Effects of Bupleuri Radix, Paeoniae Radix or Uncariae Ramulus et Uncus on experimental hyperlipidemia were studied. Hyperlipidemia was induced on male Wistar rats by feeding them on high cholesterol diet for one week, as previously described by the authors. Blood lipid profile was varified on these rats by measuring total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL) and low density lipoprotein cholesterol(LDL). Then, the rats were followed by feeding normal diet. At the same time, methanol extracts of the three individual herb medicines were orally administered to the rats for 4 weeks and the parameters above mentioned were monitered. Methanol extract of Bupleuri Radix reduced the TC value significantly at 2 week point and Paeoniae Radix reduced the TC value at 4 week point in compare to control group, suggesting the antihyperlipidemic effect of the two herbal medicines in vivo. The extract of Uncariae Ramulus et Uncus however did not show antihyperlipidemic effect in our experiment.

Development of hyperlipidemic animal model is prerequisite for the screening of drugs lowering the blood lipoprotein level. In this study, two kinds of diet regimens were tried for the hyperlipidemic effect. The model will be used for the screening of current prescriptions to a variety of diseases related to hyperlipidemia by practioner of traditional herbal medicine. Fifteen male Wister rats of 200g were divided into three groups, and given normal, or either one of the two high cholesterol diets for up to 2 weeks. Regimen HC-1 and regimen HC-2 contained I% and 2% cholesterol in diet, respectively. Total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL) and lower density lipoprotein cholesterol (LDL) levels in the serum were measured before the start of the diet, at 1 week after the start, and at 2 week point. Both regimens resulted in hyperlipidemia with the typical characteristics of increase in TC and LDL, and reduction in HDL, and showed no observable side effects such as diarrhea. In conclusion, both of HC-1 and HC-2 regimens appeared appropriate as diet regimens for the hyperlipidemic rat model.

Hyperlipidemia was induced on male Wistar rats by feeding high cholestetrol diet for 7 days. Serum lipid profile was verified on these rats by measuring total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL), and low density lipoprotein cholesterol (LDL). Then, the diet was changed to normal. At the same time, hexane, chloroform, methanol and water extract of Paeoniae Radix were given orally on daily basis, and the changes in the serum lipid profile were assessed for 4 weeks. Methanol extract of Paeoniae radix decreased TC level at 1. 2, and 4 week point significantly, and water extract decreased TC level at 4 week point significantly comparing with the control group.

Three herbal medicine recipes were tried on the animal model of diet induced hypercholesterolemia to screen for the lipid-lowering effect. The recipes adopted were based on the prescription for atherosclerosis-related symptoms by folk-medicine practioners. Hyperlipidemia was induced in rats by giving the high lipid/cholesterol diet for 7 days. Then, the recipes started with the normal diet. Blood was sampled at 1,2,3 and 5 week-points of the treatment, and levels of total cholesterol, triglyceride, high density lipoprotein and lower density lipoprotein were measured. Dae-shiho-tang decreased total cholesterol level significantly. Hwangryun-haedok-tang and Samhwang-sasim-tang slightly decreased total cholesterol level, although it was not statistically significant.

In our previous report, the water extract of Bupleuri radix showed the lowering effect of total cholesterol level in the experimentally induced hyperlipidemic rats. In order to elucidate the active ingredient(s), the water extract were dialyzed, ad two fractions, high molecule fraction and low molecule fraction, were obtained. These two fractions were tested on the experimentally induced hyperlipidemia in rats for lowering effect of serum lipoprotein contents. Hyperlipidemia was induced on male Wistar rats by feeding high cholesterol diet for 7 days. After oral administration of above samples for 4 weeks, serum lipids profile was monitored by measuring total cholesterol, triglyceride, high density lipoprotein cholesterol and low density lipoprotein cholesterol level. Low molecule fraction decreased total cholesterol and triglyceride levels significantly (p<0.05) at 4 week point when they were compared with the control group.

We previously showed that a methanol extract of Paeoniae radix decreased total cholesterol level in rats with hyperlipidemia. In order to isolate the active ingredient(s), the methanol extract of Paeoniae radix was fractionated with chloroform/methanol(4:1) solution and isolate into soluble part and insoluble part of the methanol extract. Above two parts were tested on the experimentally induced hypercholesterolemia in rats for the lowering effect of serum lipoprotein contents. Hyperlipidemia was induced on male Wistar rats by feeding high choleserol diet for 7 days. After oral administration of above samples for 4 weeks, serum lipid profile was verified on these rats by measuring total cholesterol, triglyceride, high density lipoprotein cholesterol and low density lipoprotein cholesterol. The chloroform/methanol(4:1) soluble part and insolule part showed lowering activity of total cholesterol level and triglyceride level at 4 week point significantly(p<0.01 and p<0.05) compare with the control group and the soluble part was more effective.

Active component was sought among three herb medicines, which are used in combination as a traditional medicine prescribed for patients with hyperlipidemia related diseases. Antihyperlipidemic effect of this remedy has previously been shown model by the authors on the animal model. Hyperlipidemia was induced on male Wistar rats by keeping them on high lipid diet for one week, as previously described by the authors. Blood lipid profile was verified on these rats by measuring total cholesterol (TC), triglyceride (TG), high density lipoprotein (HDL) and low density lipoprotein (LDL). Then, the diet was changed to normal. At the same time, methanol extracts of Scutellaris baicalensis Georg.(radix), Coptidis japonica Makino(rhizoma) or Rhei koreanum Nakai(rhizoma) were given on daily basis, and changes in the blood lipid profile were monitered for 4 weeks. Methanol extract of Scutellaris baicalensis Georg, significantly reduced the TC value, implying the in vivo antihyperlipidemic effect.
