        Attenuation of Experimental Autoimmune Hepatitis in Mice with Bone Mesenchymal Stem Cell-Derived Exosomes Carrying MicroRNA-223-3p

        MicroRNA-223-3p (miR-223-3p) is one of the potential microRNAs that have been shown to alleviate inflammatory responses in pre-clinical investigations and is highly encased in exosomes derived from bone mesenchymal stem cells (MSC-exosomes). MSC-exosomes are able to function as carriers to deliver microRNAs into cells. Autoimmune hepatitis is one of the challenging liver diseases with no effective treatment other than steroid hormones. Here, we examined whether MSC-exosomes can transfer miR-223-3p to treat autoimmune hepatitis in an experimental model. We found that MSC-exosomes were successfully incorporated with miR-223-3p and delivered miR-223-3p into macrophages. Moreover, there was no toxic effect of exosomes on the macrophages. Furthermore, treatments of either exosomes or exosomes with miR-223-3p successfully attenuated inflammatory responses in the liver of autoimmune hepatitis and inflammatory cytokine release in both the liver and macrophages. The mechanism may be related to the regulation of miR-223-3p level and STAT3 expression in the liver and macrophages. These results suggest that MSC-exosomes can be used to deliver miR-223-3p for the treatment of autoimmune hepatitis.

      • Establishment of Paclitaxel-resistant Breast Cancer Cell Line and Nude Mice Models, and Underlying Multidrug Resistance Mechanisms in Vitro and in Vivo

        Background: Breast cancer is a common malignant tumor which affects health of women and multidrug resistance (MDR) is one of the main factors leading to failure of chemotherapy. This study was conducted to establish paclitaxel-resistant breast cancer cell line and nude mice models to explore underlying mechanisms of MDR. Methods: The breast cancer drug-sensitive cell line MCF-7 (MCF-7/S) was exposed in stepwise escalating paclitaxel (TAX) to induce a resistant cell line MCF-7/TAX. Cell sensitivity to drugs and growth curves were measured by MTT assay. Changes of cell morphology and ultrastructure were examined by optical and electron microscopy. The cell cycle distribution was determined by flow cytometry. Furthermore, expression of proteins related to breast cancer occurrence and MDR was tested by immunocytochemistry. In Vivo, nude mice were injected with MCF-7/S and MCF-7/TAX cells and weights and tumor sizes were observed after paclitaxel treatment. In addition, proteins involved breast cancer and MDR were detected by immunohistochemistry. Results: Compared to MCF-7/S, MCF-7/TAX cells had a higher resistance to paclitaxel, cross-resistance and prolonged doubling time. Moreover, MCF-7/TAX showed obvious alterations of ultrastructure. Estrogen receptor (ER) expression was low in drug resistant cells and tumors while expression of human epidermal growth factor receptor 2 (HER2) and Ki-67 was up-regulated. P-glycoprotein (P-gp), lung resistance-related protein (LRP) and glutathione-S-transferase-${\pi}$ (GST-${\pi}$) involved in the MDR phenotype of resistant cells and tumors were all overexpressed. Conclusion: The underlying MDR mechanism of breast cancer may involve increased expression of P-gp, LRP and GST-${\pi}$.

        Adsorptive Granulocyte and Monocyte Apheresis in the Treatment of Ulcerative Colitis: The First Multicenter Study in China

        Background/Aims: Patients with active ulcerative colitis (UC) have elevated levels of activated myeloid-derived leukocytes as a source of inflammatory cytokines. The selective deple-tion of these leukocytes by adsorptive granulocyte/monocyte apheresis (GMA) with an Adacolumn should alleviate inflammation, promote remission and enhance drug efficacy. However, studies have reported contrasting efficacy out-comes based on patients` baseline demographic variables. This study was undertaken to understand the demographic features of GMA responders and nonresponders. Methods: This was a multicenter study in China involving four institutions and 34 patients with active UC. Baseline conventional medications were continued without changing the dosage. The treatment efficacy was evaluated based on the endo-scopic activity index and the Mayo score. Results: Thirty of the 34 patients completed all 10 GMA treatment sessions. The overall efficacy rate was 70.59%. The receiver operating characteristic analysis showed that the area under the curve was approximately 0.766 for a Mayo score of ≤5.5 with 0.273 specificity and 0.857 sensitivity (Youden index, 0.584) for GMA responders. No GMA-related serious adverse events were observed. Conclusions: The overall efficacy of GMA in patients with active UC who were taking first-line medications or were corticosteroid refractory was encouraging. Addition-ally, GMA was well tolerated and had a good safety profile. (Gut Liver 2017;11:216-225)

        The Synthesis and Research of Glass Fiber Felts Coated with Graphene Oxide/Phenolic Resin Binder

        Glass fiber felts (GFFs) coated with reduced graphene oxide (RGO)/phenolic resin (PF) binder was prepared by in-situ reduction reaction, subsequently spraying, and curing process. The effects of the RGO content on the thermal, antiablative, hydrophobic, and mechanical properties of GFFs were studied. Fourier-transform infrared (FT-IR) spectra, X-ray diffraction (XRD) and electronic differential system (EDS) analysis revealed that the graphene oxide (GO) was reduced to RGO by phenol during the in-situ polymerization. Thermogravimetry (TG) and DTG analytical diagram demonstrated that the thermal stability of GFFs was also significantly improved with the addition of GO due to the barrier effect of the lamellar structure. Also, by addition of 0.4 wt% of GO, hydrophobicity was improved by 39 % and the tear strength was improved by 57 % owning to its unique structure and the effective external load transfer. Moreover, the compatibility with PF matrix was improved, endowing good anti-ablative, excellent hydrophobicity and mechanical properties for GFFs.

        Step-size adaptive parametric level set method for structural topology optimization

        In the structural topology optimization (STO), the step-size of the parametric level set method (PLSM) using the explicit scheme must satisfy the Courant-Friedrichs-Lewy (CFL) condition to ensure numerical stability. However, much larger step-sizes are arbitrarily used to speed up the convergence. For this reason, a narrowband in the velocity field is defined, and the step-size adaptive parametric level set method (SAPLSM) is proposed, which multiplies different step-sizes for the velocity of different nodes. The SAPLSM satisfies the CFL condition not only on the narrowband, but also on the entire design domain. Furthermore, a narrowband annealing (NA) scheme based on “annealing” is proposed to dynamically adjust the maximum step-size during the iterations. Numerical experimental results of several benchmark problems in two-dimensional minimum compliance show that: (1) The SAPLSM is more stable than PLSM under large step-sizes and complex problems. (2) The NA scheme not only accelerates the convergence of SAPLSM but also alleviates mesh dependence.

        The effects of LNG-tank sloshing on the global motions of FLNG system

        This paper addresses a study of inner-tank sloshing effect on motion responses of a Floating Liquefied Natural Gas (FLNG) system, through experimental analysis and numerical modeling. To investigate hydrodynamic characteristics of FLNG under the conditions of with and without LNG-tank sloshing, a series of numerical simulations were carried out using potential flow solver SESAM. To validate the numerical simulations, model tests on the FLNG system was conducted in both liquid and solid ballast conditions with 75% tank filling level in height. Good correlations were observed between the measured and predicted results, proving the feasibility of the numerical modeling technique. On the verified numerical model, Response Amplitude Operators (RAOs) of the FLNG with 25% and 50% tank filling levels were calculated in six degrees of freedom. The influence of tank sloshing with varying tank filling levels on the RAOs has been presented and analyzed. The results showed that LNG-tank sloshing has a noticeable impact on the roll motion response of the FLNG and a moderate tank filling level is less helpful in reducing the roll motion response.

        Inhibition of Long Noncoding RNA SNHG15 Ameliorates Hypoxia/Ischemia-Induced Neuronal Damage by Regulating miR-302a-3p/STAT1/NF-κB Axis

        Purpose: Ischemic brain injury results in high mortality and serious neurologic morbidity. Here, we explored the role of SNHG15in modulating neuronal damage and microglial inflammation after ischemia stroke. Materials and Methods: The hypoxia/ischemia models were induced by middle cerebral artery occlusion in mice and oxygenglucosedeprivation/reoxygenation (OGD/R) in vitro. Quantitative real-time PCR (qRT-PCR) and Western blot were conducted todetermine the levels of SNHG15, miR-302a-3p, and STAT1/NF-κB. Moreover, gain- or loss-of functional assays of SNHG15 and miR-302a-3p were conducted. MTT assay was used to evaluate the viability of HT22 cells, and the apoptotic level was determined byflow cytometry. Furthermore, enzyme-linked immunosorbent assay was performed to detect oxidative stress and inflammatorymediators in the ischemia cortex and OGD/R-treated BV2 microglia. Results: The SNHG15 and STAT1/NF-κB pathways were both distinctly up-regulated, while miR-302a-3p was notably down-regulatedin the ischemia cortex. Additionally, overexpressing SNHG15 dramatically enhanced OGD/R-mediated neuronal apoptosisas well as the expression of oxidative stress and inflammation factors from microglia. In contrast, knocking down SNHG15 or overexpressingmiR-302a-3p relieved OGD/R-mediated neuronal apoptosis and microglial activation. Moreover, the rescue experimenttestified that overexpressing miR-302a-3p also attenuated SNHG15 up-regulation-induced effects. In terms of the mechanisms,SNHG15 sponged miR-302a-3p and activated STAT1/NF-κB as a competitive endogenous RNA, while miR-302a-3p targetedSTAT1 and negatively regulated the STAT1/NF-κB pathway. Conclusion: SNHG15 was up-regulated in the hypoxia/ischemia mouse or cell model. The inhibition of SNHG15 ameliorates ischemia/hypoxia-induced neuronal damage and microglial inflammation by regulating the miR-302a-3p/STAT1/NF-κB pathway.

        Optimization for the direction of arrival estimation based on single acoustic pressure gradient vector sensor

        The optimization techniques are explored in the direction of arrival (DOA) estimation based on single acoustic pressure gradient vector sensor (APGVS). By analyzing the working principle and measurement errors of the APGVS, acoustic intensity approaches (AI) and the minimum variance distortionless response beamforming approach based on single APGVS (VMVDR) are deduced. The radius to wavelength ratio of the APGVS must be not bigger than 0.1 in the actual application, otherwise its DOA estimation performance will degrade significantly. To improve the ro-bustness and estimation performance of the DOA estimation approaches based on single APGVS, two modified proce-ssing approaches based on single APGVS are presented. Simulation and lake trial results indicate that the performance of the modified approaches based on single APGVS are better than AI and VMVDR approaches based on single APGVS when the radius to wavelength ratio is not bigger than 0.1, and the two modified DOA estimation methods have excel¬lent estimation performance when the radius to wavelength ratio is bigger than 0.1.

      • Overexpression of Cyclooxygenase-1 Correlates with Poor Prognosis in Renal Cell Carcinoma

        The aim of this study was to evaluate expression of COX-1 in renal cell carcinoma (RCC) and its prognostic value. mRNA of COX-1 was detected in 42 paired RCC and adjacent normal tissues with quantitative realtime polymerase chain reaction (qRT-PCR). Expression of COX-1 was also evaluated in 196 RCC sections and 91 adjacent normal tissues with immunohistochemistry. Statistical analysis was performed to assess COX-1 expression in RCC and its prognostic significance. The results of qRT-PCR showed mRNA levels of COX-1 in RCC tissues to be significantly higher than that in adjacent normal tissues (p < 0.001). Immunohistochemical assays also revealed COX-1 to be overexpressed in RCC tissues (p < 0.001). Statistical analysis demonstrated high expression of COX-1 was correlated with tumour size (p = 0.002), pathological stage (p = 0.003), TNM stage (p = 0.003, 0.007, 0.027, respectively), and tumour recurrence (p < 0.001). Survival analysis indicated patients with high expression of COX-1 had shorter survival time (p < 0.001), and COX-1 was an independent predictor. This is the first study to reveal overexpression of COX-1 in RRC and point to use as a prognostic marker in affected patients.
