Gastric sarcoidosis is a rare disease accounting for 0.1-0.9 % of all sarcoidosis. We report our experience with a patient with lung and stomach sarcoidosis accompanied by an enlarged abdominal lymph nodes. A sixty-year-old female patient visited our hospital with an abnormal chest x-ray. The patient has no smoking history and no noticeable medical history. There were no fever, cough, dyspnea, nor weight changes. Physical examination showed clear breathing sound and no palpable lymph node. The chest x-ray showed reticular opacity in the right lower lung zone and chest computed tomography indicated that it was an interstitial thickening in the right middle and right lower lobes. A soft tissue density nodule, 9 mm in diameter, neighboring inferior pulmonary vein was also found. Furthermore, multiple enlarged lymph nodes were observed in the left gastric vessel area, which could be suspected as metastatic lymphadenopathy of intra-abdominal malignancy. Abdominal computed tomography showed that the enlarged lymph node spanned over left gastric area, splenic artery area, greater omentum, and gastro-colic trunk, with no mass or thickening in the walls of the stomach. Esophagogastroduodenoscopy revealed a satellite-shaped shallow whitish discoloration accompanied by mucosal irregularity in the anterior wall side of lower body and mucosal depression in the greater curvature side of lower body. Biopsies on both lesions showed chronic active gastritis with foreign-body type granuloma and Sydney grade 1 Helicobacter pylori. Laparoscopic biopsy on intra-abdominal lymph node was conducted. It was negative in mycobacterium tuberculosis and non-tuberculous mycobacterium, confi rming multiple well-formed non-caseating granulomas, satisfying our diagnosis as sarcoidosis. The patient was prescribes with methylprednisolone and follow-up CT 6 months after treatment indicated signifi cant improvement.

<P><B>Abstract</B></P> <P>Liquid phase deposition (LPD), which is a method to directly form a titanium dioxide (TiO<SUB>2</SUB>) film on a substrate, is the most practical method for applying TiO<SUB>2</SUB> films to medical devices because it is performed at lower temperatures than other methods. The TiO<SUB>2</SUB> films to be applied to medical devices should offer excellent antibacterial effect, but should be stable to normal cells and have appropriate strength. In this research, we observed that the size, shape, and density of TiO<SUB>2</SUB> particles varied with the nucleation duration in LPD and confirmed that these results caused changes in several properties including the mechanical properties, cytotoxicity and antibacterial effect of TiO<SUB>2</SUB> films. From the analysis of these results, we established the conditions for the preparation of TiO<SUB>2</SUB> films that are suitable for medical devices and suggest a new approach to the study of TiO<SUB>2</SUB> films prepared by LPD.</P> <P><B>Highlights</B></P> <P> <UL> <LI> In the LPD method, the growth behavior of TiO<SUB>2</SUB> particles with the continuous nucleation was investigated. </LI> <LI> The mechanical properties and biocidal effect of TiO<SUB>2</SUB> film increased with the continuous nucleation. </LI> <LI> The conditions for the preparation of TiO<SUB>2</SUB> films suitable for medical devices were established. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

Background : Platycodon grandiflorum is a perennial plant and a member of Campanulaceae family. Since ancient times, they have been using P. grandiflorum as an important medicinal plant in Korea. Platycodin D is the most abundant saponin derived from P. grandiflorum and pharmacologically active component. Cytochrome P450s (CYPs) are important enzymes in the saponin biosynthesis. CYP is, in general, the terminal oxidase enzymes and essential roles in saponin biosynthesis pathway by hydroxylation or oxidaition of triterpene skeletons. Methods and Results : We tried to identify CYP genes related to saponin biosynthesis of P. grandiflorum through RNA-seq analysis. The sequencing was performed using Illumina Hi-Seq platform after cDNA library preparation. The produced reads were assembled using CLC Genomics Workbench software (CLC Bio, Inc.). We obtained 122,663 contigs and found 191 putative CYP genes. Familes of CYP716, CYP708, CYP93 and CYP51 were selected as putative saponin biosynthesis related gene families using phylogenetic relationship analysis. Conclusion : The results in this study could help to find the CYPs related to saponin biosynthesis pathway of P. grandiflorum.

UGT is a glycosyltransferase and act on the final step of the secondary metabolite biosynthesis. Methods and Results : We tried to identify UGT genes related to saponin biosynthesis of P. grandiflorum through RNA-seq analysis. The sequencing was performed using Illumina Hi-Seq platform after cDNA library preparation. The produced reads were assembled using CLC Genomics Workbench software (CLC Bio, Inc.). We obtained 122,663 contigs and found 137 putative UGT genes. Familes of UGT71, UGT73, and UGT74 were selected as putative saponin biosynthesis related gene families using phylogenetic relationship analysis. qPCR condition about UGT73 is preheating 94℃ 180 sec, denaturation 94℃ 60 sec, annealing 53℃ 60 sec, extension 72℃ 90 sec, final extension 72℃ 600 sec, 45 cycles repeated. Conclusion : The results in this study could help to find the UGTs related to saponin biosynthesis pathway of P. grandiflorum.
