        KRICT Pervaporation 분리막

        A PVA pervaporation membrane, which has preferential selectivity for water, was introduced. For the separation of ethanol-water mixtures above 90 wt% of ethanol, the membrane exhibited selectivity above 50 and permeability of 0-0.4 ℓ/hr·㎡, The membrane surface area required for the production of 360 ㎏/hr absolute ethanol from 93 wt% ethanol-water mixture by the pervaporation process using the membrane was estimated as 163 ㎡. In this case, the average ethanol concentration was calculated as 13.5 wt%. The energy cost of pervaporation process was compared with that of azeotropic distillation process using benzene as an entrainer. As a result, it is confirmed that the energy cost of pervaporation process is merely 115 of that of azeotropic distillation process.

        Improved Production Efficiencies of Various Adeno-Associated Virus (AAV) Serotypes and a Novel Universal AAV Titration Method

        AAV는 매우 안전하고 효율적인 유전자전달방법으로 인정되어 왔다. 그러나, AAV가 가진 생물의약품으로서 단점은 효율적이고 재현성 높은 생산법이 취약하고, 또한 다양한 혈청형을 간단한 한 가지 공통적 방법으로 신뢰성 있게 정량하는 방법이 개발되어야 하는 것이다. 따라서, 본 연구에서는 AAV2와 아데노바이러스를 동시에 감염하는 종래의 생산법에 의한 효율성과 새로운 생산법, 즉 AAV2 감염 후 pHelper 플라스미드를 transfection 하는 방법을 통한 생산효율성을 비교하였고, HEK293과 293T를 생산세포주로 하여 시간에 따른 생산효율성도 분석하였다. 그 결과 AAV2와 pHelper DNA를 포함한 새로운 생산법은 기존의 방법에 비해 10배 이상 높은 생산효율성을 보였고, 293T에서 AAV2를 10 MOI로 감염한 후 5일째에 가장 높은 생산효율성을 보였는데, 생산세포 한개 당 1.61×10? virus genomes (v.g.)을 생산하는 결과였다. 따라서 이 생산조건을 다른 혈청형 생산에 적용한 결과, 모든 혈청형에서 생산세포주 한 개 당 10? v.g. 이상을 생산하는 효율성을 보였다. 한편, 다양한 AAV 혈청형을 한 가지 공통적인 방법으로 정량하기 위해 the universal PCR 프라이머를 제작하였고, 그것을 이용하여 신뢰성 높고 10개 분자까지도 증폭이 가능한 결과를 모든 혈청형에서 얻었다. 그러므로 이 한 쌍의 정량용 the universal 프라이머는 임상시험용 아데노바이러스벡터에 존재하는 AAV오염을 검출하는 것에도 사용 가능하다. Adeno-associated virus (AAV) has been considered to be a very safe and efficient gene delivery system. However, the major obstacles to therapeutic usage of AAV have been to achieve highly efficient and reproducible production processes, and also to develop a reliable quantifying method of various serotypes with a simple protocol. We compared the efficiency of the conventional production protocol of AAV2 and adenovirus (Ad) co-infection to that of a new method containing AAV2 infection followed by pHelper transfection. We tested HEK293 and 293T, and further examined the time-dependent changes of AAV2 production. The new method of AAV2 and pHelper DNA gave about ten times higher production efficiency than that of the conventional protocol. The highest production efficiency in 293T was achieved as 1.61 × 10? virus genomes (v.g.)/cell by the new method of 10 MOI of AAV2 infection and 5 days post-infection. This protocol of the highest efficiency was then applied to produce various AAV serotypes and showed the efficiencies higher than 10? v.g./cell. Next, we designed the universal PCR primers of highly conserved regions for various AAV serotypes to develop a simple and reliable titration method. The universal primers could amplify all the tested AAV serotypes with similar sensitivities by ten molecular copies. Therefore, this pair of universal primers can be further utilized to detect AAV contaminants in therapeutic adenoviral vectors.

        Renal failure caused by a wide variety of etiological factors is characterized by progressive and irreversible deterioration of nephrons in the kidney. Despite the diversity of renal diseases, the appearance of inflammatory cells at the site of tissue injury is the hallmark of almost all renal diseases. Humoral and cell-mediated immune responses and soluble mediators play pivotal roles in the progression of renal injury. The utility of IL-10, a cytokine with potent anti-inflammatory activity, in the treatment of glomerulosclerosis in FGS/Kist mice in which renal disease occurs naturally with agin was studied. The group treated with recombinant anenoviruses encoding IL-10 was shown to have significant suppression of glomerular sclerotic index and reduced proteinuria when compared to LacZ or sham controls. Practical therapies are not currently available, but candidates for molecular therapeutics for renal diseases include vector-based gene therapy and the use of molecular compounds such as anti-inflammatory cytokines. There is a compelling need to improve vector systems, to control transgene expression, and to achieve cell-specific or localized targeting in gene therapy for renal disease.

        Pervaporation 공정 Scale - Up 에 있어서의 분리막 표면적의 계산방법

        이성분 혼합물의 pervaporation에 있어서, 실험실적 규모로 측정된 공급액 조성에 따른 투과액 조성 변화 및 투과속도 변화 자료를 이용하여 scale-up시에 필요로 하는 분리막 표면적과 투과액의 평균 농도를 산출할 수 있는 방법을 제안하였다. In pervaporation of binary mixtures, a method has been proposed for the calculations of membrane surface area and average permeate composition when scaling up. For the method required are data on permeate composition and permeate flux measured as a function of feed composition in laboratory-scale experiments.

        For remove Volatile Organic Compounds(VOCs) in water which occur water pollution, siloxane-imide copolymer membranes with perfluorinated side group were prepared and were investigated good physical properties and perm-selectivities. As getting the portion of siloxanediamine in copolymer membrane, it has the higher selectivities and the better permeation properties of VOCs. and was found the superior permeation characteristic of the hydrophobic toluene.

      • 불소기를 포함한 폴리이미드막의 혼합에 따른 물성과 기체투과특성

        The effects of incorporation of fluorinated alkyl side groups into polyimide membranes were investigated in terms of their physical and gas permeation properties. Four polyimides with fluorinated side groups and four polyimides without the side groups were prepared by polycondensation of 2-(perfluorohexyl)ethyl-3, 5-diamino benzoate (PFDAB) and m-PDA with four aromatic dianhydrides(6FDA, ODPA, BTDA and PMDA), respectively. It was found that the incorporation of fluorinated side groups into the polyimide membranes decreased their T_(g)s, surface free energies, solubility parameters and fractional free volume(FFV)s and therefore, enhanced the permeabilities for CO₂, O₂, N₂ and CH₄ gases but reduced the selectivities for CO₂/ CH₄, O₂ /N₂ , CO₂/N₂ gas pairs depending upon the structure of dianhydride monomers.

        MMA 와 FA 공중합체 분리막 제조 및 기체투과 특성

        methyl methacrylate(MMA)와 fluoro acrylate(FA)를 공중합하여 제조한 분리막으로 다양한 기체들에 대한 기체투과 실험을 수행하였다. 특히 분리막을 통한 기체투과도에 미치는 불소기의 영항을 FA의 함량을 변화시키면서 조사하였다. FA의 함량이 증가함에 따라 분리막의 표면 자유에너지는 상당히 감소함을 알 수 있었으며, 그 결과로 기체의 투과도와 선택도가 특이한 값을 나타내는데 큰 영향을 미침을 알 수 있었다. P(MMA-FA)분리막들의 투과도는 H₂, CO₂, O₂, N₂, CH₄ 등 다양한 기체들에 대하여 PMMA 분리막에 비하여 2-20배 이상의 값을 나타내었고, 선택도는 PMMA 분리막들과 거의 동등한 값을 유지하였다. FA의 최적 함량은 1-5 wt%의 범위인 것을 알 수 있었다 . The gas permeation was experimentally carried out for the various gases with membranes made of methyl methacrylate(MMA) and fluoro acrylate(FA) copolymer. Particularly the effect of fluoro functional group on the gas permeability of the membrane was investigated with various FA contents. It turned out that the surface free energy of the membrane was significantly decreased as the FA content increased, resulting in very unique properties with respect to the permeability and the selectivity of gas. P(MMA-FA) membranes showed that its permeability was 2 to 20 times higher than that of the PMMA membrane for the typical gases of H₂, CO₂, O₂, N₂ and CH₄ while its selectivity was kept to be almost comparable to those of the PMMA membrane. The optimum content of FA was found to be in the range of 1 to 5 wt%.
