Peroxidase Activity and Isoenzyme Pattern Change in Two Shoot Initiating Hormonal Levels

담배 callus 배양시 kinetin과 IAA가 shoot형성과 peroxidase 활성 및 isoenzyme pattern 변화에 미치는 영향을 조사하였다. 담배 callus를 $5{\times}10^{-7}$ Kinetin/$10^{-8}\;M$ IAA 또는 $10^{-5}\;M$ kinetin/$10^{-5}M$ IAA 배지에서 15일 정도 배양하였을 때 shoot형성이 시작되었다. Shoot 형성 초기의 peroxidase 활성은 위의 두 경우 모두 대조군 ($5{\times}10^{-7}\;M$ kinetin/$10^{-5}\;M$ IAA) 보다 증가하였으 며 $10^{-8}\;M$ IAA 배지에서 배양한 callus 에서는 $10^{-5}\;M$ kinetin 배지에서 배양한 callus 보다 shoot 형성시 약 두배가량 높은 peroxidase 활성도를 나타냈다. 또한 shoot형성시 $10^{-8}\;M$ IAA callus에서 peroxidase의 활성도 증가는 cathodic isoperoxidase의 증가와 일치 하였으며 $10^{-5}\;M$ kinetin callus에서는 anodic isoperoxidase의 증가와 일치 하였다. 이와 같은 사실로 보아 shoot 형성과정에 있어서 total peroxidase의 양보다는 isoperoxidase의 종류가 중요한 역할을 하는 것으로 사료된다. The peroxidase activities of shoot intiating calli were higher than the normal non-shoot forming calli at the initial stage of shoot formation. It was also noted that the activity of peroxidase in $10^{-8}\;M$ IAA cultures was about twice higher than that of $10^{-5}M$ kinetin cultures. The increased enzyme activity in $10^{-8}\;M$ IAA cultures was due to the increased amount of cathodic isoperoxidases while anodic isoperoxidases were responsible for the increase of peroxidase activity in $10^{-5}M$ kinetin cultures. The results indicate that kinds or groups of isoperoxidases not over-all concentration of enzymes are important in shoot initiating process.

Tobacco callus grown on 5×10^(-7) M kinetin/10^(-8) M IAA or 10^(-5) M kinetin/10^(-5)IAA formed shoots after 15-20 days of culture. The total peroxidase activities in shoots were lower than those of non-shoot-forming callus. However, cathodic isoperoxidases were increased significantly in shoots formed on 10^(-8) M IAA media while anodic isoperoxidases were increased in shoots formed on 10^(-5) M kinetin media as is the case of shoot initiating callus. The gene expression patterns of isoperoxidases in shoot and non-shoot-forming tissue were also examined by rabbit reticulocyte lysate in vitro translation system. These studies indicate that fraction of translatable poly(A)isoperoxidase mRNA was increased considerably in shoots formed on 10^(-5) M kinetin, whereas poly(A)^+ isoperoxidase mRNA contents were twice higher than poly(A)isoperoxidase mRNAs in non-shoot-forming callus. At the present time, at least five isoperoxidases could be detected from the translation mixture of total cellular RNA. Preliminary translation data indicate that less cathodic isoperoxidases and more anodic isoperoxidases were expressed from poly(A)^- RNAs than from poly(A)^+ RNAs.

인두염 환아에서 A군 연구균 검출의 임상적 점수제의 유용성

목 적 : 소아 만성 질환의 원인이 되기도 하는 A군 연구균에 의한 인두염을 정확히 진단하여 항생제 투여 여부를 결정하는 것은 매우 중요하며 시간이 소요되는 인두 배양 결과가 나오기 전 조기 진단을 위해 여러 방법들이 연구되어 왔다. 이는 무분별한 항생제 사용으로 인한 경제적 손실과 균내성의 문제가 대두되면서 더욱 강조되어 온 내용이다. 임상적 증상을 기초로 한 점수제로 진단의 효용을 평가한 과거의 연구가 있으나 혈액 검사 소견을 첨가한 점수제 변형으로 진단적 유용성을 평가해 보고자 하였다. 방 법 : 1998년 1월부터 2000년 12월까지 인두통과 상기도 감염 증상으로 경희대학교병원 외래를 방문한 환아 45명에 대하여 1997년 발표된 Breese의 9가지 항목을 수정 보완하여 임상증상, 나이, 이전의 항생제 사용력, 진찰 소견에 더불어 백혈구수와 그 분율에 대한 검사를 실시하여 각각 점수화하여 결과를 예측하였으며, 인두 배양 검사를 실시한 후 점수제의 결과와 비교하였다. 결 과 : 45명의 환아 중 A군 연구균 배양 양성인 경우는 20명, 음성인 경우 25명이었으며, 배양 양성인 환아 중 그 점수 분포상 70 백분위수에 해당하는 30점을 기준으로 하여 배양 검사 결과와 비교하여 민감도 35%, 특이도 96%, 양성예측도 88%, 음성예측도 65%의 결과를 얻었다. 결 론 : 각 항목별 소견상 A군 연구균 감염을 시사하는 의미있는 결과를 보이지는 않았고 혈액 소견을 첨부함으로써 1997년의 결과에 비하여 특이성을 높이고 위양성율을 의미 있게 감소시켰다. 비교적 감수성은 낮으나 특이성이 96%로 우리나라와 같이 균분리율이 낮은 경우에서는 이 점수제가 유용할 것으로 사료된다.
Isoperoxidases and Translatable Isoperoxidase mRNAs in Shoots and Non-shoot-forming Tobacco Callus

담배 callus를 $5{\times}10^{-7}\;M$ kinetin/$10^{-8}\;M$ IAA 배지 또는 $10^{-5}\;M$ kinetin/$10^{-5}\;M$ IAA 배지에서 15~20일 배양했을 때 shoot이 형성되었다. 두 조건에서 형성된 shoot에 있어서 peroxidase의 활성도는 shoot을 형성하지 않은 callus 보다 낮았다. 또한 $10^{-8}\;M$ kinetin 배지에서 형성된 shoot에서는 cathodic isoperoxidases의 양이 증가한 반면 $10^{-5}M$ Kinetin 배지에서 형성된 shoot에서는 anodic isoperoxidases가 증가하였다. 담배 callus의 mRNA를 분리하여 in vitro translation을 행한 결과 최소한 5개의 isoperoxidases mRNA가 존재함을 확인하였으며, $poly(A)^-RNA$에서도 peroxidase가 합성되었다. 정상배지에서 4주 배양한 callus에서는 $poly(A)^+$ RNA와 $poly(A)^-$ $RNA{\mu}g$ 당 거의 같은 양의 peroxidase가 합성되었으나, $10^{-5}M\;M$ kinetin 배지에서 형성된 shoot에서는 $poly(A)^+$ RNA에서보다 $poly(A)^-$ RNA에서 더많은 peroxidase가 합성되었다. 또한 $poly(A)^-$ RNA에서는 $poly(A)^+$ RNA보다 cathodic isoperoxidases가 더 적게 translation되는 사실도 확인하였다. Purpose: The purpose of this study was to examine the effect of a blood glucose control protocol for medical intensive care unit (ICU) patients. Methods: The subjects were recruited from medical ICU adult patients whose blood glucose levels exceeded 200 mg/dL in two consecutive tests. The experimental group (n=62) received the modified Yale (MY) insulin protocol, whereas the control group (n=64) was treated with the conventional insulin therapy methods. Results: In the experimental group, the mean blood glucose levels (p<.001) and the time to reach the target range of glucose (p<.001) decreased significantly while the incidence rates of a target range of glucose of 100-140 mg/dL (p<.001) increased significantly as compared to the control group. However, no statistically significant differences were found in the incidence of hypoglycemia(p=.644), or the number of glucose tests (p=.236) between the groups. The length of stays in the ICU (p=.001), ventilator care days (p=.038), and the Sequential Organ Failure Assessment (SOFA) score (p=.029) in the experimental group were significantly lower than those of the control group. Conclusion: Application of the protocol was effective in improving the state of blood glucose control in medical ICU patients. Therefore, this protocol is expected to be used as a part of nursing intervention in critical care nursing.
감각통합치료가 아스퍼거 아동의 전정.고유감각 처리능력에 미치는 효과

Objective : This study verifies the effects of sensory integrative (SI) therapy on vestibular- and proprioceptive sensory (BPS) processing ability of a child with Asperger Syndrome (AS). Method : A boy who is 11 years and 2 months old took the Functional Independence Measure for Children (Wee-FIM), Canadian Occupational Performance Measure (COPM), Short Sensory Profile, Bruininks-Oserestky Test of Motor Proficiency-2 (BOT-2), and Test of Playfulness (ToP) for the baseline. The child participated in 3 evaluation sessions and 8 therapy sessions based on the AB research design. Duration of each session is 50 min and the therapy session is divided into 40 minutes for treatment and 10 minutes for evaluation. Since the vestibular sense and proprioception build up one's ability of postural control, several tests were employed to evaluate the child' postural control as outcome measure; distance from front leg of chair to heel of the child with sitting (C-H distance), angle between trunk and thigh (hip joint angle) with sitting, and the 'prone-extension posture' which is a subtest of Clinical Observation of Motor and Postural Skill (COMPS) to examine postural control embodied with integration of reflex and BPS processing. Result : During the therapy, average data of the C-H distance is decreased from 27.33cm to 11.69cm, average data of the hip joint angle is also decreased from $43.3^{\circ}$ to $20^{\circ}$, and average time for the prone-extension posture is increased from 13.15seconds to 24.84seconds. Conclusion : This result indicates that the ability to postural control in sitting and to maintain the prone-extension posture can be improved by sensory integrative therapy, with enhanced BPS processing.

To isolate a novel antifungal compound, we obtained 100 kinds of endolichenic fungi from Korean Lichen & Allied Bioresources Center and examined their antifungal capability. Three fungi Usnea rigidula (2326), Parmotrema pseudotinctorum (2202) and Myelochroa sp. (2292) showed high antifungal activity against Candida albicans when they grew in both liquid and solid media. We extracted the culture supernatants of these three fungi with chloroform and then with ethyl acetate. Chloroform fraction exhibited the highest antifungal activities when those fractions were examined for the growth inhibition of Candida albicans with disc diffusion method. The chloroform faction was on further analysis with C18 column chromatography to see whether the inhibitors are already known or not. Two peak fractions were collected from 4-day culture extract for Usnea rigidula and from 6-day culture extract for Parmotrema pseudotinctorum on the HPLC. A peak fraction from chloroform extracts of 4-day culture filtrate of Parmotrema pseudotinctorum showed higher antifungal activities against C. albicans and C. glabrata than another peak fraction. It appears that the antifungal materials are relatively nonpolar as usnic acid often found in lichenic fungi.

『Gyu-hab-han-hwon』 is a book of exemplary correspondences which were sent and received between the women of both families that were supposed to get marry their son and daughter in accordance with the decorum of Chosun Dynasty. Therefore, through this book, we can understand how our ancestors communicated with a medium of letter in the time of important situations and events of life, and how the situation and events affected the contents and formality of the correspondences. We can also glace at the tradition of wedding ceremony through the correspondences which were written to the parents of the women-writers' or to the family of the women-writers' husband in the middle of or after the marriage of their children. This study examined the correspondences of 『Gyu-hab-han-hwon』 which has no previous research yet. First, the present study surveyed their contents, and then, defined their category as a formal type Eongan. In addition, with the investigation of the correspondences after the Chosun dynasty, I could affirm that the tradition of formal type Eongan continues to exist to date. However, I cannot develop the discussion further than the above statements because the exact bibliography of 『Gyu-hab-han-hwon』 is still not proved. This needs to be approved by more systematic investigations of related researchers.
