A novel esterase gene, est01, was successfully unearthed from a biogas digester microbiota metagenomic library. The 1,194 bp est01 gene encodes a protein of 44,804 Da (designated Est01). The amino acid sequence of Est01 shows only moderate (33%) identity to a lipase/ esterase. Phylogenetic analysis and biochemical characterization confirmed that Est01 is a new member of family VIII esterases. The purified Est01 from recombinant Escherichia coli BL21 (DE3) showed high hydrolytic activity against short-chain fatty acid esters, suggesting that it is a typical carboxylesterase rather than a lipase. Furthermore, the Est01 was even active at 10°C (43% activity remained), with the optimal temperature at 20°C, and had a broad pH range from 5.0 to 10.0, with the optimal pH of 8.0. These properties suggest that Est01 is a cold-adaptive esterase and could have good potential for low-temperature hydrolysis application.
Taguchi Approach for Anti-heat Stress Prescription Compatibility in Mice Spleen Lymphocytes In Vitro

Heat stress (HS) may induce immunosuppression as well as inhibit the proliferation of lymphocytes. This study evaluated the effects on immune function of our prescription on splenic lymphocytes under HS as well as its compatibility. The effects of four herbal extracts from Agastache rugosa, Atractylodes lancea, Cortex Phellodendri, and Gypsum Fibrosum on heat treated splenic lymphocytes were investigated and the compatibility of the prescription was also explored by using the Taguchi method. This study revealed changes in proliferation by traditional Chinese medicines of splenic lymphocytes after HS. Proliferation in the HS group was significantly lower than the control group. Under HS, the effects of higher concentrations of Agastache rugosa (100 and 200 μg/mL), Atractylodes lancea (100 and 200 μg/mL), Cortex Phellodendri (50 and 100 μg/mL) and Gypsum Fibrosum (100 and 200 μg/mL) caused a significant increase on ConA/LPS-induced proliferation of lymphocytes than lower concentrations. We, therefore, conclude that the prescription of traditional Chinese medicines may recover splenic lymphocytes from the immunosuppression induced by HS. The Taguchi design, which allows rapid and high efficiency for the selection of the best conditions for our prescription on HS-treated splenic lymphocytes, demonstrated that Agastache rugosa (200 μg/mL), Atractylodes lancea (200 μg/mL), Cortex Phellodendri (100 μg/mL) and Gypsum Fibrosum (100 μg/mL)were the optimal conditions for the prescription. The validation experiment confirmed that our composition in optimum extraction conditions enhanced effects on ConA or LPS-stimulated lymphocytes under HS. The results showed that the Taguchi optimization approach is a suitable method for optimization of the composition of prescription.
Pipeline transport of heavy crudes as stable foamy oil

A new idea was proposed to transport heavy crudes as foamy oil at normal temperature. The effects offoaming agent type and concentration, foam stabilizer type and concentration, oil–water volume ratioand temperature on the formation and stability of foamy oil were evaluated. The foamy oil properties anddrag characteristics of foamy oilflow in small diameter pipes were investigated. The results indicate thatthe prepared stable foamy oil could be characterized as non-Newtonian power lawfluid. The predictedpressure drops were in good agreement with the measured ones. The significant dynamic viscosityreduction rates were obtained.

The new Modified-690Gd alloy, namely as Nie30Cr-(10-x) Fe-xGd (x ¼ 0.5, 1.0, 1.5,2.0, 3.0 wt%) for function structure integrated thermal neutron shielding has been prepared and characterized. The Modified-690Gd alloy was mainly composed of g austenite matrix and (Ni, Cr, Fe)5Gd precipitated along grain boundaries. The new Modified-690Gd alloy had great mechanical properties, which had the tensile strength exceeding 620 MPa and the elongation being above 50%. Meanwhile, this alloy had excellent weldability and good corrosion resistance in boric acid. The new Modified-690Gd alloy is expected to be a kind of high efficiency thermal neutron shielding materials

In high power laser facility, irreversible damage on fused silica optics, induced by laser irradiation or processing, seriously affects the service life of optics. Therefore, the work of inhibiting damage growth has been carried out in various countries. In our work, an integrated multi-station system is designed to detect and mitigate surface damage on fused silica. The process of processing fused silica optics include UV laser conditioning, surface damage detection and surface damage mitigation with CO2 laser. UV laser conditioning pre-initiates surface damage on fused silica optics with the laser flux less than Laser-Induced Damage Threshold (LIDT). Images of surface damage acquired from camera are processed by improved global threshold segmentation algorithm to extract damage information. Finally, CO2 laser is applied to process the damage with specific morphology to enhance the laser damage resistance. This integrated multi-station system saves the repeated optics installation time between the workstations with the positioning accuracy of 20 μm. Furthermore, the damage with diameter of 10 μm is mitigated to prolong service life of processed fused silica optics. The efficient and accurate integrated multi-station system is of great significance for off -line detecting and mitigating surface damage of fused silica optics in high power laser facility.

Dysregulation of wild-type p53 turnover is a key cause of hepatocellular carcinoma (HCC), yet its mechanism remains poorly understood. Here, we report that WD repeat and SOCS box containing protein 2 (WSB2), an E3 ubiquitin ligase, is an independent adverse prognostic factor in HCC patients. WSB2 drives HCC tumorigenesis and lung metastasis in vitro and in vivo. Mechanistically, WSB2 is a new p53 destabilizer that promotes K48-linked p53 polyubiquitination at the Lys291 and Lys292 sites in HCC cells, leading to p53 proteasomal degradation. Degradation of p53 causes IGFBP3-dependent AKT/mTOR signaling activation. Furthermore, WSB2 was found to bind to the p53 tetramerization domain via its SOCS box domain. Targeting mTOR with everolimus, an oral drug, significantly blocked WSB2-triggered HCC tumorigenesis and metastasis in vivo. In clinical samples, high expression of WSB2 was associated with low wild-type p53 expression and high p-mTOR expression. These findings demonstrate that WSB2 is overexpressed and degrades wild-type p53 and then activates the IGFBP3-AKT/mTOR axis, leading to HCC tumorigenesis and lung metastasis, which indicates that targeting mTOR could be a new therapeutic strategy for HCC patients with high WSB2 expression and wild-type p53.

An optimized space vector pulse width modulation (SVPWM) method with common mode voltage elimination and neutral point potential balancing is proposed for an open-end winding induction motor. The motor is fed from both of the ends with two neutral point clamped (NPC) three-level inverters. In order to eliminate the common mode voltage of the motor ends and balance the neutral point potential of the DC link, only zero common mode voltage vectors are used and a balancing control factor is gained from calculation in the strategy. In order to improve the harmonic characteristics of the output voltages and currents, the balancing control factor is regulated properly and the theoretical analysis is provided. Simulation and experimental results show that by adopting the proposed method, the common mode voltage can be completely eliminated, the neutral point potential can be accurately balanced and the harmonic performance for the output voltages and currents can be effectively improved.

The development of the gut is controlled and modulated by different interacting mechanisms, such as genetic endowment, intrinsic biological regulatory functions, environment influences and last but no least, the diet influence. In this work, we compared the fecal microbiota of breast-fed (BF), formula-fed (FF), and mixed-fed (MF) infants from Hebei Province, China. By using high-throughput 16S rDNA sequencing analyses, we found some differences in gut microbiota in the three groups. Firmicutes and Proteobacteria were the dominant bacteria at the phylum level in the three groups, where FF infants showed a significant depletion in Bacteroidetes (p < 0.001) and Actinobacteria (p < 0.05). Enterobacteriaceae was the dominant bacteria at the family level in the three groups, but FF infants showed higher Enterobacteriaceae enrichment than BF and MF infants (p < 0.05); the abundance of the Bifidobacteriaceae was only 8.16% in the feces of BF infants, but higher than in MF and FF infants (p < 0.05). The number of genera detected (abundance >0.01%) in BF, MF, and FF infants was only 15, 16, and 13, respectively. This study could provide more accurate and scientific data for the future study of infant intestinal flora.

A Collaborative Optimization (CO) methodology for ring-stiffened composite material pressure hull of underwater vehicle is proposed. Structural stability and material strength are both examined. Lamination parameters of laminated plates are introduced to improve the optimization efficiency. Approximation models are established based on the Ellipsoidal Basis Function (EBF) neural network to replace the finite element analysis in layout optimizers. On the basis of a two-level optimization, the simultaneous structure material collaborative optimization for the pressure vessel is implemented. The optimal configuration of metal liner and frames and composite material is obtained with the comprehensive consideration of structure and material performances. The weight of the composite pressure hull decreases by 30.3% after optimization and the validation is carried out. Collaborative optimization based on the lamination parameters can optimize the composite pressure hull effectively, as well as provide a solution for low efficiency and non-convergence of direct optimization with design variables.
