An aqueous two-phase system (ATPS) containing an ionic liquid (1-(2-methoxyethyl)-3-methylimidazolium bromide) and three organic salts (K3C6H5O7, (NH4)3C6H5O7, and K2C4H4O6) at different temperatures was designed. Binodal data were correlated using two empirical equations, and tie-line data were fit with the utilization of Bancroft and Othmer-Tobias equations. In the systems investigated, three conclusions were drawn from the study of the phase-forming ability of salt through effective excluded volume, Gibbs free energy of ions, and the phase diagram. First, if the same cations of salt were present, the ability of salt to form phases increased with increasing valence of the anion. Second, the larger the effective excluded volume, the stronger the aforementioned ability of salt in forming phases. Third, salt had more ability to form phases if cations (or anions) contained in the salt possessed higher negative Gibbs free energy when the cation (anion) of the salt was the same. The effect of temperature on ATPSs was also investigated. It was found that it was easier to form ATPSs at lower temperature, and the tie-line slope showed growing absolute values as the temperature was decreased.

Diabetic nephropathy (DN) is the leading cause of end-stage failure of the kidney, but the efficacy of currently available strategies for the prevention of DN remains unsatisfactory. In this study, we investigated the effects of free anthraquinones (FARs) extract, which was extracted from the rhubarb and purified by macroporous resin DM130 with gradient mixtures of ethanol/water as the lelution solvents, in high glucose-cultured glomerular mesangial cells (MCs). The cell proliferation was determined by CCK-8 assay, the levels of TGF-${\beta}1$, CTGF, ColIV and FN proteins in the supernatant of MCs were measured by ELISA assays, and the mRNA levels of these four genes were detected by RT-PCR. The results showed that the increased proliferation of MCs, the mRNA levels and protein expression of TGF-${\beta}1$, CTGF, ColIV and FN induced by high glucose were inhibited after the treatment with the FARs extract. This indicated that FARs extract could inhibit cell proliferation and the expression of main extracellular matrix induced by high glucose in MCs. The FARs extract exhibited potential values for prophylaxis and therapy of DN.

A multiscale method is presented for analysis of thin slab structures in which the microstructures can not be reduced to two-dimensional plane stress models and thus three dimensional treatment of microstructures is necessary. This method is based on the classical asymptotic expansion multiscale approach but with consideration of the special geometric characteristics of the slab structures. This is achieved via a special form of multiscale asymptotic expansion of displacement field. The expanded three dimensional displacement field only exhibits in-plane periodicity and the thickness dimension is in the global scale. Consequently by employing the multiscale asymptotic expansion approach the global macroscopic structural problem and the local microscopic unit cell problem are rationally set up. It is noted that the unit cell is subjected to the in-plane periodic boundary conditions as well as the traction free conditions on the out of plane surfaces of the unit cell. The variational formulation and finite element implementation of the unit cell problem are discussed in details. Thereafter the in-plane material response is systematically characterized via homogenization analysis of the proposed special unit cell problem for different microstructures and the reasoning of the present method is justified. Moreover the present multiscale analysis procedure is illustrated through a plane stress beam example.

A strain smoothing meshfree formulation with stabilized conforming nodal integration is presented for modeling the consolidation process in saturated porous media. In the present method, nodal strain smoothing is consistently introduced into the meshfree approximation of strain and pore pressure gradient variables associated with the saturated porous media. Meanwhile, in order to achieve a consistent numerical implementation, a smoothing approximation of the meshfree shape function within a nodal representative domain is also proposed in the stiffness construction. The resulting discrete system of equations is all expressed in smoothed nodal measures that are very efficient for numerical evaluation. Subsequently the space-time fully discrete equations are further established by the generalized trapezoidal rule for time integration. The effectiveness of the proposed meshfree consolidation analysis method is systematically illustrated by several benchmark problems.

Low back pain, triggered by intervertebral disc degeneration (IVDD), is one of the most common causes of disability and financial expenditure worldwide. However, except for surgical interventions, effective medical treatment to prevent the progression of IVDD is lacking. This study aimed to investigate the effects of circKIF18A, a novel circRNA, on IVDD progression and to explore its underlying mechanism in IVDD. In this study, we found that oxidative stress was positively correlated with nucleus pulposus cell (NPC) senescence in IVDD and that circKIF18A was downregulated in IVDD and attenuated senescent phenotypes such as cell cycle arrest and extracellular matrix degradation in NPCs. Mechanistically, circKIF18A competitively suppressed ubiquitin-mediated proteasomal degradation of MCM7, and the protective effects of circKIF18A on NPCs were partially mediated by MCM7 under oxidative stress. Intradiscal injection of adenoviral circKIF18A ameliorated IVDD in a rat model. This study revealed that circKIF18A regulates NPC degeneration by stabilizing MCM7 and identified a novel signaling pathway, the circKIF18A-MCM7 axis, for anti-senescence molecular therapy in IVDD.
An efficient Galerkin meshfree analysis of shear deformable cylindrical panels

A Galerkin meshfree method is presented for analyzing shear deformable cylindrical panels. Based upon the analogy between the cylindrical panel and the curved beam a pure bending mode for cylindrical panel is rationally constructed. The meshfree approximation employed herein is characterized by an enhanced moving least square or reproducing kernel basis function that can exactly represent the pure bending mode and thus meets the requirement of Kirchhoff mode reproducing condition. The variational form is discretized using the efficient stabilized conforming nodal integration with a smoothed nodal gradient based curvature. The resulting meshfree formulation satisfies the integration constraint for bending exactness. Moreover, it is shown here that the smoothed gradient preserves several desired properties which are valid for the standard gradient obtained by direct differentiation, such as partition of nullity and reproduction of a constant strain field. The efficacy of the proposed approach is demonstrated by two benchmark cylindrical panel examples.
Dual-loss CNN: A separability-enhanced network for current-based fault diagnosis of rolling bearings

Current-based mechanical fault diagnosis is more convenient and low cost since additional sensors are not required. However, it is still challenging to achieve this goal due to the weak fault information in current signals. In this paper, a dual-loss convolutional neural network (DLCNN) is proposed to implement the intelligent bearing fault diagnosis via current signals. First, a novel similarity loss (SimL) function is developed, which is expected to maximize the intra-class similarity and minimize the inter-class similarity in the model optimization operation. In the loss function, a weight parameter is further introduced to achieve a balance and leverage the performance of SimL function. Second, the DLCNN model is constructed using the presented SimL and the cross-entropy loss. Finally, the two-phase current signals are fused and then fed into the DLCNN to provide more fault information. The proposed DLCNN is tested by experiment data, and the results confirm that the DLCNN achieves higher accuracy compared to the conventional CNN. Meanwhile, the feature visualization presents that the samples of different classes are separated well.

Oxidative stress-induced mitochondrial dysfunction is implicated in the pathogenesis of intervertebral disc degeneration (IVDD). Sirtuin 3 (SIRT3), a sirtuin family protein located in mitochondria, is essential for mitochondrial homeostasis; however, the role of SIRT3 in the process of IVDD has remained elusive. Here, we explored the expression of SIRT3 in IVDD in vivo and in vitro; we also explored the role of SIRT3 in senescence, apoptosis, and mitochondrial homeostasis under oxidative stress. We subsequently activated SIRT3 using honokiol to evaluate its therapeutic potential for IVDD. We assessed SIRT3 expression in degenerative nucleus pulposus (NP) tissues and oxidative stressinduced nucleus pulposus cells (NPCs). SIRT3 was knocked down by lentivirus and activated by honokiol to determine its role in oxidative stress-induced NPCs. The mechanism by which honokiol affected SIRT3 regulation was investigated in vitro, and the therapeutic potential of honokiol was assessed in vitro and in vivo. We found that the expression of SIRT3 decreased with IVDD, and SIRT3 knockdown reduced the tolerance of NPCs to oxidative stress. Honokiol (10 μM) improved the viability of NPCs under oxidative stress and promoted their properties of anti-oxidation, mitochondrial dynamics and mitophagy in a SIRT3-dependent manner. Furthermore, honokiol activated SIRT3 through the AMPKPGC- 1α signaling pathway. Moreover, honokiol treatment ameliorated IVDD in rats. Our study indicated that SIRT3 is involved in IVDD and showed the potential of the SIRT3 agonist honokiol for the treatment of IVDD.
Characterization of Squamosa Promoter Binding Protein-LIKE genes in wheat

Common wheat (Triticum aestivum) is an allohexaploid (AABBDD), with Triticum urartu (AA) and Aegilops tauschii (DD) as its A and D genome donors. Squamosa Promoter Binding Protein-Like (SPL) genes play important roles in plant development; however, a systematic study of this gene family in wheat is still lacking. Here, we mined the wheat genomes for SPLs and found 19 SPL genes each from the T. urartu and Ae. tauschii genomes. We also identified 58 SPL genes from the hexaploid wheat genome. We then compared the conservation and divergence of SPLs between the two diploid species in gene structures, tissuespecific expression patterns and responses to salt stress, drought and low temperature. For functional studies, two hexaploid wheat SPLs, TaSPL3 (with miR156 site mutated) and TaSPL6, were overexpressed in Arabidopsis and found to be involved in flowering time and biomass accumulation. In addition, a mutant allele (TaSPL6-D R ) with a 47 bp duplication upstream of the SBP domain was found to be associated with maturity date and grain number per spike among landraces from a Chinese wheat mini-core collection. Collectively, our work provides useful knowledge on the sequences and functions of wheat SPL genes that may help future functional studies and molecular breeding.
