      • Dye-Ligand Affinity Chromatography를 이용한 Staphylococcus aureus의 장독소 B 분리

        Staphylococcal enterotoxins, which cause staphylococcal food poisoning, have been purified by several methods. These procedures have usually included various combinations of ion-exchange chromatogram and gel filtration; such procedures can be time-consuming and may also result in low recoveries of enterotoxins. Although high recoveries of staphylococcal enterotoxins have been reported with chromatofocusing, this technique is expensive and cannot easily be adapted for large-scale purification. In this study we applied a simple, single-step procedure for the purification of Staphylococcal enterotoxin B (SEB) from culture supernatant fluids, namely, dye ligand affinity chromatography in which dye is coupled to an agarose support matrix. Dye ligand affinity chromatography has been used to purify a wide range of enzymes and some blood proteins. We used ten kinds of dye ligand affinity columns. The results showed that SEB had the binding ability to Red A, Reactive Green 19, Reactive Green 5, Reactive Brown 10 and Cibacron Blue 3GA-Agarose. We had the highest resolution by using the Reactive Green 19 dye affinity column. The purified SEB produced a single band when subjected to sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE) and western blot using anti-SEB. We could also confirm that the purified SEB produced superantigenic effect. Therefore, it could be concluded that SEB was produced purely in single step process by dye-ligand affinity chromatography. Such a method will form the basis of a high-yielding, rapid means of purifying the staphylococcal enterotoxins that can easily be adapted to large-scale production.

        PET 초극세사 염색에서 분산염료의 친화력과 온도 의존성

        본 연구에서는 PET 초극세사의 분산염료 등온흡착염색에서 염료의 분배계수와 표준친화력의 시료 굵기에 따른 영향성과 확산계수의 온도 의존성을 알아보았다. 염색은 해도형 초극세사 0.2, 0.06, 0.01 dpf 급과 일반 극세사 0.5 dpf 급을 anthraquinone계 염료 C.I. Disperse Blue 56으로 무한염욕조건 100, 110, 120, 130 °C에서 등온염색을 진행하였다. 그 결과 Nernst type과 잘 일치하는 분산염료의 시료 굵기별 등온흡착곡선을 얻었다. 해도형 PET 초극세사의 해성분 용출은 NaOH 수용액을 이용한 PET 알칼리 가공을 통하여 이루어졌다. 염색에 사용된 분산염료는 흡광도측정을 토대로 고온에서 장시간 염색 진행시 안정성이 확보됨을 확인하였다. 초극세사의 염색은 결론적으로 동일 온도에서 굵기가 감소함에 따라 시료 표면적의 증가로 인해 분배계수 및 표준친화력은 증가하였다. 모든 굵기에서 염색현상은 발열반응이기 때문에 염색 온도가 증가할수록 분배계수가 감소하였고 이에 따라 표준친화력은 동일 굵기에서 대략 9% 감소하였다. 또한 굵기가 감소할수록 동일 온도에서 최대로 염착되는 염료 농도가 증가하였고, 염색 과정에서 발생되는 염색열은 0.01 dpf 초극세사에서 -10.64 kcal/mol로 가장 낮게 나타났다. 시료 굵기가 감소할수록 염액과 접촉 가능한 표면적은 증가하였지만, 그에 반해 초극세사 사이의 공간이 조밀하게 되어 염액이 원활히 이동하기 어렵게 된다. 그 때문에 확산계수는 시료의 굵기가 극세화될수록 감소하였다. 이러한 경향성은 고온으로 갈수록 더 뚜렷하게 나타났으며. 시료의 굵기가 증가할수록 확산계수의 온도 의존성은 130 °C에서 40%까지 증가하였다. PET microfibers with various diameters (0.5, 0.2, 0.06, and 0.01 dpf) were dyed with a dispersed dye (C.I. Disperse Blue 56) at various temperatures. The dyeing process was conducted under infinite dyebath conditions at constant temperatures. The effects of the dyeing temperature and diameter on the partition coefficient, affinity, and diffusion coefficient of disperse dyes were studied. The curve of isotherms was fitted well to Nernst-type model in a large range of initial dye concentrations. At the same temperature, the partition coefficient and affinity decreased with increasing sample diameter due to the increase in surface area. At all deniers, the partition coefficient and affinity decreased with increasing temperature because the dyeing process is an exothermic reaction. In addition, the decrease in radius of the sample gives rise to a decrease in the heat of dyeing. The fine diameter of the sample resulted in an increased surface area but decreased space between the microfibers. Consequently, decreasing the diameter of the microfibers leads to a decrease in the diffusion coefficient. At the same diameter, the diffusion coefficient increased with increasing temperature because of rapid dye movement and the large free volume of the sample inside. In addition, thermal dependence of the diffusion coefficient increased when the diameter of the sample increased.

        This study has examined and compared the physical properties and dyeing behaviors in pretreated with emulsion dyeing. The results are as follows ; 1. The crystalline growth of PET fiber treated with 99% solvent/1% water emulsion was greater than in solvent alone treatment and untreated. 2. Equilibrium dye absorption and initial rate of dyeing of fiber treated with 99% solvent/1% water emulsion was greater than in solvent alone treatment and untreated. 3. The diffusion and dye affinity of PET fiber treated with 99.5% solvent/0.5% water was greater than in solvent alone treatment and untreated.

      • Structural effects of fluorescent dye on dyeing affinity and whiteness of paper

        A series of novel stilbene-triazine derivatives containing aminoben-zenesulfonic acid groups and amine groups were synthesized and screened to evaluate their dyeing affinity and whiteness properties. The UV absorptions of the derivatives were in the range of 240-360 nm. The absorption peaks of the cis-isomers and trans-isomers were in 281- 291 nm and 320-360 nm, respectively. Their fluorescence emission peaks of the derivatives were located in the range of 400-460 nm. The whiteness data indicated that all the stilbene-triazine-aminobenzenesulfonic acid derivatives showed excellent whitening effect.

      • Structural effects of fluorescent whitening agents on dyeing affinity and whiteness of paper

        Fluorescent whitening agents are applied to textiles and papers to enhance their whiteness and brightness through absorbing ultraviolet light region and emit blue fluorescence. In this study, a series of stilbene-triazine derivatives were synthesized through 3steps nucleophilic substitution reaction using two types of aniline and two amine substituents. The stilbene-triazine derivatives were characterized by MALDI-TOF MS analysis, UV-vis absorption, fluorescence emission spectroscopy. We evaluated their dyeing affinity and whiteness properties to the paper. The UV absorptions of the derivatives were in the ultra violet region (300-370nm) and their strong fluorescence emission peaks of the derivatives were in the blue region (400-460 nm). The whiteness data indicated that all the derivatives showed excellent whitening effect compared to undyed paper. In addition, when Aniline-2,5-disulfonic acid and Morpholine were introduced, the whiteness and dyeing affinity was most improved.

        고감성 의류용 수분감응형 인텔리전트 소재의 물성

        본 연구는 수분감응형섬유와 이들로 만들어진 니트의 흡습성, 건조특성 그리고 수분자발신장성과 같은 물성과 이들 니트소재의 여러 가지 염색시간과 온도에 따른 염색특성을 조사한다. 이를 위해서 HEF와 spandex를 심사로 사용하고 일반 PET와 aerocool 필라멘트를 커버링사로 사용한 3가지의 커버링사 시료를 준비하고 이들을 사용하여 3가지의 니트시료를 편직하고 이들을 염색온도와 시간을 달리하여 염색을 실시하였다. 3가지 니트시료의 흡습성, 건조특성 그리고 가역적 수축현상 등의 물성을 측정하고 실의 특성과 함께 분석하였으며 이들 니트시료의 착용성을 FAST 시스템을 사용하여 측정하고 분석하였다. 그리고 이들 니트시료의 염착성, 색차 그리고 세탁견뢰도 등과 같은 염색특성을 측정하고 수분감응형 니트소재의 염색공정특성을 알아보기 위해 염색시간과 온도에 따른 이들 염착특성을 분석하였다. This paper investigates the physical properties such as water absorption, drying and hygral expansion of moisture responded transformable fibers and their knitted fabrics including dyeing characteristics according to the different dyeing time and temperature. For this purpose, three kinds of covered filament yarn specimens using HEF and spandex as core and PET, aero cool as a covering filament were prepared. The knitted fabric specimens were made using three kinds yarn specimens and dyed with different dyeing temperature and time. The moisture absorption, drying and hygral expansion of three kinds of knitted fabric specimens were measured and discussed with yarn structure. The wearing performance of these knitted fabric specimens were also measured and discussed with yarn characteristics using FAST system. Finally, the dyeing characteristics such as dye affinity, color difference and color fastness to washing of these knitted fabric specimens were measured and discussed with different dyeing temperature and time for examining dyeing process performance of these moisture responded transformable knitted fabrics.

Their fluorescence emission peaks of the derivatives were located in the range of 400-460 nm. The whiteness data indicated that all the stilbene-triazine-aminobenzenesulfonic acid/coumarin derivatives showed excellent whitening effect.
