Objective: Tibetan pigs, an excellent species unique to China, face serious threats, which in turn affects the development and utilization of the outstanding advantages of plateau hypoxia adaptability and reduces their genetic diversity. Therefore, a discussion of measures to conserve this genetic resource is necessary. The method, based on genetic diversity, genetic divergence and total genetic contribution rate of population, reflects the priority conservation order and varies depending on the three different purposes of conservation. Methods: We analyzed mitochondrial DNA control region (D-loop) variation in 1,201 individuals from nine Tibetan pig populations across five provinces and downloaded 564 mtDNA D-loop sequences from three indigenous pig breeds in Qinghai, Sichuan, and Yunnan Provinces distributed near the Tibetan pigs. Results: We analyzed three different aspects: Changdu Tibetan pigs have the highest genetic diversity, and from the perspective of genetic diversity, the priority conservation is Changdu Tibetan pigs. Hezuo Tibetan pigs have the highest genetic contribution, so the priority conservation is Hezuo Tibetan pigs in the genetic contribution aspect. Rkaze Tibetan pigs were severely affected by indigenous pig breeds, so if considering from the perspective of introgression, the priority conservation is Rkaze Tibetan pigs. Conclusion: This study evaluated genetic diversity and comprehensively assessed conservation priority from three different aspects in nine Tibetan pig populations.

Objective: Evidence from previous reports indicates that pig domestication in East Asia mainly occurred in the Mekong region and the middle and downstream regions of the Yangtze River. Further research identified two new origin centers for domestic pigs in the Tibetan Plateau and the islands of Southeast Asia. However, due to the small sample size of Tibetan pigs, details of the origin and spread of Tibetan pigs has not yet been established. Methods: We analyzed mitochondrial DNA control region (D-loop) variation in 1,201 individuals from nine Tibetan pig populations across five provinces. Comprehensive Tibetan pig samples were taken to perform the most detailed analysis of Tibetan pigs to date. Results: The result indicate that Rkaze pigs had the lowest level of diversity, while Changdu pigs had the highest diversity. Interestingly, these two populations were both in the Tibetan Plateau area. If we calculate diversity in terms of each province, the Tibetan Plateau area had the lowest diversity, while the Chinese province of Gansu had the highest diversity. Diversity gradient analysis of major haplotypes suggested three domestication centers of Tibetan pigs in the Tibetan Plateau and the Chinese provinces of Gansu and Yunnan. Conclusion: We found two new domestication centers for Tibetan pigs. One is in the Chinese province of Gansu, which lies in the upstream region of the Yellow River, and the other is in the Chinese province of Yunnan.

Backgrounds: Previous studies were mainly focused on the effect of catechins and caffeine, the main pharmacological ingredients of green tea. However, the ordinary way of taking green tea is by drinking the tea water. Hence the benefits of water extractives of green tea were investigated in the current study. Methods: H2O2 was applied to induce the oxidative stress on PC12 cells. The neuroprotective effects of green tea against oxidative stress were determined by observing the biological behaviors of PC12 cells. Results: With the pre-treatment of light green tea, the survival and proliferation of oxidative stressed PC12 cells were significantly enhanced. H2O2 induced PC12 cell apoptosis was completely reversed. Consistently, the expression of JNK was up-regulated while the caspase-3 was down-regulated. Conclusion: These results suggested that drinking green tea, particularly the light green tea, performed neuroprotective effects against oxidative stress.

A novel reactive fluorescent dye [2-morpholine-4-(6-chlorine-1,3,5-s-triazine)-amino]fluorescein based on 5-aminofluorescein was synthesized by electrophilic substitution. The photophysical properties, solvent effect, pH value sensitivity, and metal ions responsibility of this new fluorophore were investigated. Compared with 5-aminofluorescein, the novel fluorophore exhibited stronger fluorescence and longer lifetime. The fluorescence property of the new dye was obviously affected by different solvents and pH values, and it showed stronger fluorescence in the protic solvents or an alkaline environment. Moreover, the fluorescent intensity could be enhanced by the formation of complex with metal ions especially with Mg2+. The results show that the fluorescent dye is a promising efficient sensor for solvents, protons, and metal ions.
Dynamic analysis for gene expression profiles of endothelial colony forming cells under hypoxia

Previous studies have shown that endothelial colony forming cells (ECFCs) play an important role in the neovascularization of tumors. Hypoxia is emphasized as an important promoter of angiogenesis. However, little is known about genome-wide transcriptional regulation of ECFCs under hypoxic conditions. In this study, gene expression profiles in ECFCswere evaluated under hypoxic conditions for 3, 6, 12, 24,and 48 h, using Affymetrix U133 plus 2.0 chip microarray. 1,103 hypoxia-regulated genes were filtered, with 379(0.693 %) genes up-regulated and 724 (1.32 %) genes downregulated. Most of the up-regulated genes were involved in apoptosis, cell proliferation, or metabolic processes, while most of the down-regulated genes were involved in cell adherence,cell cycle,DNAandmRNAmetabolic processes,multi-cellular organism development, protein metabolic processes, response to stress, signal transduction, or transport. This expression profile is ECFC-specific, because it is significantly different from those of endothelial cells and smooth muscle cells under hypoxic conditions. Moreover, hypoxia-regulated apoptosis in ECFCs is mainly related with the mitochondrial pathway (p53-BAX-Caspase-9) and the death receptor pathway (FASCaspase-8-Caspase-3). MAPK pathway is activated in ECFCs under hypoxic conditions. The differentially expressed genes of ECFCs were identified under hypoxic conditions, and related with cell apoptosis, cell cycle and MAPK pathways, shedding light on the mechanism of angiogenesis.
Study on Effect Mechanism of Flow Characteristics Deviation on Low Frequency Oscillations

This paper focus on low frequency oscillations induced by the flow characteristics deviation of steam turbine. The causes and features of flow characteristics deviation are analyzed and described first. The coupling model between thermodynamic system and power system was established for time-domain simulation analysis, in an attempt to find out the effect mechanism of flow characteristics deviation on low frequency oscillations. The effect of flow characteristics derivation on the responses characteristics of turbo-generator is analyzed through simulation. Results indicate that the flow characteristics deviation is apt to cause low frequency oscillations. Both the amplitude and the speed of oscillations are related to the extent of flow characteristics deviation.

Radiation therapy is an important treatment for head and neck squamous cell carcinoma (HNSCC). However, how to promote radiation sensitivity in HNSCC remains a challenge. This study aimed to investigate the radiosensitizing effects of fenofibrate on HNSCC and explore the underlying mechanisms. HNSCC cell lines CNE-2 and KB were subjected to ionizing radiation (IR), in the presence or absence of fenofibrate treatment. Cell growth and survival, apoptosis and cell cycle were evaluated. In addition, CNE-2 cells were xenografted into nude mice and subjected to IR and/or fenofibrate treatment. The expression of cyclinB and CDK1 was detected by Western blotting. Our results showed that fenofibrate efficiently radiosensitized HNSCC cells and xenografts in mice, and induced apoptosis and G2/M arrest via reducing the activity of the CDK1/cyclinB1 kinase complex. These data suggest that fenofibrate could be a promising radiosensitizer for HNSCC radiotherapy.
Motion planning of a steam generator mobile tube-inspection robot

Under the influence of nuclear radiation, the reliability of steam generators (SGs) is an important factorin the efficiency and safety of nuclear power plant (NPP) reactors. Motion planning that remotely manipulates an SG mobile tube-inspection robot to inspect SG heat transfer tubes is the mainstream trendof NPP robot development. To achieve motion planning, conditional traversal is usually used for baseposition optimization, and then the A* algorithm is used for path planning. However, the proposedapproach requires considerable processing time and has a single expansion during path planning andplan paths with many turns, which decreases the working speed of the robot. Therefore, to reduce thecalculation time and improve the efficiency of motion planning, modifications such as the matrixmethod, improved parent node, turning cost, and improved expanded node were proposed in this study. We also present a comprehensive evaluation index to evaluate the performance of the improved algorithm. We validated the efficiency of the proposed method by planning on a tube sheet with square-typetube arrays and experimenting with Model SG.

Objective: In order to improve fermentation quality of natural grasses, their silage preparation and fermentation quality in meadow steppe (MS) and typical steppe (TS) were studied. Methods: The small-scale silages and round bale silages of mixed natural grasses in both steppes were prepared using the commercial lactic acid bacteria (LAB) inoculants Chikuso-1 (CH, Lactobacillus plantarum) and cellulase enzyme (AC, Acremonium cellulase) as additives. Results: MS and TS contained 33 and 9 species of natural grasses, respectively. Stipa baicalensis in MS and Stipa grandi in TS were the dominant grasses with the highest dry matter (DM) yield. The crude protein (CP), neutral detergent fiber and water-soluble carbohydrate of the mixed natural grasses in both steppes were 8.02% to 9.03%, 66.75% to 69.47%, and 2.02% to 2.20% on a DM basis, respectively. All silages treated with LAB and cellulase were well preserved with lower pH, butyric acid and ammonia-N content, and higher lactic acid and CP content than those of control in four kinds of silages. Compared with CH- or AC-treated silages, the CH+ AC-treated silages had higher lactic acid content. Conclusion: The results confirmed that combination with LAB and cellulase may result in beneficial effects by improving the natural grass silage fermentation in both grasslands.
