        Streptomyces coelicolor의 Protoplast Fusion 방법개발

        Streptomyces속 균주개발의 수단으로서 이용할 목적으로 원형질체 융합방법의 확립을 시도하였다. 특히 융합빈도를 높이고 실험을 간편화하는데 역점을 두었다. 원형질체의 형성 및 재생빈도는 균의 배양시간에 따라 변하였는데 대수기에서 수확한 균체로부터 가장 높은 빈도의 수율을 얻었다. 원형질체의 형성은 다른 용균효소를 사용하지 않고 Lysozyme 단독처리 만으로도 충분히 가능하였고 원형질체의 세포막 재생은 Monolay법 보다는 Overlay법이 훨씬 좋은 결과를 주었다. Monolay법은 1.8%, Overly법은 14%의 재생빈도를 나타냈다. 본 실험에서 PEG1000 (50% W/V)를 사용한 원형질체 융합방법으로 얻은 S. coelicolor의 재조합체의 빈도는 $1.8 {\times} 10^{-2}$이었다. Attempts were made to optimise protoplast formation and regeneration methods to improve the protoplast fusion frequencies of Streptomyces coelicolor. The yields of protoplast formation and regeneration were varied with different growth phase of the culture. Maximum yields were obtained when cells were taken from the late logarithmic phase. Protoplast formation reached almost its maximum with lysozyme treatment at a concentration of 2mg/ml without any other lytic enzyme. A high frequency of protoplast regeneration was accomplished by overlay method: the method gave 14% recovery of regenerated protoplast versus 1.8% recovery for monolay method. A recombinant frequency of 1.8X10^-2 was obtained by protoplast fusion using PEG 1000(50% w/v).

        마우스에서 diazinon, toxaphene 과 endrin 단독 혹은 그 혼합물 독성의 대사

        The effects of mixtures of diazinon(DA;5mg/kg), toxaphene(TOX;40mg/kg) and/or endrin(END; 5mg/kg) on the hepatic mixed-function oxygenase(MFO) system were stuided in ICR mice(18~22g) by oral intubation daily for 7 days. In general, TOX and TOX-containing mixtures were found to induced the metabolism of aminopyrine(22~60%), aniline(42~85%), phenacetin(145~194%) and benzo [a]pyrene(158~210%), and pentobaribtal biotransformation in the 9,000g liver supernatants and to increased the hepatic cytochrome p-450 contents(47~89%). Results of these may be, at least in part, associated with the MFO system. TOX pretreatment increased the aliesterase activity in the serum and liver homogenates and supernatants by 23~145%. The toxicity of TOX and TOX-containing mixtures would be lower than that of diazinon because of TOX-induced increase in the metabolism of diazinon(DA) or diazioxon(DO) and capability of TOX to stimulate the metabolism of diazinon and diazioxon and provide a pool of non-critical enzymes. These results suggest that this information might be helpful in the evaluation of the potential hazard due to occupational and/or environmental exposures to pesticides and their mixtures.

        쥐(rats)의 full-thickness skin grafts에 대한 hyperbaric oxygen과 α-tocopherol의 효과

        To document that effects of hyperbaric oxygen(HBO) and ${\alpha}-tocopherol$ on full-thickness skin grafts in rat, we performed full-thickness skin grafts bilaterally on each rats. The HBO-treated rats were received HBO twice daily for 90 minutes at 2 ATA. Surgical control rats were not treated with HBO. ${\alpha}-tocopherol$ treated rats were received the agent via oral gastric tube daily for 3 days preoperative and a fourth dose 1 to 2 hours postoperative. HBO plus ${\alpha}-tocopherol$ treated rats were received HBO and ${\alpha}-tocopherol$ as mentioned above. Biopsy specimens were taken from each rat at the time of grafting and on days 2, 4, 7, 10, 14, 21, and 28, then were processed for tissue-concentration of total glutathione(GSHt), oxidized/reduced glutathione level, and thiobarbituric acid-reactive substance(TBARS) levels. The percentage of viable graft on day 10 ranged from 67 to 93%, and was not significantly different among the each other groups. The percentage of viable graft were, however, higher in HBO plus ${\alpha}-tocopherol$ treated rats(78.6%) than in HBO alone treated rats(59.1%), ${\alpha}-tocopherol$ alone treated rats(66.7%) and surgical control rats(58.2%). TBARS concentration had a significant increase from preoperative concentration at day 2, and peak concentration at day 4(p<0.01). Concentration then decreased to preoperative concentration at day 28. GSHt concentration of free skin graft had a similar patteren of change in four groups and decreased significantly from preoperative concentration at day 2, returning to preoperative concentration by day 7(surgical control, HBO-treated, and ${\alpha}-tocopherol-treated$, alone) and 28(HBO plus ${\alpha}-tocopherol-treated$). Percentage of the concentration of reduced glutathione decreased in surgical control, HBO-treated and, ${\alpha}-tocopherol-treated$(p<0.05), and HBO plus ${\alpha}-tocopherol-treared$(p<0.01) on day 7 after surgery, whereas the concentration of oxidized increased significantly in HBO-treated(p<0.05), ${\alpha}-tocopherol-treated$(p<0.05), and HBO plus ${\alpha}-tocopherol-treated$(p<0.01).

        Phytomitogen에 의한 토끼 임파구의 blast transformation I. 유사분열에 미치는 배지, 유사분열촉진물질 및 배양시간의 효과

        The present study has been carried out to investigate the optimal condition on lymphocyte blastogenesis of rabbit lymphocytes, whole blood culture and microculture system in conjunction with a semiautomatic multiple sample harvester(SAMSH) was used to study the In vitro optimal condition of rabbit lymphocytes. Data were presented to show many variables that are involved in studying the phytohemagglutinin(PHA) and lipopolysaccharide(LPS) response of rabbit lymphocyte in a microculture system. Analysis indicated that the conditions for optimal PHA as measured by incorporation of $^3H$-TdR include: (1) use of RPMI-1640 as culture medium. (2) use of $6{\mu}g$ of PHA, per culture. (3) 48-hours culture period. Conditions for optimal stimulation with LPS mitogen were similar to those used for PHA.

        기니픽그에서의 실험적 trichothecene (T-2) 독소중독증

        Guinea pigs were administrated with T-2 toxin at a rate of 1 and 0.6mg/kg body weight per day for 21 days to study the immunological and pathological effects of T-2 toxin in guinea pigs. Blood was collected before T-2 treatment and on days 7, 14 and 21 of the trial for hematological and biological examinations and for the mitogen assay using lymphocytes. Myeloid: erythroid ratios were examined from the fernur bone marrow samples taken a day before T-2 toxin treatment began, on day 12 and at death. Guinea pigs received with 1mg/kg body weight of T-2 toxin daily showed leukopenic, lymphopenic and anemic signs on day 7 and 14. The mitogenic responses to the T-cell mitogen, Concanavalin A and B-cell mitogens, lipopolysaccharide were significantly depressed on day 7. Histologically, marked cellular damages including karyorrhexis and depletion of lymphocytes were observed in the actively dividing cells of the gastrointestinal tract, lymph node, spleen and bone marrow of guinea pigs.

        Vomitoxin의 면역 독성효과

        The present study were carried out to investigate the effects of Vomitoxin(Deoxynivalenol, DON) induced dysregulation of IgA synthesis. The data presented here demonstrate that exposure to dietary DON increases serum IgA and concurrently decreases IgG and IgM. Peyer's path(PP) and splenocytes from mice fed dietary DON produced increased IgA in mitogen stimulated and macrophage(MØ) perform essential role for Ig synthesis and was affected by the presence of DON. The synthesis and secretion of IgM and IgG under the presence of DON do not require T cell helper activity, whereas those of IgA do require such activity. These results suggest that dietary DON alters the regulation of IgA production and may act as an immunostimulator.

A microculture system in conjunction with a semiautomatic multiple sample harvester(SAMSH) was used to study the in vito optimal condition of guinea pig lymphocytes. Data were presented to show many variables that are Involved in studying the responses of guinea pig lymphocyte in a microculture system to the stimulation of Concanavalin A(Con A) and lipopolysaccharide (LPS). Analysis indicated that the conditions for optimal Con A as measured by incorporation of $^3H$-TdR include : (1) use of RPMI-1640 as culture medium, (2) use of $6{\mu}g$ of Con A, per culture, (3) use of $1{\times}10^6$ cells per culture. Conditions for optimal stimulation with LPS mitogen were similar to those used for Con A.

        The present study was carried out to investigate the optimal conditions on blastogenesis of guinea pig lymphocytes. A microculture system in conjuction with a semiautomatic multiple sample harvester was used to study the in vitro optimal condition of guinea pig lymphocytes. Careful analysis of lymphocyte transformation to concanavalin A (Con A) and lipopolysaccharide (LPS) mitogen determined optimal conditions as: (a) 10% fetal bovine serum in RPMI-1640 medium (b) 48-hour culture period.

        스위칭 손실 감소에 의한 단상 부스트 컨버터의 효율개선

        본 논문은 고주파 단상 부스트 컨버터의 스위칭 손실을 감소시킬 수 있는 새로운 방식을 제안한다. 이것은 기존의 부스트 컨버터에 별도의 스위칭 장치를 첨가한 형태를 가진다. 원래의 고속 스위칭 장치는 변함없이 전력변환을 수행하지만, 새로이 부가된 장치는 저속으로 동작하면서 고속 스위칭 소자에 흐르는 전류의 대부분을 우회시켜 스위칭 손실을 감소시킨다. 제안된 방법의 제어시스템은 매우 간단하다. 제어기는 멀티바이브레이터, 비교기 및 AND 게이트로 구성되고, 스위칭 소자의 오프 지속시간이 멀티바이브레이터에 의해 일정하게 유지되므로 최대 스위칭 주파수는 별도의 클럭 발생기 없이 제한된다. 본 논문에서는 제안된 컨버터의 형상, 설계 등을 언급하고 컴퓨터 시뮬레이션을 사용하여 제안된 방법의 스위칭 손실 감소, 효율향상에 관한 유효성을 입증한다. This paper proposes a new technique for improving the efficiency of single phase high frequency switch mode boost converter. This converter includes an additional boost converter that follows the main hish frequency switching device. The additional converter, which is controlled at lower frequencies, bypasses almost all the current in the main switch and the high frequency switching loss is greatly reduced. Both switching devices are controlled by a simple method; each controller consists of a one-shot multivibrator, a comparator and an AND gate, and the maximum switching frequency can be limited without any clock generator. The converter works cooperatively in high efficiency and acts as though it were a conventional high frequency switch mode converter with one switching device. This paper describes the proposed converter configuration, design, and discusses the steady state performance concerning the switching loss reduction and efficiency improvement. and the proposed method is verified by computer simulation.

      • Artificial Traffic Signal Light using Fuzzy Rules

        기존 교통신호등은 최적 교통신호주기기능을 상실했다. 기존 교통신호주기는 현시 교통주기와 30% -45% 가 일치하지 않고 있다. 본 논문에서는 평균 주행속도를 개선하고 평균 승용차 대기시간을 단축하기위해서 전자 교통신호등을 본 논문에서 제안한다. 본 논문에서는 실제 교통상황의 교통량, 출발 지연시간, 자동차속도 등의 40가지 센서 입력조건들을 저장하는 방법을 연구 중이다. 지능형 퍼지 기법을 이용하면, 서로 다른 10개의 교차로에 서 최 적의 녹색시간을 예측할 수 있다. 컴퓨터 모의실험 결과, 자동차 길이를 고려하지 않은 기존 교통신호등보다 . 오프셋 및 평균 자동차 대기시간을 줄일 수 있음이 입증되었다. The conventional traffic light loses the function of optimal traffic signal cycle. And so, 30-45% of conventional traffic signal cycle is not matched to the present traffic signal cycle. In this paper proposes electro sensitive traffic light using fuzzy rules which will reduce the average vehicle waiting time and improve average vehicle speed. This paper is researching the storing method of 40 different kinds of sensor input conditions. Such as, car speed, delay· in starting time and the volume of cars in the real traffic situation. It will estimate the optimal green time in the 10 different intersections using Intelligent fuzzy method. Computer simulation results prove that reducing the average vehicle waiting time and offset better than fixed signal method which doesn't consider vehicle length.
