        Interspecific hybrid between autotetraploid Sorghum(S. bicolor(L.) Moench) and Johnsongrass (S. halepense(L.) Pers) were produced using hand and plasticbags emasculation. The objectives of this research were to study cytogenetics and POD spectrum of the hybrids. The forage value of interspecific hybrid was also performed. The results showed as follows; 1. The average chromosome configuration per cell at meiotic M I of F_1 hybrid's PCMs was 0.17 Ⅰ+14.11Ⅱ+0.028Ⅲ+2.87Ⅳ+0.004Ⅴ+0.007Ⅵ+0.0007Ⅶ. At anaphase Ⅱ, unsynchronized distribution was observed expect laggards, it was negatively and significantly correlated with the number of chain quadrivalents of parent sorghum (r=-0.8718). 2. The POD spectrum of interspecific hybrid was tendency to that of Johnsongrass, and exhibited characteristic bands of Johnsongrass. Index of spectrum similarity between F_1 and parent Sorghum, Johnsongrass were 58.00%, 68.69%, respectively. The genomic model of Johnsongrass and the way of transferring desirable genes to cultivated sorghum from Johnsongrass were also discussed.

        Wnt/β-Catenin Promotes the Osteoblastic Potential of BMP9 Through Down-Regulating Cyp26b1 in Mesenchymal Stem Cells

        BACKGROUND: All-trans retinoic acid (ATRA) promotes the osteogenic differentiation induced by bone morphogenetic protein 9 (BMP9), but the intrinsic relationship between BMP9 and ATRA keeps unknown. Herein, we investigated the effect of Cyp26b1, a critical enzyme of ATRA degradation, on the BMP9-induced osteogenic differentiation in mesenchymal stem cells (MSCs), and unveiled possible mechanism through which BMP9 regulates the expression of Cyp26b1. METHODS: ATRA content was detected with ELISA and HPLC–MS/MS. PCR, Western blot, and histochemical staining were used to assay the osteogenic markers. Fetal limbs culture, cranial defect repair model, and micro–computed tomographic were used to evaluate the quality of bone formation. IP and ChIP assay were used to explore possible mechanism. RESULTS: We found that the protein level of Cyp26b1 was increased with age, whereas the ATRA content decreased. The osteogenic markers induced by BMP9 were increased by inhibiting or silencing Cyp26b1 but reduced by exogenous Cyp26b1. The BMP9-induced bone formation was enhanced by inhibiting Cyp26b1. The cranial defect repair was promoted by BMP9, which was strengthened by silencing Cyp26b1 and reduced by exogenous Cyp26b1. Mechanically, Cyp26b1 was reduced by BMP9, which was enhanced by activating Wnt/b-catenin, and reduced by inhibiting this pathway. b-catenin interacts with Smad1/5/9, and both were recruited at the promoter of Cyp26b1. CONCLUSIONS: Our findings suggested the BMP9-induced osteoblastic differentiation was mediated by activating retinoic acid signalling, viadown-regulating Cyp26b1. Meanwhile, Cyp26b1 may be a novel potential therapeutic target for the treatment of bone-related diseases or accelerating bone-tissue engineering.

        Effects of Xylanase on Performance, Blood Parameters, Intestinal Morphology, Microflora and Digestive Enzyme Activities of Broilers Fed Wheat-based Diets

        The study was conducted to investigate the effects of different levels of xylanase on performance, blood parameters, intestinal morphology, microflora and digestive enzyme activities of broilers. The wheat-based diets were supplemented with 0, 500, 1,000, 5,000 U/kg xylanase. Xylanase supplementation significantly (p<0.05) improved the feed:gain ratio of broilers from 1 to 21 d and 1 to 42 d. Supplementing 500 U/kg and 1,000 U/kg xylanase improved (p<0.05) the villus height and the ratio of villus height to crypt depth in the small intestine. Excess supplementation of xylanase (5,000 U/kg) increased the villus height in the ileum (p<0.01) and the ratio of villus height to crypt depth in the duodenum and ileum (p<0.05). The microflora in the ileum and caecum, digestive enzyme activities in the small intestine and the concentrations of serum glucose, uric acid, insulin and IGF-I were not affected by the supplementation of xylanase. Excess level of xylanase (5,000 U/kg) had a tendency to induce the multiplication of E. coli and total aerobes. The results suggested that supplementing 500 U/kg and 1,000 U/kg xylanase was beneficial for broilers and excess xylanase supplementation resulted in no further improvement or negative effects.

      • A Busbar Integrated SiC-based Converter with Embedded Heat-pipes

        In this paper, a bus-bar integrated packaging technology for press-pack power modules is proposed. The proposed packaging technology features high power-density, even pressure distribution among chips, and low stray parameters. The structure design and fabrication flow process are analyzed and experiments are made to verify the feasibility of proposed solution. The critical challenge about the package is related to thermal management. To reduce the thermal stress of chips, a heat-pipe integrated heatsink is developed and finite-element-method (FEM) simulations are used to evaluate the cooling performance of the proposed heatsink. The results demonstrate that the proposed heatsink has a relatively low cost and works effectively. It paves the way to better packaging design and more efficient thermal management method for high power devices.

        Effects of the costoclavicular block versus interscalene block in patients undergoing arthroscopic shoulder surgery under monitored anesthesia care: a randomized, prospective, non-inferiority study

        Background: Recent studies have reported that costoclavicular blocks (CCBs) can consistently block almost all branches of the brachial plexus while sparing the phrenic nerve and provide effective analgesia after shoulder surgery. We aimed to compare the efficacy of the CCB with that of the interscalene block (ISB) as the sole blocking technique for shoulder surgery.Methods: A total of 212 patients undergoing elective arthroscopic shoulder surgery were randomized to receive an ISB or CCB based on a non-inferiority design. All patients received titration sedation with propofol under monitored anesthesia during surgery. The primary outcomes were the proportion of patients with complete motor blockade of the suprascapular nerve (SSN) and incidence of hemidiaphragmatic paralysis (HDP). The secondary outcomes included block-related variables, complications, and postoperative pain scores. Results: The proportion of patients with complete motor blockade of the SSN at 20 min between the CCB and ISB groups (53% vs. 66%) exceeded the predefined non-inferiority margin of −5%, but was comparable at 30 min (87% vs. 91%). The CCB resulted in a significantly lower incidence of HDP (7.55% vs. 92.45%), Horner’s syndrome (0% vs. 18.87%), and dyspnea (0% vs. 10.38%) than the ISB. None of the patients experienced failed blocks or required conversion to general anesthesia. Pain scores were comparable between the groups.Conclusions: Ultrasound-guided CCBs may be comparable to ISBs, with fewer unfavorable complications in patients with impaired lung function undergoing arthroscopic shoulder surgery.

        Emerging nanoparticulate drug delivery systems of metformin

        Background Driving the conventional drug in new applications has emerged as a research hotspot for disease treatment. Metformin (MET) is conventionally used for the treatment of type II diabetes. It has also been found to be a versatile molecule with wide biological functions, such as losing weight. anti-aging and anticancer activity. Rational design of nanoparticulate drug delivery systems (nano-DDS) could significantly improve drug delivery efficiency. Recently, a wide range of nano- DDS has been developed to improve the delivery efficacy of MET or to perform as versatile nanoplatforms for efficient drug delivery. Area covered In this review, we outline the emerging trends in advanced nano-DDS of MET, focusing on nano-DDS of MET for diabetes therapy, nano-DDS of MET for anticancer therapeutics, and nano-DDS of MET for other therapeutic aims. Expert opinion Despite the great progression in nano-DDS of MET, there’s still a long way to truly put the conventional drug in new applications. Several important issues should be fully taken into consideration, such the manufacturing cost and economic burdens for patients, the biocompatibility and long-term toxicity of carrier materials, scale-up preparation difficulties, as well as the species gap between human beings and animal models.

        The present study was designed to isolate and characterize novel chemical constituents of the stem bark of Juglans mandshurica Maxim. (Juglandaceae).The chemical constituents were isolated and purified by various chromatographic techniques. The structures of the compounds were elucidated on the basis of spectral data (1D and 2D NMR, HR-ESI-MS, CD, UV, and IR) and by the comparisons of spectroscopic data with the reported values in the literatures. Two long chain polyunsaturated fatty acids (1 and 2) were obtained and identified as (S)-(8E, 10E)-12-hydroxy-7-oxo-8, 10-octadecadienoic acid (1) and (S)-(8E, 10E)-12-hydroxy-7-oxo-8,10-octadecadienoic acid methyl ester (2). To the best of our knowledge, this is the first report on the isolation and structural elucidation of the two new conjugated ketonic fatty acids from this genus.

        First-order Generalized Integrator Based Frequency Locked Loop and Synchronization for Three-Phase Grid-connected Converters under Adverse Grid Conditions

        This paper presents an alternative frequency adaptive grid synchronization technique named HDN-FLL, which can accurately extract the fundamental positive- and negative-sequence components and interested harmonics in adverse three-phase grid voltage. The HDN-FLL is based on the harmonic decoupling network (HDN) consisting of multiple first order complex vector filters (FOCVF) with a frequency-locked loop (FLL), which makes the system frequency adaptive. The stability of the proposed FLL is strictly verified to be global asymptotically stable. In addition, the linearization and parameters tuning of the FLL is also discussed. The structure of the HDN has been widely used as a prefilter in grid synchronization techniques. However, the stability of the general HDN is seldom discussed. In this paper, the transfer function expression of the general HDN is deduced and its stability is verified by the root locus method. To show the advantages of the HDN-FLL, a simulation comparison with other gird synchronization methods is carried out. Experimental results verify the excellent performance of the proposed synchronization method.

        The effect of melatonin on cardio fibrosis in juvenile rats with pressure overload and deregulation of HDACs

        The effect of melatonin on juveniles with cardio fibrosis is poorly understood. We investigated whether HDACs participate in the anti-fibrotic processes regulated by melatonin during hypertrophic remodeling. Abdominal aortic constriction (AAC) was employed in juvenile rats resulting in pressure overload-induced ventricular hypertrophy and melatonin was subsequently decreased via continuous light exposure for 5 weeks after surgery. AAC rats displayed an increased cross-sectional area of myocardial fibers and significantly elevated collagen deposition compared to sham-operated rats, as measured by HE and Masson Trichrome staining. Continuous light exposure following surgery exacerbated the increase in the cross-sectional area of myocardial fibers. The expression of HDAC1, HDAC2, HDAC3, HDAC4 and HDAC6 genes were all significantly enhanced in AAC rats with light exposure relative to the other rats. Moreover, the protein level of TNF- was also upregulated in the AAC light exposure groups when compared with the sham. However, Smad4 protein expression was unchanged in the juveniles’ hearts. In contrast, beginning 5 weeks after the operation, the AAC rats were treated with melatonin (10 mg/kg, intraperitoneal injection every evening) or vehicle 4 weeks, and sham rats were given vehicle. The changes in the histological measures of cardio fibrosis and the gene expressions of HDAC1, HDAC2, HDAC3, HDAC4 and HDAC6 were attenuated by melatonin administration. The results reveal that melatonin plays a role in the development of cardio fibrosis and the expression of HDAC1, HDAC2, HDAC3, HDAC4 and HDAC6 in cardiomyocytes.

        Antioxidant enzyme activity and microRNA are associated with growth of Poa pratensis callus under salt stress

        Kentucky bluegrass (Poa pratensis L.) is an important species of turfgrass that is commonly planted on golf courses and landscapes all over the world. It is sensitive to salt stress; however, details relating to its molecular mechanisms of salt resistance are not available. We, therefore, analyzed the changes in growth, antioxidant enzyme activity, and microRNA expression in the callus 1 week after treatment with 200 mM NaCl for 12 h, 24 h, 48 h, 96 h, and 144 h. The results demonstrated that callus growth declined and thiobarbituric acid-reactive substance production and cell membrane permeability increased. Treatment with salt increased ascorbate peroxidase, glutathione reductase, superoxide dismutase, catalase, peroxidase, and polyphenol oxidase activity. Changes in the expression levels of microRNAs were observed under salt treatment. The expression of miR162, miR173, miR391, miR408, miR773, and miR857 increased by 70% after 24 h of salt treatment, after which it declined to a level similar to that of the control. The expression level of miR775 and miR827 decreased by 20% after 24 h, and then further decreased by 80% after 144 h. The expression level of miR841 increased by 50% after 24 h of salt treatment, and then stabilized. In contrast, salt treatment increased the expression of the auxin response factors ARF6, ARF8, ARF10, and ARF16 in the callus from 12 to 144 h of salt treatment, during which the expression increased twofold. Gene expression analysis indicated that salt-responsive gene families were regulated by microRNAs in the callus under salinity stress. The activity of antioxidant enzymes is also changing. MiR841 is considered to be a positive regulator of antioxidant enzyme biosynthesis. The present investigation elucidates the manner in which P. pratensis responds to salt stress in the callus, and could be used to inform further studies on the molecular mechanisms of stress tolerance.
