        흰점박이꽃무지(Protaetia brevitarsis) 성충의 우화되는 시기의 표준화

        This study was conducted to determine and to standardize the time of emergence of Protaetia brevitarsis adults, through four breeding experiments, including one which selected the dominant individuals of Protaetia brevitarsis. The breeding experiments were divided into two groups with hibernating and non-hibernating Protaetia brevitarsis by placing 100, 150, 200, 250 and 300 Protaetia brevitarsis in each breeding box (50 cm × 35 cm × 15 cm) filled with 12 L of fermented sawdust. Breeding tests were with four replicates each for 3 months. The test period was classified into four stages: Exp. 1 (Sep 1, 2019 to Nov 30, 2019), Exp. 2 (Dec 1, 2019 to Feb 29, 2020), Exp. 3 (Mar 1, 2019 to May 31, 2020) and Exp. 4 (Jun 1, 2020 to Aug 31, 2020). The survey items included average dead number, average emergence number, average mortality, and average emergence rate. In Exp. 1 and 2, the changes in Protaetia brevitarsis adults were similar. In Tables 1 and 2, the allegorical changes of adult white-spotted flowers were similar. In addition, the average dead number, average emergence number, average mortality, and average emergence rate showed better results in the hibernation state than in both Exp. 1 and 2. Exp. 3 and 4 showed a pattern of change different from the results of Exp. 1 and 2 and these characteristics were superior in the hibernation states compared to the non-hibernation states. In conclusion, this result was found to be most appropriate as a standardization of the emergence time of 200 Protaetia brevitarsis treatments through four breeding experiments in the hibernating state.

        Comparison of clay and charcoal as feed additives for Protaetia brevitarsis (Coleoptera: Scarabaeidae)

        The white-spotted chafer, Protaetia brevitarsis (Coleoptera: Scarabaeidae), has been traditionally used in Korea as a medicine for preventing liver-related diseases and suppressing liver cancer. Therefore, this insect is economically important and is commercially reared and sold in Korea. Recently, P. brevitarsis was listed as a temporal food ingredient by the Korean Ministry of Food and Drug Safety. Given the increasing economic importance of this beetle, we have sought to improve rearing conditions for its commercial production. In this study, we compared the effects of two food supplements, clay and charcoal, on the growth of second instar larvae of P. brevitarsis. Clay and charcoal are generally known as good adsorbent for removal of contaminating substances in insect feed. We fed second instar P. brevitarsis larvae a commercial diet consisting of fermented sawdust with seven different combinations of clay and/or activated charcoal, and measured their effects on weight gain for approximately 17 wk until larvae pupated. We found that addition of clay at 2.5% w/w of the fermented sawdust diet had no negative effect on weight gain of second instar P. brevitarsis larvae and thus may improve the quality of P. brevitarsis as a commercial food.

        흰점박이꽃무지에 발생하는 병원성 곰팡이 Metarhizium anisopliae에 대한 옻나무 추출액의 항진균 효과

        흰점박이꽃무지(Protaetia brevitarsis) 유충은 동아시아에서 식용 및 약용으로 널리 사용되고 있다. 흰점박이꽃무지 수요 증가로 인해 유충의 대량 사육 방법이 필요하지만 좁은 공간에서 대량으로 유충을 사육할 경우 곤충 병원성 질병 감염에 취약해진다. Metarhizium anisopliae는 흰점박이꽃무지 유충에 치명적인 곤충 병원성 곰팡이 중 하나이다. 이를 저해하기 위해 항진균 성분이 함유된 옻나무(Rhus verniciflua Stokes) 추출액을 첨가하였고, 연구 결과 1% 옻나무 추출액을 첨가한 경우 녹강균에 의한 유충의 치사율이 유의미하게 낮았다. 또한 일반톱밥을 급여한 대조구에 비해 옻나무 추출액 처리구에서 유충의 무게가 더 무거웠고 유충 기간이 짧아지는 것을 확인하였다. 따라서 이 연구를 통해 곤충 병원성 곰팡이에 대한 옻나무 추출액의 항진균 효과를 확인할 수 있었으며 흰점박이꽃무지 유충 사육 시 옻나무 추출액을 첨가하여 급여한다면 병원성 곰팡이에 의한 유충의 치사율을 낮추고 증체량을 높이는 데 효과가 있을 것이다. The white-spotted flower chafer (Protaetia brevitarsis) larva is one of the insects widely-used for edible and medicinal purposes in Eastern Asia. As a result of increasing demand for P. brevitarsis, massrearing systems in domestic farms have become necessary. However, the mass-rearing of larvae under confined rearing conditions could provide conditions unsuitable for preventing entomopathogenic diseases. Metarhizium anisopliae is the strongest fatal entomopathogenic fungus against P. brevitarsis. For inhibition of M. anisopliae, we used a Rhus verniciflua Stokes extract that has antifungal components. We investigated the inhibitory effect of the R. verniciflua extract at 1%, 5%, and 10% concentrations. The results showed that a 1% R. verniciflua extract added to sawdust produced a significantly low P. brevitarsis mortality rate. Moreover, extract-treated groups were heavier and had a shorter larval period than those of the untreated group. Consequently, we suggest that using an R. verniciflua extract can reduce the P. brevitarsis fatality rate from entomopathogenic fungi (e.g. M. anisopliae), resulting in more effective mass-rearing systems for P. brevitarsis.

        Effect of photoperiod and temperature on the reproductive responses of Protaetia brevitarsis

        In the present study, we investigated the effects of temperature and photoperiod on oviposition of Protaetia brevitarsis . The effects of long- and short-day cycles on oviposition and egg hatching of P. brevitarsis were investigated at different temperatures. Three male-female pairs were confined to oviposition chambers maintained at 20°C, 25°C, 30°C, and 35°C, with 16L:8D and 8L:16D photoperiod. Oviposition was observed at all temperatures. The total number of eggs laid per female was between 46.8 and 110.8, and the optimal temperature for oviposition and fertility was between 20°C and 30°C. Furthermore, it was difficult for the eggs to hatch at 35°C. Fewer eggs were laid under short photoperiod than under long photoperiod at all temperatures. Hatching success was 93.5% at 20°C, 90.9% at 25°C, 71.5% at 30°C and 37.3% at 35°C under long-day(16L:8D) condition and . Temperature had a strong effect on the time to hatching. Neither oviposition nor subsequent egg hatching was influenced by photoperiod and temperature. The information obtained will be useful for mass rearing P. brevitarsis.

        Development and oviposition characteristics of Protaetia brevitarsis (Coleoptera: Cetoniidae) by additional feeding Korean black raspberry marc and blueberry marc

        The food source of white-spotted flower chafer, Protaetia brevitarsis is important to economical performance. The P. brevitarsis larva were feed by mulberry and oak fermented sawdust. The effect of additional additives, Korean black raspberry (called bokbunja) marc and blueberry marc to fermented sawdust feed on the development and oviposition characteristics were investigated. The development periods of P. brevitarsis was decreased from 165.7 d to 40.0 d (mulberry sawdust) and 169.3 d to 39.0 d (oak sawdust), but the survival rate of larva was not affected by the addition of Korean black raspberry marc to fermented sawdust. However, blueberry marc addition to the fermented sawdust did not significant effect on the development period and survival rate of P. brevitarsis. The percentage of larva weight over 2.5 g was tend to increase by addition of Korean black raspberry marc and/or blueberry marc. The number of oviposition was also tend to increase by adding additional feed to sawdust but there was no significant different.

Therefore, it is easy to get, and the price is comparatively low. However, they still have meaningful amount of nutrients. With four different kinds of feed and three temperature conditions on the third instar larvae of beetles, the body weight change was tracked for 14 wk. We concluded that 27.5℃ is the optimal temperature to rear the third instar larvae among three temperatures (25, 27.5, and 30℃). Among four different feeds, conventional fermented oak saw dust with rice chaff and soybean cake was the best feed for larval weight gain during 14 wk. However, feed with soybean cake at 30 ℃ was the best condition for rearing P. brevitarsis larvae when temperatures and feeds were compared at the same time.

        흰점박이꽃무지(Protaetia brevitarsis)의 식품원료화를 위한 전처리 조건 확립

        본 연구에서는 흰점박이꽃무지를 식품원료화 하기 위한 전처리 조건을 설정하기 위해 세포독성, 이취감소 및 살균조건을 확립하였다. 유충에 살균 후 공급되는 세 가지 사료(참나무톱밥, 볏짚, 목초) 중에서 참나무톱밥을 공급하여 사육한 유충의 분말이 세포독성이 없었고, 유충의 발육단계를 크게 3령 유충, 용화직전 번데기, 번데기로 구분 지은 후 각각의 분말이 가지는 세포독성 유무를 확인한 결과에서 세포독성은 나타나지 않았다. 세포독성 검사 결과를 토대로 식품원료화하기 위한 유충의 표준사료는 세포독성이 전혀 나타나지 않고 전국 사육농가 중 가장 많이 이용하고 있는 발효 참나무 톱밥을 표준사료로 선정하였다. 흰점박이꽃무지의 분말전처리 조건으로 증기로 5분 동안 살균처리를 통해 사료 내의 미생물을 제거한 참나무톱밥을 급여하고 3일간 절식상태로 배변유도 후, 중장에 존재하는 내용물의 잔여도가 현저히 감소됨을 확인하였다. 또한 관능평가를 통해 3일간의 배변된 유충의 장 내용물의 잔여물이 감소함으로써 유충이 가지는 고유한 이취와 색깔이 눈에 띄게 개선되는 효과를 확인할 수 있었다. 배변 유도된 유충을 115℃, 0.9 kgf/cm³의 고온고압멸균을 5분간 실시한 후 동결건조 한 분말에서 세균 및 진균을 포함하는 미생물이 완전히 제거됨으로서 식품원료로서의 안전성을 확보할 수 있었다. The pharmacological efficacy of Protaetia (P.) brevitarsis larvae has been described in the Dongui Bogam. It is believed that the larvae are particularly useful for hepatic disorders. However, natural aversion has made it difficult to consume these larvae as food. Thus, we sought to make an eatable form of the larvae by establishing optimal conditions for larvae preparation. Larvae were selectively bred, sterilized, and a powder of larvae generated by freeze-drying. Afterward, the CellTiter 96<SUP>®</SUP> AQueous Non-Radioactive Cell Proliferation Assay (MTS) with the RAW 264.7 cell line was used to validate the safety of the powder as a food ingredient. We determined that oak sawdust sterilized by water vapor for 5 minutes could be used for larvae feed, and a feeding for 3~5 days followed by a fasting for 3 days were optimal conditions for larvae preparation. In addition, sterilization of larvae at 115oC and 0.9 kgf/cm³ (to avoid contamination of pathogenic bacteria and fungi) was successfully applied in the production of edible powder from P. brevitarsis. The optimized processes established in our experiments can be used in the industrial production of P. brevitarsis as a food ingredient.
