        Vitamin E가 철분 투여로 인한 rat의 지질 과산화에 미치는 영향

        This study was designed to investigate the effect of dietary vitamin E on the lipid peroxidation by dietary iron-injected to male rats. Sprague-Dawely strain male rats were divided into three experimental groups, namely control, iron injected and iron-vitamin E injected groups. The control group was fed with normal diet; the iron injected group was given normal diet and while injected intraperitoneally 30mg of ferric hydroxide/100g of body weight 20 times every 3 days. The iron-vitamin E injected group was intraperitoneally administered 30mg of ferric hydroxide/100g of body weight 20 times every 3 days and vitamin E every day with the dose of 5IU(5mg)/100g body weight. All experimental groups were maintained for 60 days with feeding on the respective ratio. The results obtained from this experiment were summarized as following: 1. The net weight gain was significantly decreased by the iron injection, but much increased by the vitamin E injection. 2. The contents of unsaturated fatty acid in phospholipid in liver, kidney, muscle and serum were decreased by the iron injection, but increased by the vitamin E injection. 3. The increment of malondialdehyde contents was induced by the iron overloading, but significantly decreased by the vitamin E injection. Therefore, it is suggested that dietary iron administration to male rats facilitates the lipid peroxidation in vivo and vitamin E has the inhibiting effect on lipip peroxidation process by iron.

        This study was designed to elucidate the effects of iron, a well known catalyst of lipid peroxidation, on the contents of phospholipids, unsaturated fatty acids composed in phospholipid molecules and their derivatives, prostaglandins, and the composition changes of fatty acids contained in phospholipids. Iron decreased the contents of phospholipids and its components of unsaturated fatty acids. Catalytic action of iron decreased the composition rates of linoleate and linolenate composed in phospholipid molecules, while that of arachidonate was inclined to increase. The content of arachidonate was increased and that of prostaglandins was decreased without regard to increase the precursor of prostaglandins. It may be concluded that the decreases of prostaglandins and the increase of arachidonate are due to inhibition of the activities of enzyme systems responsible for prostaglandin synthesis by lipid peroxides produced by the catalyst of iron.

        In the former method for determining plasma free unsaturated fatty acids developed by Huh and Kim, it always showed relatively high values. The cause of high values was known to be a contamination of phospholipids in the extract of fatty acids. Some modifications were given. Plasma filtrate extracted with ordinary liped solvent was treated by chilled cetone to eliminate the interfering materials, phospholipids, followed by extraction of fatty acids with the solvent mixture. The defect of the former method could be completed by this modification giving sufficient results.

        집쥐와 닭 뇌의 γ-Aminobutyric acid 함량

        Current interest in ${\gamma}$-aminobutyric acid (GABA) has arisen from the convergence of several independent line of investigation leading to the demonstration that this and related substances are normal products of brain metabolism and that GABA has an important physiological action upon brain function as well as upon certain peripheral nervous structures. The interest for neurophysiologists has been enhanced by the importance of the discovery for the role of humoral mediator of synaptic transmission or regulator of neuronal activity in the central nervous system, particularly if it may shed some elight upon the nature of central inhibitory processes. In accordance with such an interest and importance, this work was performed in order to standardize the normal content as a preliminary investigation of so-called night active and daytime active animals GABA content in their brains when they are exposed to light and darkness. The method, through which the estimation has made in this work, was paper chromatographic method developed by Maynert and Klingman for the estimation of GABA content in animal tissues. The results obtained are summerized as follows: 1) GABA content in the cerebral cortex of house rat ranged from 90 to $310{\mu}g/gm$ of wet weight. 2) The content of GAGA ranging from 130 to $510{\mu}g/gm$ of wet weight was occurred from midbrain of the rat. 3) GABA content was ranged from 30 to $150 {\mu}g/gm$ of wet weight of the rat cerebellum. 4) The contents of fowl cerebral cortex, midbrain, and cerebellum are estimated as ranging 230-590, 250-620, $50-280{\mu}g/gm$ of wet weight, respectively. As a result, it may be concluded that among three brain tissues of both animals the midbrain is the highest region in GABA content. Fowl brain, on the other side, contains more higher GABA content than the house rat brain does.

        Rat에 있어서 탈지사료 및 철분투여가 다불포화 지방산, vitamin E 및 그 유도체에 미치는 영향

        This study was designed to elucidate the effects of administration with dietary iron and fat-free diet on the contents of unsaturated fatty acid in phospholipid molecules, vitamin E contents and malondialdehyde contents in liver, kidney, muscle and testis of the male rats. The rats were divided into 3 experimental groups, namely, control, iron injection and fat-free diet administration groups. The control group was fed with normal diet, iron injection group injected intraperitoneally 20mg of ferric hydroxide/100g of body weight 20 times every 3 days and fat-free diet group administered lipid extraction diet with hexane in normal diet. All experimental groups were maintained for 60 days with feeding on the respective ration. The results obtained were summarized as follows: 1. In the mean contents of unsaturated fatty acid in phospholipid of liver, kidney, muscle and testis among groups, control group was 21.31mg/g, 19.38mg/g, 1.67mg/g, 13.68mg/g, iron injection group was 13.83mg/g, 16.53mg/g, 0.71mg/g, 10.11mg/g and fat-free diet group was 21.07mg/g, 19.38mg/g, 1.49mg/g and 13.40mg/g, respectively. 2. In the mean contents of vitamin E in liver, kidney, muscle and testis among groups, control group was 6.77mg/g, 1.93mg/g, 0.12mg/g, 0.17mg/g, iron injection group was 3.16mg/g, 0.86mg/g, 0.07mg/g, 0.09mg/g and fat-free diet group was 7.41mg/g, 1.50mg/g, 0.11mg/g and 0.16mg/g, respectively. 3. In the mean contents of malondialdehyde in liver, kidney, muscle, testis and serum among groups, control group was 11.29nM/0.1g, 23.25nM/0.1g, 42.47nM/0.1g, 7.01nM/0.1g, 4.33nM/ml, iron injection group was 34.98nM/0.1g, 40.55nM/0.1g, 72.21nM/0.1g, 12.26nM/0.1g, 11.27nM/ml and fat-free diet group was 8.07nM/0.1g, 20.63nM/0.1g, 39.92nM/0.1g, 6.95nM/0.1g and 4.27nM/ml, respectively.

        Caffeine, 철분 및 vitamin E 혼합투여시 rat의 혈액과 간조직내에서 혈액화학성분과 지질 및 단백질 구성성분의 변화

        This study was conducted to identify the effects of caffeine or combinations of caffeine and iron or vitamin E on the lipid and protein components or blood chemistry levels of the serum as well as the total homogenate, mitochondrial and microsomal fraction of the rat(Sprague-Dawley, female) liver. Chronic test were conducted to determine those effects. The chronic test was conducted by dividing rats into 5 groups according to the type of drugs and dosages administrated as follows; the control(group A), and group B was given 25mg/kg caffeine orally once daily for 30 days, group C was given 50mg/kg caffeine orally once daily for 30 days, group D was given 25mg/kg caffeine and orally ferric chloride once daily for 30 days and group E was given 25mg/kg caffeine and 25mg/kg vitamin E once daily for 30 days. The concentrations of glucose, urea nitrogen, uric acid, creatinine, total protein, albumin, A/G ratio, triglyceride, total cholesterol, HDL-cholesterol, free fatty acid, phospholipid as well as the activities of alanine aminotransferase(ALT), aspartate aminotransferase(AST) and alkaline phosphatase(ALP) were measured in the serum of each experimental groups. The concentrations of the carbonyl group and malondiaidehyde(MDA) and the patterns of the SDS-PAGE(Sodium Dodecyl Sulfate - Polyacrylamide Gel Electrophoresis) and fatty acid compositions in free fatty acids and phospholipids were analyzed to determine the oxidative damages and metabolic changes on the lipid and protein components in the serum, and total homogenate, mitochondrial and microsomal fractions of the rat liver. The results obtained from this study were summarized as follows; 1. Body weights of groups B, C, D and E were significantly decreased(p < 0.01) in comparison with that of the control in the chronic test. 2. The concentrations of serum glucose in groups B(124.5mg/dl), C(130.1mg/dl), D(122.1mg/dl), E(119.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(101.5mg/dl). But, there were no significant differences in the concentrations of urea nitrogen, uric acid, creatinine, total protein, albumin and A/G ratio in comparison to that of the control. 3. The concentrations of total cholesterol and HDL-cholesterol in serum of groups B(69.6, 53.4mg/dl), C(73.0, 56.3mg/dl), D(68.9, 51.1mg/dl) and E(68.2, 51.3mg/dl) were significantly higher(p < 0.01) in comparison to that of the control(52.6, 38.8mg/dl). On the other hand, the concentrations of triglyceride in serum of groups B(45.0mg/dl), C(40.4mg/dl), D(33.8mg/dl) and E(47.2mg/dl) were significantly lower(p < 0.01) in comparison to that of the control(66.2mg/dl). There were no significant differences in the activities of ALT, AST and ALP in comparison to that of the control. 4. The concentrations of free fatty acid and phospholipid in serum of groups B(45.7, 154.4mg/dl), C(50.0, 167.2mg/dl), D(52.5, 148.4mg/dl) and E(41.1, 159.2mg/dl) were higher(p < 0.01) in comparison to that of the control(35.2, 125.3mg/dl). And the concentrations of the carbonyl group and malondialdehyde in serum of group D(1.82, 0.52nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(1.53nM/mg protein). 5. The concentrations of carbonyl group in total homogenate, mitochondrial and microsomal fraction of group D(1.45, 0.94, 1.67nM/mg protein) were significantly higher (p < 0.01) in comparison to the control(1.16, 0.66, 1.27nM/mg protein). And the concentrations of malondialdehyde in the total homogenate, mitochondrial and microsomal fraction of group D(6.70, 6.10, 1.36nM/mg protein) were significantly higher(p < 0.01) in comparison to the control(5.17, 3.64, 0.68nM/mg protein). 6. As the analytical results of the fatty acid compositions of free fatty acid in serum, the proportions of stearic acid and arachidonic acid of groups B(16.52, 12.62%), C(17.52, 15.18%), D(19.73, 13.47%) and E(17.62, 13.28%) were significantly higher(p < 0.01) in comparison to the control(14.75, 7.88%), but the pr

        Effect of Administration of Iron on the Lipid Concentrations in the RBC Membrane and Plasma

        철분투여가 용혈성 빈혈에 미치는 영향을 알아보기 위하여 흰쥐의 사료내에 철분을 혼합급여 또는 근육내에 주사하면서 60일간 사육하고 체중은 10일 간격으로 측정하였으며 적혈구의 취약성 실험과 적혈구막 및 혈장내의 vitamin E 함량과 인지질의 함량을 비색방법으로 측정한 결과 다음과 같은 성적을 얻었다. 증체량 측정에서는 A군의 190g에 비하여 C군 155g, B군 142g, D군 112g으로 철분함량이 높은 군에서 낮은 증가를 보였으며 적혈구 취약성 실험 결과 5마리 암수 평균값이 각각 A군 7.63% 및 2.68%, C군 10.84% 및 16.75%, B군 25.54% 및 33.62%, D군 47.23% 및 38.35%로 철분함량에 비례하여 각 실험군 사이에 유의한 증가를 나타내었다. 또한 적혈구막의 vitamin E 및 인지질의 함량은 5마리 암수 평균값이 각각 A군에서 18.54mg/dl 및 17.43mg/dl 그리고 120.55mg/dl 및 113.94mg/dl, C군에서 14.99mg/dl 및 10.56mg/dl 그리고 108.54mg/dl 및 99.97mg/dl, B군에서 13.10mg/dl 및 11.02mg/dl 그리고 91.67mg/dl 및 87.45mg/dl, D군에서 6.86mg/dl 및 8.87mg/dl 그리고 79.78mg/dl 및 70.12mg/dl로서 철분 투여량에 비례적으로 감소하였다. 혈장내의 vitamin E 및 인지질의 함량은 A군, C군, B군, D군의 순으로 각각 함량이 2.51mg/dl 및 31mg/dl, 1.94mg/dl 및 21mg/dl, 1.37mg/dl 및 20mg/dl 그리고 1.37mg/dl 및 15mg/dl로 철분 투여량에 비례적으로 감소하는 경향을 나타내었다.

        BB 랫드 및 streptozotocin이 투여된 랫드에서 vitamin E와 insulin 병합 투여 영향 I. 지질과 단백질의 산화성 손상 및 혈청내 효소 활성도에 미치는 영향

        The present study, to evaluate the effect of vitamin E on the oxidative stress in STZ-treated rat and BB rat, was investigated the biochemical enzyme activity in the serum, and malondialdehyde and carbonyl group in the RBC membrane, liver and microsomal fraction after vitamin E and/ or insulin treatment. Results obtained through the experiments were summarized as follows; 1. Effect of vitamin E and/or insulin treatment in STZ-treated rat 1) Lipid peroxidation level in RBC membrane, liver and microsomal fraction was significantly decreased in vi. tamin E and/or insulin treatment group, and especially more significantly decreased in vitamin E with insulin treated group. 2) Protein oxidation level in RBC membrane, liver and microsomal fraction was significantly decreased in vitamin E and/or insulin treatment group. And it was especially more significantly decreased in RBC membrane and liver of vitamin E with insulin treated group. 3) In the enzyme activity in the serum, the activity of AST and ALT was not altered in all experimental group. The increased ALP activity in STZ-treated group was significantly decreased in insulin treated group and vitamin E with insulin treated group. 4) Decreased level of albumin and creatinine after STZ treatment was significantly increased in vitamin E and/or insulin treated group. 5) Level of glucose, cholesterol and triacylglycerol in serum: Glucose level was not significantly different in vitamin E treated group compared to STZ control group. But it was significantly different in the insulin treated group and vitamin E with insulin treated group compared to STZ control group. The cholesterol content in the serum was significantly increased in STZ control group compared to normal control group. And except low dose vitamin E treatment group, it was significantly decreased in vitamin E and/or insulin treated group compared to STZ control group. The triacylglycerol content in the serum was significantly decreased in STZ control group and increased in high dose vitamin E treated group and vitamin E with insulin treated group. But it was not significantly different in low dose vitamin E treated group and insulin treated group compared to STZ control group. 2. Effect of vitamin E and/or insulin treatment in BB rat 1) Lipid peroxidation level in liver was decreased by vitamin E with insulin treatment compared to insulin treatment. But it was not different in microsomal fractions. 2) Protein oxidation level in liver and microsomal fraction was decreased by vitamin E with insulin treatment compared to insulin treatment only in microsomal fractions. These results suggest that the combination treatment of vitamin E and insulin could prevent the oxidative change of lipid and protein of the RBC membrane, liver and microsomal fraction in STZ-treated rats and BB rats.
