        Identification of a novel prognostic signature composed of 3 cuproptosis-related transcription factors in colon adenocarcinoma

        Background Since the mechanism of cuproptosis was recently revealed, many molecules related to this pathway have been widely concerned and exploited to have prognostic potential. However, it is still unknown whether the transcription factors related to cuproptosis could be competent as tumor biomarkers of colon adenocarcinoma (COAD). Objective To analyze the prognostic potential of cuproptosis-related transcription factors in COAD, and validate the representative molecule. Methods Transcriptome data and patients’ clinical parameters were obtained from the TCGA and GEO database. 19 cuproptosis genes were identified through literature consulting. Cuproptosis-related transcription factors were screened by COX regression analyses. Multivariate Cox regression was applied to construct the signature. Prognostic effects were evaluated by Kaplan Meier survival analyses and ROC analyses. KEGG, GO, and ssGSEA analyses were performed for function prediction. 48 COAD tissues were collected for immunohistochemistry stain to observe the expression level and prognostic value of E2F3. qRT-PCR was performed to detect mRNA expression levels, while cell viability assay was applied to detect the response of COAD cells to elesclomol treatment. Results A novel signature based on 3 prognostic transcription factors related to cuproptosis was successfully established and verified. Patients in the low-risk group tended to have better overall survival and lower immune phenotype scores than those in the high-risk group. Meanwhile, we also constructed a nomogram based on this signature and predict 10 candidate compounds targeting this signature. As an essential member of this signature, E2F3 was confirmed to be overexpressed in COAD tissues and was associated with poor prognosis of COAD patients. Importantly, CuCl2 and cuproptosis inducer elesclomol treatment could increase the expression of E2F3 in COAD cell while the overexpression of E2F3 significantly enhanced the resistance of COAD cells to elesclomol treatment. Conclusion Our research has identified a new prognostic biomarker and provides some innovative insights into the diagnosis and therapy of patients with COAD.

      • A Novel Fixed Twin-Rotor Unmanned Aerial Vehicle with Variable Angle Louver Rudder

        A novel fixed twin-rotor unmanned aerial vehicle (UAV) based on the variable angle louver rudder (VALR) is developed in this work. In view of the mechanical characteristics under the action of the louver rudder, specific kinematics and dynamics models are derived. The aerodynamic characteristics of the UAV are studied through computational fluid dynamics simulation, and a ground measurement system is designed to analyze the aerodynamic characteristics of the prototype. An attitude control model is proposed, and maneuvers such as take-off, landing, hovering, and braking are realized through flight experiments, which verify the feasibility of the prototype design. This research shows that the new layout with the VALR system, combined with the advantages of rudder control and vector control, can reduce the structural complexity and deadweight of the UAV while ensuring flight stability and improving maneuverability. Moreover, the novel UAV has higher energy efficiency and economy.

        Experimental study and analysis on physicochemical properties of coal treated with clean fracturing fluid for coal seam water injection

        In the process of coal seam water injection, the components of different fracturing fluids have differenteffects on coal. To analyze the effect of water injection fracturing fluid on the physical and chemical propertiesof coal, we studied the relationship between guar-based fracturing fluid and cationic clean fracturingfluid (C-VES). Microscopic experiments show that guar-based fracturing fluid has a certain viscosityeffect on coal, while C-VES can adsorb and capture the primary coal dust in coal. FTIR analysis shows thatC-VES has a great change on the content of coal functional groups, and guar-based fracturing fluid haslittle effect on coal. Industrial analysis shows that C-VES can better reduce the ash content of coal andhas a certain impact on the thermal efficiency of coal than guar based fracturing fluid. Combustion characteristicexperiment shows that guar-based fracturing fluid does not affect the combustion effect of coal,and C-VES can expand the exothermic temperature range of coal but has little effect on the main combustionof coal. Molecular dynamics simulation results show that C-VES has the best wetting effect oncoal. This research provides theoretical guidance for dust prevention and improving the use and conversionefficiency of coal.

        Porcine parvovirus nonstructural protein NS1 activates NF-κB and it involves TLR2 signaling pathway

        Background: Porcine parvovirus (PPV) is a single-stranded DNA virus that causes porcine reproductive failure. It is of critical importance to study PPV pathogenesis for the prevention and control of the disease. NS1, a PPV non-structural protein, is participated in viral DNA replication, transcriptional regulation, and cytotoxicity. Our previous research showed that PPV can activate nuclear factor kappa B (NF-κB) signaling pathway and then up-regulate the expression of interleukin (IL)-6. Objectives: Herein, the purpose of this study is to determine whether the non-structural protein NS1 of PPV also has the same function. Methods: Real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR), enzyme-linked immunosorbent assay, western blot, immunofluorescence assay and small interfering RNA (siRNA) were used. Results: Our findings demonstrated that PPV NS1 protein can up-regulate the expression levels of IL-6 and tumor necrosis factor-alpha in a dose-dependent manner. Moreover, PPV NS1 protein was found to induce the phosphorylation of IκBα, then leading to the phosphorylation and nuclear translocation of NF-κB. In addition, the NS1 protein activated the upstream pathways of NF-κB. Meanwhile, TLR2-siRNA assay showed TLR2 plays an important role in the activation of NF-κB signaling pathway induced by PPV-NS1. Conclusions: These findings indicated that PPV NS1 protein induced the up-regulated of IL-6 expression through activating the TLR2 and NF-κB signaling pathways. In conclusion, these findings provide a new avenue to study the innate immune mechanism of PPV infection.

        Let A_1 and A_2 be n × n nonzero complex matrices, denote a linear combination of the two matrices by A = c_1A_1 + c_2A_2, where c_1, c_2 are nonzero complex numbers. In this paper, we research the problem of the linear combinations in the general case. We give a sufficient and necessary condition for A is an involutive matrix and s+1-potent matrix, respectively, where A_1 is a tripotent matrix, with A_1A_2 = A_2A_1.       • Big Data Analytics of Multi-Relationship Online Social Network Based on Multi-Subnet Composited Complex Network

        Online social networks such as Twitter and Facebook are becoming popular form of social information networks. There are frequently many kinds of relationships in an online social network. Complex Network acting as one kind of big data technologies is often used to analyze users' social activities. By studying the Douban network, which is a representative multi-relationship online social network in China, big data of friendship relationship and book comments similar relationship are crawled through network topology measurement software, from the perspective of topological characteristics of complex network, the basic topologies of the two relationship networks constructed individually by the two relationships are analyzed. Based on these, a multi-relationship online social network based on Multi-subnet Composited Complex Network Model is constructed through loading book comments similar relationship subnet to follower relationship subnet, and accurate understanding of topologies of Douban multi-relationship network is obtained. These findings provide a deep understanding on the evolution of multi-relationship online social network, and can provide guidelines on how to build an efficient multi-relationship online social network evolution model.

      • A High Security Architecture for LTE Core Network Server

        With the expansion of wireless network technologies and the emergence of novel mobile applications, the 3rd Generation (3G) communication is moving to the 4th Generation (4G) communication. Comparing to preceding versions, one elementary difference is that 4G wireless networks will operate entirely on the TCP/IP, which would cause greater risks in terms of safety and reliability. In this paper, a new high security architecture for Long Term Evolution (LTE) core network server is proposed and designed. There are asynchronous array of simple processors and two physically isolated high-speed system buses in the security architecture, which ensure only one bus can be connected to array of processors at the same time. Experiment results show that the security architecture can effectively prevent external threats from accessing network resources.

        Experimental Analysis of the Mechanical Properties of Austenitic S30408 Stainless Steel Welded Joints at Low Temperatures

        Austenitic S30408 stainless steel exhibits good low-temperature resistance and good welding performance. This steel is often used in liquefied natural gas stainless steel storage tanks. During the construction process, the tank wall is primarily connected by butt weld joints. Because welded joints are easily affected by temperature, low-temperature weld cracking can reduce the safety of structures. To study the cryogenic mechanical properties of austenitic S30408 stainless steel welded joints at low temperatures, the low-temperature mechanical properties of austenitic S30408 stainless steel base metal and welded joint components were studied by tensile tests from − 60 to 20 °C and scanning electron microscopy analysis of fractures at various temperatures. The results show that when the temperature decreases, the stress–strain curve of base metal components changes from a power function type to an inverted "s" type; in addition, secondary hardening occurs. The yield strength and tensile strength of the welded joint and base metal increased with decreasing temperature, and the elongation and reduction of area decreased. The plastic deformation capacity of the welded joint was significantly lower than that of the base metal, and there were obvious inclusions in the microstructure.

        Single-cell characterization of macrophages in uveal melanoma uncovers transcriptionally heterogeneous subsets conferring poor prognosis and aggressive behavior

        Uveal melanoma (UM) is the most frequent primary intraocular malignancy with high metastatic potential and poor prognosis. Macrophages represent one of the most abundant infiltrating immune cells with diverse functions in cancers. However, the cellular heterogeneity and functional diversity of macrophages in UM remain largely unexplored. In this study, we analyzed 63,264 single-cell transcriptomes from 11 UM patients and identified four transcriptionally distinct macrophage subsets (termed MΦ-C1 to MΦ-C4). Among them, we found that MΦ-C4 exhibited relatively low expression of both M1 and M2 signature genes, loss of inflammatory pathways and antigen presentation, instead demonstrating enhanced signaling for proliferation, mitochondrial functions and metabolism. We quantified the infiltration abundance of MΦ-C4 from single-cell and bulk transcriptomes across five cohorts and found that increased MΦ-C4 infiltration was relevant to aggressive behaviors and may serve as an independent prognostic indicator for poor outcomes. We propose a novel subtyping scheme based on macrophages by integrating the transcriptional signatures of MΦ-C4 and machine learning to stratify patients into MΦ-C4-enriched or MΦ-C4-depleted subtypes. These two subtypes showed significantly different clinical outcomes and were validated through bulk RNA sequencing and immunofluorescence assays in both public multicenter cohorts and our in-house cohort. Following further translational investigation, our findings highlight a potential therapeutic strategy of targeting macrophage subsets to control metastatic disease and consistently improve the outcome of patients with UM.
