HCV-Specific Interleukin-21+CD4+ T Cells Responses Associated with Viral Control through the Modulation of HCV-Specific CD8+ T Cells Function in Chronic Hepatitis C Patients

        Guo-Hua Feng,Fu-Sheng Wang,Ji-Yuan Zhang,Qing-Lei Zeng,Lei Jin,Junliang Fu,Bin Yang,Ying Sun,Tianjun Jiang,Xiangsheng Xu,Zheng Zhang,Jinhong Yuan,Liyuan Wu 한국분자세포생물학회 2013 Molecules and cells Vol.36 No.4

Interleukin-21 (IL-21)+CD4+ T cells are involved in the immune response against hepatitis B virus (HBV) by secreting IL-21. However, the role of IL-21+CD4+ T cells in the immune response against chronic hepatitis C (CHC) virus infection is poorly understood. This study aimed to investigate the role of IL-21+CD4+ T cells in CHC patients and the potential mechanisms. The study subjects in-cluded nineteen CHC patients who were grouped by viral load (low, < 106 RNA copies/ml, n = 8; high, > 106 RNA copies/ml, n = 11). The peripheral frequency of HCV-specific IL-21+CD4+ T cells was higher in the low viral load group and was negatively correlated with the serum HCV RNA viral load in all CHC patients. Meanwhile, IL-21+ cells accumulated in the liver in the low viral load group. In vitro, IL-21 treatment increased the expression of proliferation markers and cytolytic molecules on HCV-specific CD8+ T cells. In summary, these findings suggest that HCV-specific IL-21+CD4+ T cells might contribute to HCV control by rescuing HCV-specific CD8+ T cells in CHC patients.

Core prescription pattern of Chinese herbal medicine for depressive disorders in Taiwan: a nationwide population-based study

        Diem Ngoc Hong Tran,I-Hsuan Hwang,Fun-Jou Chen,Yuan-Pu Tseng,Ching-Mao Chang,Shih-Jen Tsai,Jen-Lin Yang,Ta-Peng Wu,Chung-Hua Hsu,Fang-Pey Chen,Yen-Ying Kung 한국한의학연구원 2021 Integrative Medicine Research Vol.10 No.3

Background: Depressive disorders (DD) affect not only mood and behavior but also various physical functions. Traditional Chinese medicine (TCM) has been shown to have some benefits in treating DD. However, one formula or one single herb might be not show high efficacy when used to treat depression. Thus, this study aimed to examine the core prescription pattern of Chinese herbal medicine (CHM) among patients with DD in Taiwan as a reference for related research and clinical applications. Methods: All patients, who had been diagnosed with major depressive disorder or minor depression or dysthymia without any other baseline diseases and had at least one CHM outpatient clinical visit from 2002 to 2011, were extracted from three randomly sampled cohorts, namely the 2000, 2005 and 2010 cohorts of the National Health Insurance Research Database (NHIRD) of Taiwan. The collected data was analyzed to explore the patterns of herbal products. Results: There were 197,146 patients with a diagnosis of DD and of these 1806 subjects had only a diagnosis of DD and utilized CHM. The most common formula was Gan-Mai-Da-Zao-Tang (12.19%), while Suan-Zao-Ren (3.99%) was the most commonly prescribed single herb. The core pattern of prescriptions consisted of a combination of Gan-Mai-Da-Zao-Tang, Jia-Wei-Xiao-Yao-San, Chai-Hu-Jia-Long-Gu-Mu-Li-Tang, He-Huan-Pi, Yuan-Zhi and Shi-Chang-Pu. Conclusions: This study describes the CHM core prescription pattern used to treat patients in Taiwan with DD and it is a potential candidate for study in future pharmacological or clinical trials targeting DD.

Analysis and application of zeolite in microspeaker box

        Yuan-Wu Jiang,Dan-Ping Xu,권중학,Zhi-Xiong Jiang,김준형,황상문 대한기계학회 2019 JOURNAL OF MECHANICAL SCIENCE AND TECHNOLOGY Vol.33 No.8

Recently, miniaturization has become a hot topic in microspeaker research and has attracted considerable attention. However, the acoustic quality of microspeakers-evaluated in terms of the sound pressure level (SPL)-declines if their dimensions are reduced. This study discusses the application of zeolite in the microspeaker box without causing a change in speaker dimensions. The equivalent circuit method and Langmuir adsorption isotherm have been used to analyze SPL of the microspeaker box. Based on the analysis results obtained in this study, microspeaker samples have been manufactured and their effectiveness has been experimentally verified. Simulation and experimental results reveal that the zeolite can improve SPL by approximately 3.5 dB over a frequency range of 100–1000 Hz.

      • Association of GSTP1 and RRM1 Polymorphisms with the Response and Toxicity of Gemcitabine-cisplatin Combination Chemotherapy in Chinese Patients with Non-small Cell Lung Cancer

        Yuan, Zhi-Jun,Zhou, Wen-Wu,Liu, Wei,Wu, Bai-Ping,Zhao, Jin,Wu, Wei,He, Yi,Yang, Shuo,Su, Jing,Luo, Yi Asian Pacific Journal of Cancer Prevention 2015 Asian Pacific journal of cancer prevention Vol.16 No.10

Background: Previous studies showed that genetic polymorphisms of glutathione S-transferase P1 (GSTP1) were involved in glutathione metabolism and genetic polymorphisms of ribonucleotide reductase (RRM1) were correlated with DNA synthesis. Here we explored the effects of these polymorphisms on the chemosensitivity and clinical outcome in Chinese non-small cell lung cancer (NSCLC) patients treated with gemcitabine-cisplatin regimens. Materials and Methods: DNA sequencing was used to evaluate genetic polymorphisms of GSTP1 Ile105Val and RRM1 C37A-T524C in 47 NSCLC patients treated with gemcitabine-cisplatin regimens. Clinical response was evaluated according to RECIST criteria after 2 cycles of chemotherapy and toxicity was assessed by 1979 WHO criteria (acute and subacute toxicity graduation criteria in chemotherapeutic agents). Results: There was no statistical significance between sensitive and non-sensitive groups regarding the genotype frequency distribution of GSTP1 Ile105Val polymorphism (p>0.05). But for RRM1 C37A-T524C genotype, sensitive group had higher proportion of high effective genotype than non-sensitive group (p=0.009). And according to the joint detection of GSTP1 Ile105Val and RRM1 C37A-T524C polymorphisms, the proportion of type A (A/A + high effective genotype) was significantly higher in sensitive group than in non-sensitive group (p=0.009). Toxicity showed no correlation with the genotypes between two groups (p>0.05). Conclusions: Compared with single detection of genetic polymorphisms of GSTP1 Ile105Val or RRM1 C37A-T524C, joint detection of both may be more helpful for patients with NSCLC to receive gemcitabine-cisplatin regimens as the first-line chemotherapy. Especially, genetic polymorphism of RRM1 is more likely to be used as an important biomarker to predict the response and toxicity of gemcitabine-cisplatin combination chemotherapy in NSCLC.

Detection of Pseudomonas aeruginosa Carried a New Array of Gene Cassettes within Class 1 Integron Isolated from a Teaching Hospital in Nanjing, China

        Yuan Wu,Hui Li,Jun Li,Zu Hu Huang 한국미생물학회 2008 The journal of microbiology Vol.46 No.6

We report here novel array of gene cassettes found in single variable region of class 1 integron disseminated in Pseudomonas aeruginosa isolated from a teaching hospital in Nanjing, Jiangsu Province, China. 29 of 47 (61%) P. aeruginosa strains were confirmed haboured class 1 integron, and all the positive strains have the same variable region confirmed by PCR and RFLP methods. The variable region contained an unreported order of four gene cassettes aac(6’)-II-aadA13-cmlA8-oxa-10. Of those, cmlA8 gene was a variant of cmlA5 encoding non-enzymatic protein which putatively confer resistance to chloramphenicol. Susceptibility testing revealed multidrug-resistant mechanisms were involved in the class 1 integron positive clinical isolates. And the class 1 integron located on an about 15 kb transferable plasmid was certified by conjugation experiment and plasmid DNA analysis. The macro restriction profile indicated those clinical strains were clonally related.

Rhaponticin suppresses the hypoxia-induced factor-1 alpha-mediated aggressive phenotype of tongue squamous cell carcinoma

        Wu Yuan,Wan Xiaowen,Shao Yisen,Wang Wei,Huang Wenquan,Zhu Jiajun,Jiang Lin 대한독성 유전단백체 학회 2024 Molecular & cellular toxicology Vol.20 No.2

Emerging evidence suggests that rhaponticin, a stilbene monomeric compound isolated from North China rhubarb, has been shown to exhibit significant biological activity against tumors. However, the anticancer effects and mechanisms of rhaponticin in tongue squamous cell carcinoma (TSCC) remain elusive. We investigated the changes of migration and invasion abilities and EMT progression of TSCC cells treated with different concentrations of rhaponticin under hypoxia, as well as the possible mechanisms, in order to initially explore the effects of rhaponticin on the biological characteristics of TSCC cells under hypoxia. The number of cell migration and invasion was prominently increased, E-cadherin protein was down-regulated, and N-cadherin and HIF-1α protein expression was elevated under hypoxia. Rhaponticin intervention strikingly prevented the increased abilities of migration and invasion and EMT of TSCC cells under hypoxia. This was followed by further validation finding that rhaponticin indeed leads to reduced HIF-1α post-transcriptional activity. Mechanistically, rhaponticin may bind to aryl-hydrocarbon nuclear translocator (ARNT) domain of HIF-1α.Rhaponticin repressed the invasion and migration abilities and EMT process of TSCC cells under a hypoxic environment in vitro by targeted suppression of HIF-1α. On the Surface Tension and Coulomb Energy of Neutron-star Ma

Yuan-Bin Wu 한국물리학회 2014 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.65 No.6

We briefly report the results from a study of the structure, the surface and the Coulomb energyof the core-crust transition layer of neutron stars with global charge neutrality. The results showthat the surface tension is a power-law function of the baryon number density in the core region. We also present analyses of the influence of the gravitational field on the transition layer andthe instability against Bohr-Wheeler surface deformations in the case of neutron stars obeyingglobal charge neutrality. If the core-crust transition is assumed to occur at the nuclear densitycore 2.7×1014 g cm−3, the instability sets the upper limit to the crust density, critcrust 1.2×1014g cm−3.

        Yuan Chen,Xiaoqing Wu,Junfu Wei,Hailiang Wu 한국섬유공학회 2020 Fibers and polymers Vol.21 No.11

In order to nondestructively prepare polyamine modified polyethylene glycol terephthalate (PET) fabric, codepositionof catechol and polyethyleneimine onto PET fiber was conducted by the means of mussel-inspired method. Thesurface morphology, composition of woven fabric were investigated by FESEM and ATR-FTIR, XPS, respectively, whoseresults demonstrated successful modification has taken place on PET fiber surface. The TG-DSC data proved that the meltingtemperature and decomposition temperature increased from 253.78 oC and 394.08 oC to 255.01 oC and 398.21 oC,respectively. The surface polarity was characterized through the dynamic contact angle. The results indicated that the fabricdisplayed optimal surface properties at the PEI-CA mass ratio of 1:1. The modified PET fabric was utilized to prepare thefiber reinforced epoxy composite and its mechanical performances including tensile, flexural and short beam shear strengthwere evaluated. The obtained results indicated that the interlaminar shear strength (ILSS) has been significantly improved by57.02 %. The novel way to modify fabrics has great potential in recycling waste fabric for preparing fiber reinforcedcomposite.
