Atractylodes japonica is a perennial plant belonging to the Compositae family, distributing in the alpine region of central and northern of Korea. It has been reported that A. japonica has immune enhancement, anti-inflammatory, antitumor, and diuretic effects. In this investigation, we evaluated the growth of probiotic strains including Lactobacillus plantarum, Streptococcus thermophilus, and Lactobacillus helveticus under fermentation conditions using 30% fermented ethanol extract of A. japonica (AJE), and based on this, the anti-inflammatory effects of AJE and its fermented products which are fermented with Lactobacillus plantarum (AJE-LPF) and Streptococcus thermophilus (AJE-STF) in lipopolysaccharide (LPS)-induced RAW264.7 and BV2 cells. The three probiotic strains grew significantly with fermentation time, and in particular, the growth of them increased markedly under high concentration conditions compared to low concentration conditions. In addition, the fermented products under high concentration condition with AJE-STF markedly decreased the LPS-induced production of nitric oxide (NO) compared to those effect of other products in both RAW264.7 and BV2 cells.

Two different extraction methods were used to evaluate the medical value of common rue, Ruta graveolens L. (RGL). The results of our 2,2-diphenyl-1-picrylhydrazyl and 2,2'-azino-bis-3- ethylbenzothiazoline-6-sulphonic acid assays indicated that the antioxidant activity of RGL essential oil extract obtained through steam distillation was very low, whereas ethanol (EtOH) extracts of RGL showed higher antioxidant activity. RGL essential oil was extracted by steam distillation and characterized by GC/MS analysis. Furthermore, EtOH extracts of RGL were obtained under reflux and analyzed by HPLC/PDA. The GC/MS results indicated that the ketone compounds 2-undecanol acetate, nonyl cyclopropanecarboxylate, and 2-nonanone accounted for more than 70% of the composition of RGL essential oil. The HPLC/ PDA analyses indicated that the RGL extracts were rich in phenolic compounds such as protocatechuic acid, rutin, psoralen, xanthotoxin, and bergapten, among which rutin was the most abundant. Collectively, our results demonstrated that RGL contains high levels of phenolic compounds and could thus be commercialized as a valuable plant-derived antioxidant.
섬쑥부쟁이와 쑥부쟁이의 항산화 및 지방세포 분화 억제 효과

본 연구에서는 섬쑥부쟁이 (Aster glehni)와 쑥부쟁이(Aster yomena)의 항산화 효과 및 항비만 효과를 검정하기하여 물 (-W)과 에탄올 (-E)추출물을 이용하여 실험을 실히 하였다. 각 추출물을 총 페놀 및 총 플라보노이드를 함량을 분석하고 DPPH와 ABTS+를 이용하여 항산화 활성을 비교 분석하였다. 또한 총 페놀 및 총 플라보노이드를 함량과 항산화 활성의 상관관계도 분석하였다. 항산화 물질로 알려진 총 페놀과 플라보노이드 함량은 섬쑥부쟁이의 70% 주정 추출물에서 각각 45.42 mg/g와 40.32 mg/g으로 다른 추출물에 비해 높은 함량을 나타내었다. 항산화 효과의 경우 또한 ABTS+의 IC50값이 섬쑥부쟁이의 70% 주정 추출물에서 30.1 μg/mL로 가장 우수하였다. 총 페놀 및 총 플라보노이드를 함량과 항산화 활성의 상관관계도 유의적으로 통계처리 되어 추출물의 함량이 항산화 활성에 영향이 있다는 것을 보여주었다. 또한, 섬쑥부쟁이와 쑥부쟁이의 항비만 효능 평가를 위하여 지방세포인 3T3-L1의 분화 억제를 확인하였다. 추출물을 농도별로 처리하여 지방분화를 Oli-Red O 염색약으로 염색하여 비교하여 보았을 때, 섬쑥부쟁이의 물 추출물에서 70.49%의 가장 낮은 지질 축적율을 나타내었다. 이상의 결과에서, 섬쑥부쟁이의 총 페놀 및 총 플라보노이드를 함량, 항산화 활성과 지방분화 억제 활성이 우수하게 나타났다. 이에 본연구결과를 바탕으로 섬쑥부쟁이가 안정성이 검증된 항비만 기능성 식품이나 의약품 소재로써 활용될 수 있을 것으로 기대된다. Purpose: Aster glehnii (AG) and Aster yomena (AY) are medicinal plants that belong to the family Compositea and grow widely in Korea. Plants in the genus Aster have been used to treat snakebite wounds or bruises in oriental medicine. This study compared the effects of anti-oxidants and anti-adipocyte differentiation according to the species (the aerial parts of AG and AY). Methods: AG and AY were extracted using 70% ethanol (-E) and water (-W) at room temperature. The anti-oxidant activities were measured by total phenol contents (TPC), total flavonoid contents (TFC), DPPH and ABTS+ assay. In addition, correlation analysis was performed for the anti-oxidant compounds and effect. The level of anti-adipocyte differentiation was assessed using an oil red O assay on pre-adipocytes. Results: AG-W showed higher TPC (6.92 μg/mL) and AG-E presented higher TFC (8.22 μg/mL) than the other extracts. Furthermore, AG-E exhibited higher radical scavenging activity in the DPPH and ABTS+ assay (IC50: 104.88 and 30.06 μg/mL). In the cytotoxicity assay, AG and AY extracts at concentrations less than 100μg/mL were non toxic. AG-W reduced the lipid accumulation of 3T3-L1 cells significantly after differentiation (70.49%) compared to the other extracts. Conclusion: These results show that the water extract of AG has anti-oxidant effects and reduces the differentiation of 3T3-L1 cells. Therefore, AG has utility as a functional food material for its anti-oxidant activities and ability to prevent lipid accumulation.

Properties of Hydrogel Composites for The Inkjet-printed In Vitro Alveolar Model

There have been needs for in vitro lung models to rapidly develop new drug or to analyze the association between harmful factors (i.e. virus, fine dust, and chemicals) and various lung diseases. In particular, alveolus, the smallest unit in which gas exchange occurs in the lung, is very thin tissue composed of three layers: vascular endothelial cells, basement membrane, and epithelial cells. Our research group succeeded in fabricating inkjet-printed alveolar model with thin and three-layered structure. We have used collagen as the basement membrane material. However, due to its weak physical properties, shrinkage and degradation of the collagen hydrogel have occurred during the long-term culture of tissues. In this study, we blended collagen type Ⅰ, gelatin methacryloyl, and polyacrylamide to fabricate hydrogels with good cell affinity and high mechanical properties. The blended hydrogel had higher mechanical properties than collagen hydrogel. Most of the cells in blended hydrogel were alive after encapsulation, but cell spreading could not be observed like in the collagen hydrogel. Additional study will be conducted for improvement of biocompatibility.
