      • 肝吸蟲症 診斷에 있어서 ELISA법의 應用에 관한 硏究

        Human clonorchiasis is one of the most common trematode diseases and of very inportant public health porblems in Korea. Many techniques have been deviced for immunological diagnosis of clonorchiasis. But, these methods have been recognized as low specificity and sensitivity. In the present study the enzyme-linded immunosorbent assay(ELISA) test was demonstreted in the diagnosis of human clononchiasis using clonorchis VBS(veronal buffer solution) skin test antigen and peroxidase conjugated anti-human IgG, and ortho-phenylenediamine as a substrate by microminor modifications. The results obtained are as follows : 1. In the preliminary study optimal dilutions of conjugate, stock antigen and serum were 1/5,000, 1/400 respectively. 2. The O.D.(optical density) values of the ELISA in the sera of clonorchiasis cases using stock antigen and VBS skin test antigen were 1.347±0.728 and 2.332±0.420 respectively, and the mean O.D. value in the sera of clonorchiasis cases using VBS skin test antigen was higher than that of stock antigen in sensitivity. 3. During the storage in a refrigerator, the mean O.D. value of the ELISA in 138 sera of clonorchiasis cases was 1.674±0.601 in the storage for 1 month and that in 244 sera of clonorchiasis casse was 1,494±0.884 from 6 months to 3 years. 4. In the distribution of the O.D. Values of the ELISA in the sera of clonorchiasis cases, 115 (83.3%) out of 138 sera in the storage for 1 month and 161 (66.0%) out of 244 sera in the storage from 6 months to 3 years showed higher O.D. value than 1.000(O.D. at 488nm) which is referedn to the positive serum. 5. The O.D. values of the ELISA in the sera were compared with the degree of infection by E.P.G. (eggs per gram of feces) counts in the stools of the clonorchiais cases and the regression equation was applied for the evaluation: y=0.004x+1.6637 (x : 1/100 of E.P.G; Y : O.D. values) There was no correlation between the O.D. values of the ELISA and the number of egg In the results, the application of the ELISA in the diagnosis of human clonorchiasis using VBS skin test antigen within 1 month period storage seemed to be higher than stock antigen with long period storage in sensitivity. In conclusion, the present the present study suggests that the ELISA test can be applied in the diagnosis of human clonorchiasis as applicable and reproducible test in comparison with other immunological tests, especially in the sere-epidemiological study.

      • 방전간극내의 기체 압력변화에 따른 질소레이저의 동작특성

        질소레이저의 반복률과 출력 특성에 미치는 요소는 여러 가지가 있는바 방전간극내 기체압력 역시 커다란 영향을 미치고 있다. 본 연구는 이를 위하여 펄스형 전원, 다중 평행판 측전기형 Blumlein 전송회로, 고속고압 방전간극 그리고 길이 66cm의 레이저관으로 구성된 레이저를 제작하였으며 방전간극내에 주입된 질소기체의 압력을 1bar, 1.4bar, 1.8bar, 2.2ba로 변형시켜 가며 동작특성을 측정하였다. MCA와 검출기를 사용하여 측정된 최적동작특성은 방전간극거리 1.12mm에서 방전간극내 질소기체압력이 2.2bar, 레이저관내의 질소기체 압력이 60torr이었고 반복률과 안정도는 각각 90Hz, 2% 이었다. We constructed a nitrogen laser system which was consisted of pulse type high voltage power source, multiple parallel capacitor type Blumlein transmission line circuit, rapid high voltage spark gap, 66cm long laser tube and synchronized trigger system and studied on the operational characteristics of this laser when the pressure of nitrogen gas filled in the spark gap was changed into four steps as 1bar, 1.4bar, 1.8bar, 2.2bar. Using the MCA and silicon detector, the optimum operational characteristics were obtained to be nitrogen gas pressure of 2.2bar in spark gap spacing of 1.12mm, nitrogen gas pressure of 60 Torr in laser tube, repetition rate of 90Hz and stability of 2%.

      • Blumlein 전송회로에 의해 선전리되는 질소레이저의 동작특성 연구

        The design and construction of nitrogen laser preionized with a Blumlein transmission line circuit is described. For the power supply, we have used a pulse type high voltage power source that can be produce 10 joule energy in a pulse. The condition of maximum output power is observed at N_2 gas pressure of 100 torr, spark gap distance of 6.0 mm, repetition rate of 30Hz with preionization, and the stability increases by 0.7%. The maximum increment of output power is achieved at pressure of 200 torr when preionization is used. Finally we can increase the output power by approximately 60% by using this work, and the E/P value is 133 V㎝^(-1) torr^(-1).

        A preionized nitrogen laser with average output power of 3.0 mW in repetition rate of 30 Hz was constructed to be used as a pump for dye lasers such as rhodamine 6G, rhodamine B, coumarin 120 and coumarin 102. Under the optimum dye concentrations and wavelengths, dye laser average output power of rhodamine 6G, rhodamine B, coumarin 120 and coumarin 102 were obtained to be 1.6 mW, 1.1 mW, 1.1 mW respectively. According as nitrogen laser average output power was increased up to 76% by preionization effect, dye laser average output powers of rhodamine 6G, rhodamine B, coumarin 120 and coumarin 102 were increased up to 23%, 38%, 57% and 71% respectively. The nitrogen laser system used for this research can be utilized in dye lasers covering the entire visible spectra and if the nitrogen laser output power is improved, the output power of the coumarin 102 dye laser can be improved higher than that of the rhodamine 6G dye laser.

      • 肝吸蟲 感染家兎에 있어서 感染强度, 經過 및 治療에 따른 ELISA抗體價의 變動에 관한 實驗的 硏究

        Human clonorchiasis is one of the most common trmatode diseases and of very important pubic health problems in Korea. In the present study, the changing patterns of specific IgG antibody was demonstrated by the enzyme-linked immunosorbent assay(ELISA) in the sera rabbits infected with clonorchis sinensis. The O.D> (optical density) values of the ELISA in the sera were merwured according to intensity and course of infections and tendency of decrease after chemotherapy. The ELISA was performed with veronal buffered saline antigen of clonorchis, peroxidase conjugated antirabbit IgG and ortho- phenylendiamine as a substrate by micro method. In the 3 groups of rabbits infected with 50(A group), 500(B group) or 100×5(C group) metacercariae, the sera were prepared biweekly up to 18th week before chemotherapy, and biweekly up to 24th week after chemotherapy. The results were as follows: 1. O.D. values increased gradually with lapse of time before treatment. The O.D. values indicated firstly 1.000 or more in 8 weeks after infection in A group and 4 weeks after infection in both B and C groups. The O.D. values reached plateau at 10 weeks after infection in A group and 8 weeks after infection in both B and C groups. Regression equations between the O.D. values of the ELISA and period(week) after infection among 2 groups were as fllows, y=0.09x+0.319 in A group y=0.08x+ 0.826 in B group and y=0.08x+0.807 in C group. 2. The O.D. values in A group which had been treated decreased gradually and became preinfection level at 24 weeks after chemotherapy. On the contrary the O.D. values in A group which has not been treated showde plateau. The O.D. values in both B and C groups which had been cured decreased gradually and became preinfection level at 24 weeks after chemotherapy.

      • 電極間隙 變化에 따른 室素 LASER의 光出力 最適條件에 관한 硏究

        In this paper, we investigated the optical-power propensities form superradient nitrogen laser by varying the electrode-gap-spacing of laser tube by use of Blumlein transmission line which was constructed from 0.05 mm thick Al plates and 0.189 mm thick Mylar films; its capacitance was 90 nF identically. The water-cooled electrodes in laser tube are 660 mm in length and connected to the storage capacitor and the transmission line. We obtained the optimum condition of the nitrogen laser at electrode-gap-spacing of 3 mm, spark gap separation of 7.0 mm, laser tube pressure of 120 torr, charging voltage of 3850 volts and the repetition of 31 Hz with these conditions the values of E/P was about 108 volt cm^-1 torr^-1.
