흰쥐에서 Nitric Oxide 합성 억제가 Bradykinin의 승압 및 감압효과에 미치는 영향

Bradykinin has been known to elicit a pressor effect when administered centrally, and a depressor effect when administered peripherally. The present study was aimed at investigating whether the blood pressure response to bradykinin is dependent on the endogenous generation of nitric oxide (NO). Effects of NG-nitro-L-arginine methyl ester (L-NAME) on the pressor and depressor responses to intracerebroventricularly and intravenously injected bradykinin (5nmol/rat), respectively, were examined in anesthetized rats. Neither the pressor response nor the depressor response was affected by acute parenteral treatment with L-NAME. The pressor and depressor effects of bradykinin were also noted in rats chronically supplemented with L-NAME in drinking water for 4 weeks. Bradykinin caused a relaxation of the isolated thoracic aorta in vitro, which was not affected in the presence of L-NAME. However, bradykinin failed to cause a relaxation of the aorta isolated from rats chronically treated with L-NAME. These findings suggest that endogenous generation of NO may not completely account for the blood pressure responses to bradykinin in rats.
Dopamine 및 Prostaglandin 억제제가 쥐의 기초대사율에 미치는 영향

It is well known that the basal metabolic rate is closely related to the thyroid hormone secretion. This study was undertaken to investigate the effects of dopamine and prostaglandin inhibitor on basal metabolic rate in rats. 30 male rats were divided into indomethacin, bromocriptine and indomethacin with bromocriptime groups, and drugs were administered orally for 14 days. The measurements of basal metabolic rates were made on three groups of rats by indirected calorimetry using a oxygen consumption apparatus. The following results were obtained. 1. Body weight was slightly increased in indomethacin group, but almost no changed in bromocriptine and indomethacin with bromocriptine groups. 2. Basal metabolic rate was almost no changed in indomethacin group, but after 6th day, there is a tendency to decrease in bromocriptine and indomethacin with bromocriptine groups. 3. On the 14th day, basal metabolic rate was significantly decreased in domethacin with bromocriptine group as compared with indomethacm group.
신성고혈압 흰쥐에서 측뇌실내 Captopril이 Bradykinin의 중추승압효과에 미치는 영향

Effects of intracerebroventricular captopril on the central pressor response to bradykinin were examined in normotensive and 2-kidney, 1 clip (2KIC) Goldblatt hypertensive rats. The experiment was done under pentobarbital (50 ㎎/㎏. IP) anesthesia. Captopril (1 ㎎)and bradykinin (5 nmol) were administered into the right lateral cerebral ventricle, while arterial blood perssure and heart rate were continuously monitored throughout the experiment. Intracerebroventricular captopril did not affect the blood pressure within 10 minutes in both normotensive and 2KIC hypertensive rats, while it significantly augmented the central pressor response to bradykinin in the hypertensive rats. These observations suggest that an elevation in endogenous kinin levels is without effect on arterial pressure in normotensive and 2KIC hypertensive rats. However, the kallikreinkinin system in the central nervous system plays a role in the regulation of blood pressure, possibly maintaining the hypertension, in 2KIC hypertensive rats.
