Cyclin-dependent kinase 2 (CDK2) is a member of serine/threonine protein kinases, which initiates the principal transitions of the eukaryotic cell cycle and is a promising target for cancer therapy. The present study was designed to inhibit cdk2 gene expression to induce cell cycle arrest and cell proliferation suppression. Here, we constructed a series of RNA interference (RNAi) plasmids which can successfully express small interference RNA (siRNA) in the transfected human cells. The results showed that the RNAi plasmids containing the coding sequences for siRNAs down-regulated the cdk2 gene expression in human cancer cells at the mRNA and the protein levels. Furthermore, we found that the cell cycle was arrested at G0G1 phases and the cell proliferation was inhibited by different siRNAs. These results demonstrate that suppression of CDK2 activity by RNAi may be an effective strategy for gene therapy in human cancers. [BMB reports 2010; 43(4): 291-296]

Structural behaviors of sustainable hybrid columns under compression and flexure

Structural behaviors of a sustainable hybrid column with the ultra high performance cementitious composites (UHPCC) permanent form under compression and flexure were studied. Critical state and failure stage characters are analyzed for large and small eccentricity cases. A simplified theoretical model is proposed for engineering designs and unified formulas for loading capacity of the hybrid column under compression and flexure loads are derived, including axial force and moment. Non-linear numerical analysis is carried out to verify the theoretical predictions. The theoretical predictions agree well with the numerical results which are verified by the short hybrid column tests recursively. Compared with the traditional reinforced concrete (RC) column, the loading capacity of the sustainable hybrid column is improved significantly due to UHPCC confinements.

Grapholita molesta (Busck) (Lepidoptera: Tortricidae) is a notorious pest of many Rosaceae crops worldwide. Enhancement of trap efficiency of its sex pheromone was devised by addition of E-β-ocimene and E-β-farnesene. The addition of E-β-ocimene or E-β-farnesene to sex pheromone increased electroantennogram response of male G. molesta compared to sex pheromone alone. Blend of pheromone and E-β-ocimene or E-β-farnesene in 1:0.1 increased the upwind flight and landing behaviors. Furthermore, field experiments showed that sex lures with Eβ-ocimene, or /and E-β-farnesene, enhanced trapping efficiency compared to sex pheromone alone. These results may provide the basis for the development of efficient pest management systems against G. molesta using plant volatiles and insect sex pheromones.

With the development of tourist industry and diversification of a traveling way, the guesthouse seems to be a standard organization of the international hotel chain, different from the general hotels. Moreover, its unique spaces for communications attract a large number of young tourists to the guesthouse. However, with some problems learned from the communication spaces in some guesthouses, it is necessary to research characteristics of particular guesthouses and then vitalize those characteristics for young tourists’ interests. Therefore, a case study was conducted on three guesthouses located in Seoul. Analysis results are as follows: First, five design characteristics of communication spaces are interactivity, regional culture, variability, openness, and sensory stimulation. Second, based on the research from arrangement types, interactivity ranks the highest in intensive type and blended type, and in stand-alone type, regional culture shows the highest. Third, with the design characteristics examined from three places, interactivity is well-reflected in Jung-gu and Mapo-gu, and regional culture is effectively expressed in Jongro-gu.
Crashworthiness analysis on existing RC parapets rehabilitated with UHPCC

In recent year, the coat layer drops and the rebar rust of bridge parapets, which caused the structural performance degradation. In order to achieve the comprehensive rehabilitation, ultra high performance cementitious composites is proposed to existing RC parapet rehabilitation. The influence factors of UHPCC rehabilitation includes two parts, i.e., internal factors related with material, such as UHPCC layer thickness, corrosion ratio of rebars, fiber volume fraction, and external factors related with the load, such as impact speeds, impact angles, vehicle mass. The influence of the factors was analyzed in this paper based on the nonlinear finite element. The analysis results of the maximum dynamic deformation and the peak impact load of parapets revealed the influence of the internal factors and the external factors on anti-collision performance and degree degradation. This research may provide a reference for the comprehensive multifunctional rehabilitation of existing bridge parapets.

Background T -cell acute lymphoblastic leukemia (T-ALL) is considered a malignant tumor with a high mortality rate. To combat this disease, exploring the mechanism of T-ALL progression is urgently needed. Krüppel-like factors (KLFs) are known as the transcription factors and mediate series of biological processes. KLF9 is a member of the KLF family which could serve as a tumor suppressor gene in most solid tumors. GEO Database analysis showed that KLF9 expression in normal T cells was higher than T-ALL cell lines and patients. However, the possible role of KLF9 in T-ALL progression is still unclear. Objective To uncover the possible eff ects of Krüppel-like transcription factor 9 (KLF9) on the progression of T-Acute lymphoblastic leukemia (T-ALL). Results The expression of KLF9 was low in human T-ALL cells. KLF9 suppressed the viability of T-ALL cells. In addition, KLF9 stimulated the apoptosis as well as autophagy of T-ALL cells. Mechanically, KLF9 suppressed AKT/mTOR pathway in T-ALL cells. Conclusion KLF9 suppressed viability and promoted autophagy as well as apoptosis in T-ALL cells by inhibiting AKT/ mTOR pathway.
SKF96365 impedes spinal glutamatergic transmission-mediated neuropathic allodynia

Spinal nerve injury causes mechanical allodynia and structural imbalance of neurotransmission, which were typically associated with calcium overload. Storeoperated calcium entry (SOCE) is considered crucial elements-mediating intracellular calcium homeostasis, ion channel activity, and synaptic plasticity. However, the underlying mechanism of SOCE in mediating neuronal transmitter release and synaptic transmission remains ambiguous in neuropathic pain. Neuropathic rats were operated by spinal nerve ligations. Neurotransmissions were assessed by whole-cell recording in substantia gelatinosa. Immunofluorescence staining of STIM1 with neuronal and glial biomarkers in the spinal dorsal horn. The endoplasmic reticulum stress level was estimated from qRT-PCR. Intrathecal injection of SOCE antagonist SKF96365 dose-dependently alleviated mechanical allodynia in ipsilateral hind paws of neuropathic rats with ED50 of 18 μg. Immunofluorescence staining demonstrated that STIM1 was specifically and significantly expressed in neurons but not astrocytes and microglia in the spinal dorsal horn. Bath application of SKF96365 inhibited enhanced miniature excitatory postsynaptic currents in a dosage-dependent manner without affecting miniature inhibitory postsynaptic currents. Mal-adaption of SOCE was commonly related to endoplasmic reticulum (ER) stress in the central nervous system. SKF96365 markedly suppressed ER stress levels by alleviating mRNA expression of C/ EBP homologous protein and heat shock protein 70 in neuropathic rats. Our findings suggested that nerve injury might promote SOCE-mediated calcium levels, resulting in long-term imbalance of spinal synaptic transmission and behavioral sensitization, SKF96365 produces antinociception by alleviating glutamatergic transmission and ER stress. This work demonstrated the involvement of SOCE in neuropathic pain, implying that SOCE might be a potential target for pain management
