Background & Objective : Articular cartilage is a potential target for drugs designed to inhibit the activity of matrix metalloproteinases (MMPs) to stop or slow the destruction of the proteoglycan and collagen in the cartilage extracellular matrix. The purpose of this study was to investigate the effects of Aralia cordata Thunb. in inhibiting the release of glycosaminoglycan (GAG), the degradation of collagen, and MMP activity in rabbit articular cartilage explants.Methods : The cartilage-protective effects of Aralia cordata Thunb. were evaluated by using glycosaminoglycan degradation assay, collagen degradation assay, colorimetric analysis of MMP activity, measurement of lactate dehydrogenase activity and histological analysis in rabbit cartilage explants culture.Results : Interleukin-1a (IL-1a) rapidly induced GAG, but collagen was much less readily released from cartilage explants. Aralia cordata Thunb. significantly inhibited GAG and collagen release in a concentration-dependent manner. Aralia cordata Thunb. dose-dependently inhibited MMP-3 and MMP-13 expression and activities from IL-1a-treated cartilage explants cultures when tested at concentrations ranging from 0.02 to 0.2 mg/ml. Aralia cordata Thunb. had no harmful effect on chondrocytes viability or cartilage morphology in cartilage explants. Histological analysis indicated that Aralia cordata Thunb. reduced the degradation of the cartilage matrix compared with that of IL-1a-treated cartilage explants.Conclusion : These results indicate that Aralia cordata Thunb. inhibits the degradation of proteoglycan and collagen through the downregulation of MMP-3 and MMP-13 activities without affecting the viability or morphology of IL-1a-stimulated rabbit articular cartilage explants.

Objectives : The differentiation of osteoblasts is controlled by various growth factors and matrix protein expressed in bone. The aim of this study was to investigate the effects of many herbs medicine(KHBJs) for bone healing that induces osteogenic activity in human osteoblast-like SaOS-2 cells. Methods : The osteogenic effects of KHBJs were evaluated by using cell proliferation(WST-8) assay, alkaline phosphatase(ALP) activity assay, colorimetric analysis of vascular endothelial growth factor(VEGF) expression in human osteoblast like SaOS-2 cell. Also, osteogenic activity of KHBJ fractions(KHBJB and KHBJR) by activity guided fractionation were evaluated. Results : About 7 KHBJs had effect on the proliferation of osteoblast like SaOS-2 cells, and dose-de- pendently increased alkaline phosphatase(ALP) activity. KHBJs markedly increased expression for VEGF. Frac- tionated KHBJs(KHBJB or KHBJR) not enhanced more than KHBJs on osteogenic activity in SaOS-2 cells. Conclusions : This study found that 7 KHBJs had effect on proliferation, ALP activity, and VEGF ex- pression in osteoblast like SaOS-2 cells. These results propose that KHBJs can play an important role in osteoblastic bone formation, and may possibly lead to the development of bone-forming drugs.

Background : Angiogenesis is the proliferation of a network of blood vessels emanating from pre-existing vessels, supplying nutrients and oxygen and removing waste products. Angiogenesis occurs in a variety of normal physiologic and pathologic conditions and is regulated by a balance of stimulatory and inhibitory angiogenic factors. Excessive angiogenesis should be suppressed. However, if blood supply is insufficient, it should be encouraged. Hyul-Mek(血脈) or Hyul-Rark(血絡), known as blood vessels in western medicine, is deeply related to Chung-Ki-Hyul(精 氣 血). The goal of this study is to review the effects of herbal medicines on angiogenesis that is involved in wound healing and enhancement of blood supply. Methods : We conducted a systematic and comprehensive literature search for the identification, retrieval, and bibliographic management of independent studies to locate information on the topic. A computerized search of the published literature of Korea(KISS, RISS), China(CNKI), Japan(Kampo medicine, etc), and western countries(MEDLINE) was performed, and further supplemented with manual searches of print sources(1999 to 2003). Results : The herbal medicines with angiogenic activity were mainly found among herbs that carry replenish Shin-Cheng(補腎益精), foster Eum and improve the circulation of blood(養陰活血), or warm and circulate Kyung-Rark(溫經通絡). In particular, herbs with improve the circulation of blood and clear blood(活血化瘀) activity contain a significant amount of tannin, saponin, and pyrazine. Conclusion : Replenish Ki-Hyul(補氣血) and circulate Kyung-Rark(通經絡) could contribute to the induction of angiogenesis because various growth factors and proliferation, differentiation, and migration of vascular endothelial cells are involved in angiogenic activity.
