        The Effects of Waste Leachate on the Eco-Physiological Characteristics of Populus euramericana

        Populus euramericana has been identified as a possible species for use for phytoremediation of landfills. To identify the effects of waste leachate on the growth and physiological characteristics of Populus euramericana. four different treatments were applied to Populus euramericana seedlings: leachate solution (100% leachate). 25% dilution (75% leachate: 25% water. v/v), 50% dilution (50% leachate: 50% water. v/v) and control (100% tab water) were applied to Populus euramericana. Treatment with waste leachate significantly stimulated Populus euramericana height. diameter at root collar and biomass production relative to the water control. Chlorophyll contents. photosynthesis and transpiration of leachate irrigated-trees were significantly higher than those of water control. These results suggested that poplar could be a suitable species for phytoremediation in landfills because these species showed good growth performance and were capable of taking up waste leachate.

        Hydroxycinnamoyl-coenzyme A shikimate/quinate hydroxycinnamoyltransferase (HCT), PeHCT was cloned from Populus euramericana. The predicted amino acid sequence revealed that PeHCT contained a conserved HXXXDG motif that was found in acyltransferase. The in vitro substrate was determined using recombinant PeHCT. The recombinant PeHCT showed substrate-specificity towards shikimic acid. In addition, PeHCT displayed higher expression in the stem than in the leaf, and its expression increased during the growth stage of the leaf. Results indicate PeHCT could be involved in lignin biosynthesis by formation of p-coumaroyl shikimate.

        돈분 침출수를 처리할 때 포플러의 수액이동

        We examined sapflow of two populus tree species (Populus alba x Populus glandulosa and Populus euramericana) grown under livestock waste leachate treatment, and measured relationship between sapflow and. Sapflow of P. euramericana was 1.5 times greater than that of P. alba x P. glandulosa from September to late December. However, sapflow of P. euramericana and P. alba x P. glandulosa was similar after late January. The relationship between the mean temperature and sapflow was relatively high. There was a decrease in sapflow tendency of the two poplar species during the late October when the mean temperature was low. Trends on relationship between the relative humidity and sapflow showed low relationship. However, data showed weak relationship between sapflow and precipitation. During the period of less precipitation, sapflow of P. alba x P. glandulosa has no relationship. Sapflow of two populus species was still active during non-growing season. P. euramericana was considered as a better species than P. alba x P. glandulosa, in terms of phytoremediation. 이태리포플러와 현사시를 대상으로 돈분 침출수를 처리하고 수액이동량을 측정, 기상자료와 조사, 분석하였다. 9월부터 12월 말까지 이태리포플러가 현사시 보다 평균 1.5배 많은 양의 수액이동을 하였다. 그러나 1월 이후 두 수종의 수액이동의 차이가 점점 감소하다가 1월 말 이후에는 거의 비슷한 경향을 보였다. 수액이동은 기온이 높으면 증가하는 경향을 보였으며 두 포플러 클론 모두 평균 기온이 급격히 낮아지는 10월 말부터 급격히 수액이동이 감소하였다. 강수량과 수액이동의 관계는 휴면기동안 강수변화가 적어서 뚜렷한 경향을 찾기 어려웠다. 일반적으로 휴면기로 들어가는 9월 이후에도 두 수종 모두 수액이동이 여전히 이루어졌다. 수종 간에는 이태리포플러가 수액이동량이 현사시보다 평균적으로 높았지만 급격히 감소되는 온도에는 영향을 더 받는 것으로 보아 현사시보다 기온에 민감한 것으로 간주된다. 휴면기동안 두 수종의 수액이동량은 각각 이태리포플러가 2,288 L, 현사시가 1,501 L였다. 이로 미루어 축산폐수 제거 목적의 포플러가 식재 될 때 이태리포플러가 더 적합할 것으로 판단된다.

        생활(生活)쓰레기 매립지(埋立地) 침출수(浸出水)가 이태리 포플러와 자작나무 묘목(苗木)에 미치는 영향(影響)(II)

        이 연구는 대표적인 속성수인 이태리 포플러와 자작나무가 쓰레기 매립지주변의 조림 수종으로서 적당한가를 평가하기 위해서 수행되었다. 두 수종 묘목에 대한 침출수 관수 효과를 알아보기 위해서 침출수 원액, 50% 침출수, 25% 침출수 그리고 대조구(수도물) 등 4가지 침출수로 다르게 희석하여 약 3개월 정도 관수했다. 이태리 포플러와 자작나무 공통적으로 묘고, 근원경, 물질생산량 모두 침출수 원액을 관수한 처리구가 다른 처리구보다 좋았다. 침출수 원액으로 관수한 묘목의 엽록소 지수, 광합성능력, 증산량이 수도물로 관수한 대조구보다 모두 유의하게 높음을 보여주었다. 이는 비록 3개월이라는 짧은 기간의 결과이긴 하지만, 이들 두 수종은 매립지 주변의 조림, 복구를 위해서 좋은 추천 수종임을 보여주고 있다. Populus euramericana and Betula platyphylla var. japonica have been identified as possible species for use for phytoremediation of landfills. To identify the effects of waste leachate on growth and physiological characteristics in Populus euramericana and Betula platyphylla var. japonica, four different treatments were applied to seedlings for 3 months: leachate solution(100% leachate), 50% dilution(50% leachate: 50% water, v/v), 75% dilution(25% leachate: 75% water, v/v) and control(tab water) were applied to these two species. Treatment with waste leachate significantly stimulated both Populus euramericana and Betula platyphylla var. japonica height, diameter at root collar and biomass production relative to the water control. Chlorophyll contents, photosynthesis and transpiration of leachate irrigated-trees were significantly higher than those of water control. These results suggested that these two species could be suitable species for phytoremediation in landfills because these species showed good growth performance and were capable of taking up waste leachate.

        Effect of Steam Explosion Pretreatment on Bioethanol Production from Populus euramericana as Alternative Feedstock

        Populus euramericana is emerging as a viable feedstock for producing bioethanol from renewable resources. Steam explosion pretreatment of P. euramericana can solubilize a significant portion of the hemicellulosic component and enhance the glucose conversion of the remaining cellulose for fermentation into ethanol. In this study, steam explosion condition of P. euramericana is performed in a steam explosion reactor at severity log Ro 4.02 and severity log Ro 4.37. Glucose conversion varied from 72.3% to 80.1% of steam exploded P. euramericana at severity log Ro 4.02 and severity log Ro 4.37. Ethanol yields(%) based on sugar content after enzymatic hydrolysis after 48 h fermentation ranged from 87.0% to 88.4%. As a result, from 100 g of raw material, 14.0 g of ethanol are recovered of 47.3 g available cellulose content. This research of steam explosion pretreatment was a promising method to improve glucose conversion and ethanol yield for bioethanol production.

        새만금간척지에 식재한 포플러 클론의 생리적 특성

        본 연구에서 일반 토양에 비해 무기양분 함량이 적고 배수성이 좋지 않은 새만금간척지 토양에 식재 된 포플러수종의 생리적 특성을 조사하였다. 7월과 8월에, 이태리포플러 클론인 Venziano, L.Avanzo와 미루나무 교잡종 클론인97-18, 97-19은 생리적인 특성의 차이를 보였다. 이태리포플러 클론이 미루나무 교잡종 클론에 비해 생리적 특성이 우수하였다. 특히 Venziano 클론의 순광합성률은 각각 평균적으로 18.62 μmol CO<sub>2</sub> m<sup>-2</sup> · s<sup>-1</sup>, 17.11μmol CO<sub>2</sub> m<sup>-2</sup> · s<sup>-1</sup>로 다른 클론들과 비교해서 높은 수치를 보였다. 반면, 미루나무 교잡종인 97-18클론의 7월 광색소 함량은 2.37 mg · g<sup>-1</sup>FW으로 가장 높은 수치를 보였으나, 8월에는 이태리포플러 클론보다 낮은 함량을 보였다. 미루나무 교잡종은 8월에 순광합성률, 광색소함량 등 전반적인 생리적 활성이 떨어졌다. 결론적으로, 새만금간척지에 식재된 포플러 묘목의 생리적인 특성은 이태리포플러 종인 Venziano 클론이 우수하였다. This research aims to study the physiological characteristics of poplar clones in Saemanguem reclaimed land where has lower minerals and bad drainage than those in paddy field. In July and August, different poplar clones including Populus euramericana clone (Venziano and L.Avanzo) and deltoides hybrid clone (97-18 and 97-19) showed different physiological characteristics. Physiological characteristics of P. euramericana clones were better than those of P. deltoides hybrid clones. Especially, the average net photosynthetic rate (A<sub>net</sub>) of clone Venziano were 18.62 μmol CO<sub>2</sub> m<sup>-2</sup> · s<sup>-1</sup> and 17.11μmol CO<sub>2</sub> m<sup>-2</sup> · s<sup>-1</sup>, respectively. Those were the highest numerical values among other poplar clones. On the other, the total chlorophyll and carotenoid contents of P. deltoides hybrid clones were higher than those of P. euramericana clones in July. Particularly, the total chlorophyll content of clone 97-18 was 2.37 mg · g<sup>-1</sup>FW. In August, those of P. deltoides hybrid clones decreased than in July. In term of the physiological activity such as net photosynthetic rate and stomatal conductance of P. deltoides hybrid clones were much lower in August than in July. As a result, physiological characteristics of P. euramericana Venziano clone was superior than the other clones in Short rotation coppice (SRC) in Saemanguem area.

        이태리포푸라 I-214 엽육조직(葉肉組織)에서 원형질체(原形質體) 분리(分離)에 미치는 몇가지 요인(要因)

        이태리포푸라 I-214 (Populus euramericana cv. I-214)의 기내배양(器內培養)한 엽육조직(葉肉組織)에서 원형질체(原形質體) 분리(分離)에 미치는 몇가지 요인(要因)에 대(對)해 조사(調査), 검토(檢討)하였다. 기내(器內)에서 배양(培養)된 아(芽)를 다량(多量)으로 증식(增植)하기 위한 배지(培地)는 MS 기본배지(基本培地)에 $0.1mg/{\ell}$의 BAP를 첨가(添加)한 것이 가상 좋은 성적을 나타냈다. 엽(葉) 1g 당(當) $2.4{\times}10^6$개의 가장 높은 원형질체(原形質體) 분리(分離) 빈도를 나타낸 것은 Cellulase R-10 2 %, Macerozyme R-10 0.8 %, Hemicellulase 1.2 %, Driselase 2.0 %, Pectolyase Y-23 0.05 %에 DTT와 MES 완충액을 첨가(添加)한 후 삼투압 안정제로 0.6 M의 Mannitol을 넣고 pH를 5.6으로 조정한 효소용액(酵素溶液)이었다. CPW 용액(溶液)으로 세정(洗淨)한 후 0.6 M의 Sucrose 용액(溶液)에 처리(處理)한 것이 회수율(回收率) 51.8 %로 가장 높게 나타났다. A method isolating Populus euramericana cv. I-214 mesophyll protoplasts was developed to facilitate application of genetic engineering techniques to this species. The suitable medium for shoot multiplication in vitro was MS basal medium with $0.1mg/{\ell}$ BAP. The effects of several factors influencing protoplast isolation could be evaluated quickly by using leaf in vitro and known volumes of maceration and washing media. The best yields of mesophyll protoplasts were obtained using leaves in vitro in 2.0% Cellulase R-10, 0.8% Macerozyme R-10, 1.2% Hemicellulase, 2.0% Driselase, 0.05% Pectolyase Y-23, and O.6M Mannitol in addition to DTT and MES buffer adjusted to pH 5.6. Over $2.4{\times}10^6$ protoplasts per gram of leaf were produced using these conditions. For protoplast purification, the most favorable sucrose concentration of floating solution was 0.6M after washing them with CPW solution. This method of screening factors affecting protoplast isolation could be applicable to other species.
