        Exact solution for free vibration of curved beams with variable curvature and torsion

        For the purpose of investigating the free vibration response of the spatial curved beams, the governing equations are derived in matrix formats, considering the variable curvature and torsion. The theory includes all the effects of rotary inertia, shear and axial deformations. Frobenius' scheme and the dynamic stiffness method are then applied to solve these equations. A computer program is coded in Mathematica according to the proposed method. As a special case, the dynamic stiffness and further the natural frequencies of a cylindrical helical spring under fixed-fixed boundary condition are carried out. Comparison of the present results with the FEM results using body elements in I-DEAS shows good accuracy in computation and validity of the model. Further, the present model is used for reciprocal spiral rods with different boundary conditions, and the comparison with FEM results shows that only a limited number of terms in the resultant provide a relatively accurate solution.

      • BCR/ABL mRNA Targeting Small Interfering RNA Effects on Proliferation and Apoptosis in Chronic Myeloid Leukemia

        Background: To investigate the effects of small interference RNA (siRNA) targeting BCR/ABL mRNA on proliferation and apoptosis in the K562 human chronic myeloid leukemia (CML) cell line and to provide a theoretical rationale and experimental evidence for its potential clinical application for anti-CML treatment. Materials and Methods: The gene sequence for BCR/ABL mRNA was found from the GeneBank. The target gene site on the BCR/ABL mRNA were selected according to Max-Planck-Institute (MPI) and rational siRNA design rules, the secondary structure of the candidate targeted mRNA was predicted, the relevant thermodynamic parameters were analyzed, and the targeted gene sequences were compared with BLAST to eliminate any sequences with significant homology. Inhibition of proliferation was evaluated by MTT assay and colony-formation inhibiting test. Apoptosis was determined by flow cytometry (FCM) and the morphology of apoptotic cells was identified by Giemsa-Wright staining. Western blotting was used to analyze the expression of BCR/ABL fusion protein in K562 cells after siRNA treatment. Results: The mRNA local secondary structure calculated by RNA structure software, and the optimal design of specific siRNA were contributed by bioinformatics rules. Five sequences of BCR/ABL siRNAs were designed and synthesized in vitro. Three sequences, siRNA1384, siRNA1276 and siRNA1786, which showed the most effective inhibition of K562 cell growth, were identified among the five candidate siRNAs, with a cell proliferative inhibitory rate nearly 50% after exposure to 12.5nmol/L~50nmol/L siRNA1384 for 24,48 and 72 hours. The 50% inhibitory concentrations ($IC_{50}$) of siRNA1384, siRNA1276 and siRNA1786 for 24hours were 46.6 nmol/L, 59.3 nmol/L and 62.6 nmol/L, respectively, and 65.668 nmol/L, 76.6 nmol/L, 74.4 nmol/L for 72 hours. The colony-formation inhibiting test also indicated that, compared with control, cell growth of siRNA treated group was inhibited. FCM results showed that the rate of cell apoptosis increased 24 hours after transfecting siRNA. The results of annexinV/PI staining indicated that the rate of apoptosis imcreased (1.53%, 15.3%, 64.5%, 57.5% and 21.5%) following treamtne with siRNAs (siRNA34, siRNA372, siRNA1384, siRNA1276 and siRNA1786). Morphological analysis showed td typical morphologic changes of apoptosis such as shrunken, fragmentation nucleus as well as "apoptotic bodies" after K562 cell exposure to siRNA. Western blot analysis showed that BCR/ABL protein was reduced sharply after a single dose of 50nmol/L siRNA transfection. Conclusions: Proliferation of K562 cells was remarkbly inhibited by siRNAs (siRNA1384, siRNA1276 and siRNA1786) in a concentration-dependent manner in vitro, with effective induction of apoptosis at a concentration of 50 nmol/L. One anti-leukemia mechanism in K562 cells appeared that BCR/ABL targeted protein was highly down-regulated. The siRNAs (siRNA1384, siRNA1276 and siRNA1786) may prove valuable in the treatment of CML.

        Electronic Structure, Phase Transition, and Elastic Properties of ScC under High Pressure

        The structural properties and the phase transition for scandium carbide (ScC) have been studied in NaCl (B1), CsCl (B2), ZB (B3), WZ (B4), NiAs (B81), WC (Bh), and Pmmn structures by using the pseudopotential plane-wave method in the framework of the density functional theory. Our theoretical results show that the most stable structure is the B1 phase, contrary to the result of Rahim et al. The phase transitions B1 ! Pmmn and Pmmn ! B2 are predicted at 83.7 and 109.7 GPa, respectively. At the same time, we find that the B3, B4, B81, and Bh phases are not stable over the whole pressure range considered. In particular, the elastic constants of Pmmn-ScC under high pressure are obtained successfully. The effects of pressure on the elastic properties of B1-ScC and Pmmn-ScC are also predicted. The Debye temperatures and the sound velocities of these two structures are estimated from the elastic constants, and by analyzing G/B, the brittle-ductile behavior of ScC is assessed. In addition, the density of states of B1-ScC at high pressures is also discussed.

      • Screening for MiRNAs Related to Laryngeal Squamous Carcinoma Stem Cell Radiation

        Objective: To use microarray chip technology for screening of stem cell radiation related miRNAs in laryngeal squamous cell carcinoma; study and explore the relationship of miRNAs with radiosensitivity of laryngeal squamous cells. Method: After conventional culture and amplification of the laryngeal squamous carcinoma cell line Hep-2, CD 133+ cells were screened out with combination of isolated culture of stem cell microspheres and FACS for preparation of laryngeal cancer stem cells. After radiation treatment, miRNAs of laryngeal squamous carcinoma stem cells before and after radiation were enriched and purified. After microarray hybridization with mammalian miRNA and scanning of fluorescence signal, the miRNAs of laryngeal squamous carcinoma stem cells before and after radiation was subject to differential screening and clustering analysis. Real-time quantitative RT-PCR was used to verify part of the differentially expressed miRNAs. Results: 70 miRNAs related to laryngeal cancer stem cell radiation with 2-fold difference in expression were screened out, in which 62 were down-regulated and 8 were up-regulated. Fluorescent quantitative RT-PCR results were consistent with miRNAs chip results. Conclusion: Some miRNAs may be involved in self-regulation with laryngeal squamous carcinoma stem cell radiation.

        The whole plant of Euphorbia helioscopia is an important traditional Chinese medicine. Fom its BuOH soluble extract,one new lactam (1), three new terpenoids (2-4) including a new naturally occurring compound, and three known compounds were isolated. Their structures were identified by spectroscopic evidences. In particular, the absolute configurations of side chain of compounds 1 and 2 were determined using computational methods.

        Distinct expression patterns and functions of the pygopus genes in amphioxus and zebrafish early embryogenesis

        The Wnt/b-catenin transcriptional activation complex requires the adapter protein Pygopus (Pygo), which links the basal transcription machine to b-catenin to regulate the transcription of target genes. Previous studies revealed that the Pygo genes were broadly involved in the embryonic development of Drosophila, Xenopus and mouse, however, their functions in the embryonic development of amphioxus and zebrafish remain to be investigated. Here, we cloned and characterized one Pygo gene from amphioxus Branchiostoma belcheri (BbPygo) and two fromzebrafish Danio rerio (DrPygo1 and DrPygo2). The spatial–temporal expression profiles showed that all these genes were maternally expressed. BbPygo expression was distributed ubiquitously in the amphioxus embryos before the gastrula stage and became restricted in the pharyngeal region in late stages. Similar to BbPygo, DrPygo2 was also expressed throughout whole embryos before the early gastrula stage, but limited in the pharyngeal region and central nervous system (CNS) in subsequent developing stages. Further knockdown of the DrPygo2 resulted in developmental defects of the body axis, CNS and cartilage segments in zebrafish. In contrast, the expression signal of the DrPygo1 gene was not detectable using the in situ hybridization method, which is consistent with its extremely low expression level detected by RTqPCR. The expression patterns and function investigations revealed in this study suggest that the three Pygo genes probably function differentially though all of them show a conserved maternal expression pattern during the early embryonic stages of amphioxus and zebrafish.

        A Comparative Study of Camouflage Printing Color Matching Based on Monitor and Paper Card

        To improve the accuracy of color reproduction for camouflage printing, a new color matching method, namedmonitor-paper-fabric, was proposed by importing color management into textiles, which matched the printing pastesaccording to the color of the paper card printed by the ink-jet printer after color management. Not like the traditional colormatching method, it matched the printing pastes according to the color from computer monitor after color correction. Twocolor matching methods were analyzed by comparing the color difference. It was found that the “monitor-paper-fabric” colormatching method could be considered as a convenient and feasible color matching method. Most color differences betweenthe monitor and the fabric in the camouflage pattern were reduced to lower than 4, except for color blocks 5 and 6. Inaddition, the color differences of six color blocks between the paper and the fabric were all less than 3.5, and were lower thanthose between the monitor and the fabric. The color consistency between the paper and the fabric was better than thatbetween the monitor and the fabric.

      • A Composition Method of Logical Function Block Chain

        ForCES is a new open programmable architecture, it separates control and forwarding, it also abstracts resources of Forwarding Elements (FE) into some Logical Function Blocks (LFB), Control Element (CE) can reconfigure the forwarding function of FEs by recomposing their LFB chains. Firstly, A SDN architecture based on ForCES is proposed. Then, based on the formalization of the concept of LFB and the traditional I/O matching algorithm, an LFB chain composition method is proposed. This method can combine a series of LFBs to an LFB chain according to special application request. Based on the LFB chain composition algorithm, an improved algorithm is proposed, which can improve the composition efficiency. At last, an example is provided to illustrate how this method works, and a simulation is given to compare the efficiency of the base algorithm and the improved algorithm.
