        ORiginal Article : Silencing of CXCR4 Inhibits Tumor Cell Proliferation and Neural Invasion in Human Hilar Cholangiocarcinoma

        Background/Aims: To evaluate the expression of CXC motif chemokine receptor 4 (CXCR4) in the tissues of patients with hilar cholangiocarcinoma (hilar-CCA) and to investigate the cell proliferation and frequency of neural invasion (NI) influenced by RNAi-mediated CXCR4 silencing. Methods: An immunohistochemical technique was used to detect the expression of CXCR4 in 41 clinical tissues, including hilar-CCA, cholangitis, and normal bile duct tissues. The effects of small interference RNA (siRNA)-mediated CXCR4 silencing were detected in the hilar-CCA cell line QBC939. Cell proliferation was determined by MTT. Expression of CXCR4 was monitored by quantitative real time polymerase chain reaction and Western blot analysis. The NI ability of hilar-CCA cells was evaluated using a perineural cell and hilar-CCA cell coculture migration assay. Results: The expression of CXCR4 was significantly induced in clinical hilar-CCA tissue. There was a positive correlation between the expression of CXCR4 and lymph node metastasis/NI in hilar-CCA patients (p<0.05). Silencing of CXCR4 in tumor cell lines by siRNA led to significantly decreased NI (p<0.05) and slightly decreased cell proliferation. Conclusions: CXCR4 is likely correlated with clinical recurrence of hilar-CCA. CXCR4 is involved in the invasion and proliferation of human hilar-CCA cell line QBC939, indicating that CXCR4 could be a promising therapeutic target for hilar-CCA. (Gut Liver 2014;8:196-204)

        T-stress solutions for cracks in rectangular plates with multiple holes

        The elastic T-stress is increasingly being recognized as an important second parameter to the stress intensity factor for fracture and fatigue assessments. In this paper, the mutual or M-contour integral approach is employed in conjunction with the Boundary Element Method (BEM) to determine the numerical T-stress solutions for cracks in plates with multiple holes. The problems investigated include plates of infinite width with multiple holes at which single or double, symmetric cracks have grown from. Comparisons of these results are also made with the corresponding solutions of finite plates with a single hole. For completeness, stress intensity factor solutions for the cracked geometries analyzed are presented as well. These results will be useful for failure assessments using the two-parameter linear elastic fracture mechanics approach.

        Solar-powered electrocoagulation treatment of wet flue gas desulfurization wastewater using dimensionally stable anode and induced electrode

        Solar-powered electrocoagulation (EC) process is proven to be an alternative option for wet flue gas desulfurization (WFGD) wastewater treatment, due to simultaneous removal of multiple pollutants efficiently and reduce the operation costs significantly. Rapid and stable photoelectricity response is necessary for the removal efficiency of pollutants (eg. COD and turbudity), especially in low solar irradiation intensity. In this paper, dimensionally stable anode (DSA) and operation voltages in EC process driven by solar cells were investigated, for the purpose of the optimized removal of pollutants, including COD, turbidity, (free residual chlorine and chlorine dioxide). The results show that the removal efficiency of COD and turbidity can respectively reach 59.12% and 39.11% within 30 min of illumination time (the enhanced solar radiation = 915.8 W m<SUP>-2</SUP>), when Ti-based plates were adopted. The concentration of free residual chlorine and chlorine dioxide can reach 2.70 and 5.31 mg L<SUP>-1</SUP>, indicating that chlorine ions present in WFGD wastewater have been converted into active chlorine partly. Solar-powered EC equipped with Ti-based plates have a potential prospect in EC process for WFGD wastewater treatment.

        High-frame-rate Video Denoising for Ultra-low Illumination

        In this study, we present a denoising algorithm for high-frame-rate videos in an ultra-low illumination environment on the basis of Kalman filtering model and a new motion segmentation scheme. The Kalman filter removes temporal noise from signals by propagating error covariance statistics. Regarded as the process noise for imaging, motion is important in Kalman filtering. We propose a new motion estimation scheme that is suitable for serious noise. This scheme employs the small motion vector characteristic of high-frame-rate videos. Small changing patches are intentionally neglected because distinguishing details from large-scale noise is difficult and unimportant. Finally, a spatial bilateral filter is used to improve denoising capability in the motion area. Experiments are performed on videos with both synthetic and real noises. Results show that the proposed algorithm outperforms other state-of-the-art methods in both peak signal-to-noise ratio objective evaluation and visual quality.

        A Kalman Filter based Video Denoising Method Using Intensity and Structure Tensor

        We propose a video denoising method based on Kalman filter to reduce the noise in video sequences. Firstly, with the strong spatiotemporal correlations of neighboring frames, motion estimation is performed on video frames consisting of previous denoised frames and current noisy frame based on intensity and structure tensor. The current noisy frame is processed in temporal domain by using motion estimation result as the parameter in the Kalman filter, while it is also processed in spatial domain using the Wiener filter. Finally, by weighting the denoised frames from the Kalman and the Wiener filtering, a satisfactory result can be obtained. Experimental results show that the performance of our proposed method is competitive when compared with state-of-the-art video denoising algorithms based on both peak signal-to-noise-ratio and structural similarity evaluations.

        Evolution of Bond Distortion in $C_{60}$ by an Electron Uptake

        When an electron is transferred to $C_{60}$, the bond structure is distorted due to the electron-lattice interaction and a polaron-like state is formed. The evolution process of the bond distortion is studied by the dynamical equation of atomic lattice, and time-dependent changes of the bond lengths are determined. Then it can be estimated that the relaxation time to form the polaron-like state is a fraction of a picosecond.

        A heat shock protein 70 protects the green peach aphid (Myzus persicae) against high-temperature stress

        Heat shock proteins (HSPs) are encoded by Hsp genes and are important in insect tolerance to heat stress. The green peach aphid, Myzus persicae, is an important agricultural pest. The functions of Hsp genes in the thermal tolerance of M. persicae are unknown. This study identified an Hsp70 gene (MpHsp70a) and analyzed its role in protection against high-temperature stress. MpHsp70a encoded a protein consisting of 659 amino acid residues. The protein had three signature motifs of the HSP70 family and was predicted to be localized in the cytoplasm. The highest expression level of MpHsp70a was in adults, and differences in the mRNA levels between apterous and alate adults were not significant. Exposure to high temperatures (30, 35 and 40 ◦ C) for one hour and treatment with 40 ◦ C for different times (0.5, 1 and 2 h) all resulted in a greatly elevated expression level of MpHsp70a, suggesting that the gene is heat-inducible. The transcriptional level of MpHsp70a was suppressed by injection with double-stranded RNA (dsRNA), and knockdown of MpHsp70a significantly increased the suscep tibility of apterous adults to 40 ◦ C. These results indicate that MpHsp70a is required for tolerance to hightemperature stress in M. persicae. Our findings highlight the molecular mechanism underlying Hsp70-mediated thermal adaptation in M. persicae.

        Realgar transforming solution-induced differentiation of NB4 cell by the degradation of PML/RARa partially through the ubiquitin–proteasome pathway

        PML/retinoic acid receptor alpha (RARa), as ahallmark of acute promyeloid leukemia (APL), is directlyrelated to the outcome of clinical APL remedy. It isreported that arsenicals can effectively degrade PML/RARa, such as arsenic trioxide and realgar. However, thehigh toxicity or insolubility have hampered their clinicalapplications. Realgar transforming solution (RTS) wasproduced from realgar by bioleaching process in our lab. Previous studies demonstrated that RTS had a significantanti-cancer ability on chronic myeloid leukemia throughoncoprotein degradation. The capacity of RTS on treatingAPL is what is focused on in this study. The results showedthat RTS had a noticeable sensitivity in NB4 cell, and RTSremarkably down-regulated PML/RARa expression andinduced cell differentiation. Further, RTS could accumulatePML/RARa into the nuclear bodies and then executedegradation, which could be reversed by proteasomeinhibitor MG132. The results also exhibited that thereduction of RTS-induced PML/RARa expression accompaniedby the elevation of ubiquitin and SUMO-1 proteinexpression. Finally, PML and SUMO-1 had been demonstratedto be co-localized after RTS treatment byimmunofluorescence co-localization assay and immunoprecipitationassay. In conclusion, these results suggestedthat RTS-induced cell differentiation may attribute to thePML/RARa degradation partially through the ubiquitin–proteasome pathway.

        Translocation and Phosphorylation of Calcyclin Binding Protein during Retinoic Acid-induced Neuronal Differentiation of Neuroblastoma SH-SY5Y Cells

        For better understanding of functions of the Calcyclin Binding Protein (CacyBP) and exploring its possible roles in neuronal differentiation, the subcellular localization of human CacyBP was examined in retinoic acid(RA)-induced and uninduced neuroblastoma SH-SY5Y cells. Immunostaining indicated that CacyBP was present in the cytoplasm of uninduced SH-SY5Y cells, in which the resting Ca^(2+) concentration was relatively lower than that of RA-induced cells. After the RA induction, immunostaining was seen in both the nucleus and cytoplasm. In the RA-induced differentiated SH-SYSY cells, CacyBP was phosphorylated on serine residue(s), while it existed in a dephosphorylated form in normal (uninduced) cells. Thus, the phosphorylation of CacyBP occurs when it is translocated to the nuclear region. The translocation of CacyBP during the RA-induced differentiation of SH-SY5Y cells suggested that this protein might play a role in neuronal differentiation.

      • Intervention Effects of Nedaplatin and Cisplatin on Proliferation and Apoptosis of Human Tumour Cells in Vitro

        Objective: To study synergistic effects of nedaplatin and cisplatin on three human carcinoma cell lines (esophageal carcinoma cell line Eca-109, ovarian carcinoma Skov-3 and cervical carcinoma Hela). Methods: Inhibition effects were evaluated by MTT assay and cell apoptosis was detected by flow cytometry. In addition, changes of Ki-67, Bax and Bcl-2 at mRNA and protein levels were quantified by RT-PCR and Western blotting. Results: Growth inhibition in each cell lines was dose-dependent after exposure to nedaplatin or cisplatin alone. The interaction of the two drugs was synergistic at higher concentrations according to the median-effect principle. The inhibition rates with nedaplatin, cisplatin and combined treatment were $41.9{\pm}4.1%$, $47.4{\pm}2.9%$, $52.5{\pm}0.9%$(Eca-109), $39.0{\pm}1.26%$, $45.0{\pm}1.45%$, $56.2{\pm}1.44%$ (Skov-3) and $44.8{\pm}2.11%$, $46.9{\pm}0.99%$, $56.6{\pm}1.83%$ (Hela) respectively, with increase in apoptosis. Compared with the nedaplatin or cisplatin alone treatment group, the combinative treatment group's Ki-67 and bcl-2 mRNA (protein) expression was decreased while that of Bax mRNA (protein) was increased. Conclusion: Compared to the effects of nedaplatin or cisplatin alone at high concentrations, combination of nedaplatin and cisplatin at low concentrations proved to be much more effective for inhibition of proliferation and the induction of apoptosis in the Eca-109, Skov-3 and Hela cell lines.
