        Metastasis associated genomic aberrations in stage II rectal cancer

        Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

        Cadmium induces apoptosis in primary rat osteoblasts through caspase and mitogen-activated protein kinase pathways

        Exposure to cadmium (Cd) induces apoptosis in osteoblasts (OBs); however, little information is available regarding the specific mechanismsof Cd-induced primary rat OB apoptosis. In this study, Cd reduced cell viability, damaged cell membranes and induced apoptosis in OBs. We observed decreased mitochondrial transmembrane potentials, ultrastructure collapse, enhanced caspase-3 activity, and increasedconcentrations of cleaved PARP, cleaved caspase-9 and cleaved caspase-3 following Cd treatment. Cd also increased the phosphorylationof p38-mitogen-activated protein kinase (MAPK), extracellular signal-regulated kinases (ERK)1/2 and c-jun N-terminal kinase (JNK) in OBs. Pretreatment with the caspase inhibitor, N-benzyloxycarbonyl-Val-Ala-Asp-fluoromethylketone, ERK1/2 inhibitor (U0126), p38 inhibitor(SB203580) and JNK inhibitor (SP600125) abrogated Cd-induced cell apoptosis. Furthermore, Cd-treated OBs exhibited signs of oxidativestress protection, including increased antioxidant enzymes superoxide dismutase and glutathione reductase levels and decreased formationof reactive oxygen species. Taken together, the results of our study clarified that Cd has direct cytotoxic effects on OBs, which are mediatedby caspase- and MAPK pathways in Cd-induced apoptosis of OBs.

        A Coumarin-based Fluorescent Sensor for Selective Detection of Copper (II)

        Cu (II) detection is of great importance owing to its significant function in various biological processes. In this report, we developed a novel coumarin-based chemosensor bearing the salicylaldimine unit (2) for Cu2+ selective detection. The results from fluorescence spectra demonstrated that the sensor could selectively recognize Cu2+ over other metal cations and the detection limit is as low as 0.2 μM. Moreover, the confocal fluorescence imaging in HepG2 cells illustrated its potential for biological applications.

        A 2D/2D BiPO4/g-C3N4-B Z-type heterojunction for enhanced photocatalytic degradation of dye pollutants

        A 2D/2D BiPO4/g-C3N4-B nano-sheet heterojunction photocatalyst was synthesized via a simple coprecipitation method at room temperature using glacial acetic acid as solvent, which showed excellent activity toward the degradation of rhodamine B (RhB). The heterojunction showed much higher efficiency of separation and transfer of photogenerated carriers compared to that of its constituents. Moreover, the spectral response range of BiPO4 was effectively broadened after the combination of g-C3N4-B and BiPO4. Consequently, a 97.3% degradation of RhB within 25 min by BiPO4/g-C3N4-B heterojunction photocatalyst under visible light irradiation was observed. The difference in work functions of BiPO4 and g-C3N4-B was evident from UPS characterization, which led to the bending of the energy band and the establishment of an internal electric field at the interface of the heterojunction. Therefore, the synthesized direct Z-type BiPO4/g-C3N4-B heterojunction enhanced the oxidation-reduction ability by promoting the effective separation of photogenerated carriers.

        Structural and Ferromagnetic Properties SnO2/TiO2 Nanotubes Obtained by Electrospinning

        SnO2/TiO2 (Sn/Ti = 5/1) nanotubes were prepared by the electrospinning method. The morphology was characterized by scanning electron microscope (SEM) and transmission electron microscope (TEM). The results showed that the inner diameter of prepared materials was about 100 nm and the wall thickness was about 10 nm. The results of X-ray diffraction (XRD) and high resolution transmission electron microscope (HRTEM) showed that SnO2/TiO2 nanotubes had a mixed phase of SnO2 rutile and TiO2 anatase structures and no impurity phases. The magnetic properties of the SnO2/TiO2 nanotubes were characterized by a superconducting quantum interferometer (SQUID). The results indicated that the samples exhibited room temperature ferromagnetism which may be attributed to the interface between TiO2 and SnO2 phases.

        Experimental study on mechanical performances of lattice steel reinforced concrete inner frame with irregular section columns

        Based on the test on a 1/2.5-scaled model of a two-bay and three-story inner frame composed of reinforced concrete beams and lattice steel reinforced concrete (SRC) irregular section columns under low cyclic reversed loading, the failure process and the features of the frame were observed. The subsequence of plastic hinges of the structure, the load-displacement hysteresis loops and the skeleton curve, load bearing capacity, inter-story drift ratio, ductility, energy dissipation and stiffness degradation were analyzed. The results show that the lattice SRC inner frame is a typical strong column-weak beam structure. The hysteresis loops are spindle-shaped, and the stiffness degradation is insignificant. The elastic-plastic inter-story deformation capacity is high. Compared with the reinforced concrete frame with irregular section columns, the ductility and energy dissipation of the structure are better. The conclusions can be referred to for seismic design of this new kind of structure.

      • Poor Prognosis Significance of Pretreatment Thrombocytosis in Patients with Colorectal Cancer: a Meta-Analysis

        Background: Recently, several studies have reported that elevated platelet counts may be associated with the poor prognosis of colorectal cancer. However, conclusions remain controversial. This meta-analysis was therefore designed to analyze and evaluate the prognostic role of preoperative or pretreatment thrombocytosis in patients with colorectal cancer. Materials and Methods: We searched PubMed, EMBASE, the Cochrane Library and Web of Science to March 29th, 2015. The citation lists of included studies were also hand-searched to identify further relevant trials. To investigate the association between thrombocytosis and prognosis of colorectal cancer, the 1-year, 3-year and 5-year survival of each studies were obtained. The odds ratio (OR) with its 95% confidence interval (CI) was used to evaluate the relation of overall survival (OS) between thrombocytosis and normal platelet counts (PLT). Likewise, disease free survival (DFS) was obtained and evaluated. The analysis was performed and assessed using Review Manager 5.2. Results: A total of 14 studies (N=5,566 participants, 11 including 4,468 for OS, 6 including 1,533 for DFS) were included in this meta-analysis, of which seven (N=3810) defined thrombocytosis as a platelet count ${\geq}400{\times}10^9/L$, and 375 (9.8%) patients exhibited pretreatment thrombocytosis. Thrombocytosis have a close relationship with the poor OS of colorectal cancer compared with normal PLT, with the pooled ORs of 1-year, 3-year and 5-year survival being 0.41 [95% CI 0.34-0.51; P<0.001], 0.28 [95% CI 0.21-0.38; P<0.001] and 0.26 [95% CI 0.20-0.34; P<0.001], respectively. For DFS, the same results were showed as the pooled ORs of 1-year, 3-year and 5-year survival respectively being 0.34 [95% CI 0.24-0.50; P<0.001], 0.31 [95% CI 0.23-0.43; P<0.001] and 0.25 [95% CI 0.18-0.34; P<0.001]. Conclusions: This meta-analysis indicated that thrombocytosis may predict poor prognosis for patients with colorectal cancer, and platelet counts may be a cost-effective and noninvasive marker.

      • A SANDWICH-INJECTION METHOD FOR MICROCHIP ELECTROPHORESIS

        In microchip electrophoresis (μ-CE), sample injection is generally achieved through cross, double-T, or T-form injector structures. In these reported approaches, the separation efficiency and detection sensitivity of μ-CE is significantly influenced by the shape and size of the sample plug introduced into the separation channel or sample leakage in separation phase. Here, we present a sandwich-injection method for controlling discrete sample injection in μ-CE. This method involves four accessory arm channels in which symmetrical potentials are loaded to form a unique parallel electric field distribution at the intersection of sample and separation channels. The parallel electric field effectuate a virtual wall to confine the shape of a sample plug and depress the spreading of the sample plug at the junction of sample and separation channels, and also prevent sample leakage during separation step. The key features of this method are the ability to inject well-defined sample plugs at the original sample concentration and the ability to control the sample plug size. The virtues of the novel injection technique were demonstrated with numerical models and validated with fluorescence visualizations of electrophoretic experiments.

        Genome-wide identification of the class III aminotransferase gene family in rice and expression analysis under abiotic stress

        Aminotransferases are pyridoxal 50-phosphatedependent enzymes that play crucial roles in plant growth, development, and responses to abiotic stress. The class III aminotransferase family (ATIII family) is an important subfamily. However, no characterization of rice ATIII genes has been previously reported. Using available rice genome sequence information, we identified 12 japonica and 13 indica ATIII genes that were randomly localized on chromosomes 2, 3, 4, 5, 7, 8, and 11. Information provided by the Plant Genome Duplication Database revealed that four japonica and four indica ATIII genes are the results of segmental duplications, and two japonica and six indica genes resulted from tandem duplications. A phylogenetic analysis of the ATIII genes in japonica, indica and Arabidopsis enabled the classification of the genes into six different groups, and the characteristics were established before the monocot-dicot and japonica–indica split. An analysis of the Ka/Ks, divergence time and average indel length suggested the diverse selection styles of the duplicated gene pairs. Gene structure and motif analyses revealed that the ATIII gene family has experienced extensive divergence. Real-time PCR was performed to examine the expression pattern of the japonica ATIII genes in response to various abiotic stresses including drought, salt, and cold. The results suggested that most of the genes were differentially up- or down-regulated in rice seedlings in response to at least one stress factor, which indicates the key role of the rice ATIII gene family in responding to abiotic stresses. These results provide a basis for elucidating the roles of the ATIII genes and their further functional analysis under abiotic stresses.
