      • The MTHFR C677T Polymorphism and Prostate Cancer Risk: New Findings from a Meta-analysis of 7306 Cases and 8062 Controls

        Methylenetetrahydrofolate reductase (MTHFR) is an essential enzyme involved in folate metabolism; a single nucleotide polymorphism (SNP) C677T has been reported to be linked with altered incidences of several diseases. We here conducted a meta-analysis of 15 published epidemiological studies with a total of 7306 cases and 8062 controls to evaluate its association with prostate cancer risk with overall and subgroup analyses. No statistical relationship was found overall with any genetic model (TT vs. CC: OR = 0.80, 95%CI = [0.62, 1.04], P = 0.094; CT vs. CC: OR = 0.97, 95%CI = [0.84; 1.12], P = 0.667; Dominant: OR = 0.94, 95%CI = [0.82; 1.07], P = 0.343; Recessive: OR = 0.81, 95%CI = [0.64; 1.04], P = 0.104), but after the exclusion of several studies, we could observe the homozygote TT to confer less susceptibility to prostate cancer in carriers; moreover, different effects of the polymorphism on prostate cancer risk was detected from subgroup analysis stratified by participants' residential region: significant reduced prostate cancer risk was found to be associated with the polymorphism from Asian studies (TT vs. CC: OR = 0.47, 95%CI = [0.33; 0.67], P < 0.001; CT vs. CC: OR = 0.73, 95%CI = [0.60; 0.90], P = 0.002; Dominant: OR = 0.67, 95%CI = [0.56; 0.82], P < 0.001; Recessive: OR = 0.55, 95%CI = [0.40; 0.76], P < 0.001) while studies from Europe indicated a slight increased risk under dominant model with marginal significance (OR = 1.14, 95%CI = [0.99; 1.30], P = 0.064). Moreover, the protective effect of the polymorphism against prostate cancer was also shown by studies performed in yellow Asians (TT vs. CC: OR = 0.48, 95%CI = [0.31; 0.75], P = 0.001; CT vs. CC: OR = 0.68, 95%CI = [0.51; 0.90], P = 0.006; Dominant: OR = 0.63, 95%CI = [0.48; 0.82], P < 0.001; Recessive: OR = 0.57, 95%CI = [0.39; 0.84], P = 0.004). We propose that these phenomena should be viewed with the consideration of folate metabolism profile and different gene background as well as living habits of different populations, and more relevant studies should be conducted to confirm our hypothesis and provide a comprehensive and clear picture concerning this topic.

        Berberine Inhibits HEp-2 Cell Invasion Induced by Chlamydophila pneumoniae Infection

        This study investigated the inhibitory effects of berberine on Chlamydophila (Chlamydia) pneumoniae infection-induced HEp-2 cell invasion and explored the possible mechanisms involved in this process. C. pneumoniae infection resulted in a significant increase in HEp-2 cell invasion when compared with the control cells (P<0.01) in a Matrigel invasion assay. This enhanced cell invasion was strongly suppressed by berberine (50 μM) (P<0.01). In a cell adhesion assay, the infection-induced HEp-2 cell adhesion to Matrigel was also significantly inhibited by berberine (P<0.01). C. pneumoniae infection was found to promote HEp-2 cell migration remarkably (P<0.01), which was markedly suppressed by berberine (P<0.01) in the cell migration assays. There were no statistically significant differences in the expression of matrix metalloproteinase-1 (MMP-1) and MMP-9 in the infected cells and berberine did not change the expression of MMP-1 and MMP-9. These data suggest that berberine inhibits C. pneumoniae infection-induced HEp-2 cell invasion through suppressing HEp-2 cell adhesion and migration, but not through changing the expression of MMP-1 and MMP-9.

        Safe Range of Retaining Pressure for Three-dimensional Face of Pressurized Tunnels based on Limit Analysis and Reliability Method

        Based on the existing research, the 3D active and passive failure mechanisms of a pressurized tunnel face are constructed. The nonlinear failure criterion is introduced into the limit analysis by virtue of the tangent technique. By neglecting the randomness of soil parameters, the collapse pressure of active failure and the blowout pressure of passive failure, namely the lower and upper limit of retaining pressure, are obtained along with the failure ranges. On account of the randomness of soil properties, a three-dimensional stochastic model of a pressurized tunnel face is established, and the optimal range of retaining pressure of the pressurized tunnel face is presented on the basis of the multiple correlated failure modes. Taking Changsha Metro Line 2 as an example, the good agreement of the measured data in situ with the reasonable range of retaining pressure verifies the presented method. The combination of limit analysis and reliability method can provide more scientific and reasonable support parameters for the construction of pressurized tunnels in the future.

        Critical effects of long non-coding RNA on fibrosis diseases

        The expression or dysfunction of long non-coding RNAs (lncRNAs) is closely related to various hereditary diseases, autoimmune diseases, metabolic diseases and tumors. LncRNAs were also recently recognized as functional regulators of fibrosis, which is a secondary process in many of these diseases and a primary pathology in fibrosis diseases. We review the latest findings on lncRNAs in fibrosis diseases of the liver, myocardium, kidney, lung and peritoneum. We also discuss the potential of diseaserelated lncRNAs as therapeutic targets for the clinical treatment of human fibrosis diseases.

        The production technology and properties of an aluminum alloy processed by equal channel angular pressing

        Distribution of equivalent stress and equivalent strain was analyzed in the ECAP process based on material flow laws. Simulation results showed that severe plastic deformation occurred at the corner of the cavity, indicating a stress concentration at the equal channel angle. Distribution of stress and strain at the same section was not uniform. Equivalent stresses on the surface at the inside corner of the part were larger. Experimental results showed that forming speed and lubrication conditions had a great influence on quality of parts. The parts were in good condition with no wrinkle and rupture after ten ECAP passes. EBSD analyses showed that the original coarse grains were refined and uniformly distributed, and the average grain size of the cross section was about 620 nm after ten ECAP passes. With increasing the number of ECAP passes, the proportion of small angle grain boundaries decreased gradually and the ratio of large angle grain boundaries increased. Tensile strength and hardness increased sharply, while the elongation decreased sharply after the first ECAP pass. With succeeding the ECAP passes, the tensile strength, hardness and the elongation all increased slowly.

        Diversity and composition of microbiota during fermentation of traditional Nuodeng ham

        The microbial community is one of the most important factors in shaping the characteristics of fermented food. Nuodeng ham, traditionally produced and subjected to 1–4 years of fermentation, is a dry fermented food product with cultural and economic significance to locals in southwestern China. In this study, we aimed to characterize the microbiota and physicochemical profiles of Nuodeng ham across different stages of fermentation. Ham samples from each of the four years were analyzed by sequencing bacterial 16S rRNA gene and fungal internal transcribed spacer sequence, in order to characterize the diversity and composition of their microflora. A total of 2,679,483 bacterial and 2,983,234 fungal sequences of high quality were obtained and assigned to 514 and 57 genera, respectively. Among these microbes, Staphylococcus and Candida were the most abundant genera observed in the ham samples, though samples from different years showed differences in their microbial abundance. Results of physicochemical properties (pH, water, amino acid, NaCl, nitrate and nitrite contents, and the composition of volatile compounds) revealed differences among the ham samples in the composition of volatile compounds, especially in the third year samples, in which no nitrite was detected. These results suggest that the structure and diversity of microbial communities significantly differed across different stages of fermentation. Moreover, the third year hams exhibits a unique and balanced microbial community, which might contribute to the special flavor in the green and safe food products. Thus, our study lends insights into the production of high quality Nuodeng ham.

        This paper describes a case study of applying 3D human simulation to the design of a u-assembly machine which is a micro semi-automatic machine. Two types of assembly machine are tested and compared. The purpose of our research is to find out a better design considering ergonomics through the implementation of 3D human simulation and RULA analysis. The worker's working postures are analyzed during the simulation of assembly process.

      • Predictive Potential of Glutathione S-Transferase Polymorphisms for Prognosis of Osteosarcoma Patients on Chemotherapy

        Objective: To evaluate the predictive value of glutathione S-transferase (GST) gene polymorphisms for the prognosis of osteosarcoma patients receiving chemotherapy. Methods: A total of 159 patients were included in our study between January 2005 and December 2007., with follow-up until January 2012. Genotyping was based upon the duplex polymerase-chain-reaction with the PCR-CTPP method. Results: At the time of diagnosis, 15.4% of the patients presented with metastasis, while 22.3% developed metastasis during follow-up. At the time of final analysis on January 2012, the median follow-up was 45.5 months. Patients with null GSTM1 and GSTT1 had a higher event free survival rate than non-null genotype, but no significant association was found between the two genotypes and prognosis of osteosarcoma. Individuals with GSTP1 Val/Val genotype tended to live shorter than with the IIe/IIe genotype, and we found a significantly higher risk of death from osteosarcoma (adjusted HR=2.35, 95% CI=1.13-4.85). Conclusion: The GSTP1 gene polymorphism may have an important role in the prognosis of osteosarcoma patients with chemotherapy. Further analyses with larger samples and more genes encoding metabolizing and DNA repair enzymes are warranted.

      • Mathematical Simulation of Spray Forming Process

        In this paper, an integrated approach for modeling the entire spray forming process is presented and the program for the simulation is written by hand. The whole spray forming process can be divided into four calculate process and the basis for the analysis is classical k-ε turbulence model which used to simulate the flow field of gas formed in the chamber. The other three mathematical models are atomization model,deposition model and solidification model. In the atomization model coupled the flow field of gas and formation、velocity and location of droplet. By means of integrate above mathematical model the process of spraying forming was simulated in detail.

      • KAI1/CD82 and MRP1/CD9 Serve as Markers of Infiltration, Metastasis, and Prognosis in Laryngeal Squamous Cell Carcinomas

        Objective: The current study explored the expression of KAI1/CD82 and MRP1/CD9 and its significance in laryngeal squamous cell carcinoma (LSCC). Methods: The expression levels of KAI1/CD82 and MRP1/CD9 in 100 LSCC tissue specimens, as well as in 30 para-LSCC non-carcinomatous tissue specimens randomly taken from the patients, were assessed using the quantitative polymerase chain reaction (Q-PCR) and immunohistochemistry and correlations with pathological parameters of LSCC and their influence on survival function were analyzed. Results: KAI1/CD82 and MRP1/CD9 showed basically consistent changes in both mRNA and protein expression. Their expression in the 30 LSCC specimens was significantly lower compared with that in the corresponding non-carcinous tissues (P < 0.01 or 0.05), notably correlating with TNM stage, differentiation degree, clinical stage, and lymphatic metastasis (P < 0.01 or 0.05), but not gender, age, and LSCC growth sites (P > 0.05). The median survival of patients with positive KAI1/CD82 and MRP1/CD9 protein expression was longer than that of patients with negative protein expression (P < 0.01 or 0.05). KAI1/CD82 protein expression negatively correlated with MRP1/CD9 protein expression in LSCC (${\chi}^2$= 31.25, P < 0.01). Conclusion: KAI1/CD82 and MRP1/CD9 may jointly participate in the development of LSCC. They may serve as the markers for judging the infiltration, metastasis, and prognosis of LSCC.
