        A Comparative Study on Ternary Low-Platinum Catalysts with Various Constructions for Oxygen Reduction and Methanol Oxidation Reactions

        Three types of ternary low-platinum nanocatalysts, alloy PdPtIr/C, core–shell PdPt@PtIr/C and Pd@PtIr/C, have been prepared, and their catalytic behaviors toward methanol oxidation reaction (MOR)/oxygen reduction reaction (ORR) are comparatively investigated via cyclic voltammetry and chronoamperometry analysis in an acidic medium. Through a two-step colloidal technique, the synthesized core–shell structured catalyst PtPd@PtIr/C with alloy core and alloy shell show the best catalytic activity toward MOR and the best poisoning tolerance. The alloy PdPtIr/C catalyst prepared via a one-step colloidal technique exhibits the best performance toward ORR among the three catalysts. All the three catalysts are characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD) and other characterization techniques.

        Downregulation of LINC01508 contributes to cisplatin resistance in ovarian cancer via the regulation of the Hippo-YAP pathway

        Background: Some long non-coding RNAs (lncRNAs) have been found to contribute to cisplatin resistance. Here, we identified a novel lncRNA that was downregulated in cisplatin- resistant to ovarian cancer (OC) cells and aimed to examine the contribution of LINC01508 to cisplatin resistance in OC cells. Methods: Differences in the lncRNA expression profile between OV2008 and C13K cells were assessed by lncRNA expression microarray. The expression of LINC01508 in ovarian epithelial cells, four OC cells, and OC, benign ovary tumor and normal ovary, cisplatin- resistant and non-resistant OC specimens were evaluated by quantitative real-time polymerase chain reaction (qPCR). The role of LINC01508 in OC cisplatin-resistant was evaluated by cell counting kit-8 (CCK-8), flow cytometry, colony formation, wound healing, Transwell, and tumor growth inhibition study in vivo. The clinical associations of LINC01508 in OC were evaluated using correlation analysis. The effects of verteporfin (VP) on cisplatin were explored to reveal the function of the hippo-YAP pathway on the cisplatin tolerance of C13K. Results: LINC01508 was downregulated in cisplatin-resistant OC cells and platinum- resistant OC tissue (p<0.01). LINC01508 downregulation was correlated with tumor size, residual tumor, and platinum resistance. The overexpression of LINC01508 improves in vitro and in vivo sensitivity to cisplatin while predicts the poor overall survival which need further follow-up research. The increased level of LINC01508 could suppress the cisplatin resistance of OC cells through the inhibition of the hippo-YAP pathway. Conclusions: The study proposes that dysregulation of LINC01508 expression results in resistance of OC to cisplatin through the inhibition of the hippo-YAP pathway.

        Identification of Quantitative Trait Loci (QTLs) for Flowering Time Using SSR Marker in Maize under Water Stress

        The selection of reduction of anthesis-silking interval (ASI) in maize breeding is an efficient way to develop maize varieties more tolerant to dry growing conditions. Characterization of the quantitative trait loci (QTL) that controlled the flowering time will be helpful for selection in maize breeding. In this study, flowering time of individuals in a 234 F_(2:3) family, derived from the cross between inbred lines X178 and B73, was evaluated under well-watered and water-stressed conditions at the same location. SSR (microsatellite) was used to identify flowering time QTL. The results showed that the broad-sense heritability for male flowering time (MFT), female flowering time (FFT) and ASI were 0.72, 0.72 - 0.74 and 0.40 - 0.42, respectively, and ASI was significantly correlated to FFT. Under water-stressed condition, 9, 6 and 6 QTLs were identified for MFT, FFT and ASI, respectively, and individual QTL accounted for approximately 2.88% - 31.65% of the phenotypic variation. Some QTLs for MFT were mapped overlapping with those for FIT and ASI. One QTL on chromosome 9 (near nc134) had the strongest effect on MFT, FTT and ASI. It was suggested that the epistasis contributed to the phenotypic variation of flowering time.

      • Effects of Home Nursing Intervention on the Quality of Life of Patients with Nasopharyngeal Carcinoma after Radiotherapy and Chemotherapy

        Background: The effects of home nursing intervention on the quality of life in patients with nasopharyngeal carcinoma (NPC) after radiotherapy and chemotherapy are unclear. According to the characteristics of nursing home patients with nasopharyngeal carcinoma, we should continuously improve the nursing plan and improve the quality of life of patients at home. Materials and Methods: We selected 180 patients at home with NPC after radiotherapy and chemotherapy. The patients were randomly divided into experimental and control groups (90 patients each). The experimental group featured intervention with an NPC home nursing plan, while the control group was given routine discharge and outpatient review. Nursing intervention for patients was mainly achieved by regular telephone follow-up and home visits. We use the quality of life scale (QOL-C30), anxiety scale (SAS) and depression scale (SDS) to evaluate these patients before intervention, and during follow-up at 1 month and 3 months after the intervention. Results: Overall health and quality of life were significantly different between the groups (p<0.05), Emotional function score was significantly higher after intervention (p<0.05), as were cognitive function and social function scores after 3 months of intervention (p<0.05). Scores of fatigue, nausea and vomiting, pain, appetite and constipation were also significantly different between the two groups (p<0.05). Rates of anxiety and depression after 3 months of intervention were 11.1%, 22.2% and 34.4%, 53.3%, the differences being significant (p<0.05). Conclusions: NPC home nursing plan could effectively improve overall quality of life, cognitive function, social function (after 3 months) of patients, but improvement regarding body function is not suggested. Fatigue, nausea and vomiting, pain, appetite, constipation were clearly improved. We should further pursue a personalized, comprehensive measurements for nursing interventions and try to improve the quality of life of NPC patients at home.

        Effects of Lactobacillus plantarum SCS2 on blood glucose level in hyperglycemia mice model

        In this study, the hyperglycemia mice model was established with 1-week high sugar and fat diet plus with 70 mg/kg body weight of streptozotocin injection for 3 days. Sixty male Kunming mice of 3 weeks old in a specific-pathogen-free grade were divided into six groups randomly, which includes normal group (NG), prevention group (PG), treatment group for low dose (TGL), middle dose (TGM), high dose (TGH), and model group (MG). NG and MG mice were fed with sterile physiological saline (10 mL/kg body weight). PG mice were fed with the concentration of 6.0 × 109 CFU/mL L. plantarum SCS2 suspensions from the second to third week. TGL, TGM, and TGH mice were fed with the concentration of 2.0 × 109, 4.0 × 109, and 6.0 × 109 CFU/mL L. plantarum SCS2 suspensions (10 mL/kg body weight), respectively from fourth to tenth week. The results showed that the fasting and postprandial 2 h blood glucose levels of TGH mice were reduced by L. plantarum SCS2 significantly (p < 0.05) as compared with MG. The body weight of TGH mice came to normal level at tenth week. Content of K+ in plasma of TGH mice was increased and contents of Na+ and Cl− in the plasma of TGH mice were decreased as compared with MG. Meanwhile, content of glycogen in TGH mice was reduced. However, the effect of L. plantarum SCS2 on the prevention of hyperglycemia in PG mice was not significant as compared with NG mice during the experiment. These results suggested that L. plantarum SCS2 showed a hypoglycemic effect on hyperglycemic mice model.

        Metabolomic analysis of biochemical changes in the tissue and urine of proteoglycan-induced spondylitis in mice after treatment with moxibustion

        Background: Moxibustion is widely used in East Asian countries to manage the symptom of rheumatic diseases. The aim of this study was to identify potential metabolic profiles of moxibustion on relieving ankylosing spondylitis (AS) mice through UHPLC-Q-TOF/MS metabolomic study. Methods: Thirty-two female Balb/c mice were randomized into healthy control (HC), AS model, moxibustion at acupuncture points (MA) in AS model, and moxibustion at non-acupuncture points (MNA) AS model groups. Moxibustion was administered daily at GV4, bilateral BL23 and bilateral ST36 acupuncture points for four weeks in the MA group. The overall health status, the thickness of hind paws and the tissue concentrations of IL-1β, PGE2, IL-6 and TNF-α were assessed. The UHPLC-Q-TOF/MS was used to explore the perturbations of endogenous metabolites in tissue and urine of AS model mice intervened by moxibustion. Results: Compared with the AS group, the overall health status was significantly improved after 4-week moxibustion intervention (p < 0.05). The results also showed that MA significantly reduced the levels of paw thickness and decreased the levels of four cytokines in the tissue (p < 0.01). Thirty-seven endogenous metabolites identified by the OPLS-DA were considered to be contributing to therapeutic effects of moxibustion. Moreover, metabolic pathway analysis further revealed that the identified metabolites were mainly involved in TCA cycle, Lipid metabolism, Amino Acid metabolism, Intestinal flora metabolism and Purine metabolism. Conclusions: UHPLC-Q-TOF/MS based metabolomics approach, as a novel and powerful tool, can help us to gain the insights into potential mechanisms of action of moxibustion for AS.

      • An Updated Meta-analysis Between the Association of XRCC1 Arg399Gln Polymorphism and Hepatocellular Carcinoma Risk

        Background: Various studies have evaluated the relationship between X-ray repair cross-complementing group 1 (XRCC1) Arg399Gln polymorphism and hepatocellular carcinoma (HCC) risk, but the conclusions have been inconsistent and underpowered. The purpose of this updated meta-analysis was to examine whether XRCC1 Arg399Gln polymorphism confers susceptibility to HCC. Methods: Eligible studies extracted from PubMed, Embase, Cochrane Library, VIP (chinese) and CNKI (chinese) up to November 2013 were included in the study. Pooled odds ratio (OR) together with their 95% confidence interval (CI) were estimated to evaluate XRCC1 Arg399Gln polymorphism and HCC risk. Results: Finally, 21 studies with 4,170 cases and 5,030 controls were involved in our meta-analysis. The results demonstrated that there was significant association between Arg399Gln polymorphism and HCC risk under two contrast models in overall populations (AG vs GG: OR=1.265, 95%CI=1.036-1.545, p=0.021; AA+AG vs GG: OR=1.240, 95%CI=1.021-1.506, p=0.030). In subgroup analyses, significant association was found in Asians (A vs G: OR=1.175, 95%CI=1.013-1.362, p=0.033; AG vs GG: OR=1.317, 95%CI=1.070-1.622, p=0.009; AA+AG vs GG: OR=1.289, 95%CI=1.055-1.575, p=0.013) and Caucasians (A vs G: OR=0.591, 95%CI=0.361-0.966, p=0.036; AA+AG vs GG: OR=0.468, 95%CI=0.234-0.934, p=0.031). Conclusions: The results suggest that XRCC1 Arg399Gln polymorphism may increase HCC risk especially among Asians. However, XRCC1 Arg399Gln polymorphism might act as a protective role against HCC among Caucasians.

        Neoadjuvant Chemotherapy-Guided Bladder-Sparing Treatment for Muscle-Invasive Bladder Cancer: Results of a Pilot Phase II Study

        Purpose Reduced quality of life after cystectomy has made bladder preservation a popular research topic for muscle-invasive bladder cancer (MIBC). Previous research has indicated significant tumor downstaging after neoadjuvant chemotherapy (NAC). However, maximal transurethral resection of bladder tumor (TURBT) was performed before NAC to define the pathology, impacting the real evaluation of NAC. This research aimed to assess real NAC efficacy without interference from TURBT and apply combined modality therapies guided by NAC efficacy. Materials and Methods Patients with cT2-4aN0M0 MIBC were confirmed by cystoscopic biopsy and imaging. NAC efficacy was assessed by imaging, urine cytology, and cystoscopy with multidisciplinary team discussion. Definite responders (≤ T1) underwent TURBT plus concurrent chemoradiotherapy. Incomplete responders underwent radical cystectomy or partial cystectomy if feasible. The primary endpoint was the bladder preservation rate. Results Fifty-nine patients were enrolled, and the median age was 63 years. Patients with cT3-4 accounted for 75%. The median number of NAC cycles was three. Definite responders were 52.5%. The complete response (CR) was 10.2%, and 59.3% of patients received bladder-sparing treatments. With a median follow-up of 44.6 months, the 3-year overall survival (OS) was 72.8%. Three-year OS and relapse-free survival were 88.4% and 60.0% in the bladder-sparing group but only 74.3% and 37.5% in the cystectomy group. The evaluations of preserved bladder function were satisfactory. Conclusion After stratifying MIBC patients by NAC efficacy, definite responders achieved a satisfactory bladder-sparing rate, prognosis, and bladder function. The CR rate reflected the real NAC efficacy for MIBC. This therapy is worth verifying through multicenter research.

        Effects of Wheel-Rail Impact on the Fatigue Performance of Fastening Clips in Rail Joint Area of High-Speed Railway

        In order to study the effects of wheel-rail impacts on the fatigue damage of the fastening clips at the rail joints of a high-speed railway, field tests were carried out on the wheel-rail impacts at the rail joints of a high-speed railway in China. On the basis of the track irregularities at the rail joints obtained from field measurements, a nonlinear vehicle-track coupling dynamics three-dimensional (3D) model with consideration of Timoshenko beam elements and a refined finite element model of WJ-8 fastening system were constructed. And combined with the theory of cumulative damage and the method of fatigue analysis, the effects of wheel-rail impact at the rail joints on the fatigue life of fastening clips were analyzed. The results showed that the impacts of the wheel-rail in the joint area caused high-frequency fluctuations in the stress-time curves of the clips, which significantly increased the amplitude of stress cycle and increased a lot of stress cycles, of which the amplitudes were reached at a large level; the clips in the area where a rail joint was located suffered the most serious fatigue damage and had the shortest fatigue life; in the actual operation line, besides the clips in the rail joint area, it is also necessary to be concerned about the fatigue performance of fastening clips near the rail joint area.
