      • Xanthomonas malvacearum 돌연변이주(突然變異株)의 Heteropolysaccharide 생산성(生産性)에 관(關)하여

        Xanthan gum의 생산력(生産力)이 있는 Xanthomonas SNUF-560 균주(菌株)에서 자외선(紫外線) 조사방법(照射方法)으로 생산능(生産能)이 우수(優秀)한 변이주(變異株)를 선택(選擇)해서 동결건조법으로 보존(保存)했으며 PDA배지(培地)에서 2주(週)마다 이식(移植)하여 생산실험(生産實驗)에 사용(使用)하였다. 최적발효배지(最適醱酵培地)의 검토(檢討)에서 탄소원(炭素源)으로 sucrose 40g/l, 질소원(窒素源)으로 glutamic acid 10mM, 유기산(有機酸)으로 succinic acid 10g/l, $PO_4\;^{-3}\;65mM$일때 각각 좋은 결과(結果)를 얻었으며, 최종적(最終的)으로 배지(培地) 1과 배지(培地) 2에서 각각 1,050cp(12g/l), 900cp(10.5g/l)를 얻었으며 조성(組成)은 다음과 같다. 배지(培地) 1 = sucrose 40g/l, glutamate 10mM, $PO_4\;^{-3}\;65mM(KH_2PO_4\;12.65g/l)$, citrate 2g/l, $MgSO_4{\cdot}7H_2O\;0.2g/l$, $H_3BO_3\;0.006g/l$, ZnO 0.006g/l $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$ 배지(培地) 2 : sucrose 40g/l, $(NH_4)_2SO_4\;2g/l\;PO_4\;^{-3}\;65mM\;(KH_2PO_4\;12.65g/l)$ succinate 10g/l, $MgSO_4{\cdot}7H_2O\;0.02g/l$ $H_3BO_3\;0.006g/l$ $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$. The mutant with high productivity, X. malvacearum SNUF 560-6, was acquired from the X. malvacearum SNUF 560 with low productivity by UV-light irradiation. It was preserved is lyophilized stock culture and it was transferred to PDA slant to maintain viability fortnightly. Fermentations were started by retransfering to MY agar slant from PDA stok culture. The experiments for optimal xanthan gum production were studied in a chemically defined medium. Of the carbon and nitrogen sources tested, 0.4% sucrose medium and 10mM glutamic acid medium yielded the highest xanthan gun production respectively. The addition of 10g/l succinic acid stimulated xanthan gum production. Also 65mM $PO_4\;^{-3}\;(12.6g/l\;KH_2PO_4)$ was effective on xanthan gum production. Finally, medium 1 and medium 2 which have high xanthan gum production potencies were achieved in this stud. The components of medium 1 and medium 2 were as follows: Medium 1 : sucrose 40g/l glutamate 10mM $PO_4\;^{-3}\;54mM\;(KH_2PO_4\;12.65g/l)$ Citrate 2g/l $MgSO_4{\cdot}7H_2O\;0.2g/l$ $H_3BO_3\;0.005/l$ ZnO 0.006/l $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$ Medium 2 : $Sucrose\;40g/l\;(NH_4)_2SO_4\;2g/l$ $PO_4\;^{-3}\;65mM\;(KH_2PO_4\;12.65g/l)$ Succinate 10g/l $MgSO_4{\cdot}7H_2O\;0.02g/l$ $H_3BO_3\;0.06g/l$ ZnO 0.006g/l $FeCl_2{\cdot}6H_2O\;0.0024g/l$ $CaCO_3\;0.02g/l$.

        국내 쇼핑관광지 이미지형성에 관한 연구 -명동, 인사동, 동대문, 남대문, 이태원지역을 중심으로-

        This study is to propose that the shopping tourism as an independent sector is an important source of the domestic tourism industry recognized as the next-generation growth engine, on the basis of the shopping destination image. In the pursuit of this aim, we approached the issue from behavioral aspect focusing on results of conceptual difference between inbound tourists’ satisfaction and attitude of shopping destination image, which is not adequately addressed in the existing research. Specifically, we designed the structural model on relationship between shopping destination image, satisfaction, attitude and behavior intention of inbound tourists. As per the results, the shopping tourists’ satisfaction and attitude reacted differently according to shopping destination image types. Especially the shopping tourists’ behavior intention was influenced more by results of attitude than satisfaction on indirect dimension. Finally, this study is conceptually meaningful in presenting the different path of the shopping image types on satisfaction and attitude. In addition, this study suggests some useful implications for tourism industry workers and policy makers.

        정신지체아동의 K-ABC와 K-SIB-R 결과 차이에 대한 비교 분석

        이 연구는 정신지체아동의 K-ABC와 K-SIB-R의 하위요소에 대한 결과와 두 검사 간의 상관관계를 비교·분석하는데 목적이 있다. 부산과 대구 지역에 거주하며 치료실을 이용하고 있는 만 2세부터 만 12세 5개월까지의 정신지체아동 176명을 대상으로 K-ABC와 K-SIB-R 검사를 실시하였다. 연구 결과, K-ABC와 K-SIB-R의 하위요소에 대한 정신지체아동의 검사결과는 성별과 연령 집단별 간에 차이가 없었으며, 또한 두 검사의 하위요소 간에는 높은 상관관계가 나타났다. 즉 두 검사 도구는 정신지체아동 판별에 타당하며, 검사에 대한 결과들을 교육적 지원 등의 활용에 가능하다는 것을 이 연구에서는 시사하고 있다. The purpose of this study was compared and analyzed that Korean K-ABC and K-SIB-R results for sub-elements and the correlation between the two tests for mental retarded children. The K-ABC and K-SIB-R were examined for 176 mental retarded children who were 2 year to 12 years 5months and were studying at clinics in Busan and Daegu. The results of this study were that: (a) results of the subtests in the K-ABC for mental retarded children were no difference between gender and age; (b) the corellation of the subtests of the K-SIB-R was validated to test for mental retarded children; and (c) the correlation between K-ABC and K-SIB-R was meaningful, however the correlation of the K-ABC and the inappropriate test which was a part of the K-SIB-R was not suitable statistically. This study concluded that both of the two tests, K-ABC and K-SIB-R had to be tested to get validation to diagnosis and evaluation for mental retarded children.

      • 液浸培養에 依한 酒精製造에 對하여 : (第Ⅱ報) 培地選定

        1. 液浸培養에 適當한 9種類培地로 前報에서 選定한 Aspergillus inuii, A. niger NRRL 337 및 330 을 使用하여 糖化力을 檢定하였다。 2. 培地中 C.D兩培地가 振動培養 및 Aeration培養에 있어서 다 優秀한것 같으나 一致되지않는 結果를 發見하였다。 卽 原料를 甘藷와 玉蜀黍를 組成分으로 할때에 이들 糖化力에는 何等의 影響을 주지 않었었다。 3. 培地組成分에 앞서서 培地中의 溶存酸素量이 이들 酵素生産力에 至大한 影響을 주는것 같다。 4. 本實驗에 있어서는 前報와는 달리 A. niger NRRL 337이 A. inuii보다 優秀한것 같다。 1) The saccharifying activities of the previously reported strains, Aspergillus inuii, Aspergillus niger NRRL 337 and 330 were examined for nine kinds of culture media suitable for a submerged culture, under the culture conditions of shaking and aeration. 2) Although both media C and D seem to be superior to other culture media under the conditions of shaking and aeration, the result does not coincide. That is, the saccharifying activities were not affected to any extent by the difference of raw material, sweet potato and corn. 3) The dissolved oxygen in the culture media has greater effect on the production of enzymes than the composition of the media does. 4) Aspect from the previous report, A niger NRRL 337 seems to be better in saccharifying activities than A. inuii in these experiments.
