MicroRNA Expression Profile Analysis Reveals Diagnostic Biomarker for Human Prostate Cancer

Prostate cancer is a highly prevalent disease in older men of the western world. MicroRNAs (miRNAs) are small RNA molecules that regulate gene expression via posttranscriptional inhibition of protein synthesis. To identify the diagnostic potential of miRNAs in prostate cancer, we downloaded the miRNA expression profile of prostate cancer from the GEO database and analysed the differentially expressed miRNAs (DE-miRNAs) in prostate cancerous tissue compared to non-cancerous tissue. Then, the targets of these DE-miRNAs were extracted from the database and mapped to the STRING and KEGG databases for network construction and pathway enrichment analysis. We identified a total of 16 miRNAs that showed a significant differential expression in cancer samples. A total of 9 target genes corresponding to 3 DE-miRNAs were obtained. After network and pathway enrichment analysis, we finally demonstrated that miR-20 appears to play an important role in the regulation of prostate cancer onset. MiR-20 as single biomarker or in combination could be useful in the diagnosis of prostate cancer. We anticipate our study could provide the groundwork for further experiments.

This is the first report on using Catathelasma ventricosum for production of fruiting body and lignocellulosic enzymes. To improve the laccase activity and productivity of mushroom, the substrate was added with different supplements (eight aromatic compounds, Mn2+, and Cu2+). Based on the results, all these supplements can improve the laccase activity and productivity of C. ventricosum, and it seems that there is a critical value of laccase activity that affects the productivity of C. ventricosum. In addition, when Penicillium decumbens was inoculated into the substrate that had been cultivated with C. ventricosum for 20 days, the highest values of laccase activity, FPA activity, and productivity of C. ventricosum were obtained. Moreover, the laccase activity showed a positive correlation with the productivity of C. ventricosum. Finally, the effect of Mn2+, Cu2+, and P. decumbens on laccase activity was investigated by response surface methodology (RSM).

Two genes coding for eukaryotic translation initiation factors, eIF4E.a and eIF4E.c, were isolated from twelve accessions of Chinese cabbage (Brassica rapa L. ssp. pekinensis). Polymorphism analysis revealed that 94and 142 polymorphic sites were characterized from allele of BraeIF4E.a and BraeIF4E.c which produced complex haplotype structures. Six novel haplotypes were characterized from the two alleles respectively. Among the six novel haplotypes of BraeIF4E.a, three loss-of-function mutations were identified in which a conserved single nucleotide deletion mutation cause the early termination of BraeIF4E.a coding product; while for six new BraeIF4E.c haplotypes, their coding product show amino acid substitution mutations on non-conservative amino acid residues which might affect TuMV infection in Chinese cabbage.

Tissue-specific gene expression is regulated by epigenetic modification involving trans-acting factors. Here, we identified that the human MAGEB16 gene and its mouse homolog, Mageb16, are only expressed in the testis. To investigate the mechanism governing their expression, the promoter methy-lation status of these genes was examined in different samples. Two CpG islands (CGIs) in the 5` upstream region of MAGEB16 were highly demethylated in human testes, whereas they were methylated in cells without MAGEB16 expression. Similarly, the CGI in Mageb16 was hypomethylated in mouse testes but hypermethylated in other tissues and cells without Mageb16 expression. Additionally, the expression of these genes could be activated by treatment with the demethylation agent 5`-aza-2`- deoxycytidine (5`-aza-CdR). Luciferase assays revealed that both gene promoter activities were inhibited by methylation of the CGI regions. Therefore, we propose that the testis-specific expression of MAGEB16 and Mageb16 is regulated by the methylation status of their promoter regions. [BMB Reports 2014; 47(2): 86-91]
Annual Financial Impact of Thyroidectomies for Nodular Thyroid Disease in China

A large proportion of patients with thyroid nodules in China undergo thyroidectomy in order to get confirmatory histology diagnosis. The financial impact of this modality remains to be investigated. To evaluate rationality of performing thyroidectomy without a routine FNA preoperatively from the economic perspective, we conducted a retrospective, observational study of all archival thyroidectomies with records of cost per stay (CPS), cost per day (CPD) and length of stay (LOS) from 2008 to 2013 in the First Affiliated Hospital of Nanjing Medical University. We compared all the parameters between cancer and non-cancer thyroidectomies. We recruited 6, 140 thyroidectomies with valid records of CPS, CPD and LOS in this period. The CPS of cancer thyroidectomy was significantly higher than non-cancer thyroidectomy. The percentage of cancer thyroidectomy increased from 26.5% to 41.6%. The percentage of annual cost of cancer thyroidectomies rose from 30.2% to 45.2%. The LOS for cancer and non-cancer thyroidectomy decreased while the CPD increased in the past six years. The estimated national cost in 2012 for all thyroidectomies would be USD 1.86 billion with USD 1.09 billion for non-cancer thyroidectomies. We have witnessed great improvement in the healthcare for patients with thyroid nodules in China. However, given limited healthcare resources, currently thyroid FNA for more precise preoperative diagnosis may help to curb the rapidly increasing demand in healthcare costs in the future for nodular thyroid disease in China.
Analysis of Key Genes and Pathways Associated with Colorectal Cancer with Microarray Technology

Objective: Microarray data were analyzed to explore key genes and their functions in progression of colorectal cancer (CRC). Methods: Two microarray data sets were downloaded from Gene Expression Omnibus (GEO) database and differentially expressed genes (DEGs) were identified using corresponding packages of R. Functional enrichment analysis was performed with DAVID tools to uncover their biological functions. Results: 631 and 590 DEGs were obtained from the two data sets, respectively. A total of 32 common DEGs were then screened out with the rank product method. The significantly enriched GO terms included inflammatory response, response to wounding and response to drugs. Two interleukin-related domains were revealed in the domain analysis. KEGG pathway enrichment analysis showed that the PPAR signaling pathway and the renin-angiotensin system were enriched in the DEGs. Conclusions: Our study to systemically characterize gene expression changes in CRC with microarray technology revealed changes in a range of key genes, pathways and function modules. Their utility in diagnosis and treatment now require exploration.
Bonded-cluster simulation of tool-rock interaction using advanced discrete element method

The understanding of tool-rock interaction mechanism is of high essence for improving the rock breaking efficiency and optimizing the drilling parameters in mechanical rock breaking. In this study, the tool-rock interaction models of indentation and cutting are carried out by employing the discrete element method (DEM) to examine the rock failure modes of various brittleness rocks and critical indentation and cutting depths of the ductile to brittle failure mode transition. The results show that the cluster size and inter-cluster to intra-cluster bond strength ratio are the key factors which influence the UCS magnitude and the UCS to BTS ratio. The UCS to BTS strength ratio can be increased to a more realistic value using clustered rock model so that the characteristics of real rocks can be better represented. The critical indentation and cutting depth decrease with the brittleness of rock increases and the decreasing rate reduces dramatically against the brittleness value. This effort may lead to a better understanding of rock breaking mechanisms in mechanical excavation, and may contribute to the improvement in the design of rock excavation machines and the related parameters determination.
Plasmonic Nanoantennae Fabricated by Focused Ion Beam Milling

We show a novel approach to fabricate plasmonic nanoantennae based on a maskless focused ion beam nanoring patterning process. Antenna nanoarrays with desired outlines are achieved by precisely controlling the geometric parameters during the milling process. Various nanoantenna designs of bow-tie, nanoclusters (pentamers), and ellipsoid shaped satellites surrounded particle lattices arerealized. The whole fabrication method is programmable and monolithic since only a one-step milling process is involved. The opticalproperties are experimentally characterized. Such nanoantennae may find extensive applications in chemical/bio-sensing due toremarkably enhanced near field intensity at the plasmon resonance.

<P><B>Abstract</B></P> <P>To solve the problems of membrane material tolerance and membrane fouling, a micro/nano hierarchial structure with low surface energy was constructed above the exterior surface of a ceramic hollow fiber membrane using ZnO nanorod arrays and 1H,1H,2H,2H-perfluorodecyltriethoxysilane (PDTS) coatings, which make it superhydrophobic and self-cleaning. The surface morphology, chemical functional groups, and water contact angel of the modified membranes were identified. The results show that large quantity of ZnO nanorods possess desirable characteristics (<I>i.e.</I> superhydrophobicity, exceptional thermal and mechanical stability, and water contact angle of 160.12°) were detected on the ceramic membrane. The novel membrane shows excellent self-cleaning performance and good desalination ability in the utilization of vacuum membrane distillation (VMD) system for high-salinity water desalination.</P> <P><B>Highlights</B></P> <P> <UL> <LI> • Ceramic membrane is modified by imitating of superhydrophobic biological surfaces. </LI> <LI> • The micro/nano hierarchical structure with low surface energy endows the superhydrophobicity. </LI> <LI> • The P-ZN-CHF membrane obtains antifouling ability. </LI> <LI> • The modified membrane shows excellent performances during vacuum membrane distillation. </LI> </UL> </P>
