        Upregulation of miR-223 in the rat liver inhibits proliferation of hepatocytes under simulated microgravity

        Long-term spaceflight affects numerous organ systems in the body, including metabolic dysfunction. Recently, ample evidence has demonstrated that the liver is a vulnerable organ during spaceflight. However, the changes in hepatocyte proliferation and cell cycle control under microgravity remain largely unexplored. In the present study, we first confirmed that the serum levels of aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase, biochemical markers of liver function, were altered in rats under tail suspension (TS) conditions to simulate microgravity, as shown in previous reports. Next, we demonstrated that the cell proliferation activity, determined by Ki67, PCNA and PH3, was significantly decreased at the different TS time points (TS for 14, 28 and 42 days) compared with that in the control group. Consistently, the positive cell cycle regulators Ccna2, Ccnd1, Cdk1, Cdk2 and cyclin D3 were also significantly decreased in the TS groups as shown by quantitative real-time PCR and western blotting analysis. Subsequent analysis revealed that the aberrant hepatocyte proliferation inhibition under simulated microgravity was associated with the upregulation of miR-223 in the liver. We further found that miR-223 inhibited the proliferation of Hepa1-6 cells and identified CDK2 and CUL1 as its direct targets. In addition, the decreased expression of CDK2 and CUL1 was negatively correlated with the level of p27 in vitro and in vivo, which may have been responsible for retarding hepatocyte proliferation. Collectively, these data indicate that upregulation of miR-223 was associated with the inhibition of liver cell growth and reveal the role of miR-223 in rat hepatocyte proliferation disorders and the pathophysiological process under simulated microgravity.

        Grub polypeptide extracts protect against oxidative stress through the NRF2-ARE signaling pathway

        Grub polypeptide extracts (GPEs) have antioxidant effects; however, their underlying molecular mechanisms are unknown. This study explored the antioxidant molecular mechanism of GPE via the nuclear factor-erythroid 2-related factor 2 (NRF2)-antioxidant response element (ARE) signaling pathway in C2C12 muscle satellite cells exposed to oxidative stress. The effects of GPE/or H2O2 on C2C12 were investigated by the MTT (3- (4,5-dimethylthiazol-2-yl)-2,5- diphenyltetrazolium bromide) viability assay and immunofluorescence and small interfering RNA (siRNA) analyses. The cell viability, cell damage, intracellular reactive oxygen species (ROS) levels, and NRF2 signaling pathways related to proteins were measured. GPE significantly increased the antioxidant capacity of cells, evident by increased cell viability and decreased lactate dehydrogenase leakage, DNA damage, malondialdehyde content, and ROS level. GPE also markedly increased mRNA expression levels and activities of antioxidant enzymes including superoxidase 1 and 2, catalase, and glutathione peroxidase. In addition, GPE increased the gene and protein expression of NRF2 and heme oxygenase 1 by promoting NRF2 translocation from the cytoplasm to the nucleus and activating NRF2-ARE signaling pathways. The antioxidant effects of GPE through these signaling pathways were further confirmed by NRF2-specific siRNA silencing. Thus, GPE enhances antioxidant capacity and alleviates oxidative damage of C2C12 cells via the NRF2-ARE signaling pathway.

        Separation of valuable metals from mixed cathode materials of spent lithium-ion batteries by single-stage extraction

        With the intensity of resource scarcities and environmental problems, the disposal and recovery of spent lithium-ion batteries, especially recovery of valuable metals, becomes vital. In this work, a method of co-extracting nickel, cobalt, manganese and being separated from lithium by single-stage solvent extraction is proposed. The extraction and separation process of D2EHPA was studied. The effects of extractant concentration, saponification percentage, extraction time and O:A on the extraction efficiency of D2EHPA were systematically studied. Nearly 100% manganese, 94% cobalt and about 90% nickel were co-extracted and separated from lithium using D2EHPA in kerosene by single-stage extraction. The maximum value of separation factors (Ni/Li, Co/Li and Mn/Li) was 13.03, 23.42 and 1904.24. The mathematical model of extraction of four ions was developed by combination of Levenberg-Marquardt method and Universal Global Optimization method. The proposed extraction model accurately fits the experimental data and helps to predict the extraction efficiency of each metal under the corresponding conditions.

        Optimization of bayberry juice spray drying process using response surface methodology

        In this study, thirteen different runs according to the central composite design-response surface methodology were used to optimize the quality parameters of spray-dried bayberry juice powder. The independent variables were different levels of inlet air temperature (145.8–174.1 °C) and maltodextrin concentration (22.9–37.0%). The responses were process yield, moisture content, hygroscopicity, glass transition temperature (Tg), total color difference (ΔE), redness, retention of phenolics and anthocyanins, and antioxidant capacity. The optimum operation conditions for the highest Tg, redness, anthocyanins retention and antioxidant capacity with the lowest ΔE and hygroscopicity were obtained at inlet drying temperature of 150 °C and maltodextrin concentration of 31%. This study revealed that by applying these optimal conditions, bayberry juice powder with a 74.16% yield, 3.15% moisture content, 10.25% hygroscopicity, 41.15 °C Tg, 24.74 ΔE, 27.45 Hunter a redness, 89.55% anthocyanins retention, 77.71% phenolics retention, and 30.19 mmol Trolox equivalents/g antioxidant capacity was produced.

        Double-Blind Compact E-cash from Bilinear Map

        Compact E-cash is the first scheme which can withdraw 2l coins within (1) operations and then store them in ( ) bits. Because of its high efficiency, a lot of research has been carried out on its basis, but no previous research pay attention to the privacy of payees and in some cases, payees have the same privacy requirement as payers. We propose a double-blind compact E-cash scheme, which means that both the payer and the payee can keep anonymous while spending. In our scheme, the payer and the bank cannot determine whether the payees of two different transactions are the same one and connect the payee with transactions related to him, in this way, the privacy of the payee is protected. And our protocols disconnect the received coin from previous transaction, then, the coin can be transferred into an unspent coin which belongs to the payee. The proposed e-cash scheme is secure within y-DDHI and LRSW assumption.

        Reliable Localization for Wireless Sensor Networks in Complex Environments

        Although localization has been widely studied for Wireless Sensor Networks (WSNs), the complex environments and the large network scale pose severe challenges and make it necessary to develop new reliable localization algorithms. In this paper, we propose a novel Multi-Hop Localization Algorithm for large-scale WSNs in complex environments. This work is based on the consideration that the localization process would encounter several kinds of adverse factors with different nature at the same time (e.g. anisotropic network characteristics, ranging uncertainty, link quality of multihop paths, etc.), which lead to obvious degradation of localization performance. Unlike most of the existing schemes, we transform the localization problem in complex environments into a hybrid constraint satisfaction problem (CSP) which is composed of three different kinds of constraints, i.e. spatial constraint, network situation constraint, and confidence constraint. Set-membership approach and interval analysis method have been utilized to deal with the CSP and determine the positions of sensor nodes. Simulation results show that our scheme is an effective and efficient approach to localization in large-scale WSNs.

        Mechanical Characteristics of Hybrid-Fiber-Reinforced Concrete Shaft Wall Structure Under Uneven Load

        To prevent damage to shaft walls in mines due to uneven loading, the mechanical properties of a shaft wall structure made of hybrid-fiber-reinforced concrete were studied. First, through orthogonal testing, the optimal mix proportion of the hybrid-fiber-reinforced concrete was obtained. Subsequently, a numerical calculation model of the shaft wall structure under uneven loading was established. The calculation results showed that the structure exhibits a tensile stress under the action of uneven loading and that its bearing capacity can be improved using the hybrid-fiber-reinforced concrete. Based on the numerical simulation results, a calculation formula for the bearing capacity of the shaft wall was obtained by regression. Finally, a model test was conducted on the shaft lining structure. The results obtained were consistent with those obtained using the numerical simulation regression formula, confirming the reliability of the numerical simulation results. This study showed that a hybrid-fiber-reinforced concrete shaft wall structure can better withstand uneven loads and has improved brittle failure characteristics. Hybrid-fiber-reinforced concrete is an excellent material for deep shaft wall structures.

        Estrogen receptor β promotes bladder cancer growth and invasion via alteration of miR-92a/DAB2IP signals

        Although early studies suggested that bladder cancer (BCa) is more prevalent in men than in women, muscle-invasive rates are higher in women than in men, suggesting that sex hormones might play important roles in different stages of BCa progression. In this work, we found that estrogen receptor beta (ERβ) could increase BCa cell proliferation and invasion via alteration of miR-92a-mediated DAB2IP (DOC-2⁄DAB2 interacting protein) signals and that blocking miR- 92a expression with an inhibitor could partially reverse ERβ-enhanced BCa cell growth and invasion. Further mechanism dissection found that ERβ could increase miR-92a expression at the transcriptional level via binding to the estrogen-response-element (ERE) on the 5′ promoter region of its host gene C13orf25. The ERβ up-regulated miR-92a could decrease DAB2IP tumor suppressor expression via binding to the miR-92a binding site located on the DAB2IP 3′ UTR. Preclinical studies using an in vivo mouse model also confirmed that targeting this newly identified ERβ/miR-92a/ DAB2IP signal pathway with small molecules could suppress BCa progression. Together, these results might aid in the development of new therapies via targeting of this ERβ-mediated signal pathway to better suppress BCa progression.

      • Computational and mathematical simulation for the size-dependent dynamic behavior of the high-order FG nanotubes, including the porosity under the thermal effects

        Some researchers pointed out that the nonlocal cantilever models do not predict the dynamic softening behavior for nanostructures (including nanobeams) with clamped-free (CF) ends. In contrast, some indicate that the nonlocal cantilever models can capture the stiffness softening characteristics. There are substantial differences on this issue between them. The vibration analysis of porosity-dependent functionally graded nanoscale tubes with variable boundary conditions is investigated in this study. Using a modified power-law model, the tube's porosity-dependent material coefficients are graded in the radial direction. The theory of nonlocal strain gradients is used. Hamilton's principle is used to derive the size-dependent governing equations for simply-supported (S), clamped (C) and clamped-simply supported (CS). Following the solution of these equations by the extended differential quadrature technique, the effect of various factors on vibration issues was investigated further. It can be shown that these factors have a considerable effect on the vibration characteristics. It also can be found that our numerical results can capture the unexpected softening phenomena for cantilever tubes.

        Sublethal and transgenerational effects of six insecticides on Bactrocera dorsalis (Hendel) (Diptera: Tephritidae)

        To clarify the effects of sublethal concentration of insecticide on the growth, development and reproduction of Bactrocera dorsalis (Hendel) (Diptera: Tephritidae), the sublethal and transgenerational effects of the LC 30 of six insecticides on adults and progeny (F 1 and F 2 generations) were studied using age-stage, two-sex life tables. Exposure to six insecticides had significant negative impacts on the life-history traits of B. dorsalis and led to reduced adult longevity and fecundity in F 0 , a longer larval stage duration in F 1 , and reduced fertility and survival. The intrinsic rate of increase (r), finite rate of increase (λ), and net reproductive rate (R 0 ) were reduced in F 1 , with the greatest reduction observed in the chlorpyrifos treatment. In the F 2 generation, the adverse effects were weakened, only the cyantraniliprole treatment had lower values than the control, and the fecundity of other treatments returned to normal. Obvious “hormesis” phenomena were not observed in any of the generations. Based on the population hazard index, chlorpyrifos had the best control effect in the early stage, and cyan traniliprole had the best control effect in the long term. These results indicate that the LC 30 of the six insecticides exert different transgenerational effects on the B. dorsalis population.
