        Highly Sensitive Detection of Low-Abundance White Spot Syndrome Virus by a Pre-Amplification PCR Method

        White spot syndrome virus (WSSV) is a major threat to the shrimp farming industry and so far there is no effective therapy for it, and thus early diagnostic of WSSV is of great importance. However, at the early stage of infection, the extremely low-abundance of WSSV DNA challenges the detection sensitivity and accuracy of PCR. To effectively detect low-abundance WSSV, here we developed a pre-amplification PCR (pre-amp PCR) method to amplify trace amounts of WSSV DNA from massive background genomic DNA. Combining with normal specific PCR, 10 copies of target WSSV genes were detected from ~10<sup>10</sup> magnitude of backgrounds. In particular, multiple target genes were able to be balanced amplified with similar efficiency due to the usage of the universal primer. The efficiency of the pre-amp PCR was validated by nested-PCR and quantitative PCR, and pre-amp PCR showed higher efficiency than nested-PCR when multiple targets were detected. The developed method is particularly suitable for the super early diagnosis of WSSV, and has potential to be applied in other low-abundance sample detection cases.

        Chinese herbal medicine (Bu-Shen-Tian-Jing Formula) for outcomes of IVF in Chinese patients with polycystic ovary syndrome: A retrospective cohort study

        Background: Polycystic ovary syndrome (PCOS) is one of the most common causes of anovulatory infertility. Chinese herbal medicine (CHM) has many advantages in treating PCOS. We conducted a retrospective cohort study to investigate the effects of CHM (Bu-Shen-Tian-Jing Formula, BSTJF) on the outcomes of IVF in Chinese patients with PCOS and the potential underlying mechanism. Methods: A total of 111 patients with PCOS who undergone IVF between November 2009 and July 2018 were included. Fifty-four patients received a three-month BSTJF therapy before controlled ovarian hyperstimulation, while the other 57 patients didn't. The data of the PCOS patients was collected. Anti-Müllerian hormone (AMH), growth differentiation factor-8 (GDF-8) levels in the follicular fluid were evaluated. Results: BSTJF helped patients with PCOS to get more retrieved oocytes (P<0.05) and fertilized oocytes (P<0.05). The clinical cumulative pregnancy rate, live birth rate, and term delivery rate were significantly higher in the same stimulated cycle of the PCOS patients with BSTJF treatment (P<0.05). No significant differences existed between the two groups in the rate of fertilization, hospitalization rate of ovarian hyper stimulation syndrome and obstetrical or neonatal complications. BSTJF significantly decreased the AMH levels in the follicular fluids (P<0.05). Conclusion: BSTJF significantly may improve the outcomes of IVF in Chinese patients with PCOS through decreasing AMH levels in follicular fluids.         Clinical Comparison of Proximal Gastrectomy With Double-Tract Reconstruction Versus Total Gastrectomy With Roux-en-Y Anastomosis for Siewert Type II/III Adenocarcinoma of the Esophagogastric Junction

        Purpose The incidence of adenocarcinoma of the esophagogastric junction (AEG) has increased in recent years, and the optimal surgical strategy for AEG remains highly controversial. We aimed to evaluate the safety and efficacy of proximal gastrectomy with double-tract reconstruction (PG-DT) for the treatment of patients with AEG. Materials and Methods We retrospectively analyzed patients with Siewert type II/III AEG between January 2013 and July 2018. Clinicopathological characteristics, survival, surgical outcomes, quality of life (QOL), and nutritional status were compared between the PG-DT and total gastrectomy with Roux-en-Y anastomosis (TG-RY) groups. Results After propensity score matching, 33 patients in each group were analyzed. There were no statistical differences between the 2 groups in terms of disease-free survival and overall survival. The surgical option was not an independent prognostic factor based on the multivariate analysis. In addition, no differences were found in terms of surgical complications. There were no significant differences in QOL assessed by the Visick grade, Gastrointestinal Symptom Rating Scale, or endoscopic findings. Furthermore, the long-term nutritional advantage of the PG-DT group was significantly greater than that of the TG-RY group. Conclusions PG-DT is a safe and effective procedure for patients with local Siewert type II/III AEG, regardless of the TNM stage.         Preparation and photoluminescence properties of the Eu2+, Sm3+ co-doped Li2SrSiO4 phosphors

        A series of Eu2+ and Sm3+ co-doped Li2SrSiO4 phosphors are prepared by the high temperature solidstate reaction. The morphology, structure and spectroscopic properties of the prepared samples are characterized by scanning electron microscopy, X-ray diffraction, diffuse reflection spectra, photoluminescence spectra and electron paramagnetic resonance spectra, respectively. The effect of Sm3þdoping concentration on the photoluminescence intensity of the prepared samples is also investigated. The results indicate that the crystal structure of Li2SrSiO4 is not changed with the Eu2+, Sm3+ co-doping. The spherical-like particle size of the obtained product is about 20-30 nm in diameter. When the Sm3+ concentration is 0.3 mol% and the Eu2+ concentration is 0.7 mol%, the phosphors show the maximum emission intensity, which is 50% higher than that of Eu2+ doped Li2SrSiO4. Excited at 420 nm, the phosphor presents a single broad emission band peaking at 558 nm, which is ascribed to the 4f65d1 / 4f7 transitions of Eu2+ and 4G5/2 / 6H5/2 and 4G5/2 / 6H7/2 transitions of Sm3+. The Commission International de I0Eclairage chromaticity coordinates of Li2SrSiO4:0.7 mol% Eu2+, 0.3 mol%Sm3+ are x = 0.28, y = 0.28.         Tuberculosis (TB) caused by Mycobacterium tuberculosis (M.tb) remains a major global health problem. Human ficolin2 (L-ficolin/P35) is a recently identified lectin complement pathway activator present in normal plasma associated with infectious diseases, however, little is known about the roles and mechanisms of ficolin2 during M.tb infection.We describe in this study for the first time L-ficolin expression levels in tuberculosis (TB) patients. We found that serum levels of ficolin2 of 107 pulmonary TB patients were much lower compared with 107 healthy controls, and recurrence TB patients have even lower levels than onset TB patients. In vitro analysis showed that Ficolin2 bound to virulent M.tb H37Rv strain much stronger than to non-virulent M. bovis BCG and M. smegmatis. Ficolin2/ficolin A bound to the H37Rv surface glycolipid portion and blocked H37Rv infection to human lung A549 cells. We further determined that the extrogenous ficolin2 had remarkable protecting effects against virulent H37Rv strain infections in C57BL/6J mice. FicolinA (ficolin2 like molecule) knockout mice had greatly increased susceptibility to the H37Rv infection. Ficolin2 activated macrophages via phosphorylation of JNK and stimulated IFN-□, IL-17, TNF-□ and nitric oxide (NO) secretions. Ficolin2 also stimulated IFN-□ production of CD8+T cells, but not CD4+T cells. The opsono-phagocytosis was promoted by ficolin2 as well. Our data demonstrate that ficolin2 can defend against virulent M.tb H37Rv infection both in vitro and in vivo mainly via activating macrophage proinflammatory cytokines IFN-□ and NO production and opsonophagocytosis, and its insufficiency is associated with more susceptible infections in human.

        Preparation and Characterization of Core/Shell-type Ag/Chitosan Nanoparticles with Antibacterial Activity

        Making use of chitosan (CS) and ethylenediaminetetraacetic acid (EDTA) as a reaction system, CS-EDTA nanoparticles were synthesized through a facile counterion complex coacervation method. Ag^+ could enter porous CS nanoparticles synthesized with this method, allowing Ag nanoparticles within chitosan nanoparticles were synthesized by reducing silver nitrate with chitosan. Because of the noncovalent interaction between CS and EDTA, the EDTA could be easily removed via dialysis against water, and pure core/shell-type Ag/CS nanoparticles could be obtained. The nanoparticles showed higher antibacterial activity toward E. coli than the active precursor Ag nanoparticles and CS.

        Population dynamics of Frankliniella occidentalis Pergrande and its predator Orius similis Zheng on common crops and surrounding plants

        Frankliniella occidentalis Pergrande is important invasive pests in China, causing damage to agricultural pro duction, and Orius similis Zheng is the dominant predator species of F. occidentalis. A two-year survey was conducted to determine the population density of F. occidentalis and O. similis, on chili (Capsicum annuum L.) and maize (Zea mays L.) crops and surrounding weed species, which included white clover (Trifolium repens L.), St. John’s wort (Hypericum beanii N. Robson), alfalfa (Medicago sativa L.) and beggarticks (Bidens pilosa L.) in Kunming, southern China. The activity of F. occidentalis on these 6 host plant species was determined using the quartile method. F. occidentalis mainly damaged plants during their flowering stage. The main activity period of F. occidentalis occurred earlier on H. beanii and T. repens than on C. annuum. The peak activity of F. occidentalis occurred in the middle of May (on T. repens). During the whole activity period, the highest thrips densities were recorded on H. beanii among all of the sampled host plant species, followed by C. annuum. The lowest density was recorded on B. pilosa. Dynamics of immature F. occidentalis were more irregular than that of adults. The highest density of O. similis was recorded on Z. mays. It was 2.27–26.43-fold (2017) and 2.01–19.09-fold (2018) higher than that on other host plant species. This study showed that F. occidentalis could migrate between C. annuum and surrounding weeds. The weeds were the main source of thrips on C. annuum. The results indicated that Z. mays can be planted around C. annuum fields as a potential banker plant, to attract O. similis to control F. occidentalis on C. annuum, T. repens and B. pilosa. The flowering period of plants and surrounding plant species has a great effect to the population activities of F. occidentalis and predator O. similis on crops.

        A GAPSO-Enhanced Extreme Learning Machine Method for Tool Wear Estimation in Milling Processes Based on Vibration Signals

        Tools are the most vulnerable components in milling processes conducted using numerical control milling machines, and their wear condition directly influences work-product quality and operational safety. As such, tool wear estimation is an essential component of NC milling operations. This study addresses this issue by proposing an extreme learning machine (ELM) method enhanced by a hybrid genetic algorithm and particle swarm optimization (GAPSO) approach for conducting tool wear estimation based on workpiece vibration signals. Here, a few feature parameters in the time, frequency, and time–frequency (Ensemble empirical mode decomposition, EEMD) domains of the workpiece vibration signals are extracted as the input of the ELM model. Then, the initialized weights and thresholds of the ELM model are optimized based on the GAPSO approach with training dataset. Finally, tool wear is estimated using the optimized ELM model with testing dataset. The effectiveness of the proposed method is verified by its application to vibration signals collected from two milling tool wear experiments (an open-access benchmark dataset and a milling tool wear experiment) by comparison to the ELM, GA-ELM, and PSO-ELM methods. The results indicate that the estimation accuracy and optimization efficiency of the proposed method outperforms that of other three methods.

        Preparation and properties of the green-emitting phosphors NaCa0.98-xMgxPO4:

        In this study, a series of the green-emitting phosphors NaCa0.98-xMgxPO4:Eu2þ 0:02 was prepared through high temperature solid-state reaction. The structure and spectroscopic properties of these samples were characterized by XRD, FT-IR, reflectance spectra and photoluminescence (PL) spectra, respectively. The effect of Mg2+ co-doping concentration on the PL intensity of as-prepared samples was also investigated. The enhanced intensities of excitation and emission band of Eu2+ and a slight red-shift of the emission peak were observed due to Mg2+ co-doping. When the x value is 0.08, the phosphors possessed the maximum emission intensity monitored at 360 nm and the emission spectrum shows a single band centered at about 500 nm. The obtained phosphors had strong absorption in near-UV zone (350–400 nm), which can match the emission of near-UV light-emitting diodes. The prepared material is potential green phosphors for white LEDs.
