data transmission modes. Experimental results from the constructed prototype verify that the proposed monitoring system can indeed achieve the desired function.
