YAP/TAZ-Mediated Upregulation of GAB2 Leads to Increased Sensitivity to Growth Factor–Induced Activation of the PI3K Pathway

        Wang, Chao,Gu, Chao,Jeong, Kang Jin,Zhang, Dong,Guo, Wei,Lu, Yiling,Ju, Zhenlin,Panupinthu, Nattapon,Yang, Ji Yeon,Gagea, Mihai (Mike),Ng, Patrick Kwok Shing,Zhang, Fan,Mills, Gordon B. American Association for Cancer Research 2017 Cancer Research Vol.77 No.7

<P>Interactions between HIPPO, YAP/TAZ, and the PI3K/AKT pathway may be therapeutically targetable, providing new approaches to treating endometrial cancers and other cancers where the HIPPO pathway is a core oncogenic driver.</P><P>The transcription regulators YAP and TAZ function as effectors of the HIPPO signaling cascade, critical for organismal development, cell growth, and cellular reprogramming, and YAP/TAZ is commonly misregulated in human cancers. The precise mechanism by which aberrant YAP/TAZ promotes tumor growth remains unclear. The HIPPO tumor suppressor pathway phosphorylates YAP and TAZ, resulting in cytosolic sequestration with subsequent degradation. Here, we report that the PI3K/AKT pathway, which is critically involved in the pathophysiology of endometrial cancer, interacts with the HIPPO pathway at multiple levels. Strikingly, coordinate knockdown of YAP and TAZ, mimicking activation of the HIPPO pathway, markedly decreased both constitutive and growth factor–induced PI3K pathway activation by decreasing levels of the GAB2 linker molecule in endometrial cancer lines. Furthermore, targeting YAP/TAZ decreased endometrial cancer tumor growth <I>in vivo</I>. In addition, YAP and TAZ total and phosphoprotein levels correlated with clinical characteristics and outcomes in endometrial cancer. Thus, YAP and TAZ, which are inhibited by the HIPPO tumor suppressor pathway, modify PI3K/AKT pathway signaling in endometrial cancer. The cross-talk between these key pathways identifies potential new biomarkers and therapeutic targets in endometrial cancer. <I>Cancer Res; 77(7); 1637–48. ©2017 AACR</I>.</P>

Effect of BiFeO3 addition on Bi2O3–ZnO–Nb2O5 based ceramics

        Xiaolian Chao,Zupei Yang,Chao Kang,Rui Gu 한국물리학회 2010 Current Applied Physics Vol.10 No.1

In this paper, low temperature sintering of the Bi2(Zn1/3Nb2/3)2O7 (b-BZN) dielectric ceramics was studied with the use of BiFeO3 as a sintering aid. The effects of BiFeO3 contents and the sintering temperature on the phase structure, density and dielectric properties were investigated. The results showed that the sintering temperature could be decreased and the dielectric properties could be retained by the addition of BiFeO3. The structure of BiFeO3 doped b-BZN was still the monoclinic pyrochlore phase. The sintering temperature of BiFeO3 doped b-BZN ceramics was reduced from 1000 ℃ to 920 ℃. In the case of 0.15wt.% BiFeO3 addition, the b-BZN ceramics sintered at 920 ℃ exhibited good dielectric properties, which were listed as follows: εr = 79 and tan δ = 0.00086 at a frequency of 1 MHz. The obtained properties make this composition to be a good candidate for the LTCC application.

Degradation of sulphapyridine by Fe-Mn binary oxide-mediated radical reactions

        Kang Wu,Zhongliang Zhang,Fei Hua,Zhou Ye,Chao Li,Youzhi Yao 한국화학공학회 2022 Korean Journal of Chemical Engineering Vol.39 No.10

Sewage containing antibiotics is harmful to the water environment. Here, a newly prepared iron manganese binary oxide (FMBO) activated persulfate (PDS) has excellent performance for the degradation of sulfapyridine (SPY) in an aqueous environment. The effects of initial pH, SPY, PDS concentration and FMBO dosage on the degradation ability of SPY were investigated. When the pH of FMBO/PDS was 4.0 and the initial concentration of PDS was 2.0mmol/L, the degradation ratio of SPY could reach 94.4%. In the system of removing organic pollutants by transition metal activated PDS, SO4 • and •OH radicals are generally generated, and •OH radical plays a major role. Through the identification of SPY intermediate products by LC/MS, the degradation mechanism was explored. The degradation pathway showed that the strong oxidation of FMBO and the activity of PDS promoted the production of hydroxyl radical and contributed to the degradation and transformation of antibiotics. This study shows that FMBO/PDS has high degradation efficiency in the degradation of SPY.

SUBMILLIMETER OBSERVATIONS OF DENSE CLUMPS IN THE INFRARED DARK CLOUD G049.40-00.01

        Kang, Miju,Choi, Minho,Bieging, John H.,Rho, Jeonghee,Lee, Jeong-Eun,Tsai, Chao-Wei IOP Publishing 2011 The Astrophysical journal Vol.743 No.2

<P>We obtained 350 and 850 mu m continuum maps of the infrared dark cloud G049.40-00.01. Twenty-one dense clumps were identified within G049.40-00.01 based on the 350 mu m continuum map with an angular resolution of about 9 ''.6. We present submillimeter continuum maps and report physical properties of the clumps. The masses of clumps range from 50 to 600 M-circle dot. About 70% of the clumps are associated with bright 24 mu m emission sources, and they may contain protostars. The two most massive clumps show extended, enhanced 4.5 mu m emission indicating vigorous star-forming activity. The clump-size-mass distribution suggests that many of them are forming high-mass stars. G049.40-00.01 contains numerous objects in various evolutionary stages of star formation, from pre-protostellar clumps to H II regions.</P>

      • Expression Profile Analysis of Zinc Transporters (ZIP4, ZIP9, ZIP11, ZnT9) in Gliomas and their Correlation with IDH1 Mutation Status

        Kang, Xing,Chen, Rong,Zhang, Jie,Li, Gang,Dai, Peng-Gao,Chen, Chao,Wang, Hui-Juan Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.8

Background: Zinc transporters have been considered as essential regulators in many cancers; however, their mechanisms remain unknown, especially in gliomas. Isocitrate dehydrogenase 1(IDH1) mutation is crucial to glioma. This study aimed to investigate whether zinc transporters are correlated with glioma grade and IDH1 mutation status. Materials and Methods: IDH1 mutation status and mRNA expression of four zinc transporters (ZIP4, ZIP9, ZIP11, and ZnT9) were determined by subjecting a panel of 74 glioma tissue samples to quantitative real-time PCR and pyrosequencing. The correlations between the expression levels of these zinc transporter genes and the grade of glioma, as well as IDH1 mutation status, were investigated. Results: Among the four zinc transporter genes, high ZIP4 expression and low ZIP11 expression were significantly associated with higher grade (grades III and IV) tumors compared with lower grade (grades I and II) counterparts (p<0.0001). However, only ZIP11 exhibited weak correlation with IDH1 mutation status (p=0.045). Samples with mutations in IDH1 displayed higher ZIP11 expression than those without IDH1 mutations. Conclusions: This finding indicated that zinc transporters may interact with IDH1 mutation by direct modulation or action in some shared pathways or genes to promote the development of glioma. Zinc transporters may play an important role in glioma. ZIP4 and ZIP11 are promising molecular diagnostic markers and novel therapeutic targets. Nevertheless, the detailed biological function of zinc transporters and the mechanism of the potential interaction between ZIP11 and IDH1 mutation in gliomagenesis should be further investigated.

      • A Simple Reversible Watermarking Scheme Based on Difference Expansion

        Chao-Ming Wu,Cheng-Kang Wu,Houshou Chen 대한전자공학회 2009 ITC-CSCC :International Technical Conference on Ci Vol.2009 No.7

A simple reversible watermarking scheme based on block-wise difference expansion is proposed. Pixels in a block are expanded with respect to a common reference point. The maximum capacity will exceed 0.5 bpp if the block size is larger than 2. According to local variances of the reference points, blocks introducing small distortion by difference expanding are selected firstly to embed watermark. In Tian’s difference expansion algorithm, the embedding of the location map causes large overhead cost, apparently in low payload region. A good lossless compression algorithm is required to reduce this cost but the complexity is increased. In the proposed algorithm, a simple all 0 pattern is used to distinguish between non-expandable and expandable blocks. In this way, the location map is no longer necessary and the complexity of the scheme will be reduced.

Tip fabrication methods of hollow metal microneedles

        Chao-Wei Dong,Jong-Yeong Jeon,Hye-Min Kang,박우태 대한기계학회 2023 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.37 No.1

Hollow microneedles can penetrate the corneum of the skin and various biological barriers. Drugs can enter through a minimally invasive path or bodily fluid can be collected from the epidermis or dermis. This makes hollow microneedles (MN) a promising tool for drug delivery and body fluid sensing in a painless manner. Here, we propose a nickel microneedle with a length of 720 μm, an inner diameter of 27 μm, and an outer diameter of 87 μm. To construct the metal hollow microneedle model, we introduce three different manufacturing methods. The three methods are laser cutting, re-dissolution by SU-8 coating, and ultrasonic blade cutting. To verify the penetration capability, the microneedle array was tested to penetrate of the rabbit skin without rupture. The microneedle array was also repeatedly used in a film puncture experiment to verify the reliability of the structure. These microneedles can be combined with syringes and other drug delivery devices to achieve accurate quantitative and controllable flow rates of drug delivery. Through mechanical test and solution delivery test, we demonstrated that the microneedle structure has strong mechanical strength and a controllable rate of drug delivery, which is feasible as a drug delivery device.
