이스트 two-hybrid 시스템을 이용한 hnRNP E1 cDNA의 클로닝과 hnRNP E1-hnRNP K 상호결합에 대한 연구

hnRNP K 단백질은 hnRNP 복합체를 구성하는 핵단백질들 중의 하나이며 시토신이 많은 RNA/DNA RNA/DNA sequence에 잘 결합한다. 이 단백질은 핵 내에서만 머무르지 않고 핵과 세포질을 왕복하는 특생을 지니고 있다. hnRNP K의 기능을 조사하기 위하여 우선 hnRNP K와 상호 결합하는 세포내 단백질을 찾아내고자 하였다. 이를 위하여 본 연구에서는 이스트 two-hybrid 시스템을 사용하여 HeLa cDNA library를 탐색하였다. 그 결과 얻은 클론들 중에는 사람의 hnRNP El (poly(rC) binding protein 1) cDNA (GenBank accession number XM_031585) 클론이 포함되어 있었다. 본 논문에서는 이스트 two-hybrid 시스템과 in vitro에서의 생화학적 실험을 통하여 hnRNP El은 hnRNP K와 특이적으로 상호 결합한다는 것을 밝혔다. The heterogeneous nuclear ribonucleoprotein K (hnRNP K) is a component of hnRNP complexes. This protein binds strongly to cytidine-rich RNA/DNA sequences. It is a nucleocytoplasmic shuttling protein. To investigate the functions of hnRNP K, I searched for hnRNP K-interacting proteins in HeLa cDNA library using a yeast two-hybrid screening system. One of the cDNA clones is identical to human hnRNP E1 (poly(rC) binding protein 1) cDNA (GenBank accession number XM_031585). In this study, hnRNP K is shown to specifically interact with hnRNP E1 in yeast two-hybrid system and in vitro biochemical assay.

응급구조학과 학생들의 비판적 사고성향이 임상실습만족도와 추천의도에 미치는 영향

본 논문은 병원임상실습관련 응급구조학과 학생들의 비판적 사고성향이 임상실습만족도와 추천의도에 나타 나는 관계를 규명하는데 있다. 자료 분석을 위하여 사용된 통계방법은 Windows용 SPSS 20.0과 AMOS 20.0을 사용 하였으며, 빈도분석, 문항내적 일치도를 측정하는 Cronbach's ⍺, 요인분석, 다중회귀분석, 구조방정식 모형 분석 등 을 실시하였다. 모든 통계분석은 유의수준 .05에서 분석하였다. 자료 수집은 2012년 5월 12일부터 8월 25일까지 조 사를 실시하여 305명을 수집된 자료를 활용하였다. 응급구조학과 학생들의 비판적 사고성향은 임상실습 만족도에 유 의한 영향을 주는 것으로 나타나 채택되었고, 비판적 사고성향이 추천의도에 유의한 영향을 주는 것으로 나타나 채 택되었으며, 임상실습 만족도는 추천의도에 유의한 영향을 주는 것으로 나타나 채택되었다. 이러한 연구 결과로 볼 때 질적으로 우수한 응급실 응급구조사의 전문 인력이 되기 위해서는 응급구조학과 학생들의 임상실습을 통해 필요 한 교육 내용과 프로그램 개발의 질적 관리가 체계적이고 지속적으로 이루어져야 할 것으로 사료된다 The purpose of this study was to examine the relationship of the critical thinking disposition of paramedic students to their satisfaction with hospital clinical practice and intention of recommendation. As for data analysis, the statistical packages SPSS WIN 20.0 and AMOS 20.0 were employed, and frequency analysis, factor analysis, multiple regression analysis and structural equation modeling were utilized. Besides, Cronbach alpha coefficients were calculated to check item internal consistency. The level of significance was all set at .05. A survey was conducted to collect data from May 12 to August 25, 2012, and the answer sheets from 305 respondents were gathered and analyzed. The critical thinking disposition of the paramedic students was found to have a significant impact on their satisfaction with clinical practice, and the hypothesis about that was accepted. Their critical thinking disposition was found to exert a significant influence on their intention of recommendation as well, and the hypothesis about that was accepted. Finally, their satisfaction with clinical practice was found to affect their intention of recommendation in a significant way, and the hypothesis about that was accepted. The findings of the study suggested that in order to nurture professional and competent paramedics, the content of clinical practice should systematically be managed on a regular basis, and prolonged research efforts should be directed into program development.

응급의료종사자들의 자기조절이 직무만족과 이직의사에 미치는 영향

본 연구는 응급의료종사자의 자기조절이 직무만족과 이직의사에 미치는 영향을 파악하고자 수행되었다. 자료 는 전국 8개 지역에서 2012년 5월부터 8월까지 4개월간 종합병원의 응급실에 종사하는 응급의료종사자 300명으로부 터 설문지를 통하여 수집하였으며 빈도분석, 요인분석, 상관관계 및 다중회귀분석으로 분석하였다. 일반적 특성에 따 라 여자가 남자보다 많았으며, 연령별로는 20대가 가장 많았다. 학력은 대졸이 전문대 및 대학원 이상 보다 많았으며, 결혼 여부에서는 미혼이 기혼보다 많았다. 고용형태에 따라서 정규직이 비정규직 보다 많은 것으로 나타났다. 자기조 절과 직무만족도 및 이직의사의 상관관계가 매우 높은 것으로 나타났다. 자기조절과 직무만족도의 결과 직무만족의 하위요인인 직무내용에 의사결정오류와 정보인식활용이 영향을 주는 것으로 나타났으며, 인간관계에서는 정보인식활 용이, 근무환경에서는 정보인식활용이 영향을 주는 것으로 나타났다. 자기조절과 직무만족이 이직의사에 미치는 영향 에 대한 결과 자기조절이 직무만족의 하위요인인 보상체계, 인간관계에 영향을 미치는 것으로 나타났으며, 또한 보상 체계는 이직의사에도 영향을 미치는 것으로 나타났다. The purpose of this study was to examine the influence of the self- regulation of paramedics on their job satisfaction and turnover intention. The subjects in this study were 300 paramedics who worked at general and larger hospitals. A survey was conducted for about four months from May 6 to August 30, 2012. As for data analysis, the statistical package SPSS WIN 18.0 was employed to make a frequency analysis, and the reliability of the instrument used was checked. A factor analysis was made, and a correlation analysis was carried out to look for connections between the variables, and a multiple regression analysis was utilized. Concerning the general characteristics of the respondents, many of the respondents were female, in their 20s, unmarried and received junior-college or higher education. Self-regulation had a very close correlation to job satisfaction and turnover intention, and self-regulation affected the subfactors of job satisfaction. Self-regulation exerted an influence on job satisfaction and turnover intention. The findings of the study are expected to suggest how to boost the job satisfaction of paramedics to step up the development of the organization that they belong to.

HnRNP L 단백질에서의 핵수송에 중요한 영역의 동정

The heterogeneous nuclear ribonucleoprotein (hnRNP) L is one of the major pre-mRNA binding proteins in human cells. It is localized in the nucleoplasm. It has not been identified whether the domain of hnRNP L is important for nuclear localization yet. For determination of the domain in hnRNP I, that localizes it to the nucleus, a series of amino terminal and carboxy terminal deletions of hnRNP L were Fused to the 3' end of the myc epitope-tagged chicken muscle pyruvate kinase cDNA. The suhcellular localization of transiently expressed polypeptides was investigated by immunofluorescence microscopy. Here, we report that an amino terminal domain of the hnRNP L protein is necessary and sufficient for nuclear localization. This amino terminal domain confers complete nuclear localization onto pyruvate kinase, a cytoplasmic reporter protein when expressed as a fusion protein.
