Transient gene expression and promoter analysis in basidiomycete

Ceriporiopsis subvermispora is a unique white rot fungus degrading plant cell wall lignin without severe loss of cellulose. Recombinant plasmids containing homologous gene expression signals fused to the coding sequence of Escherichia coli hph which encodes for hygromycin phosphotransferase were introduced in protoplasts of wild type C. subvermispora using PEG/CaCl2 protocol. A number of hygromycin B resistant strains were isolated on a screening plate containing 100㎍/ml of hygromycin B: whereas, no colonies were observed when protoplasts were treated with no DNA as a negative control. It was demonstrated that most of the isolates lost their drug resistance during successive cultivations in the presence of the same concentration of hygromycin B, but some of the isolates showed stable drug resistance after five times repeated screening. They did not lose the drug resistance even after the cultivation in the absence of hygromycin B and incorporation of the hph sequence was confirmed by specific amplification of the target sequence in PCR experiments and Southern hybridization analysis. The stable transformation system will make it possible to do molecular genetic analysis, as well as breeding of genetically modified strains, in C. subvermispora. Moreover, it was demonstrated that recombinant constructs with truncated promoter showed reduced number of the drug resistant isolates on the first screening plate, in response with the length of the remaining promoter sequence. These findings indicated that unstable drug resistance observed in these isolates should originate from transient expression of the introducing marker genes, and that a promoter assay system has been developed for the first time in basidiomycete. This system is practically not affected by the positional effect of the integrated recombinant gene in the host chromosome.

Prolonged air leakage is a major cause of morbidity in pneumothorax. When conservative management is not effective, surgery should be performed. However, surgery is not appropriate in patients with low pulmonary function. In these patients, occlusion of the airway with endobronchial blockers may be attempted under bronchoscopy. We treated two patients with prolonged air leakage using endobronchial Watanabe spigots under fibrobronchoscopy.
Development of a Test Method for Assisted Walking Trolleys

Manufacturers have recently been developing assistive walking devices with electric motors, based on rollators and walking trolleys, in Japan. These assistive devices have electric motors to assist walking on sloped surfaces. Furthermore, they have a function that automatically suppresses the speed on downhill slopes. Here a walking experiment was conducted to investigate the speed at which the suppression function needed to be applied in various walking conditions. The subjects in the experiment walked while leaning on the assistive device, applying forces of 37 (horizontal) and 204 N (vertical) on downhill slopes. Furthermore, an analysis of the walking data obtained from traditional walking trolleys revealed that 80% of the subjects walked at a speed of 2.5 km/h or more. Two experiments were conducted to develop a test method that analyzed the performance of the suppression function on downhill slopes. The proposed test method results yielded sufficient accuracy.

<P>Experimental observations on the B<SUB>4</SUB> phase of liquid crystal mixtures formed from bent-core (P-8-O-PIMB) and rod (5CB) achiral molecules reveal the stabilization of the B<SUB>4</SUB> phase with an unusually large size of chiral domains. The phenomenon is interpreted as an intrinsic property of the B<SUB>4</SUB> phase when arising below the isotropic liquid (Iso); actually direct transition between Iso and B<SUB>4</SUB> phases occurs except for only very limited regions of pure P-8-O-PIMB and 5CB molecules. We have developed a new HD-2700 (Hitachi High-Technologies Corp., Japan) scanning transmission electron microscope (STEM) that includes an automatic aberration correction function, and a large-solid-angle energy-dispersive X-ray spectroscopy detector that enables high-resolution and sensitive analysis. For observation with atomic resolution, using spherical-aberration-corrected STEM, in order that satisfactory performance of the device can be achieved readily, and within a short time, irrespective of the operator's skill level, a spherical-aberration-correction device with an automatic aberration-correction function was developed. This automatic aberration-correction function carries out the entire correction-related process (aberration measurement, selection and correction) automatically, with automatic selection of the aberrations that require correction, and automatic measurement of the appropriate corrections.

Experimental investigations of interlaminar mechanical properties for carbon fiber reinforced plastic (CFRP) laminates were carried out using aramid fiber ($Kevlar^{(R)}$-29 1000d) and carbon fiber (TR40-1K 612d, Mitsubishi Rayon) stitching. Various carbon fiber (CF) stitch densities were used to prepare a number of CF stitched CFRP laminates for double cantilever beam (DCB) tests. An insert tongue-type loading fixture, developed by the Japan Aerospace Exploration Agency (formerly the National Aerospace Laboratory of Japan), was also employed in the DCB test. Interlaminar tension tests were carried out under an out-of-plane directional loading using a single CF stitch thread in the CFRP laminates. The DCB test results clarified that the relationship between the volume fractions of the CF stitch thread ($V_{ft}$) and mode I critical energy release rate ($G_{Ic}$) showed a mostly linear function with a higher gradient than that of the $Kevlar^{(R)}$ stitched CFRP laminates. The CF stitched CFRP tension test results indicated that the consumption energy per unit area ($E_i$) was larger than that of $Kevlar^{(R)}$ stitched CFRP laminates.
