        Momordicine I alleviates isoproterenol-induced cardiomyocyte hypertrophy through suppression of PLA2G6 and DGK-ζ

        This study aimed to observe the protective effect of momordicine I, a triterpenoid compound extracted from momordica charantia L., on isoproterenol (ISO)-induced hypertrophy in rat H9c2 cardiomyocytes and investigate its potential mechanism. Treatment with 10 μM ISO induced cardiomyocyte hypertrophy as evidenced by increased cell surface area and protein content as well as pronounced upregulation of fetal genes including atrial natriuretic peptide, β-myosin heavy chain, and α-skeletal actin; however, those responses were markedly attenuated by treatment with 12.5 μg/ml momordicine I. Transcriptome experiment results showed that there were 381 and 447 differentially expressed genes expressed in comparisons of model/control and momordicine I intervention/model, respectively. GO enrichment analysis suggested that the anti-cardiomyocyte hypertrophic effect of momordicine I may be mainly associated with the regulation of metabolic processes. Based on our transcriptome experiment results as well as literature reports, we selected glycerophospholipid metabolizing enzymes group VI phospholipase A2 (PLA2G6) and diacylglycerol kinase ζ (DGK-ζ) as targets to further explore the potential mechanism through which momordicine I inhibited ISO-induced cardiomyocyte hypertrophy. Our results demonstrated that momordicine I inhibited ISO-induced upregulations of mRNA levels and protein expressions of PLA2G6 and DGK-ζ. Collectively, momordicine I alleviated ISO-induced cardiomyocyte hypertrophy, which may be related to its inhibition of the expression of glycerophospholipid metabolizing enzymes PLA2G6 and DGK-ζ.

        Strengthening Packet Loss Measurement from the Network Intermediate Point

        Estimating loss rates with the packet traces captured from some point in the middle of the network has received much attention within the research community. Meanwhile, existing intermediate-point methods like [1] require the capturing system to capture all the TCP traffic that crosses the border of an access network (typically Gigabit network) destined to or coming from the Internet. However, limited to the performance of current hardware and software, capturing network traffic in a Gigabit environment is still a challenging task. The uncaptured packets will affect the total number of captured packets and the estimated number of packet losses, which eventually affects the accuracy of the estimated loss rate. Therefore, to obtain more accurate loss rate, a method of strengthening packet loss measurement from the network intermediate point is proposed in this paper. Through constructing a series of heuristic rules and leveraging the binomial distribution principle, the proposed method realizes the compensation for the estimated loss rate. Also, experiment results show that although there is no increase in the proportion of accurate estimates, the compensation makes the majority of estimates closer to the accurate ones.

        Passive Overall Packet Loss Estimation at the Border of an ISP

        In this paper, a heuristic method that leverages packet traces captured at the entire boarder of an ISP to distinguish and estimate the overall packet loss within an ISP’s management domain (Intra_Path_Loss) and that in the outside Internet (Inter_Path_Loss) is proposed. Our method is inspired by that packet losses happened at different locations will cause different TCP sequence number patterns at the border of an ISP. Thereby, we leverage these TCP sequence number patterns to build a series of heuristic rules to estimate Intra_Path_Loss and Inter_Path_Loss, respectively. We do this work with an eye towards showing that the overall packet losses defined and estimated in this paper can provide the operators with some valuable information to help them precisely grasp the overall performance of network paths and narrow down the range of network anomalies. The proposed method is rigorously validated with simulations, and finally the results from a regional academic network JSERNET verify its effectiveness and practicability.

        Knock out of the annexin gene OsAnn3 via CRISPR/Cas9-mediated genome editing decreased cold tolerance in rice

        Plant annexins are Ca2+-dependent phospholipidbindingproteins and exist as multigene families in plants. They are implicated in the regulation of plant developmentas well as protection from environmental stresses. In thisstudy, the rice annexin gene OsAnn3 knockout was performedvia the CRISPR/Cas9 (clustered regularly interspaced shortpalindromic repeats/CRISPR associated proteins) mediatedgenome editing. Thus, mutant plantlets were successfullyobtained. We identified cold tolerance phenotype of T1mutant lines from T0 biallelic mutants using the 4~6°C for 3days cold treatment. The results showed that REC (the relativeelectrical conductivity) of T1 mutant lines was increased, andthe survival ratio of T1 mutant lines was decreased dramaticallycompared with the wild type after the exposure to coldtreatment. It was suggested that OsAnn3 was involved in coldtolerance of rice.

        Microstructure and Mechanical Properties of Mg–2.0Gd–1.2Y–1.0Zn–0.2Zr Alloy

        The microstructural evolution of Mg–2.0Gd–1.2Y–1.0Zn–0.2Zr (GWZK) alloy during homogenization and subsequentaging heat-treatment and the tensile properties of the peak-aged sample at various temperatures have been investigated. Thephase transformation during homogenization, static precipitation during aging, and deformation mechanisms at 250 °C aresystematically analyzed through XRD, TEM and EBSD measurements. The peak-aged GWZK alloy has good combinationof strength and ductility at elevated temperatures owing to its high thermal stability by the LPSO phases distributed alongthe grain boundaries. The change (19 MPa) in the yield strength of peak-aged samples between RT and 250 °C is mainlyrelated to easy gliding of dislocations on the non-basal planes due to the decrease in critical resolved shear stress of slip. Two strengthening models are applied to quantitatively describe the efect of {11 −2 0}훼precipitate plates on Orowanstrengthening corresponding to RT and 250 °C. Ignoring changes in solid solution strengthening and grain boundary strengthening of the peak-aged sample caused by increasing temperature, the calculated reduction value (16.5 MPa) in macroscopicyield strength from RT (based on the basal<a>slip mode) to 250 °C (based on the pyramidal<c+a>slip mode) is in goodagreement with the measured reduction value (19 MPa).

        Damping and transformation behaviors of Ti50(Pd50−xCrx) shape memory alloys with x ranging from 4.0 to 5.0

        The damping and transformation behaviors of Ti50(Pd50−xCrx) shape memory alloys with x ranging from 4.0 to 5.0 are systematically investigated. The damping capacity (Q−1) at the martensitic transformation is found to be inversely proportional to the square root of frequency, i.e., Q−1∝ω−0.5. A relaxation peak or shoulder is observed slightly below the martensitic transformation damping peak for compositions within the compositional crossover region (4.5 ⩽x⩽ 4.8). Furthermore, the damping capacity at the martensitic transformation is smaller within the compositional crossover region (4.5 ⩽x⩽ 4.8), compared with that of compositions at both sides (x = 4.0 and x = 5.0). These observations can be ascribed to the hysteretic motion of interfaces between different phases near the compositional crossover region.

        High mobility group box 3 promotes cervical cancer proliferation by regulating Wnt/β-catenin pathway

        Objective: High mobility group box 3 (HMGB3) plays an important role in the developmentof various cancer. This study aims to explore whether HMGB3 regulates cervical cancer (CC)progression and elucidate the underlying mechanism. Methods: HMGB3 expression in clinical patients' tumor samples were determined byreal-time quantitative polymerase chain reaction (qRT-PCR) and western blot. HMGB3overexpression/knockdown were used to investigate its function. Cell apoptosis and cyclewere detected by Annexin V/PI staining and flow cytometry. In vivo tumor model was madeby subcutaneous injection of HeLa cells transfected with shRNAs targeting HMGB3 (sh HMGB31) into the flank area of nude mice. Western blot was used to detect the levels ofβ-catenin, c-Myc, and matrix metalloproteinase-7 (MMP-7) in Hela and CaSki cells transfectedwith sh-HMGB3 or shRNAs targeting β-catenin. Results: Both messenger RNA and protein levels of HMGB3 were upregulated in CC tissuesfrom patients. High expression level of HMGB3 had positive correlation with serosalinvasion, lymph metastasis, and tumor sizes in CC patient. Functional experiments showedthat HMGB3 could promote CC cell proliferation both in vitro and in vivo. The expressionlevels of c-Myc and MMP-7 were increased, resulting in regulating cell apoptosis, cell cycle,and activating Wnt/β-catenin pathway. Conclusions: Our data indicated that HMGB3 may serve as an oncoprotein. It could be used asa potential prognostic marker and represent a promising therapeutic strategy for CC treatment.

        Mechanical Behavior of PMMA/SiO2 Multilayer Nanocomposites: Experiments and Molecular Dynamics Simulation

        Poly(methyl methacrylate) (PMMA)/nano-silica (nano-SiO2) nanocomposite film with 256 layers containing different amounts of nano-SiO2 was manufactured by a new type of micro-nano multilayer co-extrusion technology. The structure, morphology and mechanical properties of PMMA/nano-SiO2 nanocomposite films were investigated through Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM) and tensile test. Besides, molecular dynamics simulation was adopted to study the dispersion and content effect on mechanical properties of PMMA/nano-SiO2 nanocomposite film. It is demonstrated that the tensile strength and the elongation at break of the composites improved with increasing nano-SiO2 content from 0 to 5 wt%, which was in good agreement with the Molecular Dynamics simulation. And the nanofiller dispersion in the multilayer nanocomposite film was better than that in the single-layer film with equivalent thickness under the effect of torsion lamination. Overall, the best performance was found for the nanocomposites of PMMA with nano-SiO2 content of 3.5 wt%.
